1.Distribution of sodium pump α-subunit isoforms in normal sprague-dawley rats

ObjectiveTo explore the distribution of sodium pump α-subunit isoforms at both mRNA and protein level in normal Sprague-Dawley(SD) rats. MethodsWith RT-PCR (reverse transcription-polymerase chain reaction) and immunohistochemical assay, sodium pump α1-, α2-, andα3-subunit isoforms were detected at mRNA level and protein level in the myocardium, liver, kidney, adrenal gland, aortic smooth muscle, pituitary and hypothalarnus in normal rats. ResultsSodium pump α1-, α2-, and α3-subunit isoforms distribute in a tissue-specific fashion in normal SD rats. α1 and α2 are the main isoforms and α3 is much less in myocardium. α1 is more than α2 or α3 isoform at mRNA level in liver, while all of α1-,α2-, and α3-isoform are less expressed at protein level. α1-isoform is abundantly but α2 and α3 are less expressed both at mRNA and protein levels in kidney. α2isoform is just more than α1 and α3 isoforms in aortic smooth muscle. All of three isoforms are expressed abundantly in pituitary. α2 and α3 are more and α1 is less expressed in hypothalamus. ConclusionThe results of this study have revealed the distribution of sodium pump α-subunit isoforms at both mRNA and protein level in normal SD rats, which might be helpful for the further studying the physiologic and pathologic roles of sodium pump.
2.Correlation between serum procalcitonin concentration and systemic in-flammatory response syndrome score in patients with bacterial bloodstream infection

Objective To evaluate the correlation between serum procalcitonin concentration and systemic inflam-matory response syndrome (SIRS)score in patients with bacterial bloodstream infection.Methods In January-De-cember,2012,96 patients with bacterial bloodstream infection in a hospital were selected as trial group,and these patients were divided into three groups(group A,B and C)according to SIRS score;84 patients without bacterial in-fection was as control group,PCT concentration of all patients were detected,and the correlation between PCT con-centration and SIRS score was analyzed.Results Among 96 patients with bacterial bloodstream infection,7 (7.29%)died (4 were in group B and 3 in group C);there was no death case in control group.PCT concentration in control group,group A,B and C of trial group were (0.28±0.09)ng/mL,(0.63±0.13)ng/mL,(3.68±1.01)ng/mL,and(7.45±1.53)ng/mL,respectively,the difference between each group was significant(P<0.01).Pairwise comparison of four groups showed statistical difference (P<0.001).Spearman correlation analysis on PCT concen-tration and SIRS score was conducted,correlation coefficient r=0.874(P<0.001)suggested positive correlation be-tween serum PCT concentration and SIRS score.Conclusion PCT concentration in patients with bacterial blood-stream infection and SIRS score is positively correlated,PCT concentration and SIRS score can be used as two mark-ers for assessing the extent and prognosis of bacterial bloodstream infection.
3.Serum antibody to Campylobacter jejuni(CJ) in Guillain-Barre Syndrome(GBS) patients and its significance

Objective:To detect serum antibody to CJ in GBS patients with LPS purified from CJ strains (CJ LPS) and total CJ constituents (CJ T) as antigens and to deduce whether LPS is surely the epitope of CJ in the induction of CJ antibodies from the difference between the positive rates of both the two assays with CJ LPS and CJ T and from the concordance rates of the positive cases Methods:Sera from 81 GBS patients, 34 patients with other neurological diseases (OND) and 63 normal controls (NC) were tested with CJ LPS and CJ T by indirect ELISA method CJ LPS was purified by phenol water extraction method from 15 CJ strains of various Penner serotypes, including those isolated from GBS patients CJ T was made by collecting the heat stable constituents of CJ Results:The positively infective rate of GBS were 51 9% by CJ LPS and 56 8% by CJ T antigens There is significant difference between GBS and NC in both assays, as well as OND There is no difference between the positively infective rate of GBS patients detected with the two sets of antigens and between concordance rates of the positive cases Conclusion:High incidence of GBS following CJ infection was confirmed once more, and LPS is probably one of the epitopes of CJ in the induction of CJ antibodies
4.Influence of mild hypothermia therapy on expression of heat shock proteins in lymphocyte in patients with severe craniacerebral injury

Objective To explore the influence of mild hypothermia therapy on expression of heat shock protein(HSP)60,HSP70,HSP90 in lymphocyte in patients with severe craniacerebral injury.Methods 23 patients with severe craniacerebral injury were randomly divided into the mild hypothermia group (12 cases) and the normothermia group(11 cases).At the base of routine therapy,the mild hypothermia group received mild hypothermia therapy by the temperature control blanket to keep the body temperature among 32~34℃ for 4~6 d.The expression of HSP60,HSP70,HSP90 in lymphocyte were measured by Western Blot at the 1st d,4th d and 8th d after treatment.The infection incidence was observed at 7 d after treatment.And the expression of HSPs in lymphocyte at 32℃ and 37℃ in normal control group were measured respectively.Results Compared with normothermia group,the expression of HSP60 in mild hypothermia group was significantly lower at each time point(all P

6.Real world study of Dengzhan xixin injection in treatment of cerebral infarction with medication.

To analysis of Dengzhan Xixin injection (DZI) in treatment of cerebral infarction (EBHM) in the real world population characteristics and concomitant medication. By selecting the 20 hospital information system (HIS) used in the database of DZI and primary diagnosis of 2 484 cases of cerebral infarction patients information, use the Apriori algorithm to construct the model, using Clementine 12.0 analysis, cerebral infarction complicating diseases, commonly used drug combination analysis of DZI. The results showed that patients with more males than females (1.63: 1); age > 46 in older persons, treatment 7-14 days accounted for the majority of patients with hypertension, cerebral infarction, diabetes, coronary heart disease and other diseases; common drug combination can be divided into seven categories: medicine of antiplatelet therapy (aspirin, clopidogrel hydrogen), hypolipidemic drugs (atorvastatin, probucol), calcium channel blockers (cinepazide), cerebral protection drugs (laci staw), to improve cerebral circulation drugs (alprostadil), other traditional Chinese medicine injection (Shuxuetong injection, Xueshuantong), treatment with underlying disease: nifedipine, metoprolol, isosorbide dinitrate etc. The clinical cure rate and improvement rate of 97.60%. The next step needs to be combined with clinical practice, carry out analysis of effectiveness and safety of the combination scheme, and provide reference for clinical rational drug use.

7.Expression of telomerase in skin lesions of condyloma acuminatum,bowenoid papulosis,Bowen's disease and vulvar squamous carcinoma

Objective To study the expression of telomerase and its possible clinical significance in patients with condyloma acuminatum,bowenoid papulosis,Bowen's disease and vulvar squamous cell car- cinoma.Methods The human telomerase reverse transcriptase (hTERT) was detected by immunohisto- chemistry technique in all patients.Histopathological analysis was done in patients with bowenoid papulosis or Bowen's disease.Results There was a significant difference in the expression of hTERT among normal tissue,condyloma acuminatum and squamous carcinoma,and the expression rate increased successively from normal control to squamous carcinoma.In bowenoid papulosis,the expression of bTERT was significantly higher as compared to that in condyloma acuminatum,significantly lower as compared to that in squamous carcinoma,and was as the same as that in Bowen's disease.Histopathologically,the degree of cell atypia was significantly higher in Bowen's disease than that in bowenoid papulosis.Conclusions The expression of telomerase increases successively from normal skin,benign tumors to malignant carcinomas,which sug- gests that the expression of telomerase may play an important role in both cell proliferation and immortaliza- tion.
8.Rabbit model of aspergillus keratitis induced by modified corneal surface lens method

Background Animal model of fungal keratitis is an available tool to the experimental study of the pathogenesis mechanism of fungal keratitis. Current modeling methods of fungal keratitis include corneal scratching, corneal stroma injection and corneal surface lens methods. But these methods still have their own shortages. Objective This experiment was to create a fungal keratitis animal model by modifying corneal surface lens method. Methods Modified animal models of fungal keratitis were created by modified corneal surface lens method in 12 general adult New Zealand white rabbits. The filter papers soaked 108 spores / ml or A106spores / ml of spergillus fumigatus suspension were attached on the de-epithelial cornea surface and fixed with contact lens and tarsorrhaphy for 2 days, and the filter paper with physiological saline was used as control group. The symptoms of anterior segment were examined under the slit lamp in 3 ,7 and 14 days after surgery and scored based on the criteria of Dong. Corneal scraping was stained with 10% potassium hydroxide and calcofluor white stain to observed mycelium under the fluorescence microscope. Corneal tissue sections were examined by hematoxylin-eosin staining and periodic acid Schiff staining under the light microscope. The use of animal followed the Standard of Association for Research in Vision and Ophthalmology. Results Fungal keratitis models were successfully established in 6 eyes and 4 eyes in 108 spores/ml group (6/6) and 106 spores/ml group respectively. The symptom was more severer and score was higher in the eyes of 108 spores/ml group than that in 106 spores/ml group. At 3 and 7 days after surgery,the symptom scores of fungal keratitis models were higher than those of control group from 3 through 7 days with the statistically significant difference (P<0. 01) and the symptom scores of 108 spores/ml group were significantly higher than those of 106 spores/ml group (P<0. 01). At 14 days after surgery, the symptom scores of 108 spores/ml group were still higher than those of control group (P<0. 05). Fungal hyphae was seen in the corneal scrapes in 108 spores/ml group and 106 spores/ml group respectively from 3 through 7 days after surgery. Inflammatory cell infiltration, stroma cells necrosis and fungal hyphae were presented in 108 spores/ml group, and the corneal neovascularization could be observed in 108 spores / ml group 14 days later. Fungal culture revealed the positive outcome in both 3 and 7 days after surgery in 108 spores/ml group,but in 106 spores/ml group,the positive result was only in the 3rd day. Conclusion Modified corneal surface lens method is more feasible and sample in the model of Aspergillus keratitis. This animal model of Aspergillus keratitis is practical for the further study of fungal keratitis.
9.Simultaneous determination of 5 kinds of alkaloids in Kechuanning tablets by SPE-UPLC under different UV-vis wavelength.

The paper is to establish a method for simultaneous determination of 5 kinds of alkaloids in ephedra and poppy which are in Kechuanning tablets. Solid-phase extraction (SPE) was adopted in pretreatment, and a UPLC method with 2 different wavelengths had been developed: 210 nm for the detection of morphine, codeine phosphate, ephedrine hydrochloride and pseudoephedrine hydrochloride, and 251 nm for papaverine hydrochloride. The column used was Acquity UPLC BEH C18 (100 mm x 2.1 mm ID, 1.7 microm) with linear gradient elution using acetonitrile and 0.1% phosphoric acid. The flow rate was 0.4 mL.min-1, and the column temperature was 30 degrees C. The linear response range was 0.375 0 - 12.50 microg.mL-1 for morphine, 0.064 32 - 2.144 microg.mL-1 for codeine phosphate, 0.030 06 - 1.002 microg.mL-1 for papaverine hydrochloride, 1.126 - 37.52 microg.mL-1 for ephedrine hydrochloride, 0.287 8 - 9.592 microg.mL-1 for pseudoephedrine hydrochloride (r = 0.999 7). The average recoveries of these compounds were 99.26%, 100.6%, 95.29%, 100.1% and 97.48%, respectively. This is a more reasonable and credible method of quality control for Kechuanning tablets.
10.Demographic characteristics of acupuncture outpatient based on clinical investigation.

OBJECTIVEThe demographic characteristics of acupuncture outpatient in China is investigated so as to provide clinical evidence for the establishment of acupuncture indication.
METHODSClinical epidemiological table was adopted to do cluster multistage random sampling in 36 hospitals of 31 provinces (municipalities and autonomous regions) in 2008. The composition ratios of all indices were analyzed with SPSS 13.0 software. And trend analysis was carried out on the patient's demographic characteristics and the age distribution of clinical indications.
RESULTSIn the year 2008, 31 858 cases of out patients from 36 hospitals of 6 administrative regions were collected. There were totally 14 411 male patients (45.2%) and 17 447 (54.8%) female. The gender ratio of male and female was 82: 100. The average age of the patients was (50.6 +/- 17.7) years old. The number of patients over 45 years old was more obvious than groups of other ages (P<0.05). Age differences can be found in those 6 regions (P<0.05). Clinical symptoms varied according to different ages of the patients. Different characteristics of age distribution of acupuncture indications were held to exist (P=0.000).
CONCLUSIONAt the present, differences of gender, age and region can be found in out patients of acupuncture clinic. And the result shows that the number of female patients is more than male, with a large proportion of patients over 45 years old. Differences of age distribution characteristics of acupuncture indications still exist, which is considered to have the relation with the dominate indications of acupuncture and aging of the population.
