OBJECTIVETo establish a simple high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS) system for determination of capsaicin in rat plasma.
METHODSRat plasma (100 µl) was extracted with mixed solvent (ethyl acetate:acetone 85:15) after addition of 50 ng/ml internal standard, and analyzed by Agilent 6460 Triple Quadrupole LC/MS system. The detection was performed with multiple reactions monitoring using electrospray ionization. The precursor/product ion transitions were monitored at m/z 306→137 (+Ion mode) for capsaicin. Verapamil was used as the internal standard at m/z455→165.
RESULTSThe analysis time was only 1.5 min in the positive mode; the calibration curve was linear in the concentration range of 1.85-370 ng/ml. The lowest limit of quantification reached 1.85 ng/ml. The extraction recoveries were 77.34%, 70.64% and 78.02% for the three quality control concentration levels (3.7, 37, and 370 ng/ml), respectively.
CONCLUSIONThe LC-MS/MS system is simple, accurate, reliable and time-saving for determination of trace amount of capsaicin in plasma.

Objective To explore the key technology of all-round and holistic nursing care in treating the patients with ectopic pregnancy combined with hemorrhagic shock. Methods Eighty-six patients of ectopic pregnancy combined with hemorrhagic shock were treated with active rescue and infusion and comprehensive nursing care. Results Of eighty-six patients,eighty-four cases were cured, and two cases were dead. Conclusions Limited fluid resuscitation has an obvious advantage in treating uncontrolled bleeding with hemorrhagic shock. And immediate surgical therapy along with shock-resistant is the key to rescue successfully.

OBJECTIVETo explore the efficacy differences in early intervention of different acupuncture and moxibustion methods on gastrocnemius fatigue in rats induced by electrical stimulation.
METHODSFifty male SD rats were randomly divided into a control group, a model group, a hand acupuncture group, an electroacupuncture group and a moxibustion group, 10 rats in each group. Electrical stimulation of the sciatic nerve was given in the control group and gastrocnemius fatigue test was induced by electrical stimulation of the sciatic nerve in the model group after anesthesia without other treatment, but just take 6 times interval stimulation in the control group. The hand acupuncture group, the electroacupuncture group and the moxibustion group were treated with the corresponding acupoints stimulation method respectively for 20 min before gastrocnemius fatigue test, and Dazhui (GV 14) and Zusanli (ST 36) were selected. Immediately after gastrocnemius fatigue test, three or four gastrocnemius tissues at the same site on the right side were quickly taken for making specimen for transmission electron microscope (TEM). The changes of skeletal muscle ultrastructure of myofibrils, mitochondria, sarcoplasmic reticulum, glycogen particles were observed under TEM.
RESULTS(1) Muscle fibers disorder, partial mitochondrial vacuolization and glycogen particles smaller were shown in the model group. (2) No abnormalities were shown in the hand acupuncture group and the moxibustion group with mitochondrial morphology and number, which better than that in the model group, and glycogen particles increased. (3) Abnormal changes in morphology were shown in the electroacupuncture group with part of the muscle fibers derangement, Z line malalignment and a few mitochondria vacuolization.
CONCLUSIONHand acupuncture, electroacupuncture and moxibustion have the different effects on ultrastructure of gastrocoemius in rats. Acupuncture and moxibustion have shown good effects on the prevention and treatment of exercise-induced skeletal muscle cell and organelle damage and delaying exercise-induced fatigue.

During the process of electroacupuncture (EA) therapy, whether there being a corrosive effect in ac- upuncture needles was observed. Acupuncture needles were inserted into a rabbit's acupoint to perform a 12-hour electrical stimulation with three types of common EA waveform; additionally two needles were put in 0.9% sodium chloride solution with 12-hour direct current. Afterwards, environmental scanning electron microscope was applied to detect the surface physical characteristics of acupuncture needles. As a result, after a 12-hour continued electri- cal stimulation with three types of common EA waveform in the rabbit, there was no corrosive effect in acupunc- ture needles; but the direct current could cause severe corrosion in acupuncture needles. It is believed that there is no corrosion effect on acupuncture needles in current EA treatment, and some accidents reported in literature may be related to quality of EA device or improper manipulation during the treatment.

Objective To observe the effect of Huoxueyizhi slice on memory impairment and expression of Bcl-2 and Bax in hippocampus of vascular dementia(VaD)rats. Methods The imitative vascular dementia rat model was developed by repeatedly occluding the common carotid artery in combination with all abdominal injection of sodium nitroprusside(2.5 mg·kg~(-1)).The memory test, activities of SOD,MDA and Bcl-2 and Bax gene expression in hippocampus of VaD rats treated with and without Huoxueyizhi slice (1.55,3.1,6.2 g·kg~(-1)) and dihydroergotoxine (5.4 mg·kg~(-1)),continuous administration of 28 days, were measured. Results Compared with model group,Huoxueyizhi slice significantly improved memory impairment,as evaluated by shortened escape latency and increased swimming time near the platform.Meanwhile, decreased the content of MDA,increase the activity of SOD,based on the observation in hippocampal CAI region through method of the immunohistochemistry,Huoxueyizhi slice decreased the expression of the proapoptotic Bax gene,on the contrary,it increased expression of the antiapoptotic gene Bcl-2.Conclusion Huoxueyizhi slice can exert antiapoptotic effect through counter-regulating Bcl-2 and Bax gene expression.

OBJECTIVETo investigate the effects of triptolide on Notch receptor and ligand expressions in rats with adjuvant-induced arthritis (AA).
METHODSForty rats were randomly divided into normal control (NC) group, model (MC) group, methotrexate group and triptolide groups. Rat models of AA were established by an intradermal injection of 0.1 mL Freund's complete adjuvant into the right paw. Twelve days after the injection, the rats were treated with corresponding drugs for 30 days; the rats in NC group and MC group were given saline only. Paw edema volume (E), arthritis index (AI), pulmonary function, histomorphologies, and Notch receptor/ ligand expression in the lung tissue were analyzed after the treatments.
RESULTSCompared with the NC group, E, AI, Notch3, Notch4, and Delta1 expressions in the lung tissues significantly increased while pulmonary function and pulmonary expressions of Notch1, Jagged1, and Jagged2 significantly decreased the model rats (P<0.01). Compared with the MC group, triptolide-treated rats showed significantly improved pulmonary functions, increased expressions of Notch1, Jagged1, and Jagged2 and decreased expressions of Notch3, Notch4, and Delta1 in the lungs (P<0.05, P<0.01); the therapeutic effect of triptolide was better than that of methotrexate.
CONCLUSIONTriptolide can reduce inflammatory reaction and immune complex deposition to improve joint and pulmonary symptoms in rats with AA possibly by up-regulating the expressions of Notch3, Notch4, and Delta1 and down-regulating the expressions of Jagged1, Jagged2, and Notch1.
