1.The Relationship Between KRAS Status and Response of Cetuximab in Combination with Oxaliplatin,Leucovorin and Fluorouracil/Xeloda in the First Line Treatment of Advanced Colorectal Cancer

Objective This study assessed whether the best overall response rate(ORR) of cetuximab combined with oxaliplatin,leucovorin and fluorouracil/Xeloda was superior to that of this method alone as first-line treatment for metastatic colorectal cancer.The influence of KRAS mutation status was investigated.Methods Patients received cetuximab(400mg/m2 initial dose followed by 250mg/m2,wk thereafter)less than 2 times plus chemotherapy(oxaliplatin 130mg/m2 on day 1,plus leucovorin 200mg/m2 and fluorouracil as a 400mg/m2 bolus followed by a 600mg/m2 infusion during 22 hours on days 1 and 2) or chemotherapy alone.Treatment was continued until disease progression or unacceptable toxicity.KRAS mutation status was assessed in the subset of patients with assessable tumor samples.Results The confirmed ORR for cetuximab plus oxaliplatin,leucovorin and fluorouracil/Xeloda was higher than that with alone(42.86% vs 21.74%).A statistically significant increase in the odds for a response with the addition of cetuximab to oxaliplatin,leucovorin and fluorouracil/Xeloda could be established.In patients with KRAS wild-type tumors,the addition of cetuximab to oxaliplatin,leucovorin and fluorouracil/Xeloda was associated with a clinically significant increased chance of response(ORR 54.55% vs 21.74%) and a lower risk as compared with chemothrapy alone.Cetuximab plus oxaliplatin,leucovorin and fluorouracil/Xeloda was generally well tolerated.Conclusion The clinical effcacy of chemothrapy(oxaliplatin,leucovorin and fluorouracil/Xeloda) plus cetuximab is better than only chemothrapy.KRAS mutational status was shown to be a highly predictive selection criterion in the treatment decision regarding the addition of cetuximab to oxaliplatin,leucovorin and fluorouracil/Xeloda for previously untreated patients with metastatic colorectal cancer.
2.Analyzing Financial Security Situation of Disease Prevention and Control System Construction in Jiangxi

Objective: To understand current situation and existing problems in financial security of the center for disease control and prevention system in Jiangxi, to provide scientific references for advancing continuous, fast and healthy development of the system. Methods: combining three methods of questionnaire, group discussions and field surveys to investigate the situation of financial income from 112 centers of disease control and prevention in Jiangxi. Results: The government finance investment increased by year, but the funds of daily work and personnel is insufficient, public and regular business funds of some institution still depend on paid service income. Conclusion: It needs to further enhance financial structure adjustment and disease control investment, increase disease control input, change government appropriation method for disease prevention and control institutions, change the government ’ s funding mode to “purchase system” , reasonably pricing public product, the government on behalf of the people buying services according to quality and quantity from the disease prevention and control institutions.
3. Immunological Effects of Total Flavones from Leaves of Choerospondias axillaris on Mice

Objective: To investigate the effect of total flavones from the leaves of Choerospondias axillaris (TFLCA) on the immune function of normal mice and to provide the experimental basis for the reasonable application of C. axillaris. Methods: The carbon clearance method, cutaneous delayed hypersensitivity reaction method, serum hemolysin method, and index of immune organs were used to study the effect of TFLCA on the immune function of mice. Results: TFLCA could enhance the phagocytic function of mononuclear macrophage and the cutaneous delayed hypersensitivity reaction of mice, and increase the content of hemolysin antibody and the thymus index in mice. Conclusion: TFLCA could improve the celiac macrophage activity and specific immunity of mice, and TFLCA, consisting with the total flavones of Choerospondiatis Fructus (TFCF), has the effect on the immune function of mice. So both TFLCA and TFCF have the regulatory effects on the immune function of mice. © 2013 Tianjin Press of Chinese Herbal Medicines.
4. Chemical Constituents from Citrus aurantium L.

OBJECTIVE: To study the chemical constituents of Citrus aurantium L. METHODS: Chromatographic techniques were employed for isolation and purification of the constituents and their structures were determined by spectral analysis and chemical evidence. RESULTS: Sixteen compounds were obtained and identified as pyrrolezanthine-6-methyl ether (1), methyl 3-hydroxy-4-methoxybenzoate (2), 3-hydroxy-4-methoxybenzoic acid (3), methyl 3,4-dihydroxybenzoate (4), methyl 3-(4-acetoxyphenyl)propanoate (5), 4-hydroxybenzoic acid (6), scopoletin (7), 5-(hydroxymethyl)furan-2-carbaldehyde (8), hesperidin (9), naringin (10), gossypetin-8,3'-dimethylether-3-O-β-D-galactoside (11), 2-homoeriodictyol-7-O-β-D-glucopyranoside (12), 6-demethoxytangeretin (13), naringenin (14), 5,6,7,8,4'-pentamethoxyflavone (tangeretin) (15), and 5-O-desmethylnobiletin (16). CONCLUSION: Compounds 124811-13 are for the first time obtained from Citrus.

Type 1 diabetes mellitus(T1DM)is a chronic, immune-mediated disease characterised by the destruction of insulin-producing cells.The specific pathogenesis of T1DM has not been clarified.It is mainly believed that the occurrence of T1DM is caused by the joint action of genetic and environmental factors.The occurrence, development, treatment and prevention of T1DM are urgent problems to be solved.A number of studies have found that vitamin D is involved in the pathological process of many autoimmune diseases and is related to cell proliferation, differentiation, apoptosis and other mechanisms.Vitamin D may play a key role in the pathological mechanism of T1DM.Here we review the relationship between the incidence, prevention and treatment of T1DM and vitamin D.
6.Rabbit model of aspergillus keratitis induced by modified corneal surface lens method

Background Animal model of fungal keratitis is an available tool to the experimental study of the pathogenesis mechanism of fungal keratitis. Current modeling methods of fungal keratitis include corneal scratching, corneal stroma injection and corneal surface lens methods. But these methods still have their own shortages. Objective This experiment was to create a fungal keratitis animal model by modifying corneal surface lens method. Methods Modified animal models of fungal keratitis were created by modified corneal surface lens method in 12 general adult New Zealand white rabbits. The filter papers soaked 108 spores / ml or A106spores / ml of spergillus fumigatus suspension were attached on the de-epithelial cornea surface and fixed with contact lens and tarsorrhaphy for 2 days, and the filter paper with physiological saline was used as control group. The symptoms of anterior segment were examined under the slit lamp in 3 ,7 and 14 days after surgery and scored based on the criteria of Dong. Corneal scraping was stained with 10% potassium hydroxide and calcofluor white stain to observed mycelium under the fluorescence microscope. Corneal tissue sections were examined by hematoxylin-eosin staining and periodic acid Schiff staining under the light microscope. The use of animal followed the Standard of Association for Research in Vision and Ophthalmology. Results Fungal keratitis models were successfully established in 6 eyes and 4 eyes in 108 spores/ml group (6/6) and 106 spores/ml group respectively. The symptom was more severer and score was higher in the eyes of 108 spores/ml group than that in 106 spores/ml group. At 3 and 7 days after surgery,the symptom scores of fungal keratitis models were higher than those of control group from 3 through 7 days with the statistically significant difference (P<0. 01) and the symptom scores of 108 spores/ml group were significantly higher than those of 106 spores/ml group (P<0. 01). At 14 days after surgery, the symptom scores of 108 spores/ml group were still higher than those of control group (P<0. 05). Fungal hyphae was seen in the corneal scrapes in 108 spores/ml group and 106 spores/ml group respectively from 3 through 7 days after surgery. Inflammatory cell infiltration, stroma cells necrosis and fungal hyphae were presented in 108 spores/ml group, and the corneal neovascularization could be observed in 108 spores / ml group 14 days later. Fungal culture revealed the positive outcome in both 3 and 7 days after surgery in 108 spores/ml group,but in 106 spores/ml group,the positive result was only in the 3rd day. Conclusion Modified corneal surface lens method is more feasible and sample in the model of Aspergillus keratitis. This animal model of Aspergillus keratitis is practical for the further study of fungal keratitis.
8.Relationship between Personality, Psychosocial Factors and the Morbidity of Irritable Bowel Syndrome

ObjectiveTo explore the effect of personality and psychosocial factors on irritable bowel syndrome (IBS).MethodsPersonality and psychological status of 46 IBS patients and 32 healthy individuals were evaluated with Eysenck Personality Questionaire (EPQ), Symptom Checklist-90 (SCL-90) and Life Events Scale (LES).ResultsIn EPQ, E (extroversion-introversion) scores of IBS group were lower than those of control group; N (neuroticism) scores of IBS group were significantly higher than those of control group (P<0.01). In SCL-90, the scores of somatization, compulsion, anxiety, depression and etc. were higher than those of control group (P<0.05). The frequency of meeting negative life events, the quantity of stimulus of meeting negative life events and the total quantity of stimulus of meeting life events of IBS patients within one year until episode were significantly higher than those of control group (P<0.01).ConclusionPersonality psychological characteristics and life events before illness are associated with the morbidity of IBS.
9.Linkage disequilibrium and mutation rate analysis of sixteen X-STR loci.

OBJECTIVE:
To assess the patterns of linkage disequilibrium (LD) of 16 STR loci on X chromo- some and investigate the genetic stability.
METHODS:
Genomic DNA samples extracted from blood stains from 500 unrelated individuals and 885 lineage members from Eastern Chinese Han population were genotyped through multiplex amplification using IDtyperX-16 kit by our independent research followed by capillary electrophoresis. LD was assessed by PowerMarker v3.25 software and mutation rate of every locus was analyzed.
RESULTS:
LD were not found at the 16 X-STR loci. Allele mutations were observed at 10 loci. Among them, mutation rates of DXS6809 and DXS7132 were both up to 0.0048.
CONCLUSION
When the 16 X-STR loci included in IDtyperX-16 kit were used for parentage testing, product princi- ples can be applied to calculate the likelihood, but mutation should be taken into consideration in the case that the genotypes do not meet the genetic law (especially at DXS6809 and DXS7132).

10.Comparison of osteopontin and mineralization culture medium in the induction of dental pulp stem cells into osteoblasts

Objective:To compare the osteogenic differentiation of dental pulp stem cells(DPSCs)induced by osteopontin(OPN)and mineralizing culture medium(MCM).Methods:DPSCs were cultured with OPN(OPN group)and MCM(MCM group)respectively. The morphology of the DPSCs were observed under inverted microscope.The mineralize nodules were observed by alizarin red staining. RT-RCR was used to detect the mRNA expression of bone sialoprotein (BSP),Runt-related transcription factor 2(Runx-2),osteocal-cin(OCN)and collagen-1(Col-1).Results:Similar number of mineralized nodules was found in the 2 groups(P >0.05)after 28 day culture.The mRNA expression level of BSP gene in OPN group was higher than that in MCMgroup(0.864 ±0.112 and 0.514 ±0.068, P <0.05),while the expression level of Runx-2 gene in OPN group is lower than that in MCMgroup(0.186 ±0.017 and 0.324 ±0. 058,P <0.05).The expression level of Col-1 and OCN genes in both groups were similar(P >0.05).Conclusion:The capabilities of OPN and MCMin inducing osteogenic differentiation of DPSCs are similar.
