1.Effects of Simulated Microgravity on the Morphology and Growth of PC12 Cells.

Objective To observe whether the increase of oxidative stress in PC12 cells could influence the levels of protein carbonyls and nitrotyrosine and alter the cytoskeleton and cell morphology under clinostat condition,and whether the increase of content of nitrate/nitrite in cell culture medium could influence the cell proliferation and differentiation.Method Cell morphology,carbonylated actin and nitrotyrosinated tubulin,and mRNA and protein express of nNOS and iNOS were observed and determined with immunofluorescence and RT-PCR technology in clinostat rotated and control static groups.At the same time,cell density was measured and cell cycles were detected with flow cytometry.The relationship between all these changes and NOS were also analyzed.Result The levels of carbonylated and nitrotyrosinated cytoskeleton protein were altered,no obvious changes in cell morphology but neurite outgrowth after on a clinostat rotation.Cell density also increased significantly,DNA synthesis in cell cycles was shortened.Conclusion All of these results indicate that simulated weightlessness do not alter cell morphology and is beneficial to the growth of PC12 cells.The mechanism involved may be associated with the increase of NOS activity.
2.Intervention effects of fluid wax on adhesive small bowel obstruction

Objective To observe the intervention effects of fluid wax on the therapeutic course of patients with adhesive small bowel obstruction.Methods Two hundreds and eighty-eight patients with adhesive small bowel obstruction admitted into the Department of Gastrointestinal Surgery of Huzhou Central Hospital from December 2014 to June 2016 were enrolled, and they were divided into a fluid wax group and acontrol group by mechanical sampling method, each group 144 cases. The control group was treated with conventional comprehensive non-surgical treatment, in the fluid wax group, on the basis of the above conventional treatment, additionally after 2 hours of gastrointestinal decompression, the fluid wax 3 mL/kg was injected through a gastric tube that then was closed by a clip for 2 hours. The first exhaust and defecation times, the time for amelioration of abdominal pain, the time of gas-liquid flat disappearance, the length of stay in hospital, the rate of operation and the occurrence of adverse reactions were observed in the two groups.Results After treatment, the first exhaust time, the first defecation time, the time of relieving abdominal pain, the time of gas-liquid flat disappearance and the length of stay in hospital were significantly shorter in fluid wax group than those in control group [the first exhaust time (hours): 29.97±19.71 vs. 49.28±33.61, the first defecation time (hours): 60.25±28.37 vs.74.23±50.12, the time of relieving abdominal pain (hours): 35.78±20.98 vs. 51.83±25.02, the time of gas-liquid flat disappearance (hours): 71.60±39.50 vs. 90.98±57.91, the length of stay in hospital (days): 7.00±3.77 vs. 9.00±5.81, allP < 0.05], and the rate of operation in the fluid wax group was lower than that in the control group [18.75% (27/144) vs. 27.08% (39/144),P < 0.05]. No patients died in the two groups. In nearly 1 year follow-up, there were no adverse reactions associated with the study in the fluid wax group.Conclusion The intervention of fluid wax combined with conventional non-surgical methods can significantly shorten the disease course, reduce the rate of operation and the hospitalization time in patients with adhesive small bowel obstruction.

4.Gene mapping in Avellino corneal dystrophy pedigree

AIM:To point the susceptible gene in Avellino corneal dystrophy family with autosomal dominant inheritance.
●METHODS: Genomic DNA was extracted from the peripheral blood samples of all individuals of the pedigree. Several microsatellite makers were selected for gene scan in the hot regions of mutation. Linkage analysis was carried out using a Linkage software package. The haplotype data were processed using Cyrillic software to define the region of the disease gene.
●RESULTS: ln our pedigree, significant evidence of linkage was obtained at marker D5S396 and D5S393 [LOD score (Z)=3. 01, recombination fraction (θ)=0. 00]. The haplotype analysis of our pedigree was located between the microsatellite markers D5S808 and D5S638.
●CONCLUSION:The pathogenic gene of the Avellino corneal dystrophy pedigree is traced to a 11. 2 cM region in the chromosome 5q.

6.Relationship of von Willebrand factor gene single-nucleotide polymorphism with thrombosis diseases.

Recently it has been discovered that not only von Willebrand factor (vWF) decrease results in von Willebrand disease, but also vWF increase can lead to several thrombosis diseases. Plasma vWF level is affected by genetic factors. Individuals with different single nucleotide polymorphism (SNP) genotype in vWF have different susceptibility to disease; individuals with different blood group have different plasma vWF level. Environment factors also affect plasma vWF level. Understanding relationship of polymorphisms in promoter, exon and intron with thrombosis diseases contribute to prevent and cure these diseases. In this review, the relationship of SNP in promoter, exon and intron of vWF gene with thrombosis diseases is summarized.

7.Dextran-spermine polycation as a vector for gene transfection in vitro.

This work reports the properties of dextran-spermine polycation (DSP) as a gene vector and its gene transfection efficiency in vitro. Oxidized dextran was reacted by reductive amination with spermine to obtain DSP, the resulted polycation was then incubated with plasmid pEGFP to form polyplexes by electrostatic interactions. DSP formed stable polyplexes when the weight ratio (DSP/DNA) varied from 4 : 1 to 20 : 1. The particle size and zeta potential of polyplexes were in the range of 162.6 - 187.9 nm and increased from + 8.45 mV to + 39.6 mV, respectively. DSP could effectively protect condensed DNA from DNase I degradation, and it showed strong buffering capacity in a certain pH range. The highest yields of transfection efficiency were found to be as high as Lipofectamine 2000 when the polyplexes were transfected to SMMC-7721 and BHK-21 cells at the weight ratio of 8 : 1. This research indicates that dextran-spermine polycation is a highly active gene vector in vitro.

8.Preparation and characterization of cucurbitacin B sodium deoxycholate/phospholipid-mixed oral fast dissolving film and antitumor activity study.

A novel drug delivery system combining oral fast dissolving film with sodium deoxycholate/phospholipid mixed micelles was prepared to increase the absorption of cucurbitacin B that is a poor aqueous solubility substance. Encapsulation efficiency, particle size, zeta potential, polydispersity coefficient, investigated the morphology, disintegration time of oral fast dissolving film and the pharmacodynamic properties of cucurbitacin B sodium deoxycholate/phospholipid-mixed micelles before and after solidified in mice were evaluated and compared. The oral fast dissolving film prepared in this study showed a homogeneous pale yellow and could completely disintegrated in the 30 s. It could meet the requirements of rapidly disintegrating fully. The encapsulation efficiency, particle size, zeta potential, polydispersity coefficient of cucurbitacin B sodium deoxycholate/phospholipid-mixed micelles loaded in oral fast dissolving film were (43.36 +/- 2.12)%, (108.82 +/- 5.2) nm, (-34.18 +/- 1.07) mV, 0.088 +/- 0.012, respectively. The encapsulation efficiency, particle size, zeta potential, polydispersity coefficient of cucurbitacin B sodium deoxycholate/phospholipid-mixed micelles in solution were (41.26 +/- 2.22)%, (181.82 +/- 4.48) nm, (-30.67 +/- 0.81) mV, 0.092 +/- 0.012, respectively. The difference of pharmacodynamics among film of cucurbitacin B-loaded micelles, cucurbitacin B-loaded micelles and free cucurbitacin B in vivo was compared. Solubility of cucurbitacin B loaded in sodium deoxycholate/phospholipid-mixed micelles has also been greatly improved. The tumor inhibition rate of cucurbitacin B loaded in sodium deoxycholate/phospholipid-mixed micelles was significantly improved and did not change significantly before and after solidified. These showed that the sodium deoxycholate/phospholipid-mixed micelles could enhance the antitumor activities of cucurbitacin B and the stability of cucurbitacin B sodium deoxycholate/phospholipid-mixed micelles was improved significantly after solidified by oral fast dissolving film technology without pharmacodynamic properties changed significantly.
