Aim To study the mechanism of curcumin induced U251 cell apoptosis. Methods U251 cells were treated with 20～100?mol?L~ -1 curcumin for 24 h and the growth inhibition rates of U251 cells were measured with MTT method. MTT method was used to measure the caspase inhibitors effect on curcumin too. Cell apoptosis was inspected with flow cytometry (FCM) and observed using electron microscope. The protein levels of FAK in U251 cells were observed by SP immunocytochemistry. The activity of caspase-3 was measured by spectrofluorometry. Results Curcumin inhibited the proliferation of U251 cells,induced apoptosis of U251 cells. At the same time the protein levels of FAK in U251 cells decreased and the activity of caspase-3 increased significantly. The apoptosis of U251 cells was partially reversed by caspase inhibitors. Conclusion Curcumin induced apoptosis via inhibition of expression of FAK and activation of caspase-3.

Objective To compare the surgical outcomes between laparoscopic and open surgeries in treatment of ectopic oviduct pregnancy, and investigate the clinical value of laparoscopic surgery. Methods Two hundred and forty-six patients with ectopic oviduct pregnancy in our hospital from June 2009 to March 2013 were retrospectively analyzed. 134 cases were treated with laparoscopic operation and 112 with open surgery. Some parameters such as operative time, blood loss, usage of pain-killer, hospital stay were compared between both groups. Results All operations were successful. Laparoscopic surgery was shown to be superior to open operation in the parameters of operative time, blood loss, usage of pain-killer and hospital stay ( < 0.05) . Conclusion Compared with open operation, laparoscopic surgery has the advantages of less trauma and rapid postoperative rehabilitation. It may become the first line treatment for ectopic oviduct pregnancy.

Objective To describe the structure and the level of the stress of nursing students in clinic practice,and supply reference for reform of clinical teaching method.Methods Totally 100 undergraduate nursing students in clinic practice,who would graduate from nursing school and begin to work in 2013,were interviewed and investigated.Results The main stress sources were divided into five aspects:recognition,clinical nursing ability,career development,field response and exhaustion of body and mind.Conclusions The structure and level of the stress of nursing students changed a lot.Some new appropriate reform should be introduced into clinic education,in order to help nursing students to complete psychological change and role adaptation.

Objective To analyze the doctors' working enthusiasm and its influencing factors on county level hospitals.Methods 17 seminars were held in 8 counties of 3 provinces to investigate 1055 doctors with questionnaires, analyzing the three scales of working enthusiasm, working enthusiasm influencing factors, and changes since the 2009 new healthcare reform.Results Working enthusiasm comprises inner drive, working pleasure, work recognition and external recognition, and inner drive of which is found to play the most important part, with 20.55％ variance contribution.Among the six dimensions of the influencing factors, working conditions rank the highest with 19.46％ variance contribution.Conclusion Optimized measures in the healthcare reform, better working conditions and social environment can improve working enthusiasm of these doctors.

OBJECTIVETo observe the effect of Chinese medicine and pharmacy (CMP) on the mortality of senile HIV/AIDS patients as adjunctive therapy.

METHODSHIV/AIDS patients of a certain rural area of Hanna Province, who were recruited in national CMP HIV treatment trial program (NTCMTP) in 2004, were enrolled as the CMP treatment group. HIV/AIDS patients in the same village without recruiting in NTCMTP were enrolled as the non-CMP treatment group. Data related to subjects were collected from the database of NTCMTP and National HAART Reporting System. Multiple regression analysis under Cox proportional hazard model was applied to examine the risk factors for death of senile HIV/AIDS patients.

RESULTSA total of 436 HIV/AIDS were enrolled in this study, 204 in the CMP treatment group and 232 in the non-CMP treatment group. There were 70 AIDS-relative deaths in the CMP treatment group, with 8-year mortality rate of 37.74%. There were 111 AIDS-relative deaths in the non-CMP treatment group, with 8-year mortality rate of 48.34%. The 8-year mortality rate was higher in the non-CMP treatment group than in the CMP treatment group (chi2 = 5.136, P < 0.05). Results of univariate Cox proportional hazards regression analysis showed the hazard ratio in the non-CMP treatment group was 1.41 times that of the CMP treatment group (P < 0.05). Result of multivariate Cox proportional hazards regression analysis showed the hazard ratio in the non-CMP treatment group was 1.44 times that of the CMP treatment group (P < 0.05). Besides, gender and marital conditions were significantly associated with death of HIV/AIDS patients.

CONCLUSIONCMP treatment was favorable to lower the mortality rate of senile HIV/AIDS patients, and its objective evaluation awaits for further prospective study.

Objective To investigate the feasibility and clinical effect of immediate ureterovesical reimplantation after ureteral rupture during laparoscopic hysterectomy. Methods From August 2010 to December 2015, 5 cases of ureteral rupture during the operation of laparoscopic hysterectomy were treated with immediate ureterovesical reimplantation under laparoscopy. Results All operations were successfully performed without transversion to open surgery. No patients with urinary leakage occurred. The mean follow-up were 21 months (range 3-60 months) . No cases with ureteral stricture were observed. Slight urine reflux was found in two patients, of whom obvious hydronephrosis and renal damage were not found. Conclusion Immediate ureterovesical reimplantation under laparoscopy is a feasible, safe and minimal invasive method for treatment of ureteral ruputure during laparoscopic hysterectomy.

Objective To explore the impact of four different anesthetic drugs commonly used in animal experi-ments on cardiovascular system in rats.Methods Electrocardiogram ( ECG) and blood pressure were dynamically recor-ded by a BioPac MP150 system after anesthesia.In addition, the blood glucose at different time points and hepatic func-tion, kidney function, cardiac enzymes and electrolytes at the end of the test were collected.Rusults Chloral hydrate caused severe ventricular arrhythmia.Isoflurane had inhibitory effect on the heart rate.Pentobarbital sodium induced a in-crease of ECG P wave.Urethane caused J point elevation of ECG.Blood pressure in the urethane-and pentobarbital sodi-um-treated groups were increased.Chloral hydrate caused CK to be raised, while isoflurane showed the opposite effect on CK and CKMB.Alanine aminotransferase and aspartate aminotransferase in the pentobarbitol sodium and isoflurane groups were decreased.Creatinine in the chloral hydrate, pentobarbital sodium and isoflurane groups were lower, and the serum sodium and potassium were decreased in the four groups.Conclusions Chloral hydrate has obvious effect on the cardio-vascular system, and is not suitable for animal studies on cardiovascular diseases.Pentobarbital sodium, urethane, isoflu-rane can be chosen for animal studies on cardiovascular diseases.

Objective To explore the protective effect of resveratrol against oxidative damage to cultivated fibroblasts irradiated with UVB. Methods Fibroblasts from normal human skin cultured in vitro were divided into 5 groups (a normal control group, a group irradiated with UVB, a group treated with resveratrol before UVB irradiation, and a group treated after irradiation). A monolayer of fibroblasts was irradiated with UVB at 60 mJ/cm2. The vitality of the cells was measured using the methylthiazol tetrazolium (MTT) method. The activity of superoxide dis-mutase (SOD) and malondialdehyde (MDA) content were determined using enzyme biochemistry. Results Resveratrol over 100 μM inhibited the proliferation of fibroblasts. Resveratrol under 100 μM improved the proliferation of cells. The optimal concentration was 50 μM. UVB irradiation decreased the vitality of the cells and SOD activity, and it significantly enhanced MDA content. Conclusions Resveratrol treatment before or after UVB irradiation elevates the survival rate of fibroblasts, enhances the activity of SOD, and decreases MDA content. Resveratrol at low concentration could improve the proliferation of fibroblasts, and at high concentration could inhibit their proliferation. Res-veratol at 50 μM relieves the inhibited proliferation of fibroblasts damaged by UVB irradiation.

Objective To investigate the effects of essential fatty acids(EFA)on the contents of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in the brain tissues of rats. Methods A total of 30 neonatal rats, 1 month old, were randomly divided into normal, EFA deficiency and fish oil supplement groups and given different feeding stuff for 3 months respectively. Objective To construct the prokaryotic expression vector of human procalcitonin(PCT) and obtain the highly purified recombinant PCT protein.Methods PCT primers were designed based on the sequence of PCT gene from NCBI,and PCT gene was amplified by PCR.Then,the recombinant vector PCT/pET-22b(+) was constructed and transferred into E.coli BL21 to induce the expression of recombinant PCT protein.Last,the recombinant PCT protein was purified by the nickel column affinity chromatography and identified by Western blot and the colloidal gold method,respectively.Results The results of agarose gel electrophoresis showed that the product of PCR amplification was about 350 bp.The homology comparison analysis revealed that the PCT gene fragment(348 bp) was successfully inserted into pTG19-T vector,and that no any base mutation was found.When the recombinant plasmid of PCT/pET-22b(+) was digested with BamH Ⅰ and HindⅢ,two pieces of about 350 bp and 5 500 bp were obtained.SDS-PAGE showed that the recombinant PCT protein with about 14 000 of Mr was existed in soluble form,and was easily obtained by the nickel column affinity chromatography.Moreover,western blot and the colloidal gold method demonstrated that the recombinant PCT protein was successfully expressed and contained histidine label(His-tag).Conclusion The recombinant vector PCT/pET-22b(+) is successfully constructed by the recombinant DNA technology,and the recombinant PCT fusion protein is successfully obtained by the nickel column affinity chromatography.
