1.The Influence of Lonone and Ergosterol on the Growth and the Yield of CoQ_(10) in Yeast

The influence of ?-ionone and ergosterol on the growth and the yield of CoQ_ 10 in yeast was investigated. The results indicated that ?-ionone had the stimulative effect on the biosynthesis of CoQ_ 10 in yeast, and 28.3% increase of the CoQ_ 10 content in the cell was observed when the concentration of ?-ionone in the medium was 0.208?10~ -3 mol/L. Little ergosterol also had the same effect with ?-ionone, while 31.3% increase of the CoQ_ 10 content in the cell was observed when the concentration of ergosterol in the medium was 0.15?10~ -4 mol/L. However, the biosynthesis of CoQ_ 10 was inhibited when the concentration of ergosterol was 0.60?10~ -4 mol/L. Addition of ?-ionone and ergosterol to the medium resulted in 36.1% increase over the control. The results showed that the cell could effectively accumulate more CoQ_ 10 with the addition of ?-ionone and ergosterol to the medium.
2.Effects of total saponins of Panax japonicus on human leukemic HL-60 cells

OBJECTIVE: To study the effects of total saponins of Panax japonicus (TSPJ) on human leukemic HL-60 cells. METHODS: Human leukemic HL-60 cells were cultured in vitro. The cancer cell vigor was detected by using cell counting kit-8. Nitroblue tetrazolium (NBT) was used for measuring cell reduction. The cell cycle and the expression of differentiation antigen CD11b were detected by flow cytometry. RESULTS: Compared with the negative control group, TSPJ in different concentrations could decrease the vigor of HL-60 cells and the number of cells in S phase and up-regulate the CD11b expression, while the numbers of NBT positive cells and cells in G(0)/G(1) phase in the different concentrations of TSPJ-treated groups were increased. CONCLUSION: TSPJ can inhibit the HL-60 cell growth in vitro. Its mechanism may be related to inhibiting proliferation and inducing cell differentiation and cycle arrest.

Objective To evaluate the molluscicidal effect of black plastic mulching in mountainous areas of Yunnan Prov-ince. Methods In Leqiu Village,Nanjian County,Yunnan Province,the terraced fields with Oncomelania hupensis snails were divided into A,B,C,D,E groups,and of which,A,B,C,D groups,as the experimental groups,were administered with sim-ple black plastic mulching for 30 days,60 days,90 days and 120 days respectively;Group E,as the control group,was adminis-tered with 50%niclosamide ethanolamine salt wettable powder spraying. Results In Group C and D(simple black film mulching for 90 days and 120 days),no snails were found after the administration,the average density of living snails decreased by 100%, and the cost of one year was similar to that of the control group. Conclusion The molluscicidal effect of black plastic mulching is good in mountainous areas.
5.Changes in expression of phospholipase C-gamma1(tyr783) in young rat condylar cartilage during functional mandibular protraction.

OBJECTIVETo investigate the changes in the expression of phospholipase C-gamma1tyr783 (PLC-γ1tyr783) in the condylar cartilage of a young rat during functional mandibular protraction. This work also explores the function of PLC-γ1tyr783 in the rat mandibular condylar cartilage bone remodeling, which could provide experimental evidence for clinical bone ortho- pedic work.
METHODSA total of 60 four-week-old male Sprague-Dawley (SD) rats were used in this study. The rats were divided equally and randomly into experimental group and control group. The functional appliances that were fitted to the upper incisors of the animals in the experimental group were worn 24 h a day after the rats were fed for 7 d with homemade pellet feed. The animals in the experimental group, along with their matched controls, were sacrificed after 1, 3, 7, 14, 21, and 28 d. The bilateral condylar was fixed, decalcified, dehyded, and then conventional paraffin embedded. Immunohisto- chemistry of PLC-γ1tyr783 was applied to observe its express distribution and variation.
RESULTSThe expression of PLC-γ1tyr783 decreased gradually in the control group, which showed age-related changes (P > 0.05). On the 14th day, PLC-γ1tyr783 expres- sion in the experimental group was significantly higher than that in the control group. PLC-γ1tyr783 expression began to appear statistically and significantly different between the two groups (P < 0.01).
CONCLUSIONPLC-γ1tyr783 is involved in the bone remodeling process of the rat condylar cartilage after functional mandibular-protraction.

6.Expression of clock genes Bmall and Bmal2 in total leukocytes in health and Parkinson's disease

Objective To study the expression of clock genes in Parkinson's disease(PD) and also the molecular clock machinery of PD.Methods Seventeen PD patients(nine men,eight women)and sixteen agematched controls(nine men,seven women) were investigated in this study.Bload samples were collected over a 12h span at 21:00,00:00,06:00 and 09:00.Using a real-time PCR assay,the peripheral molecular clock was examined by measuring Bmall and Bmal2 expression in total leukocytes during the dark span(from 21:00 to 09:00)in PD patients and age-matched healthy controls.Results At PD,the relative abundance of Bmall was significantly lower at 21:00,00:00 and 06:00(21:00:(22.17±4.09)vs(51.14±8.31),P=0.003,00:00:(30.30±5.45)vs(100.00±24.71),P=0.008,06:00:(19.02±3.33)vs(65.61±14.11),P=0.002).The relative Bmal2 levels in PD patients were significantly less abundant than controls at 21:00 and 00:00(21:00:(48.09±7.40)vs(84.96±9.34),P=0.005;00:00:((65.85±7.88)vs(100.00±11.78),P=0.025).Conclusion These results suggest that a peripheral molecular clock is altered in PD patients.In addition,the relative abundance of Bmall and Bmal2 was significantly lower in PD patients versus control subjects,which can provide a molecular basis to help monitor disease progression and response to investigational drugs.
7.THE PERMEABILITY OF BLOOD-BRAIN BARRIER AND PLACENTA TO SELENIUM IN RAT

15 rat dams were divided into 2 groups: selenium treated group drank water containing 2 ppm selenium during pregnancy and breast-feeding period, control group drank tap water. The concentration of selenium in brain tissue for dams were 0.73 ?0.11 (Se-treated) and 0.54 ?0.09 ug/g wet tissue (control), for neonates 0.32?0.02 (Se-treated) and 0.25 ? 0.03 (control), and for 20 days old pups 0.53?0.04 (Se-treated) and 0.44?0.06 (control). The results showed that the selenium administered orally could be transfered through placenta or blood-brain barrier of fetus, young pup and adult rat.
8.Effect of interaction between CD4+ T cells and HUVECs infected with DWNV-2 on expression of inflammatory cytokines

Objective:To study the interaction of the inflammatory cytokines expression between CD4+ T cells and primary human umbilical vein endothelial cells (HUVECs) infected by dengue virus (DENV-2).Methods:PBMC was extracted from white blood cells by density gradient centrifugation,CD4+ T cells were sorted by immunomagnetic beads.The expression of CD3 and CD4 molecules on the surface of cells was detected by flow cytometry to identify the purity of CD4+ T cells.First,HUVECs were pretreated by specific-S1 P1 receptor agonist CYM-5442 for 24 h,second,infected by DENV-2 on the titer of 103 TCID50,then co-culturing with CD4+ T cells,The relative expression of NS1 partial sequence and IL-6,L-8 mRNA of HUVECs,and IL-4,IL-17,TNF-α,IFN-γ of CD4+ T cells detected by Real-time RT-PCR.IL-6 and IL-8 secreted in cultured supematant analyzed by ELISA.Results:The purity of CD4+ T cells was (98.02±0.32) %.The expression of NS1 gradually increased to 24 h (3.03±0.26,P<0.001),decreased after reaching the peak.The relative expression of NS1 in the group of co-cultured with CD4+ T cells was lower than other groups.After infection,the expression of IL-6 and IL-8 were up-regulated,and the expression of IL-6 and IL-8 at each time point was significantly increased after co-culturing with CD4+ T (P<0.01).IL-6 of CYM-5442 pretreatment group,in 24 h (28.91±2.34,P<0.05),36 h(19.36±0.1,P< 0.05) and 72 h(13.84±0.82,P<0.05) was significantly decreased,the expression of IL-8 also decreased significantly.The mRNA expression of IL-4,IL-17,TNF-α and IFN-γ in CD4+ T cells was significantly increased after co-culturing with HUVECs.After the treatment with CYM-5442 group,the expression was decreased.Conclusion:DENV-2 could infect the primary HUVECs,and the expression of NS1 was inhibited after co-culturing with CD4+ T cells.CD4+ T cells can not only enhance the activation of HUVECs infected by DENV-2,but also can be activated by the infected HUVECs infected with DENV-2.
9.Soft tissue profile of normal occlusion in the early permanent dentition:E line analysis

AIM:To explore the distance from the labrale superius and inferius of the soft tissue profile in early permanent dentition with normal occlusion to the esthetic line (E line) and the proportion among each section of E plane. METHODS: Thirty Han teenagers with normal occlusion including 15 males and 15 females with the average age of 11.5 years were selected from January 2000 to December 2005. They were examined cephalometrially to measure the distance between labrale superius (Ls) and E, labrale inferius (Li) and E, and the mean values and standard deviation of prenasale (Prn)-Ls/distance between Prn and pogonion (Pg') of soft tissue, Ls-Li/distance between Prn and Pg' and Li-Pg'/distance between Prn and Pg'. RESULTS: ①Distance of Ls-E and Li-E: Ls-E and Li-E of normal occlusion were (0.083 3?0.920 8) mm and (0.621 7?1.124 6) mm. ②Proportion of each segment to the distance of Prn and Pg': Prn-Ls/Prn-Pg', Ls-Li/Prn-Pg' and Li-Pg'/Prn-Pg' were 0.413 8?0.022 3, 0.200 9?0.023 1, 0.385 7?0.022 8, respectively. CONCLUSION: E plane analysis is a convenient and effective method in the diagnosis of soft tissue in clinic. The Ls and Li are exactly on the E plane in normal occlusion and the ratio of three sections is 2∶1∶2.

Objective:To explore the pharmaceutical care model for the patients with acute lung injury/acute respiratory distress syndrome ( ALI/ARDS) caused by inhalation of chemical agents to ensure the safety, rationality and effectiveness of drugs. Methods:According to the characteristics of drug therapy for the patients with ALI/ARDS caused by inhalation of chemical agents, the pharma-ceutical care was carried out and the suggestions on the medication were given. Results:Through the pharmaceutical care, the safety, reasonability and effectiveness were improved. And the incidence of adverse drug reactions was decreased. Conclusion: Cooperating with clinical physician, clinical pharmacists can perform active pharmaceutical service and optimize dosage regimen, which is beneficial to the safety, reasonability and effectiveness of drug therapy for the patients.
