1.Genetic Transformation of Phytoene Synthase Gene into Ginseng Callus Cells

Phytoene synthase is a rate-limiting enzyme in the pathway of carotenoid biosynthesis.To aim at establishing a transformation of Ginseng callus cells, elevating the nutritive value through encouraging the composition of corresponding carotenoid,taking Ginseng callus cells as acceptor,psy gene were transformed into cells via Agrobacterium-mediated. The factors affecting genetict ransformation were also investigated seperately from concentrations of A.tumefaciens, age of Ginseng callus cells, infection time and co-culture time. PCR,PCR-Southern and RT-PCR analysis from the transferred gene plants proved that psy gene has been transferred into Ginseng callus cells and can be expressed. The content of ?-carotene was increased by an average of 26 times.A base of improving and raising the contents of carotenoid in the Ginseng callus cell was established.
2.Placental Grb10 expression analysis for evaluation of security for blastocyst vitrification

Objective To analyze the expression of the placenta Grb10 from women conceived by transferred thawed blastocyst,and to evaluate the security of blastocysts vitrification.Methods A cross-sectional study was performed in the Department of Obstetrics and Gynecology of Fujian Provincial Maternity and Children's Hospital from January 2012 to May 2014,50 women conceived by transferring thawing blastocyst and 50 natural pregnancy control women were enrolled in this study.The expression of Grb10 protein was detected by immunohistochemistry and Western blot,and the expression of Grb10 mRNA was detected by Realtime PCR method.Results Comparison of two cases of gestational age,gestational age,fetal sex,fetal body weight,body length,head circumference,abdominal circumference,there were no significant differences(P>0.05),comparison of placental area,placental weight,the difference was statistically significant(P<0.05).Real-time PCR and Western blot results showed that,there was no significant difference in the expression of Grb10 mRNA and protein between the two groups(P>0.05).Conclusion Blastocysts vitrification may increase the area and quality of delivery of placenta,however,there was no significant change in the expression of Grb10 in placenta.
3.18F-FDG PET/CT for assessing therapeutic response to chemotherapy in patients with diffuse large B-cell lymphoma

Objective To explore the value of 18F-FDG PET/CT on the assessment of chemotherapy response in patients with diffuse large B-cell lymphoma (DLBCL). Methods 18F-FDG PET/CT was performed before and after 4 cycles of chemotherapy( R-CHOP or CHOP protocol) in 53 patients with DLBCL. The patients were divided into 3 groups: complete response group, partial response group and no response group. The therapeutic response was assessed by comparing post-treatment 18F-FDG PET/CT with pre-treatment PET/CT. Complete remission (CR) rate at the end of chemotherapy was calculated. χ2 test was performed with software SPSS 13.0. Results CR rates of complete response group, partially response group and no response group were 88.5% (23/26), 73.3% (11/15) and 8.3% (1/12), respectively (χ2=23.548, P=0.000). CR rates of the complete and partially response groups were significantly higher than those of no response group (χ2=22.656, P=0.000; χ2=11.407, P=0.001). Conclusion 18F-FDG PET/CT may be useful for the assessment of chemotherapy response in DLBCL.
4.Studies on chemical constituents in herb of Centella asiatica.

OBJECTIVETo study the chemical constituents from Centella asiatica.
METHODChemical constituents were isolated by repeated column chromatography (Toyopearl HW-40C and HPLC) and their structures were elucidated on the basis of spectroscopic method.
RESULTFive compounds were identified as: docosyl ferulates (1), bayogenin (2), 3beta-6beta-23-trihydroxyolean-12-en-28-oic acid (3), 3beta-6beta-23-trihydroxyurs-12-en-28-oic acid (4), D-gulonic acid (5).
CONCLUSIONAll of the Compounds were isolated for the first time from C. asiatica.

5.Determination of aisaticoside in Centella asiatica extract by HPLC-ELSD.

OBJECTIVEA HPLC-ELSD method was established for the simultaneous quantitative determination of asiaticoside in Centella asiatica extract.
METHODThe column was packed with 5 m HIQ C18 stationary phase. The mobile phase consisted of acetonitrile-water, eluted in gradient mode. The temperature of drift tube was 105 degrees C and the nebulizer nitrogen flow rate was 2.9 L min(-1).
RESULTThe linear ranges of the asiaticoside were 0.35-7.0 microg. The recovery of the asiaticoside in C. asiatica extract was 94.9% (RSD 1.7%).
CONCLUSIONThe method is reliable, simple, precise and could be used for the quality control of C. asiatica extract.

6.Cassae-type diterpenes from seeds of Caesalpinia minax.

Fifteen cassaen-type diterpenes were isolated from the 95% ethanolic extract of the seeds of C. minax through various chromatographic techniques. Their structures were identified on the basis of spectroscopic data as pulcherralpin (1), caesalpinin ML (2), chamaetexane C (3), chamaetexane D (4), 6β, 18-diacetoxycassan-13, 15-diene (5), neocaesalpin K (6), neocaesalpin MP (7), neocaesalpin M (8), neocaesalpin Q (9), neocaesalpin P (10), neocaesalpin R (11), caesaldekarin D (12), caesaldekarin A (13), caesaldekarin b (14), 3β,6α-diacetoxyvouacapane (15). Among them, compounds 14, 9-11 were isolated from this plant for the first time.

7.Research progress of gene recombinant mesenchymal stem cells as tumor targeting delivery vehicles.

The applications of targeting gene delivery systems in tumor therapy have attracted extensive attention of researchers in recent years, as they can selectively deliver the therapeutic gene to tumor sites, improve the success rate of gene therapy and reduce the side effects. Therefore, design and development of novel gene delivery vehicles have been a hot area of current research. Recent studies have shown that mesenchymal stem cells (MSCs) have the ability to migrate towards and engraft into the tumor sites. Therefore, these properties make them a great hope for efficient targeted-delivery vehicles in cancer gene therapy. In this review, we examine the promising of utilization of MSCs as a targeted-delivery vehicle for cancer gene therapy, and summarize various challenges and concerns regarding this therapy.

8.Purification of large-scale plasmid DNAs by selective precipitation with cetyltrimethylammonium bromide.

Following Escherichia coli lysis with alkali, cetyltrimethylammonium bromide (CTAB) was directly titrated into the supernatant. An easy and feasible technology for plasmid purification was established with the optimized proportion between the quantity of CTAB and plasmid, combined with the specific solution for DNA release and TritonX-114 for endotoxin removal. Quality detection showed that the purified plasmid was free of contamination of host RNA. The host bacterial genomic DNA, endotoxin and bacterial protein were less than 10 microg, 50 EU and 10 microg per mg plasmids, respectively. The ratio of OD260/OD280 was between 1.75-1.85. Eighty percent of the prepared plasmids were presented in the supercoiled form. The plasmid purified with this technology can satisfy all criteria stipulated by FDA. The main advantages of the technology include the avoidance of animal-derived enzymes such as ribonucleases A, Proteinase K and toxic reagents like chloroform and phenol. In addition, the technology has low cost and no pollution.

9.Extraction and characterization of total DNA from Dendrobium.

OBJECTIVETo explore the quality of DNA with three methods of DNA extraction and the influence on RAPD-PCR.
METHODthe electrophoresis of total DNA, UV spectrophotometry and RAPD analysis were carried out on DNA extracted from three different methods.
RESULTThe DNA concentration and yields were different, which greatly influenced the result of RAPD-PCR.
CONCLUSIONThe higher quality DNA from Dendrobium can be obtained with the method of CTAB-free extraction medium before total DNA was isolated.

10.BIS Monitoring on Intraoperative Awareness: A Meta-analysis

Intraoperative awareness is a very serious complication of general anesthesia.Several studies have evaluated the potential association between bispectral index (BIS) and intraoperative awareness,however,the results obtained were controversial.Therefore,we performed a meta-analysis to further assess the association between the BIS monitoring and the incidence of intraoperative awareness.A comprehensive search was conducted to identify all eligible studies from the online literature databases published prior to Feb.2017.A total of five studies with 17 432 cases and 16 749 controls were included.An odds ratio (OR) and a 95% confidence interval (CI) were calculated to examine the strength of the association.The results showed that in the overall analysis,the association between the BIS monitoring and the incidence of intraoperative awareness was not significant (OR=0.58,95% CI=0.22-1.58,P=0.29).A stratified analysis by comparing different anesthesia methods revealed that BIS monitoring group showed a lower incidence of intraoperative awareness in patients with intravenous anesthesia when compared with non-BIS monitoring group (OR=0.20,95% CI=0.08-0.49,P=0.0004),whereas there was no statistically significant difference in the incidence of intraoperative awareness between BIS and non-BIS monitoring groups in patients with inhalation anesthesia (OR=1.13,95% CI=0.56-2.26,P=0.73).In conclusion,our meta-analysis showed that BIS monitoring had no appreciable advantage in the reduction of the intraoperative awareness incidence in inhalation anesthesia,while showed a remarkable superiority in intravenous anesthesia.
