1.Effect of CO_2 Pneumoperitoneum during Laparoscopy on Migration of GFP-labeled Living Cells into the Liver through Blood Route

Objective To study the effect of CO2 pneumoperitoneum on migration of GFP-labeled living cells into the liver through blood route in rat model.Methods SD rats was inoculated intraportally with high-dose(5?106) GFP-labeled liver cells from C57BL/6 mice after cutting belly open.Pneumoperitoneum was established immediately after closing the abdominal wall.The rats were randomly divided into four groups(n=10 in each) to receive CO2 pneumoperitoneum at 5,10,or 15 mmHg,or no treatment other than cells inoculation(control).The pneumoperitoneum was maintained for 30 min.Afterwards,the rats were euthanized by cervical dislocation,and the liver of the rats was removed for fast frozen section biopsy.The expression of GFP-labeled living cells in rat livers was compared between the groups.Results No significant difference was detected in the positive expression of GFP-labeled cells between the groups(8 rats in the control group,9 in the 5 mm Hg group,9 in the 10 mm Hg group,and 10 in the 15 mm Hg group,?2=2.222,P=0.528).The mean number of GFP-positive cells in the four groups was 6.63?2.45(control),7.67?2.83(5 mm Hg),13.89?4.37(10 mm Hg),and 15.50?6.29(15 mm Hg).There was significant differences between the four groups(F=10.78,P=0.000).In addition,the numbers of GFP-positive cells in the high pressure groups(15 mm Hg and 10 mm Hg) were significantly higher than that in the low pressure group(5 mm Hg) and the control(P
2.Dynamic contrast-enhanced MRI of vertebral metastatic tumors' early diagnosis

Objective To discuss the diagnostic value of dynamic contrast-enhanced MRI in differentiating vertebral metastatic tumors from benign deseases after studying the early changes of signal intensity.Methods Twenty patients were scanned by dynamic contrast-enhanced MRI.The parameters of signal intensity were measured and calculated,and time-intensity cures(TICs)were drawn.Then,they were compared with those of normal verte- braes,the characteristics were analyzed.Results The parameters of vertebral metastatic tumors:peak signal intensi- ty,signal intensity minimum signal intensity of post-peak,peak slope,initial ratio of enhancement,maximum ratio of enhancement was respectively(50.61?11.38),(46.75?10.23),(0.73?0.12),(0.33?0.08),(1.03?0.31); that of normal vertebraes was respectively(40.53?12.12),(34.72?14.06),(0.31?0.11),(-0.25?0.05), (0.69?0.28);and ten normal cases showed negative in initial percent of enhancement,but only two abnormal ver- tebraes showed that.All of the above had significant statistical difference.Four types of TICs were concluded:early rapid rise and early rapid descent followed by steady phase(type A),persistently rise(type B),rise phase followed by steady phase(type C),rise phase followed by rapid descent phase(type D).TICs of abnormal vertebraes were respec- tively 6,5,5,5 cases;types of normal vertebraes were mainly type C,15 cases.Type A and type B and the type with the characteristic of early rapid descent phase were features of abnormal vertebraes;type C was the feature of normal ones.Conclusions Dynamic contrast-enhanced MRI could quantitatively show the characteristics of early changes of signal intensity of vertebral metastatic tumors,which were significant statistically different from those of normal ver- tebraes.Dynamic contrast-enhanced MRI was worth performing in the early diagnosis of vertebral metastatic tu- mors.
3.Study on the vaginal bacteria test by PCR and bacterial culture

Objective To study the effect of PCR and bacterial culture in vaginal bacteria test.Methods A total of 84 patients were enrolled in the department of obstetrics and gynecology, affiliated hospital of Nankai university, from February 2014 to March 2016.Vaginal secretion was collected and tested by PCR(group A, n=42), and bacterial culture(group B, n=42).Vaginal bacteria test Resultswere compared in the two groups.ResultsEnterococcus were positive in 13 cases(30.95%)in group A, 3 cases(7.14%)in group B, gardnerella vaginalis were positive in 10 cases(23.81%)in group A, 19 cases(45.24%)in group B;Corynebacterium were positive in 13 cases(30.95%)in group A, 5 cases(11.91%)in group B.Negative rate of vaginal bacterial test was 14.29% in group A, 35.71% in group B.In the above data, the differences between the 2 groups were statistically significant(P<0.05).Conclusion PCR test can effectively improve the positive rate of vaginal bacterial examination, and provide an objective basis for clinical treatment.
4.The study of differentiation potential into enteric neuron from rat bone marrow stromul cells in vitro

Objective To explore the differentiation potential of bone marrow stromal cells (BMSC) to enteric neuron in vitro and to seek proper induction methods.Methods BMSC were harves- ted from male rats and cultured in DMEM supplemented with 20% fetal bovine serum,and characterized by flow cytometry.At passage 6,BMSC were pre-induced by basic fibroblast growth factor (bFGF,10 ng/ml) for 24 h,then induced in two groups:glial cell-line derived neurotrophic factor (GDNF) group, 10 ng/ml GDNF in fetal gut condition medium (FGCM) for 10 d.Vitamin A acid (VA) group,VA, zinc in FGCM for 10 d.The expressions of neuronal markers,neural specific enolase (NSE) and neu- rofilament (NF),glial cell marker,glial fibrillary acidic protein (GFAP),enteric neuronal marker,pro- tein gene production 9.5(PGP9.5),nitric oxide synthase (nNOS),enteric neural transmitter,vasoactive intestinal peptide (VIP) were detected by fluorescent immunohistochemistry method.Results The cul- tured BMSC were CD90 positive and CD45 negative on flow cytometry.After 10 d of induction,a certain number of cells adopted neuron-like morphological changes and showed the expressions of NSE,NF, PGPg.5,nNOS and VIP without the expression of GFAP by fluorescent immunohistoehemistry method in both groups.But in GDNF group,the positive rate of NF,PGP9.5,nNOS and VIP was significantly higher than that in VA group (75.6%?8.4% vs 48.5?7.5%;57.7%?6.5% vs 35.7%?7.2% 46.6%?5.4% vs 30.5%?6.6%;72.3%?6.7% vs 40.4%?7.4%;P<0.01).Conclusion BMSC can be induced to differentate into enteric neuron in vitro by different methods.GDNF with FGCM can induce higher rate of enteric neuron like cells compared with VA etc.

Objective To investigate the status and diagnostic value of aberrant phosphatase and tensin homology deleted on chromosometen(PTEN) gene promoter in tissues, peripheral plasma and BALF of lung cancer patients. Methods We analyzed the methylation of PTEN gene in tissues, peripheral plasma and BALF by methylation specific-PCR. Objectives To screen the differentially expressed proteins in serum of population chronically exposed to arsenic in drinking water,thus to provide candidate protein biomarkers for arsenic exposure and arsenicosis.Methods Subjects were selected from the drinking water type of endemic arsenicosis areas in Shanxi province,China.Demographic characteristics,history of arsenic exposure,cigarette smoking,alcohol drinking,health and other information were collected using questionnaire.The subjects were divided into low-arsenic group (with arsenic in drinking water < 10 μg/L),medium-arsenic group( 10 - 50 μg/L),high-arsenic group( > 50 μg/L),and arsenicosis group(the drinking water with arsenic > 50 μg/L was replaced by low arsenic water < 10 μg/L).The number of cases in each group was 30.The arsenicosis patients were diagnosed according to “Standard of Diagnosis for Endemic Arsenism” (WS/T 211-2001 ).With the principle of informed consent,blood samples were collected.Differentially expressed serum proteins of different arsenic exposure groups and arsenicosis group were screened by two-dimensional differential gel electrophoresis(2-D DIGE),and further identified by mass spectrometry (MS).Results An average of (1299 ± 167) protein spots were identified in 6 gel images and 688 protein spots were discovered repeatedly in at least 5 gels.There were 33 protein spots differentially expressed among low-,medium- and high-arsenic groups P < 0.01).Fifty four protein spots were significantly different among low-,medium-,high-arsenic exposure groups and arsenicosis group(P < 0.01 ).Twenty five protein spots were selected for MS analysis,and13 protein spots were identified.Compared with low-arsenic group,the expressions of apolipoprotein A-Ⅳ,retinol binding protein,and estrogen receptor hypothalamic isoform in medium- and higharsenic exposure groups were down regulated,and the expressions of component 4A and 4B were up regulated.Compared with low-,medium- and high-arsenic groups,the expressions of beta-2-glycoprotein Ⅰ,Keratin 1,hemopexin,complement C1r subcomponent,and ficolin-3 in arsenicosis group were down regulated,and the expressions of pigment epithelial-differentiating factor,alpha-1-microglobulin and carboxypeptidase N catalytic chain were up regulated.Conclusions Chronic arsenic exposure can significantly change population's serum protein expression.Differentially expressed proteins in arsenicosis patients will not decline with the decline of arsenic in a short term.Whether or not the differentially expressed proteins identified in this study can be used as biomarkers for arsenic exposure and arsenicosis needs to be further verified.
7.Relationship Between Blood Big Endothelin Level and Contrast-induced Acute Kidney Injury in Patients With Emergent Percutaneous Coronary Intervention

To explore the relationship between blood level of big endothelin and contrast-induced acute kidney injury (CI-AKI) in patients with emergent percutaneous coronary intervention (PCI). Methods: A total of 1061 consecutive patients received emergent PCI in our hospital from 2013-01 to 2015-06 were enrolled. According to blood levels of big endothelin, the patients were divided into 2 groups: Normal big endothelin group, n=236 and Elevated big endothelin group, n=825. The baseline condition, procedural features, occurrence rate of CI-AKI and composite endpoint events at 6 and 12 months post-operation were studied which including nonfatal myocardial infarction, revascularization, stroke and all-cause death. The risk factors for CI-AKI occurrence were identified by Logistic analysis. Results: The overall occurrence rate of CI-AKI was 22.7% (241/1061). Compared with Normal big endothelin group, Elevated big endothelin group had the higher incidence of CI-AKI, increased composite endpoint events at 6 and 12 months post-operation with P=0.041, P=0.040 and P=0.021, respectively. With adjusted covariates, elevated blood level of big endothelin, no matter as a continuous variable or categorical variable had the enhanced risk of CI-AKI incidence in patients after emergent PCI. Conclusion: Elevated blood level of big endothelin may significantly increase the risk of CI-AKI in patients with emergent PCI.
8.Experimental study on cytochrome P450 enzymes after receiving ferment powder caterpillar fungus.

OBJECTIVETo study the effect of ferment powder caterpillar fungus on cytochrome P450 isozymes CYP1A2, CYP3A4 and CYP2E1.
METHODThe methods of Cocktail probe drugs were used. The rats were randomly divided into two groups. One group were given ferment powder caterpillar fungus once daily orally for ten days. Another group received orally normal saline one daily as the blank control. After ten days of treatment, the rats were given probe drugs of coffine, dapsone and chlorzoxazone and the blood was taken out by femoral catheterization. The plasma concentration of probe drugs were determined by HPLC. Data of plasma drug level-time were disposed with DAS Ver 2.0.
RESULTThe metabolism of caffeine and dapsone speeded up after receiving ferment powder caterpillar fungus, but the metabolism of chlorzoxazone was hardly changed.
CONCLUSIONIt suggested that ferment powder caterpillar fungus tended to be the inducer of CYP1A2 and CYP3A4. But the CYP2E1 was hardly affected.

9.Characteristics of Wenchuan earthquake patients with spinal injuries

Objective To investigate the characteristics of spinal injury caused by earthquake as well as to improve its therapeutic ability by comparatively studying the medical records of the inpatients with spinal injuries caused by Wenchuan earthquake and the inpatients with spinal injuries in peace time.Methods Medical records of inpatients with spinal injuries were extracted from 5.12 earthquake injury database(earthquake group) and medical records of the patients with spinal injuries firstly admitted to Daping Hospital between 2001 and 2007 were extracted from Chinese trauma database systems (control group).The epidemiological characteristics of the two groups were comparatively studied,including gender,age,injury causes,injury severity,treatments and outcomes.Results (1) The incidence of spinal injuries in the earthquake group was 16.7% (524/3145),which was significantly higher than that (6.4%,682/10644) in control group (P<0.01).(2) The female ratio in the earthquake group was significantly higher than that in the control group (57.4%:31.8%) (P<0.01).(3) The age range of the earthquake group was (47.9 ±20.9) years.The wounded number presented an undulately rising trend in the earthquake group as the growth of age,while it showed a parabolic distribution in the control group.(4)The main causes for spinal injuries were crush injury (69.3%) and multi-factor injury (2.8%) in the earthquake group,while slipping up/fall (56.0%) and traffic accidents (23.6%) were the main causes in the control group.(5) There were 82.3% of the earthquake group that were combined with multiple injuries and 48.1% combined with at least two injuries,while the control group accounted for only 28.9% and 12.0% respectively.The NISS (new injury severity score) in the earthquake group was significantly higher than that in the control group [(11.6±8.8) points vs (7.5±4.8)points](P<0.01).Meanwhile,the proportion of NISS≥16 in earthquake group was significantly higher than that in the control group as well (P<0.01).(6) The length of hospital stay of the wounded in the earthquake group was significantly longer than that in control group [(40.5±24.5)d VS (23.9±26.7)d](P<0.01).The treatment inefficiency rate in the earthquake group was higher than that in the control group (9.2% vs 4.8%) (P<0.01).Conclusions The earthquake group has a higher proportion of spinal injuries,female and elder cases,and combined injuries,a much severer injury,a longer length of hospital stay,and a higher treatment inefficiency rate,as compared with the control group.In the earthquake relief,more importance should be attached to salvage,diagnosis,transfer regulation and missed diagnosis,misdiagnosis and secondary injury should be avoided so as to improve the treatment level of the spinal injuries in earthquake.
