1.Research Progress of a Novel Pro-apoptosis Gene PNAS-4 in Gene Therapy and Its Molecular Mechanism Hypotheses.

PNAS-4 is a novel pro-apoptosis gene identified latetly. In recent years, there has been a large number of research reports on the basic studies about PNAS-4 in cancer gene therapy and gene therapy of PNAS-4 alone or combined with chemotherapy or radiotherapy manifested a good application prospect, but its molecular mechanisms to promote apoptosis is not clear yet. In this paper, recent research about PNAS-4 in cancer gene therapy is briefly reviewed, and recent hypotheses on its molecular mechanisms to promote apoptosis are especially elucidated. Based on its newly identified characteristics of structural domain, we made a point that PNAS-4 might regulate functions of some target protein related to apoptosis by deSumoylation as a new deSumoylating isopeptidase, and consequently promote apoptosis.

2.Expression level and significance of TGF-β1 and CTGF in cerebrospinal fluid of patients with subarachnoid hemorrhage

Objective To explore the expression and significance of TGF-β1 and CTGF in the occurrence and development of patients with subarachnoid hemorrhage. Methods Cerebrospinal fluid TGF-β1 and CTGF concentrations in all subjects were serially detected by ELISA. Results Mean total TGF-β1 and CTGF in patients were elevated more than that in controls at the first day(P<0.05).Thereafter the levels of TGF-β1 and CTGF fell by the third day, then rose to the second peak at the tenth day and remained significantly increase until the sixteenth day(P<0.05).The levels of TGF-β1 was positive correlated CTGF. Conclusion The expressions of TGF-β1 and CTGF in patients with subarachnoid hemorrhage showed two peaks character. After SAH, the concentration of TGF-β1 and CTGF concentration is related to chronic communicating hydrocephalus.
3.Roles of Percutaneous Coronary Intervention in Improving Left Ventricular Function, Reducing Incidence of Cardiovascular Events of Patients With Ischemic Cardiomyopathy

Objective To investigate the roles of percutaneous coronary intervention in improving left ventricular function, reducing incidence of cardiovascular events of patients with ischemic cardiomyopathy. Methods Ninety cases of ischemic cardiomyopathy patients from August 2012 to August 2015 were randomly selected, these patients were divided into two groups, the study group (n=45) and control group (n=45) according to the random number table. The control group of patients were given routine treatment, while the study group of patients were given conventional treatment combined with percutaneous coronary intervention. Results The LVEDV, LVEF of the study group were significantly higher (P<0.05), the incidence of acute myocardial infarction onset, hospitalization rates again, mortality rate 8.89% (4/45), 13.33% (6/45), 2.22% (1/45) were significantly lower than the control group 24.44% (11/45), 40.00% (18/45), 17.78% (8/45) (P<0.05) . Conclusion Percutaneous coronary intervention can improve left ventricular function and reduce the incidence of cardiovascular events of patients with ischemic cardiomyopathy.
4.Content Determination of Gallic Acid, Corilagin and Ellagic Acid in Aqueous Extract from Sanguo Decoction by HPLC

Objective To establish an HPLC method for the simultaneous determination of contents of gallic acid, corilagin and ellagic acid in aqueous extract from Sanguo Decoction. Methods The analysis was carried out on an Agilent C18 column (4.6 mm × 250 mm, 5 μm), and the mobile phase was composed of acetonitrile and 0.1%phosphoric acid aqueous with gradient elution. The detection wavelength was 260 nm. The flow rate was 1.0 mL/min at column temperature of 30 ℃. Results Gallic acid, corilagin and ellagic acid were completely separated and the peak shape was good. It showed a good linearity in the concentration range of 0.174–2.61 μg, 0.04–0.60 μg, and 0.052–0.78 μg, respectively. The average recoveries of gallic acid, corilagin and ellagic acid were 99.98%, 99.81%and 100.12%, respectively. The RSD were less than 2.0%. Conclusion This method is rapid, accurate and repeatable, which can provide references for the quality control of Sanguo Decoction.
5.Human umbilical cord blood-derived CD133 cells for treatment of gestational diabetes

BACKGROUND:Human umbilical cord blood contains a large number of hemopoietic stem cels and mesenchymal stem cels. Neonate umbilical cord blood-derived stem cels show milder immunological rejection and lower immunogenicity than peripheral blood- and bone marrow-derived mesenchymal stem cels. OBJECTIVE:To investigate the feasibility of umbilical cord blood-derived CD133 cels for treatment of gestational diabetes, so as to provide novel methods for clinical treatment of gestational diabetes. METHODS:Mouse models of gestational diabetes were established by high-fat and high-sugar diet feeding. Umbilical cord blood-derived CD133 cels were sorted by flow cytometry sorting technique and then transplanted into mouse models of gestational diabetesvia the tail vein. At 7 days after cel transplantation, serum levels of fasting blood glucose, fasting insulin, total cholesterol and triacylglycerol were measured and mouse pancreatic tissue injury and repair was observed by hematoxylin-esoin staining. Mouse insulin resistance index and pancreatic islet function were analyzed using the homeostatic model assessment. RESULTS AND CONCLUSION:Umbilical cord blood-derived CD133 cels could be isolated from umbilical cord blood monocytes using flow cytometry sorting technique, with cel purity of (90.24±2.56)%. At 7 days after cel transplantation, serum levels of fasting blood glucose, fasting insulin, total cholesterol and triacylglycerol were significantly decreased, insulin resistance index was significantly decreased, and pancreatic islet function was significantly improved in mouse models of gestational diabetes (alP < 0.05). In addition, atrophy of pancreatic tissue and infiltration of inflammatory cels were reduced. These results show that umbilical cord blood-derived CD133 cel transplantation can improve the disease condition of gestational diabetes by repairing injured pancreatic tissue, decreasing insulin resistance index and improving pancreatic islet function.

To study the alkaloid constituents in the seed of Sophora alopecuroides L. and isolation of lehmannine by transhydrogenation of sophocarbine to matrine present in the mixture. Methods Various processes of exchange, ion exchange and column chromatograph with very similar physical and chemical properties was successfully separated by transhydrogenation. Results 5 alkaloids were obtained and identified by elementary analysis, IR, MS, 1HNMR and 13CNMR as oxymatrine, oxysophoridine, matrine, sophoridine and lehmannine (12, 13-dehydromatrine). Conclusion Lehmannine was isolated from the seed of S. alopecuroides for the first time.

9.The influence of puerarin on the expression of TNF-α in rats with focal cerebral ischemia and reperfusion

Objective To study the nerve protective mechanism of puerarin on ischemic brain injured and observe the expression change of TNF-α cytokines in ischemic brain area. Methods 90 healthy male SD(Spraque-Dawley)rats were used to built the animal model of brain medium sized artery focal cerebral ischemia reperfusion with the patching methods, and the rats were randomly divided into three groups: Sham operation group(n=30),operation group(n=30),operation with puerarin interfered group(n=30).There were five different observing time in every group(6h,12h,24h,48h,72h after operation).The operation with puerarin group rats were treated with puerarin solution, the sham operation and operation group rats were injected the same volume Sodium Chloride. To observe the expression of TNF-α cytokines in different time, immunohistochemistry staining in ischemic region was used. Results Compared with the sham operation group. the expression of TNF-α cytokine in cerebral isehemic reperfusion brain tissue in the operation and the operation with puerarin interfered groups was higher(P<0.01).Compared with the operation group, the expression of TNF-αcytokine was significantly decreased in different time after cerebral ischemic-reperfusion in the operation with puerarin interfered group(P<0.01 or P<0.05).Conclusions The puerarin can restrain the expression of the TNF-α cytokine.
