1.Inhibitory effects of Zedoray Rhizome Abstracts on hepatic cell line HepG2 and their mechanisms

Aim Curcumol and Zedoary Turmeric Oil (ZTO) were prepared to liposome, which were used to study the anti-hepatoma effect, and its mechanism that was expected to provide theoretical support for the clinic anticarcinomal use of Zedoray Rhizome Abstracts.Methods Curcumol and ZTO were prepared to liposome by mechanical diffusion and ultrasonic techniques.Proliferation of HepG2 cell in vitro was observed with curcumol and ZTO by MTT assay. Apoptosis of cells was observed by staining with Hoechst 33258/PI and laser confocal scanning microscopy (LCSM). Expression of COX-2 mRNA and VEGF mRNA of HepG2 cell was investigated by RT-PCR.Results Envelopment ratios of curcumol liposome and ZTO liposome were 86.2% and 74.5%,respectively.Curcumol and ZTO inhibited proliferation of HepG2 cell by MTT assay.Compared with control group, Curcumol and ZTO increased the apoptosis rate of HepG2 cell significantly (P

Clinical practice of medical imaging technology undergraduates is a key link in the transformation from imaging technology students to imaging technologists. However, there are a series of problems leading to poor practice effect, e.g., imaging technology involves a lot of learning contents, and the teachers' level is uneven. Based on the above problems, some improvements have been implemented, including the establishment of the admission lecture system and tutor responsibility system, application of pluralistic teaching methods, standardizing the examination system of interns, flexible adjustment of practice time arrangements, training of practice teaching team, and so on. Finally, the quality and effect of undergraduate practice teaching in medical imaging technology has been improved significantly.

4.Effect of high glucose toxicity on JNK pathway, cell viability and apoptosis in pancreatic β-cell line INS-1

AIM: To investigate the effect of high glucose toxicity on JNK pathway and cell function of INS-1 cells.METHODS: Cultured INS-1 cells with or without IGF-1 exposure, were treated with glucose at 3 concentrations (5.6 mmol/L, 11.2 mmol/L and 33.3mmol/L), respectively. MTT was used to measure the cell viability. Apoptosis was determined by immuno-fluorescence and flow-cytometry analysis. The serine 270 phosphorylation of IRS and phosphorylation of JNK in INS-1 cells were detected in the presence or absence of SP600125 treatment.RESULTS: The cell viability decreased and apoptosis increased with elevated glucose concentrations. The percentage of apoptosis cells was 11.3% in 5.6 G group, 12.7% in 11.2 G group and 28.2% in 33.3 G group. There was remarkable increase in apoptosis in 33.3 G group with a 2.49-fold increase to the cells in the basal 5.6 mmol/L glucose. High glucose activated the serine 270 phosphorylation of IRS correlates with JNK phosphorylation in INS-1 cells. Using Western blotting analysis, the levels of JNK phosphorylation were 3.33 fold increased and serine 270 phosphorylation of IRS was 1.17 fold increased in 33.3 G group compared to 11.2 G group (P<0.01). IGF-1 treatment inhibited phosphorylation of JNK and IRS. SP600125 treatment completely blocked JNK phosphorylation in 11.2 G group and reduced JNK phosphorylation by 90% in 33.3 G group. In addition, SP600125 treatment partly reduced serine 270 phosphorylation of IRS by 88.3% in 11.2 G group and 80% in 33.3 G group, the viability of INS-1 cells increased and the apoptosis decreased.CONCLUSION: The toxicity of chronic high glucose, which inhibits the cells viability and induces the cell apoptosis, might be related to suppress IRS signal by activating the JNK pathway. Blocking the JNK pathway might relieve the effect of glucose toxicity to the β cell function by improving the IRS signal pathway.
5.The efficient of chemotherapy weekly in the chemoradiotherapy of cervical cancer

Objective To evaluate the clinical efficacy and safety of chemotherapy weekly in the chemoradiotherapy of cervical cancer.Methods Between Jan 2003 and Oct 2007,168 patients with stage Ⅲ B squamous cell carcinoma of the uterine cervix in Chongqing Cancer Hospital were divided into two groups randomly.The observation group,total 86 patients receiving radiotherapy concomitant with weekly cisplatin and taxol;the matched group,total 82 patients,receiving radiotherapy concomitant with cisplatin plus taxol every 3 weeks.There were no statistical difference in the clinical and pathological characteristics between the two groups. We compare the 5 years survival rate and side reaction of the two groups.Results The 5 years survival rate of the observation group and the matched group were respectively 52.3%(45/86)and 42.7%(35/82),in which there was no significant difference (P >0.05).The response rate(CR+PR)were respectively 94.2%(81/86)and 92.7%(76/82)in the observation group and the matched group,in which there was no significant difference (P >0.05).Grade Ⅲ acute gastrointestinal toxicities (nausea and vomi-ting)in the matched group were exactly higher than that in the observation group [19.5%(16/82)vs.8.1% (7/86),P <0.05]. Moreover,Grade Ⅲ myelosuppression in the matched group were exactly higher than that in the observation group [20.7%(17/82) vs.9.3% (8/86),P <0.05].there were no significant difference of the incidence of the radiocystitis,radiation proctitis and radio-dermatitis between the two groups.But the Radiotherapy completion time in the matched group were exactly longer than that in the observation group [(61.12±6.71)days vs.(54.72±4.76)days,P <0.05].Conclusion The toxicity of the observation group was lower than the matched group.Moreover the efficacy of observation group is similar to the matched group.
6.Use of Psychotropic Substances in Outpatients of Our Hospital

OBJECTIVE: To investigate the use of psychotropic substances for outpatients in our hospital.METHODS: The psychiatric substances used in the outpatients between June and December in 2006 were analyzed statistically in respect of the defined daily dose,DDDs and average daily expense etc.RESULTS: Of the total 7 286 prescriptions analyzed,the psychotropic substances prescribed totaled 12 kinds,of which,2 were typeⅠ,and 10 type Ⅱ.Estazolam,Alprazolam and Diazepam ranked at the first three places in terms of numbers of prescriptions;Estazolam,Alprazolam and Clonazepam ranked at the first three places in terms of DDDs;Zolpidem,Midazolam and Alprazolam ranked at the first three places in terms of consumption sum;Nitrazepam,Estazolam and Phenobarbital ranked at the lowest places in terms of average daily expense.CONCLUSION: Of the total psychiatric substances consumed in the outpatients of our hospital,benzodiazepines were used predominantly for their merits of low cost but proved efficacy.
7.Alteration of vascular remolding and blood pressure in rat offsprings following maternal exposure to LPS during pregnancy

Objective To explore the effect of prenatal inflammation on vascular remolding and blood pressure in mid-aged rats.Methods Time-mated pregnant Sprague-Dawley(SD)rats were randomly divided into 2 groups,received peritoneal injection of 0.79 mg/kg LPS on the gestation days 8,10,and 12,or same volume of the sterile saline at the same time points.Nine pups were randomly selected from each group for the later experiments,and the offspring were named as LPS groups for those having prenatal LPS exposure and control group for those having not.Their blood pressure was determined with a rat tail non-invasive instrument by tail-cuff method from 6 weeks old to 35 weeks old,and then their aortas were taken out for media thickness(MT),diameter of lumen(LD),and the ratios of MT/LD.The expression of proliferating cell nuclear antigen(PCNA)on the vessel was detected by ELISA.The serum level of NO and plasma endothelin-1(ET-1)were measured by nitrate reductase and radioimmunoassay respectively.Results Compared with control group,LPS groups had significantly raised blood pressure,a significantly higher ratio of MT/LD,obviously increased expression of PCNA,and markedly elevated serum ET-1.Conclusion The offspring whose prenatal rats were exposured to LPS result in vascular remolding,vasofunctional disturbances and hypertension.

10.Analysis on optimal protocol of lower extremity MR angiography in patients with diabetic foot

Objective To explore the optimal protocol of lower-extremity contrast-enhanced MRA (CE-MRA) in the evaluation of diabetic foot.Methods Twenty eight healthy volunteers were scanned by CE-MRA in crus twice with parellel imaging factor (PIF) of 3 or 4.The signal-to-noise ratio (SNR),contrast-to-noise ratio (CNR) and image quality of popliteal artery,posterior tibial artery,anterior tibial artery and peroneal artery were compared.Twenty patients with diabetic foot underwent CE-MRA by both of protocol 1 and 2 in leg,crus and foot.Protocol 1 was the traditional Care-bolus protocol and protocol 2 was the optimized K-space center filling delay-time protocol.The difference of two protocols in venous aliasing and in display of femoral artery,popliteal artery,posterior tibial artery,anterior tibial artery,peroneal artery,dorsalis pedis artery,medial plantar artery and lateral plantar artery were compared.The SNR,CNR of two different PIF sequences were compared by paired t test,and the display of artery of crus was compared by Wilcoxon.The display of vessels and venous aliasing of 2 protocols of diabetic foot patients were compared by Wilcoxon.Results In the images of healthy volunteers with PIF of 3,the SNR were 267±84,174±51,147±42;and the CNR were 232 ±83,139±51,108±39 at popliteal artery,posterior tibial artery and peroneal artery.However,in the images with PIF of 4,the SNR were 239±73,157±53,132±35;and CNR were 206±71,124±50,103±33,respectively.Both the SNR and CNR were higher in the former than in the latter(t values were 2.31 to 4.11,P<0.05).There was no significant difference in the vessel display between the different PIF volunteers (P>0.05).In the protocol 1 of patients with diabetic foot,the display of popliteal artery,posterior tibial artery,anterior tibial artery,peroneal artery,dorsalis pedis artery,medial plantar artery and lateral plantar artery,the venous aliasing in crus and foot were 3.40±0.82,2.70±0.80,2.50±1.00,2.20±0.77,2.30±0.92,2.15± 1.04,1.45±0.60,2.20± 1.01,2.20± 1.06.And in the protocol 2,they were 3.85±0.37,3.55± 0.69,3.30±0.92,2.90±0.79,3.30±0.92,3.25±0.79,1.95±1.10,3.70±0.47,3.65±0.49,respectively(P<0.05).All of these parameters of protocol 2 were superior to protocol 1.Conclusion Using a higher PIF properly,setting the personalized K-space center filling delay-time can contribute to improving the image quality of whole lower-extremity MRA in patients with diabetic foot.
