Objective To explore the clinical analysis of Compound Erlongzuoci pill combined with auricular point stickingand vitamin B12 acupoint injection therapy on nervous tinnitus of middle and old aged.Methods 80 cases of middle and old aged patients with neurogenic tinnitus treated in our hospital from November 2015 to November 2016 were selected and randomly divided into the control group and the observation group, 40 cases of patients in each group, the control group was given Auricular Pressure Combined with vitamin B12 acupoint injection, the observation group was treated with Compound Erlongzuoci pills combined with auricular point sticking therapy and vitamin B12 acupoint injection,The clinical therapeutic effect of two groups, the tinnitus severity score of patients before and after treatment, the changes of tinnitus loudness and different frequency thresholds were compared between the two groups.Results The total effective rate of the clinical observation group was significantly higher than that of the control group, the difference between the two groups was statistically significant(P<0.05).Before treatment, there is no significant difference between tinnitus severity score, tinnitus loudness and hearing threshold between the two groups,the indexes after treatment in two groups were significantly improved(P<0.05), and the observation group tinnitus severity score, tinnitus loudness and different frequency thresholds were significantly lower than the control group, the difference was statistically significant(P<0.05).Conclusion Compound Erlongzuoci pill combined with auricular senile nervous tinnitus pressure and vitamin B12 acupoint injection therapy can significantly improve the degree of tinnitus patients, the clinical effect is confirmed.

O-linked N-acetylglucosamine (O-GlcNAc) glycosylation is an important form of posttranslational protein modification,mainly intracellular.It is closely related to cellular signaling pathways,and is involved in signal transduction,gene transcription and other important biological processes.Studies have found that O-GlcNAc glycosylation is directly related with diabetic retinopathy (DR),further studies may help us to uncover the DR mechanism,and develop new strategies for the diagnosis and treatment of this disease.
4.Combined Pharmacokinetics-pharmacodynamics Study of Rabeprazole in Inhibition of Gastric Acid Secretion

Objective To investigate the pharmacokinetics ( PK ) and pharmacodynamics ( PD ) processes of rabeprazole in inhibiting gastric acid secretion with the combined PK-PD model. Methods A total of 10 healthy volunteers were given a intravenous infusion of 20 mg rabeprazole over a 30-min period. The concentration of rabeprazole in the plasma at different time points was detected by HPLC,and the PK parameters were calculated by DAS 2. 0 software. At the same time the intragastric pH was monitored over 24 hours to fit the PD parameters with indirect inhibition model. Results The main PK parameters,t1/2,Cmax,and AUC were(60. 5±17. 3)min,(1 299. 1±201. 0)ng·mL-1,and(106. 4±26. 0)mg·min·L-1, respectively.The corresponding PD parameters,Kin,Ke,IC50 and Imax were(8.200±3.362)h-1,(1.080±0.378)h-1,(0.286± 0. 129)mg·L-1 and(6. 93± 2. 15)pH,respectively. Conclusion The PK of rabeprazole in healthy volunteers conforms to one compartment model,and the PD fits the indirect response inhibition model. The equation can effectively establish the relationship between the blood drug concentration and the effect.
5.Expression of human proinsulin gene in non-?-cells by point mutation with concise PCR

The human proinsulin gene was introduced a cleanage site for furin with two point mutation by polymerase chain reaction. The mutated gene was subcloned to the retroviral vector, and transferred into HepG2 hepatocarcinoma cells. Mature insulin can be expressed and secreted into supernatant by transferred HepG2 cells. The site mutation method is concise and with no misproduction.
6.Regression between MR findings of lumbar elements and chronic low back pain

Objective To investigate the relationship between the MR findings of lumbar elements and chronic low back pain(CLBP).Methods The patients underwent lumbar MRI examinations and sent for a questionnaires of low back pain (LBP).Among them,139 patients whose questionnaires illustrated with CLBP were enrolled.The enrolled patients included 68 patients with nerve roots compression and 71 patients without.Meanwhile,198 hospital staffs underwent lumbar MRI examinations and were sentfor a LBP questionnaire.Among them,62 patients without LBP and nerve roots compression were enrolled.Categorical regression was used to analyze the relationship between MR findings and CLBP.The MR findings iucluded nerve roots compression,average disk degeneration scores(ADD),high-intensity zones (HIZ),Schmorlnodes,Modic Ⅰ change,average facet joints degeneration scores(AZZ),facet joint effusion,high T2 signal in interspinous ligament and subcutaneousparaspinal muscles edema.The regression model was used to analyze the MR imaging and CLBP.Results The regression model was statistically significant (F=9.478,P<0.01).All predictors yielded an adjusted value was 0.446.Among all predictors,nerve roots compression,ADD,AZZ,subcutaneous or paraspinal muscles edema were statistically associated with the VAS degree (P<0.05).The sum of the importance of the four predictors above was 0.983.The quantification of predicted VAS degree increased as ADD level increased.The quantification of predicted VAS degree increased to the top at the 2 AZZ level and then decreased.Nerve roots compression and Subcutaneous or paraspinal muscles edema yielded higher quantification of predicted VAS degree level.Conclusion ADD,AZZ,subcutaneousparaspinal muscles edema were probably associated with CLBP degree after adjusting for nerve roots compression.

Objective To investigate the in vitro expressions of chamotatic factor CXCL12 [also known as stromal cell-derived factor 1 (SDF-1)],the receptor of the factor,Cys-X-Cys receptor 4 (CXCR4) and vascular endothelial growth factor C (VEGF-C) in human laryngeal carcinoma cell line Hep-2 and their roles in the homing of laryngeal carcinoma. Methods The expressions of CXCR4 mRNA and protein in Hep-2 cells was detected by RT-PCR and Western blotting,the expression of VEGF-C also was evaluated before and after incubated with SDF-1 (1,10,100 ng/ml) or the antagonist of CXCR4 AMD3100. Objective: To explore the growth inhibition and apoptosis-inducing effects of apigenin in human gastric cancer SGC-7901 cells. Method: MTT and flowcytometry were used to detect the growth inhibition and cell cycle distribution in apigenin-treated SGC-7901 cells respectively. DAPI fluorescence staining and DNA ladder assay were applied to study the pro-apoptosis effects of apigenin. The expression of apoptosis relative proteins, caspase-3 and Bcl-2, were analyzed by using Western blotting. Results: Apigenin treatment significantly inhibited the growth of human gastric cancer cell SGC-7901 and markedly caused their apoptosis following activation of caspase-3 and downregulation of Bcl-2 protein expression. Conclusion: The activation of caspase-3 and downregulation of apoptosis relative protein Bcl-2 expression were the possible mechanism of apigenin induced growth inhibition and apoptosis in SGC-7901 cells.
9.Efficacy of electroacupuncture on the recovery of gastrointestinal function after laparoscopic cholecystectomy:a systematic review

Objective:To evaluate the efficacy of electroacupuncture(EA)in enhancing the recovery of gastrointestinal function after laparoscopic cholecystectomy(LC). Methods:Randomized controlled trials(RCTs)of EA treatment in the postoperative period of patients undergoing LC were searched.Studies were obtained from Excerpta Medica Database(EMBASE),PubMed,Cochrane Library,Wanfang Academic Journal Full-text Database(Wanfang),China National Knowledge Infrastructure(CNKI),China Biology Medicine Disc(CBM),and Chongqing VIP Database(CQVIP)from inception to December 10th,2022.RevMan 5.4.1 was used to perform the meta-analysis.The Cochrane tool was used to assess the risk of bias.Mean difference(MD)and confidence interval(CI)were used for statistical descriptions. Results:A total of 7 studies were included in the meta-analysis.The meta-analysis found that the EA group had a shorter time to the first flatus[P<0.001,MD=-5.32,95%CI(-6.42,-4.21)],bowel movement recovery[P<0.001,MD=-6.22,95%CI(-8.11,-4.34)],and the first defecation(P<0.001,MD=-11.08,95%CI(-15.78,-6.39)]than the control group. Conclusion:EA treatments can promote the recovery of gastrointestinal function after LC.
10.Preparation of carbon nanoparticle paclitaxel suspension and pharmacokinetic study of intraperitoneal chemotherapy.

OBJECTIVETo prepare carbon nanoparticle-paclitaxel suspension(CNPS) and to study the pharmacokinetics of intraperitoneal chemotherapy with CNPS.
METHODSSaturated absorption capacity of carbon nanoparticle suspension (CNS) and paclitaxel were detected by high performance liquid chromatography in order to prepare the above suspension. Wistar rats were randomly divided into the experimental group (A) and the control group (B), to which intraperitoneal injections of CNPS and paclitaxel were given respectively. At different time points, measure the blood samples, mesenteric lymph nodes, and intraperitoneal lavage fluid were collected to measure the concentration of paclitaxel.
RESULTSOne ml CNS could absorb 7 mg paclitaxel by maximum. The ratio of area under the curve (AUC) in the plasma of group A to group B was 0.63. The ratio of AUC in lymph nodes of group A to group B was 0.75 and that in intraperitoneal lavage fluid was 1.25. The metabolic half-life (t1/2) of paclitaxel in the plasma of group A was 1.61 times as long as that of group B. The t1/2 of paclitaxel in intraperitoneal lavage fluid of group A was 0.88 as long as that of Group B. The t1/2 of paclitaxel in lymph nodes of group A was 1.10 as long as that of Group B.
CONCLUSIONSCNS has a high absorption capacity with paclitaxel. Intraperitoneal chemotherapy by CNPS is characterized by low drug concentration in the blood, high drug concentration in the peritoneal cavity and high safety. However, the targeting and lymphatic retention effect are not significant. The mechanism warrants further investigation.
