2.Mechanism of Lead Biosorption and Biotransformation in Lead-Resistant Bacteria by In Situ Synchrotron Radiation Micro X-Ray Fluorescence and X-Ray Absorption Near Edge Structure

To investigate the mechanism of lead biosorption and biotransformation in lead-resistant bacteria in microcosmic scale, synchrotron radiation micro X-ray fluorescence (μ-SRXRF ) and X-ray absorption near edge structure ( XANES) was used to determine the element distribution characteristics and speciation of lead ( Pb) in lead-resistant bacteria strain isolated from farmland soil samples taken from a lead-zinc mine in Lanping county, Yunnan province in China. The isolated species was identified to be Arthrobacter sp. The concentration of Pb biosorpted in lead-resistant bacteria was directly determined byμ-SRXRF, and the results showed that the highest concentration was 5925μg/g and the bioaccumulate factor was 14 . 8 . Speciation of Pb in lead-resistant bacteria B2, LB ( Lysogeny Broth) medium and soil were also presented after using Pb LⅢXANES. Compared with Pb LⅢ XANES peak shape and peak position between standard samples, it was demonstrated that the Pb in lead-resistant bacteria was mainly formed by amorphous forms like PbS (58. 0%), (C17H35COO)2Pb (22. 2%), Pb5(PO4)3Cl (19. 8%) rather than organic Pb speciation which was exist in LB medium. Therefore, the isolated lead-resistant bacteria could biotransform Pb to sulfuric compounds. The biotransformation regular could be of great interest for other investigators as reference in study of bioavailability impact factor of heavy metals.
3.The biological features of adipose-derived stem cells and its application in oral tissue regeneration

BACKGROUND:In comparison with stem cel s from other tissues, adipose-derived stem cel s (ADSCs) are not only abundant and easy to harvest but also have the strong abilities to proliferate and differentiate. These make ADSCS the promising stem cel s in the field of oral tissue regeneration. OBJECTIVE:To summarize the biological features of ADSCs and the development of functions as stem cel s in the oral tissue regeneration. METHODS:We took“adipose-derived stem cel s, oral tissue regeneration, bone tissue engineering, periodontal tissue regeneration, bone defects”as the key words in Chinese and English, respectively, to retrieve the related literatures from PubMed, CNKI, Wangfang databases during 2000 to 2016 based on internet search. RESULTS AND CONCLUSION:In vitro studies on periodontal tissue regeneration have shown that ADSCs can promote the regeneration of bones, fibroblasts, blood vessels around the periodontal area, and improve the healing abilities of injured tissues. In the study of craniofacial bone regeneration, ADSCs can successful y repair massive bone loss due to the surgeries of tumor or accidents. ADSCs have a promising role in the oral tissue regeneration, but further studies wil be required because of the problems of immunogenicity and safety.
4.Re-irradiation of locally recurrent soft tissue sarcoma

The incidence of local recurrence after wide local excision and radiation of soft tissue sarcoma (STS) ranges from 5% to 20%.The optimal management of locally recurrent STS must be individualized.Approaches for retreatment include wide local re-excision followed by a variety of radiation which include external-beam radiation,brachytherapy and intraoperative electron radiotherapy.Following retreatment,the likelihood of ultimate local control ranges from 37% to 100%.However,each radiation technique could produce severe side effects,and so for selected patients,repeat irradiation may be unnecessary.
5.Effect of Ghrelin on contractility of gastric smooth muscle in guinea pigs and the related mechanism

Objective:To investigate and compare the influences of Ghrelin and growth hormone releasing peptide 6(GHRP-6) on the contractility of stomach smooth muscle in guinea pigs,and to study the related mechanism.Methods: The myenteric plexuses of gastric fundus and antrium in guinea pigs were stimulated with electrical field stimulation(EFS) to observe the influence of Ghrelin and GHRP-6 on the contractility of stomach smooth muscle.The influences of N-nitro-L-arginine(L-NNA) and L-Arginine(L-AA) on the effect of Ghrelin and GHRP-6 were studied to disclose the mechanism of the effects of Ghrelin and GHRP-6.Results: The circular muscle tissues of the gastric fundus generated on-relaxations and off-contractions when stimulating myenteric plexuses with 1-16 Hz electrical field;the on-responses induced relaxation could be reduced by L-NNA and the off-contractions induced contraction could be blocked by atropine and guanethidine.In fundic strips,ghrelin and GHRP-6 could decrease the on-response induced relaxation and increase off-response induced contraction of the muscle,with the effect of Ghrelin obviously stronger than that of GHRP-6.L-NNA could increase the effects of Ghrelin and GHRP-6-induced muscle contraction,and L-AA could decrease their effects.In the antral strips,electrical field stimulation of myenteric plexuses led to disappearance of relaxation wave,only leaving off-contractions.Both ghrelin and GHRP-6 could increase that contraction.Conclusion: Both ghrelin and GHRP-6 can promote the contractility of stomach smooth muscle in guinea pigs through stimulating myenteric plexuses of gastric fundus and antrium,which might be related to the NO pathway.
6.Analysis on Utilization of Hypoglycemic Drug in Medical Insurance Inpatients in Class A Grade Three Hospital During 2007~2009

OBJECTIVE:To evaluate the status quo and tendency of utilization of hypoglycemic drugs in medical insurance inpatients.METHODS:DDD was used to analyze the data of utilization of hypoglycemic drugs in medical insurance inpatients in class A grade three hospital during 2007~2009.RESULTS:The consumption sum of hypoglycemic drug used for medical patients increased year by year.The rates of increase were up to 14.04% for 2008 and 21.65% for 2009.The consumption sum of insulin increased rapidly.The rate of drug combination was 81.57% among which the rate of using two-drug was 68.66%.CONCLUSION:The new type of hypoglycemic drugs have become the main drugs in the hypoglycemics market.Domestic drug manufacturers should develop effective,safe,economical and be benefit for the treatment of complication so as to decrease medical costs and save the resources of medical insurance.
7.Correlation between seventy-three patients with Crohn's disease and Mycobacterium avium subspecies paratuberculosis infection

Objective To elucidate the correlation between Chinese patients with Crohn's disease (CD) and Mycobacterium avium subspecies paratuberculosis (MAP) infection.Methods The peripheral blood of 73 patients with CD (ileocolon type) and 40 healthy individuals was obtained.The monocyte layer was isolated for MAP culture.The growth of bacteria colony in the culture tube was observed every two weeks and totally for 16 weeks.By the end of culture,specific DNA of MAP (IS900) in the culture tube was detected by nested polymerase chain reaction.ANOVA was performed for measurement data analysis and chi square test was used for count data.Results After cultured for 16 weeks,no bacteria colony of MAP had been observed in 113 mycobacteral growth indicator tube (MGIT).No IS900 had been detected in the peripheral blood of all patients with CD and all healthy controls.Conclusion MAP is not correlated with the pathogenesis of CD (ileocolon type) in Chinese patients.
8.Mechanisms of childhood hemophagocytic lymphohistiocytosis:A bioinformatic analysis

Objective Hemophagocytic lymphohistiocytosis (HLH) is a life-threatening condition characterized by excessive inflammation, with a high incidence in children and a death rate of 40%.This study was to analyze the gene expression profile in child-hood HLH and explore the important pathways of childhood HLH using bioinformatic methods . Methods The childhood HLH gene ex-pression profile data GSE26050 were obtained from the Gene Expression Omnibus (GEO) database of the National Center for Biotechnolo-gy Information.Differentially expressed genes were identified with the GEO 2R online analysis tools released recently .The key pathways of the differentially expressed genes were investigated using the Kyoto Encyclopedia of Genes and Genomes ( KEGG) pathway enrichment a-nalysis. Results A total of 184 differentially expressed genes were identified , 126 upregulated and the other 58 downregulated .They were enriched in 3 pathways, including cytokine-cytokine receptor interaction , hematopoietic cell lineage and NOD-like receptor signaling pathways. Conclusion Bioinformatic tools allow the identification of the key genes and pathways associated with the development and progression of childhood HLH and point out the potential directions for researches on the mechanisms of childhood HLH .
9.Oxidative stress and interleukin-1β in acute gouty arthritis and the efficacy of probucol treatment

Objective To investigate the role of malondialdehyde(MDA),superoxide dismutase(SOD),and interleukin-1β (IL-1β) played in the pathogenesis of acute gouty arthritis and the treatment efficacy of probucol.Methods Acute gouty arthritis model in rat was established with injection of monosodium urate monohydrate (MSU) into the ankle joint cavity,then the joint swelling index was observed periodically,and MDA,SOD,IL-1β,and white blood cell count in the synovial fluid were determined by 72 h.Results The joint swelling index gradually increased,reaching a peak by 11h,and this finding was most obvious in the model group.The joint swelling index in the group treated with high-dose of probucol was significantly lower than that in the model group by 48 h(P<0.05).The joint swelling indices in the groups treated with moderate and high doses of probucol were lower than that in the model group by 72 h(P<0.05).The values of MDA in the moderate and high-dose groups were (14.45±3.11) and (11.54 ±3.10) nmol/ml respectively,being significantly lower than that in the model group[(24.46±4.27) nmol/ml,P<0.01].The amounts of SOD in the moderate and high-dose groups were (25.56±4.51) and (44.61±4.11)U/ml respectively,being higher than that in the model group[(13.32±2.02) U/ml,P<0.01].The amount of IL-1β in the high-dose group was lower than that in the model group[(15.41 ± 3.12 vs 19.87±3.99)pg,P<0.05].The white blood cell counts in the moderate and high-dose groups were (11.08±1.64)×l0~7/L and (7.43±1.52) x 10~7/L,being lower than that in the model group[(14.18±2.30)×10~7/L,P<0.01].Conclusion The levels of MDA and IL-1β in the synovial fluid were raised,while SOD was significantly decreased in acute gouty arthritis.Moderate and high-doses of probucol can effectively control acute gouty arthritis attack.The efficacy of high-dose probucol was equal to that of colchicines.
