1.Synthesize of water-solubility derivant of 6,7,4′-trihydroxyisoflavone (T2)and study of its inhibition effect on human breast cancer cell proliferation

Objective To synthesize of water-solubility derivant of 6,7,4′- trihydroxyisoflavone(T2 ),to characterize its structure and to evaluate its anti-tumor activity.Methods The sulfonic group(-SO3 H)was grafted to 3'position in loop B of T2 through sulfonation reaction,then strong aqua was added to above production,and obtained a water-solubility elevated compound named T2-SO3 (NH4 )2 through ammoniation.The construction of T2-SO3 (NH4 )2 was characterized by 1 H-NMR,13 C NMR,MS and element analysis.Its activity for killing human breast cancer cells (Hela)was analyzed by CCK-8 assay and flow cytometry.Re-sults The two kinds of water-solubility derivant of T2-SO3 H·2H2 O and T2-SO3 (NH4 )2 were obtained through above methods, and their yield rates were 96% and 75% respectively.The biological experiments showed that the anti-tumor activity of T2-SO3 (NH4 )2 was significantly enhanced.Conclusion Compared to T2,T2-SO3 (NH4 )2 exhibit higher biocompatibility and anti-tumor activity with vast biological application prospect.
2.Impact of sleep deprivation on coronary heart disease and progress in prevention and treatment with traditional Chinese medicines.

Sleep deprivation (SD) has been taken as an independent predictor for cardiovascular risks, which was closely related to the increased morbidity and mortality in coronary heart disease (CHD). In this article, after reviewing the impact of modern medical method sleep deprivation on CHD and studies on principle method recipe medicines for preventing and treating CHD, the authors observed the autonomic nerve dysfunction, hormonal metabolism dysfunction, endothelial dysfunction and inflammatory responses after sleep deprivation, which can cause or aggravate CHD. On the basis of the traditional Chinese medicine theories of "heart dominating the blood and vessels and the mind", the authors considered that traditional Chinese medicines can tonify heart and soothe the nerves, reducing all of the risk factors through multi-target and multi-pathway, and improve sleep and decrease the risk factors caused by sleep deprivation, which provides a new idea for the prevention and treatment of CHD.

Asaprimarilyused plasticizerin polyvinyl chloride,d(i2-ethylhexy)lphthalate(DEHP)is extensively used in consumer products,flooring and wall coverings,food contact applications,and medical devices.Because DEHP has been detected in waters,soils and air,and its metabolites have also been detected in human urinary,sufficient evidences show that DEHP is ubiquitous in the environment worldwide.DEHP is already in the blacklist of priority pollutants in water in China.Many researches indicate that DEHP is one kind of endocrine disrupting chemicals in animals,with carcinogenicity,teratogenicity and mutagenicity.Based on the researches and review on DEHP,this paper principally discussed the environmental level,metabolism and biological markers of DEHP,as well as the research progress on human health effects.

8.Clinical Significance of Lymphocyte Subsets Change in Breast Cancer Patients before and after Treatment

Objective To investigate the changes of lymphocyte subsets before and after the treatment with operation and chemotherapy in patients with breast cancer.Methods Using cytometer to measure lymphocyte subsets of CD3+,CD4+, CD8+,CD4+/CD8+ and NK of 25 breast cancer patients before and after treatment.The all 25 patients were treated with operation and chemotherapy after pathological diagnosis.The article compared the figures of T cell subsets before and after treatment.Results Compared with before operation and chemotherapy,the CD8+ statistically decreased after 1~3 months of treatment (t=2.57),and NK statistically increased after 3~6 (t=-2.23).After 6 months of treatment,the patients’ CD3+(t=-2.25),CD4+(t=-2.07)were statistically lower and NK (t=-3.56)was statistically higher.There was no significant difference for other items.Conclusion Lymphocyte subsets test against breast cancer patients can reflect immuni-ty level at a certain level during treatment.But how to explain the figures in different stage against treatment effectiveness and prognosis need further research.
9.Effect of leflunomide on peripheral blood CD28/CTLA-4 costimulatory molecules expression in guinea pig with EAE

Objective To investigate immunological mechanism underlying leflunomide ( LEF ) against experimental allergic encephalomyelitis ( EAE) by studying the effects of LEF on peripheral blood CD28/CTLA-4 costimulatory molecules expression in guinea pig with EAE.Methods 50 adult female guinea pigs were randomly divided into normal control group,low dose group,EAE control group,middle dose group,and high dose group, 10 guinea pigs in each group respectively.Low, medium and high dose group were treated with LEF 10 mg/kg· D, 20 mg/kg· D and 40 mg/kg· D orally, 1 times/day, to the termination of the experiment.The expression of CD28, CTLA-4 were detected by flow cytometry, incidence was recorded at the same time.Results Compared with EAE control group,high dose and middle dose group incubation period were prolonged, progress period were shorten and neurological dysfunction score decreased(P<0.05).Compared with normal control group, the expression of CD28 of EAE control group increased and the decreased expression of CTLA-4 (P<0.05).High, middle dose treatment group compared with EAE control group CD28 down regulated expression (P<0.05), but the expression of CTLA-4 increased(P<0.05).Among the treatment groups, of CD28 molecules with high dose group decreased significantly(P<0.05), the expression of CTLA-4 molecules with high dose treatment groups upregulated significantly (P<0.05).Correlation analysis showed that the EAE control group, the treatment group the CD28/CTLA-4 ratio in peripheral blood and nerve dysfunction score is proportional ( r=0.85, P=0.002; r=0.77, P=0.000).Conclusion LEF can reduce EAE guinea pig neurological symptoms, the better and the higher dose effect.Its mechanism may be through down-regulation of CD28 molecules in peripheral blood, upregulation of the expression of CTLA-4 molecule.so as to reduce the inflammatory response, relieve the clinical symptoms.
10.Clinical analysis for 18 cases of vulvar Bowen's disease

Objective To analyse the clinical and pathological characteristics,diagnosis,treatment and prognosis of vulvar Bowen's disease.Methods Clinical data including pathological characteristics,diagnosis,treatment methods and follow-up of 18 cases with vulvar Bowen's disease admitted to Cancer Hospital,Chinese Academy of Medical Sciences during January 1991 to June 2011 were retrospectively analyzed.Results The median age of the 18 patients was 37 years (range:23 to 64 years).Sixteen patients had symptoms of vulvar itching and two patients had no symptom.Five cases were single neoplasm focus and the other 13 cases were multiple focuses.The diagnosis of vulvar Bowen's disease was according to the pathological diagnosis.Its diagnostic characteristic was giant round or ovoid cells with mono nucleolus in the whole layer of epidermis.All the patients received operation,eleven with simple vulvectomy and other seven cases with lumpectomy.The median follow-up time was 123 months (range:5 to 197 months).Relapse was found in two cases.One patient relapsed five months postoperation and received vulvectomy.Another patient relapsed fifteen moths post-operation and received lumpectomy again.And they were follow-up for 192months and 55 months respectively after second operation without relapse.Conclusions The diagnostic characteristic of vulvar Bowen's disease is giant round or ovoid cell with mono nucleolus in the whole layer of epidermis,itsdiagnosis is according to the pathological diagnosis.Operation could get very good curative effect for patients with primary vulvar Bowen's disease and even for the recurrent patients.The prognosis of vulvar Bowen's disease is good.
