Medical Journal of Chinese People's Liberation Army 2001;0(09):-
Objective To investigate the effect of high glucose on the number and proliferation, migration and adhesion of peripheral endothelial progenitor cells (EPCs). Methods Total mononuclear cells (MNCs) were isolated from peripheral blood by Ficoll density gradient centrifugation, and then the cells were plated on fibronectin coated culture dishes. After 7 days of culture, several groups of attached cells were incubated with glucose in a series of concentrations (15, 25, 35, 45mmol/L) for different durations (6, 12, 24 and 48h). EPCs were characterized as adherent cells which were double positive for DiLDL uptake and lectin binding by direct fluorescent staining demonstrated under a laser scanning confocal microscope. EPCs were further documented by demonstrating the expression of KDR, VEGFR 2 and AC133 with flow cytometry. EPCs proliferation, migration and in vitro vasculogenesis activity were assayed with MTT assay, modified Boyden chamber assay and in vitro vasculogenesis kit, respectively. EPCs adhesion assay was performed by replating MNCs on fibronectin coated dishes, and then the adherent cells were counted. Results Incubation of isolated human MNCs with high glucose concentration decreased the number of EPCs, and this effect was most prominant when glucose concentration was 45mmol/L, and incubated for 24 hours (approximately 1 fold decrease, P

Objective: To investigate the effect of adhesion of Lactobacillus reuteri JCM1081 to enterocyte-like HT-29 cells on filamentous cytoskeleton. Methods:Cultured human intestinal enterocyte-like HT-29 cells were used.Cell permeability was examined in vitro by Evans blue labeled albumin and Examination of F-actin was conducted by direct immuno-fluorescence labeling using fluorescein labeled phalloidin. Results:Lactobacillus reuteri JCM1081 did not alter cell integrity and prevented the increase in permeability induced by enteropathogenic Escherichia coli infection. The distribution of F-actin were also not altered. Conclusion :Lactobacillus reuteri JCM1081 exerts a protective effect against the filamentous cytoskeleton rearrangement and permeability lesions promoted by enteropathogenic Escherichia coli infection.

Objective To investigate whether the neutrophil serine proteinase 3 (PR 3),a specific autoantigen of C ANCA,could induce human fetal glomerular endothelial cell apoptosis in vitro .Methods PR 3 was isolated from polymorphonuclear leukocytes obtained from C ANCA positive children with SLE.The cultured glomerular endothelial cells from human fetal kidneys were treated with PR 3 and apoptosis was assessed by different methods including the propidium iodide staining,DNA fragmentation analysis, and morphology observation. Results Percentage of apoptotic cells significantly increased ( P

Objective To investigate the effect of propofol on the expression of bone morphogenetic protein-7 ( BMP-7 ) mRNA in heart during ischemia-reperfusion ( I/R) in rats. Methods Thirty Wistar rats weighing 280-300 g were randomly divided into three groups of 10 animals : (1) control group; (2) I/R group and (3) propofol group. In control group sham operation was performed. Chest was opened and closed. In I/R group the anterior descending branch of the left coronary artery was reversibly blocked with a 3-0 thread. Ischemia of the myocardium was confirmed by the color of apex turning from bright to dark red. The block was maintained for 10 min, then relieved for reperfusion. In propofol group propofol 1mg?kg-1?min-1 was continuously infused iv during 10 min of myocardial ischemia. The animals were saclificed at the end of 4h reperfusion. Specimen was obtained from apex of heart for determination of expression of BMP-7 mRNA using RT-PCR. Results The expression of BMP-7 mRNA was 1.14 ?0.08 in control group, significantly higher than that in I/R group (0.9 ? 0.05) and propofol group (1 .00 ? 0.08) (P

China Pharmacy 1991;0(04):-
OBJECTIVE:To prepare and appraise eucalyptus oil?-cyclodextrin inclusion compound, and to testify the feasibility of transforming the dosage form of eucalyptus oil by clathration techniques.METHODS:The physicochemical properties of inclusion compound was identified by thin-layer chromatography(TLC),infrared spectroscopy(IR),ultra-violet spectroscopy(UV)and gas chromatograph mass spectrometer(GC-MS),respectively.Meanwhile,the changes in constituents and clathration outcomes of eucalyptus oil before and after clathration were also investigated.RESULTS:The analytic result of TLC,IR and UV showed that stable inclusion compound has been formed from eucalyptus oil and?-cy?clodextrin.The result of GC-MS demonstrated that there was no significant change in the essential components and the percentage composition of eucalyptus oil before and after inclusion.CONCLUSION:Stable inclusion compound can be made from eucalyptus oil and?-cyclodextrin meanwhile without changes in main components and the percentage composition of eucalyptus oil.
