Background and purpose:There were reports that radiotherapy might intensify angiogenesis and result in the resistance of tumor to radiotherapy.Endostar,a new type rh-endostatin,may specifically inhibit angiogenesis and tumor growth,so it may reduce the resistance to radiotherapy. This study was to evaluate the inhibitory effect of Endostar in combination with radiation compared with cisplatin(DDP) plus radiation on lung adenocarcinoma A549 in BALB/c nude mice. Methods:We established A549 lung adenocarcinoma xenograft models. When the largest diameter of tumor reached 1.0 cm,all mice were randomized into 4 groups(n=6):control,radiation alone, radiation plus DDP,radiation plus Endostar. We measured the largest diameter and the vertical diameter of the tumors at different time points after treatment. At the 16th day, mice were executed and the tumors were subjected to removal for the analysis of apoptosis, and the expression level of basic fibroblast growth factor(bFGF) mRNA by reverse transcription-polymerase chain reaction(RT-PCR) and vascular endothelial growth factor(VEGF) were also detected by immunohistochemistry. Results:The tumor growth velocity of control, radiation alone, radiation plus DDP,radiation plus Endostar were(162?6)%,(131?8)%,(104?7)% and(108?11)% respectively(P0.05).The apoptosis rates of control, radiation alone, radiation plus DDP,radiation plus Endostar were(12.2?1.1)%,(16.5?0.8)%,(24.4?1.5)% and(20.5?1.9)%,respectively(P

In order to investigate the inhibitory effects of Endostar (rh-endostatin, YH-16) in combination with radiotherapy on lung adenocarcinoma A549 in mice and the interaction mechanisms of combined therapy, the transplantation tumor models of A549 lung adenocarcinoma were established. When the largest diameter of tumor reached 1.0 cm, all nude mice were randomly divided into 4 groups: Endostar group, radiotherapy group, radiotherapy plus Endostar (combined treatment) group, and control group (n=6 in each group). The largest diameter and the vertical diameter of tumor were measured at different time points. At the 16th day, mice were executed, and the tumors were applied to analysis of rate of tumor cell apoptosis, and the expression levels of basic fibroblast growth factor (bFGF) mRNA were detected by reverse transcription-polymerase chain reaction (RT-PCR) and those of vascular endothelial growth factor (VEGF) by immunohistochemistry. The results demonstrated that the rate of tumor inhibition in combined treatment group was higher than that in other groups. And the rate of tumor cell apoptosis in combined treatment group was also higher than that in other groups. Meanwhile, the levels of bFGF mRNA and VEGF expression in combined treatment group were lower than those in other groups. It was concluded that Endostar obviously enhanced the curative effectiveness of radiotherapy on lung adenocarcinoma A549 in mice. The underlying mechanisms may involve the down-regulation of bFGF mRNA and VEGF expression to inhibit angiogenesis by Endostar and the cooperative effect of Endostar and radiotherapy to synergistically promote tumor cell apoptosis. And Endostar inhibits angiogenesis by down-regulating the expression of bFGF mRNA and VEGF.

Purpose To investigate the expression level and the role of TUSC7 in human cutaneous malignant melanoma (CMM).Methods Quantitative real time-PCR (qRT-PCR) assay was performed to detect the expression of TUSC7 in 60 cases of CMM tissues,20 cases of benign nevus,4 CMM cell lines, and one normal human epidermal melanocytes. Then overexpression of TUSC7 was performed and its role in tumor progression was explored. Results TUSC7 expression was significantly downregulated in primary CMM tissues (n =60) compared to benign nevi (n =20),which were significantly downregulated in all the four melanoma cell lines, especially in A375 cells, compared with the normal melanocytes cells (all P < 0.05).In comparison with the A375 cells transfected with the empty plasmid, those transfected with pcDNA-TUSC7 showed an obvious decrease in the proliferation and colony formation activity, while increase in the apoptosis rate (all P < 0.05).Conclusion Our results suggested that the dysregulation of TUSC7 may play an important role in the CMM progression.

Objective It has been shown that adult brain is still capable of neurogenesis which can beinhibited by activation of NMDA receptor.Since lidocaine can inhibit NMDA-mediated excitatoryueurotransmission,we aimed to investigate the interaction between lidocaine and NMDA on the proliferation ofpheochromocytoma cells which are used as a model for central neuronal cells. Methods The PC 12 ceils culturedin vitro were divided into 6 groups:(1)control group, cultured in normal DMEM complete nutrient liquidmedium;(2)NMDA group, cultured in DMEM containing 400 ?mol?L~(-1) NMDA;(3)-(6)lidocaine group, cultured in DMEM medium containing 400 ?mol L~(-1) NMDA and 10,10~2,10~3 or 10~4 ?mol?L~(-1) lidocaine. After 5day incubation, the cell cycle progression was analysed using a flow cytometer. The percentage of cells in S-phase(S-phase fraction,SPF)was determined and proliferation activity(cells in S+G_2 phase/cells in M-phase)wascalculated.Results NMDA 400 ?mol?L~(-1) significantly decreased the SPF of PC12 cells in group 2 compared tocontrol group, and proliferation activity(S+G_2 phase/M-phase)was also significantly reduced(P0.05).The SPF of PC12 cell ingroup 3 and 6(10 and 10~4 ?mol?L~(-1) lidocaine)was also significantly higher than that in NMDA group butsignificantly lower than that in control group. Conclusion NMDA inhibits proliferation of PC12 cells whilelidocaine can antagonize the inhibitory effect of NMDA and promotes proliferation and differentiation of centralneuronal cells.

Objective To observe the effects of in vitro isolated Schwann cells co-cultured with chemically acellular nerve allografts on improving repair of large facial nerve defects. Methods A total of 30 Wistar rats were equally randomized into three groups, ie, experimental group, allograft group and autograft group. Nerve defect of 12 mm in length was made in the left inferior buccal branch of facial nerve and repaired with acellular nerve allograft implanted with Schwann cells, acellular nerve allograft and fresh tibial nerve autograft respectively. At the 5th month postoperatively, the function and morpholo-gy of the regenerated nerves were observed by electrophysiological method, methylene blue staining and transmission electron microscope. Results In experimental group, the recovery rate (operation side/normal side) of amplitude of nerve-muscle action potential was (35.8±2.5)%, the lantency recovery rate (normal side/operation side) (65.8±2.9)%, the number of the regenerated axon 1 570±188 and the myelin thickness (0.383±0.031) μm. The results in the experimental group were significantly supe-rior to those in the acellular nerve allograft group (P < 0.05), with similar results to fresh nerve autograft group (P > 0.05). Conclusion Transplantation of Schwarm cells in acellular nerve allograft can im-prove repair of large facial nerve defects.

The diethyl sulfate (DES) mutagenesis was chosen for the mutagenic treatment to Phellinus igniarius, and the relationship of mutagenesis time and death rate was investigated with 0.5% DES. The differences of mycelial growth speed, liquid fermentation mycelia biomass, morphology and pigment classes of secondary metabolites production speed and antioxidant activities of metabolite products were discussed. The study displayed that DES mutagenesis could change mycelial morphology without obvious effect on mycelium growth, and the DES mutagenesis improved antioxidant activities of the active ingredients of P. igniarius and had more antioxidant activity of hypoxia/sugar PC12 nerve cells than that of P. igniarius.

Spleen kidney Yang deficiency (SKYD) diarrhea is a common syndrome in tranditional Chinese medicine (TCM). Until now, there is not an ideal SKYD diarrhea rat model for the research. In this study, we compared single factor way (method I, injecting hydrocortisone and gavaging Sennae Folium) with compound factors way(method II, gavaging adenine, improper diet, exhaustion, and gavaging Sennae Folium) on establishing SKYD diarrhea rat model. After modelling, diarrhea index, D-xylose excretory rate, NOS/cGMP signal transduction system, organ index and histopathology examination were used to evaluate the two ways. The results showed that, compared with health group, all the assessment criterias of method I and method II had significant differences (P < 0.01, 0.05). In addition, the index such as diarrhea index, NOS/cGMP signal transduction system, organ index (kidney, testis and thymus) and histopathology examination had significant differences (P < 0.01, 0.05) between method I and method II. In conclusion, the compound factors modelling method better conforms to the symptom of diarrhoea model caused by SKYD. This new modelling method provides a basis for studying on TCM astringents warming and tonifying the spleen and kidney, relieving diarrhea.
