Objective To observe the effects of AMPK activation on cancer cell apoptosis and to investigate the underlying mechanism and its significance to cancer therapy. Methods Human cervical cancer HeLa-S3 cells were divided into six groups:DMSO control,5 μmol/L CD437,CD437+Compound C,CD437+AICAR,Compound C,AICAR.Apoptosis rates in every groups induced by CD437 and other factors were determined by FACS and confirmed with clave Caspase-3 expression by Western blot analysis. Finally, apoptosis rates were detected in HeLa-S3 cells induced by the above factors after wt-LKB1 reintroduction by plasmid transfection. Results FACS results showed that treatment with 5 μmol/L CD437 for 8 hours could induce Hela-S3 cells apoptosis by (87.42±5.95) %.Co-incubation with 20 μmol/L compound C enhanced apoptotic effect of CD437 by (86.42±5.95) %,while co-incubation with 2 mmol/L AICAR markedly reduced cell apoptosis induced by CD437 [(51.04±8.26) %] (P < 0.05).Re-introduction of wt-LKB1 by plasmid transfection attenuated the protective effect of AICAR in CD437 induced apoptosis. Conclusion Endogeneous AMPK activation exerts a protective effect in HeLa-S3 ceils. This effect could be attenuated by re-introduction of wt-LKB1.

Objective To investigate the imaging features of Budd-Chiari syndrome(BCS) on 64 slice spiral computed tomography(64SCT) and the diagnostic value of 64SCT for BCS.Methods Twenty-nine patients diagnosed as BCS by 64SCT were retrospectively included into this study and all the patients were researched by digital substraction angiography(DSA).Two abdominal radiologists analyzed the CT imaging features of BCS,paying attention to the vascular lesion, the morphology abnormality of the liver and the degree of portal hypertension, with review of DSA findings. Results ①The accuracy of 64SCT for BCS was 93.1%(27/29),and there were 2 false positive cases and no false negative case. The accuracy of 64SCT for those patients with thrombosis of inferior vena cava(IVC) and(or) hepatic vein(HV) was high as compared to those with stenosis of IVC and(or) HV.②The morphology abnormality of the liver included hepatomegaly(24 cases),low attenuation(27 cases) and inhomogeneous pattern of parenchymal contrast enhancement(5 patients in arterial phase and 19 patients in portal vein phase).③The images of all the patients showed the features of portal hypertension. Conclusion The accuracy of 64SCT for BCS is satisfactory and the false negative is seldom. The 64SCT could accurately display the morphology abnormality of the liver and the compensatory circulation in BCS patients. For those patients with stenosis of IVC and(or) HV,however, the diagnostic power of 64SCT is limited.

Objective:To observe the expression of B7-H4 in experimental autoimmune myocarditis (EAM).Methods:BALB/c mice were randomly divided into 2 groups:the control group and the experimental group. The mice of experimental group were injected with myosin to establish EAM models , while the mice of control group were injected with complete Freund 's adjuvant and normal saline. All the mice were killed separately at the 14th,21st,30th and 45th day for lymphocyte proliferation assay ,hematoxylin-eosin staining, immunohistochemical staining and real-time PCR.Results:The inflammation infiltration of heart was most serious at the 14th and 21st day, then it was gradually relieved with time;the results of lymphocyte proliferation assay and real-time PCR were similar to that of the inflammation infiltration of heart ,which were in high level at the 14th and 21st day,and they were both higher than that of the control group ( P<0.05 );B7-H4 protein were only detected in the experimental group ,and it was constantly expressed during the whole experiment on the endothelium of heart with myocarditis.Conclusion:B7-H4 participates in the progress of EAM ,and it may be a new way of studying the mechanism of myocarditis.

Objective:To study and promote the quality standard for Shuangbai Quyu powder. Methods:A microscopic identifi-cation method was performed to identify Phellodendri amurensis cortex, Rhei radix et rhizoma, Platycladi cacumen and Andrographis herba. TLC was performed to identify Phellodendri amurensis cortex, Andrographis herba and Rhei radix et rhizoma. HPLC was used to determine the content of berberine hydrochloride in Phellodendri amurensis cortex. and the lower limit was determined. Results: The microscopic identification method was simple. The spots in TLC were distinct without any interference from the negative coutrol. Ber-berine hydrochloride had a good linear range of 0. 010-0. 400 μg(r=0. 999 9). The average recovery was 99. 51%, and RSD was 0. 31% (n=6). Conclusion:The method can be used for the quality control of Shuangbai Quyu powder effectively, which is beneficial to the quality standard promotion of hospital preparations.

Objective: To investigate the long-term outcome of anterior decompression and bone graft fusion for cervical spondylotic myelopathy(CSM) and factors affecting the outcome. Methods: Two hundred and forty-five patients with CSM were treated with anterior cervical decompression and auto iliac bone graft fusion, of whom 31 had a second operation between 4 months and 2 years after operation. Follow-up studies were carried out within 5 to 15 years after operation, averaging 6.8 years. Results: Function evaluation: excellent in 118 cases (48.16%), good in 71 (28.98%), passable in 35 (14.29%) and poor in 21 (8.57%). According to the 40 points score method, there was an average of 8 point increase in all cases, of which 101 were between 36 to 40 points, 54 between 31 to 35 points. Conclusion: The long-term outcome of surgical treatment for CSM is definite. Significant factors affecting the outcome include timing of operation, degree of pathology and technique of surgery.
