Objective To explore the effect of vasoactive intestinal peptide ( VIP) on the prevention and treatment of experimental autoimmune encephalomyelitis(EAE) rats by regulating the levels of IFN-γand IL-17A in brain tissue. Methods Sixty healthy female Wistar rats were randomly divided into 4 groups:normal control group, EAE control group, VIP low-dose group and VIP high-dose group.Myelin basic protein ( MBP)+complete Freurd′s adjuvant ( CFA) was used to establish an EAE model. The low and high-dose VIP groups were intraperitoneally injected with VIP 4 nmol/kg(0.2 ml) and 16 nmol/kg (0.8 ml) respectively every other day, while normal control group and EAE group with 0.8 ml saline for ten consecutive days. The incubation period, progression and peak of neurological dysfunction score ( NDS) changes of rats were recorded. The pathological changes, the GFAP+astrocyte activation in the brain at the morbidity peak of rats and the cytokine levels of IFN-γand IL-17A in brain tissue were observed. Results The incubation period was extended, the progression and peak NDS were shortened in the two VIP groups. In normal control group, there was no inflammatory cell infiltration or active astrocytes in brain tissue. The degree of infiltration of inflammatory cells and the degree of astrocyte activation in the VIP control group were significantly lower than in the EAE group. The cytokine levels of IFN-γand IL-17A in brain tissue were reduced in VIP groups. Conclusion By lowering IFN-γand IL-17A content in brain tissue, the infiltration of inflammatory cells and astrocyte activation are inhibited.VIP plays an important role in prevention and control of EAE.

We describe a modified technique for the in vitro study of human spermatozoal penetration into hamster oocytes which is easier to master and more simple to perform and needs no complicated facilities. It comprises the following aspects:1 The hamster oviduct is stained with 1% neutral red agar slics, so that the opening of the oviduct is easy to find and easy to be inserted with needle for flushing eggs.2. A special small tube,3 cm long with a diameter of 0.5 cm is used for fertilization instead of plastic dish with paraffin oil covering the media.3. Ordinary incubator is used rather than CO_2 incubator. With these procedures, the sperm penetration rates we got were 75%, 25%, 40%, 80% and 46% respectively in 5 experiments. It could meet the needs to evaluate the fertilizing capacity of human spermatozoa.

Objective To study the clinical effects on patients with brain metastases from lung cancer combined with Elemene Injection and radiotherapy. Method 86 patients with brain metastases from lung cancer were randomly divided into control group and treatment group, 43 cases in each group. Control group was given conventional radiotherapy treatment, while treatment group combined Elemene Injection with radiotherapy. Results Compared with control group, treatment group’s disease control effect was obviously better. Survival rates after treatment for six months, one year, three years were higher than control group, and the number of adverse reactions were lower. Serum MMP-2 and MMP-9 levels were significantly greater. Radiotherapy plan time and total time in hospital for treatment were significantly shorter. Conclusion The combination of Elemene Injection with radiotherapy treatment for patients with brain metastases from lung cancer is very obvious.

Ebosin is a novel exopolysaccharide produced by Streptomyces sp.139 with remarkable activity against rheumatic arthritis in vivo. In this paper, we reported effects of Ebosin on the inflammatory cytokines including interleukin-1beta (IL-1beta), interleukin-6 (IL-6) and tumor necrosis factor alpha (TNFalpha) in THP-1 cells. With the special fluorogenic peptide as substrates, the enzymatic activities of interleukin-1beta converting enzyme (ICE) and TNFalpha-converting enzyme (TACE) were inhibited by Ebosin separately. Using the real-time reverse transcription polymerase chain reaction (real-time PCR), the mRNA synthesis of the three cytokines were identified decline separately by Ebosin. The secretion quantum of three cytokines in THP-1 cells with Ebosin was lower than that of normal THP-1 cells determined by ELISA assay and Western blotting. All of these results showed that Ebosin has remarkably suppressed synthesis of the three cytokines in THP-1 cells through different pathways. The primary study of Ebosin on anti-inflammation mechanism was promoted developing the new drugs treating rheumatic arthritis.

International Journal of Cerebrovascular Diseases 2017;25(6):541-545
Blood-brain barrier (BBB) injury is an important pathological change of ischemic stroke.Occludin constitutes the major component of the tight junction of BBB, which plays a significant role in maintaining the BBB integrity.Cerebral ischemia initiates occludin degradation through the induction of the activation of inflammatory mediators, reactive oxygen, and matrix metalloproteinases, and causes the changes of tight junction cytoskeletal protein interaction, resulting in BBB integrity destruction and increased permeability. This article mainly elaborates the relationships of occludin with BBB, brain edema and hemorrhagic transformation, as well as the possibility of occludin as an intervention target for ischemic stroke.
