1.The influence of early mechanical ventilation and perfluorocarbons gasification inhalation on inflammatory factor levels in patients with severe pancreatitis

Objective To investigate the clinical effect by early mechanical ventilation and perfluorocarbons gasification inhala-tion on patients with severe acute pancreatitis (SAP) complicated with acute respiratory distress syndrome (ARDS) .Methods 34 cases of SAP and ARDS patients accepted early mechanical ventilation and vaporization of perfluorocarbon inhalation therapy (ob-servation group) ,and 34 patients received mechanical ventilation for early treatment alone (control group) ,The efficacy ,oxygenation index(PaO2/FiO2 ) and inflammatory cytokines IL-1 ,6 ,8 ,and TNF-a level of two groups were compared .Results The total effi-ciency of the observation group was 94 .12% ,which was significantly higher than the control group that was 76 .47% (P<0 .05) . After treatment ,the arterial oxygen saturation (SaO2 ) ,arterial oxygen pressure (PaO2 ) ,PaO2/FiO2 of the observation group in-creased significantly ,and higher than the control group(P<0 .05) .After treatment ,IL-1 ,6 ,8 ,TNF-αof the observation group were significantly lower than the control group(P<0 .05) .Conclusion The treatment of early mechanical ventilation with perfluorocar-bon inhalation gasification could control the inflammation in patients with SAP complicated with AROS ,which could improve the cure rate .

With the rapid development of therapeutic colon-technology,we see the daylight of its clinic use.But, at the same time,there are many social,ethical problems.We think, cloning human embryo and extracting stem-cells should be protected ethically. The fusion of the cells of human and rabbits violates the principles of life such as dignity.harmony and independence.Cloning organs of animals and transplanting them to patients can not shun the social and ethical problems and those of the law. So, the first and most important thing is to formulate the norm of the law and that of the athics.
3.Investigations on nutritional status and nutritional risk in hospitalized pediatric patients

Objectives To validate the Screening Tool for the Assessment of Malnutrition in Pediatrics (STAMP) in hospitalized pediatric patients, and to assess the nutritional status and nutritional risk using STAMP. Methods the nutritional status of hospitalized pediatric patients was investigated by adopting STAMP. The STAMP score≥4 was deifned as a high standard of nutritional risk. Results Among 1 506 hospitalized children, children with high nutritional risk accounted for 26.56%. The overall prevalence of malnutrition and stunting was 19.12%and 10.16%, respectively. There was signiifcant diffe-rence in stunting among each department (P<0.05), which was the highest in department of internal pediatrics. The proportion of patients receiving nutritional support was 10.09%. Among them, the rate of parenteral and enteral nutrition support was 7.84%and 2.92%, respectively. The ratio of parenteral nutrition to enteral nutrition was 2.68:1. After intervention to hospitalized pa-tients with high nutritional risk, the overall prevalence of high nutritional risk decreased from 26.56%at admission to 21.71%at discharge (P<0.05). Conclusions STAMP can objectively relfect the possible malnutritional risk in hospitalized pediatric patients. The nutrition condition of the pediatric patients can be improved through nutritional risk management.

5.Inhibitory effects of fluvastatin on activation of THP-1 cells induced by anti-beta2GPI/beta2GPI complex.

This study is to explore the interventional effects of fluvastatin on anti-beta2GPI/beta2GPI-induced activation in THP-1 mononuclear cells. In vitro, human mononuclear cells THP-1 were treated with fluvastatin, LPS and anti-beta2GPI/beta2GPI, then the TF expression on THP-1 cells was detected by real-time quantitative PCR (RT-qPCR) or TF activity was detected by kit. TNF-alpha mRNA and its protein expression were investigated by RT-PCR and ELISA kit. The expression of phospho-NF-kappaB p65 and inhibitory protein of NF-kappaB (IkappaB-alpha) were measured by Western blotting. The results suggested that the expression of TF and TNF-alpha on THP-1 cells was significantly up-regulated with treatment of anti-beta2GPI/beta2GPI complex (100 mg x L(-1)), compared with that of untreated cells (P < 0.05). Fluvastatin (50 mg x L(-1)) could decrease TF (mRNA and activity) expression and the level of TNF-alpha (mRNA and protein) in THP-1 cells with anti-beta2GPI/beta2GPI complex. The expression of TF and TNF-alpha was shown in a concentration-dependent manner. Moreover, anti-beta2GPI/beta2GPI complex could downregulate IkappaB-alpha levels and increase the levels of phospho-NF-kappaB p65. And these effects of anti-beta2GPI/beta2GPI complex could be blocked by fluvastatin. In conclusion, fluvastatin may interfere the expression and regulation of NF-kappaB signal transduction pathway, thereby inhibit the effects of anti-beta2GPI/beta2GPI on activation of THP-1 cells, by decreasing the expression of TF and TNF-alpha.
6.Correlation between the volume of epicardial adipose tissue and SNYTAX score in patients with coronary artery atherosclerosis

Objective To explore the correlation between the epicaridal adipose tissue (EAT)volume and the SNYTAX score in patients with coronary artery diseases and to evaluate its clinical value.Methods Epicardial fat volume of 102 patients with coronary heart disease in our hospital were measured on dual-source CT angiography images,SNYTAX scores were calculated,and level of blood lipids,blood glucose (Glu),blood pressure,renal functional parameters and body mass index(BMI)were collected.Depending on SNYTAX scores,patients were divided into three groups (mild 0-22,moderate 23-32 and severe>33).The difference of EAT volume between groups and correlation with other indicators were analyzed.With indictors statistical significant in one-factor analysis,multi-ple regression equation was constructed to evaluate the risk factors of coronary artery diseases,particularly correlation between EAT volume and SNYTAX score.Results EAT,TC,TG,HDL,HbALc,GLu and BMI were significant different between three groups. Pearson regression showed that EAT,TC,GLu and BMI were independently risk factor in relation to the value of SNYTAX.Among them,standardized regressive coefficient of the EAT volume was the highest (β=0.52).Conclusion EAT volume is significantly positively correlated with the SNYTAX score in coronary heart disease,which can be as an effective predictor for its severity and prognosis.
7.Effect of lactobacillus acidophilus on disordered intestinal flora of mice introduced by antibiotics

Objective: To establish a female SPF BALB/c mouse model of antibiotic-associated intestinal microflora disturbance and analyze the effect of lactobacillus acidophilus.Methods: Female SPF BALB/c mice were treated with ceftriaxone in drinking water for five days,and followed by oral gavage of sodium chloride as natural recovery group or lactobacillus acidophilus(108CFU/mL) as therapeutic group.After seven days the intestinal microflora of the cecum content was abstracted and computed by plate counts and 16S rDNA-based PCR-DGGE method.Results: Plate counts indicated that the number of original bacterium lacticum and bacillus bifidus decreased significantly and enterococci proliferated abnormally after antibiotic treatment.After lactobacillus acidophilus treatment,the number of physiological bacteria and enterococci go back to normal levels.DGGE profile clearly indicated that the dominant bacteria changed and the bacterial diversity of intestinal microbiota was obviously decreased in the antibiotics treated group. Conclusion: Oral gavage of lactobacillus acidophilus has adjustment effect on Ceftriaxone induced intestinal microbiota disturbance.PCR-DGGE is a rapid、effective molecule method to reveal the whole microbiota status.

Taxol could induce tumor cell apoptosis throug h activation of Fas/FasL and caspases. bcl-2 also played an important role in t axol-induced cell apoptosis. Furthermore, taxol had the similar action with lip opolysaccharide on inducing the release of bioactive substances including tumor necrosis factor, interleukin and nitric oxide. There were as well some progresse s in taxol-resistance of tumor cells.
9.Melatonin Ameliorates Liver Fibrosis Induced by Carbon Tetrachloride in Rats via Inhibiting TGF-β1/Smad Signaling Pathway

Melatonin has been reported to inhibit hepatic fibrosis and the mechanism may be correlated to its anti-oxidant effect.Nevertheless,the mechanism is not completely identified.This study was conducted to investigate the effects of melatonin on TGF-β1/Smad signaling pathway in liver fibrosis in rats.The liver fibrosis model was made by the subcutaneous injection of CCl4.The liver pathology changes were detected using hematoxylin and eosin (H&E) staining and Van Gieson (VG) staining.Serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities were measured with an autoanalyzer.Glutathione peroxidase (GPx) activities and levels of malondialdehyde (MDA) and hydroxyproline (Hyp) in liver were evaluated by spectrophotometry.Expression levels of TGF-β1,Smad2/3,phosphorylated Smad2/3 (p-Smad2/3) and Smad7 in liver were detected by immunohistochemistry and Western blot analysis.Results showed that melatonin suppressed CC14-induced liver fibrosis,along with an improvement in histological changes,significant decreases in pathologic grading sores and obvious decreases in Hyp levels in liver.Melatonin improved liver function indicated by decreased serum ALT and AST activities.In addition,melatonin exerted its anti-oxidant effects,as supported by decreased MDA levels and increased GPx activities in liver.Furthermore,melatonin inhibited TGF-β1/Smad pathway,as evidenced by decreased TGF-β1,Smad2/3 and p-Smad2/3 expression and increased Smad7 expression in liver.In conclusion,melatonin may suppress CCl4-induced hepatic fibrosis in rats via inhibiting TGF-β1/Smad pathway.It is possible for melatonin to be a potential reagent to treat and cure liver fibrosis.
10.Mutagenesis Screening of Astaxanthin-producing Phaffia rhodozyma and Fermentation Condition Optimization

The original strain was treated with UV combining LiCl. The total carotenoid yield of the strain UL-61 was 7.62mg/L, the maximal yield of astaxanthin had reached 616.8?g/g under 25℃. 8 variables including fermentation temperature, initial pH were optimized by Plackett-Burman design and Response Surface Analysis of SAS software. The temperature, initial pH and corn steep liquor were the major factors. The optimal conditions were predictted to be 16.78℃, pH4.73 and 7.06 mg/L, respectively. Under these conditions, the theroretical maximal yield of carotenoid was 3.9407 mg/L and the practical maximal yield was about 3.9261mg/L. The carotenoid production was increased by 20.4% when Phaffia rhodozyma was cultivated under the optimal conditions as compared with the control.
