1.Professor Lu Zhizheng of Clinical Examples for the Treatment of Rheumatoid Arthritis

Objective To summarize professor Lu Zhizheng's clinical experience in the treatment of rheumatoid arthritis,in order to better guide clinic. [Method]By analyzing professor Lu Zhizheng's treatment of two patients with rheumatoid arthritis of the medical record data, it summarized from the aspects of etiology and pathogenesis,syndrome differentiation and treatment.[Result]Professor Lu argued that invigorating spleen to remove dampness should be throughout the course of the treatment of rheumatoid arthritis.Two patients can reduce joint swel ing and pain,improve the appetite gradual y,and the symptoms were relieved obviously by replenishing qi and reconciling ying,adjusting ying and wei.[Conclusion]Professor Lu Zhizheng's Chinese medicine treatment of rheumatoid arthritis has significant curative effect,it is worthy of reference study.
2.Over-expression of human ribonuclease inhibitor suppresses invasion and migration of transplanted bladder cancer

Objective To investigate the effect of over-expression of human ribonuclease inhibitor suppresses invasion and migration of transplanted bladder cancer .Methods The T24 cells were stably transfected with pIRES2-EGFP-RI and pIRES2-EG-FP plasmid respectively .Using the cell transfected with pIRES2-EGFP and untransfected cell as controls .and the positive clones were screened by G418 ,respectively ;Tumor cells of the three groups at 2 × 106 were respectively injected into the back of BALB/C nude mice to establish the xenograft models .Change of micro-blood vessels in tumor tissue and expression of CD31 were detected by Immunohisto-chemical and HE staining .Immunohisto-chemical assay was used to detect the expression of RI ,MMP-2 ,MMP-9 ,E-cadherin ,N-cadherin ,Vimentin ,Snail ,Slug ,Twist in the tumors .Results Animal experiment showed that the T24-RI cells group significantly inhibited the growth of bladder cancer compared with the other two control groups .Compared with the T24 and T24 vector cells groups ,the microvessel density in tumor tissue of T24-RI group was notably reduced and the expressions of MMP-2 , MMP-9 ,N-cadherin ,Vimentin ,Snail ,Slug ,Twist significantly were decreased simultaneously ,while the expressions of RI and E-cadherin were increased .Conclusion up-regulation RI can inhibit the growth of transplanted bladder cancer in nude mice by decrea-sing the expression of invasion protein and EM T protein .
3.Application analysis of curette and stripping wire method in diabetic foot nursing

Objective To observe the clinical efficacy using curette and stripping wire method in treating diabetic foot. Methods 36 hospitalized patients diagnosed as diabetic foot were enrolled, and 18 of them were chosen as the experimental group according to random digital table method who was treated with medical therapy plus curette and stripping wire method in treating diabetic foot. The other 18 patients were named as the control group who were treated by medical treatment and routine treatment. The granulation tissue maturity, wound healing, and the differences on curative effect were compared. Results 10 cases′wound granulation tissue maturity in the experimental group got++and+++on the 10th day while 4 cases in control group. Wound healing rate of the experimental group at the 10th and 14th day were (78.6±10.5)%and (82.7±8.4)%while the control group were (43.2±8.7)%and (66.2±10.1)%. Cure rate of the experimental group was 14/18 four weeks after treatment, the inefficiency was 0, while the control group was 6/18 and 3/18. Conclusions Curette and stripping wire method is a good method to promote the dressing diabetic foot wound healing. It is superior to traditional methods, worthy of clinical application.
4.Association between XPC rs2228000 (C/T)polymorphism and the susceptibility of breast cancer:a Meta-analysis

Objective To quantitatively examine the relationship between xeroderma pigmentosum complementation C group (XPC)rs2228000 (C /T)polymorphism and the susceptibility of breast cancer. Methods The relevant case-control studies published up to December 2015 which investigated XPC rs2228000 (C /T)polymorphism and breast cancer risk were identified by searching PubMed,Cochrane Library,Chinese Biomedical Literature Data,Wanfang Database,China National Knowledge Infrastructure and VIP Database. Meta-analysis was conducted using STATA 12.0 software and odds ratio (OR)with its 95%CI were estimated. Results A total of 8 researches involving 9 case-control studies (3 850 breast cancer cases and 5 047 healthy controls) were included.The Meta-analysis showed that there was statistical association between XPC rs2228000(C /T)variance and breast cancer risk in the homozygous model (TT vs.CC:OR =1.28,95%CI:1.08-1.52,Z =2.80,P =0.005)and recessive model (TT vs.TC +CC:OR =1.23,95%CI:1.05-1.43, Z =2.64,P =0.008),but not in the allele model,heterozygote model and dominant model.In the subgroup of ethnicity and genotyping methods,the different significant correlation was existed between them under Asian and PCR-RFLP in genetic models (T vs.C:OR =1.21,95%CI:1.05-1.40,Z =2.63,P =0.009;TT vs. CC:OR =1.55,95%CI:1.13-2.13,Z =2.70,P =0.007;TT +TC vs.CC:OR =1.26,95%CI:1.02-1.55,Z =2.19,P =0.028;TT vs.TC +CC:OR =1.39,95%CI:1.04-1.87,Z =2.23,P =0.026).We also found significant association between them in subgroup of population-based controls in the homozygous model (TT vs.CC:OR =1.27,95%CI:1.02-1.57,Z =2.16,P =0.031).Conclusion XPC rs2228000 (C /T)polymorphism may be associated with the susceptibility of breast cancer,especially in Asian,and gene-type TT may increase the risk of breast cancer.
5.Association between XPC Lys939Gln (A/C) gene polymorphism and the susceptibility of gastric cancer:a Meta-analysis

Objective To explore the association between Xeroderma pigmentosum complementation C group (XPC) Lys939Gln (A/C) gene polymorphism and the susceptibility of gastric cancer.Methods By searching PubMed,Cochrane Library,Elsevier,Springer-Verlag,China National Knowledge Infrastructure,Chinese Biomedical Literature Data,VIP Database and Wanfang Database,all eligible case-control studies published up to September 2015 were selected and the quality of each article was valuated by two reviewers independently according to the inclusion and exclusion criteria.Meta-analysis was performed by using STATA 12.0 software.Odds ratio (OR) and 95% confidence interval (CI) were calculated.The text was estimated for the subgroup analysis,sensitivity analysis and publication bias test.Results A total of 7 case-control studies were included,including 2 336 cases with gastric cancer and 3 502 controls.The Meta-analysis showed that compared with the allele A,the allele C increased the risk of gastric cancer (OR =1.09,95% CI:1.01-1.18,Z =2.12,P =0.034);compared to the genotype AA,the homozygous model (CC) and dominant model (CC + AC) also increased the risk of gastric cancer (CC vs.AA:OR =1.19,95% CI:1.00-1.42,Z =2.00,P=0.046;CC+ACvs.AA:OR=1.12,95%CI:1.00-1.25,Z=2.03,P=0.042).The Meta-analysis showed the statistical significance between XPC Lys939Gln (A/C) gene polymorphism and the gastric cancer risk in subgroup of Asian people (C vs.A:OR =1.10,95% CI:1.01-1.20,Z =2.28,P =0.023;CC vs.AA:OR=l.21,95%CI:1.01-1.46,Z=2.02,P=0.043;CC +AC vs.AA:OR =1.13,95% CI:1.01-1.27,Z =2.11,P =0.035) and the source of community in the control group (C vs.A:OR =1.11,95% CI:1.01-1.21,Z=2.25,P =0.024;CC vs.AA:OR =1.23,95% CI:1.02-1.50,Z =2.12,P =0.034).Conclusion XPC Lys939G1n (A/C) gene polymorphism may be associated with the susceptibility of gastric cancer,and genotype CC,CC + AC and allele C can increase the risk of gastric cancer.
6.Overview of traditional Chinese medicine quality evaluation method based on overall research.

The establishment of quality evaluation of traditional Chinese medicine system that not only accords with Chinese medicine function characteristics but also is recognized as international medical circles, is an arduous task in urgent need of solving the current modernization of traditional Chinese medicine in the process of internationalization. It is difficult to evaluate atraditional Chinese medicine by detection of single active components in traditional Chinesemedicinewiththe western medicine quality controlmethod due to the overall effects of traditional Chinese drugs, the components of the overall diversity, targets, and the complexity of the interaction between components of unpredictable make the Long-term since, domestic and foreign scholars continue to explore and put forward a series of quality evaluation of traditional Chinese medicine to promote the development of traditional Chinese medicine. This article summarized the related academic ideas and developments to, providea new thought and perspective for the quality control of traditional Chinese medicine.

7.Construction and expression of eukaryotic expression vector of human IL-37 b gene

Objective To construct the eukaryotic expression vector pEGFP-N1/IL-37b and analyze the expression of IL-37 gene in THP-1 cells. Methods Total RNA was extracted from human peripheral blood mononuclear cells ( PB-MCs) and the coding region of IL-37b gene was amplified by RT-qPCR. Then, the gene was cloned into pEGFP-N1 eu-karyotic expression vector. After transfected the recombinant plasmid into THP-1 cells, the expression of IL-37 was detec-ted by RT-qPCR and Western blot. Results Double restriction enzyme digestion and gene sequencing showed that IL-37b gene was correctly inserted into the eukaryotic expression vector pEGFP-N1. RT-qPCR and Western blot showed that the IL-37 expression level was increased significantly (P<0. 01) after transfection in THP-1 cells. Conclusions We successful-ly constructed a novel anti-inflammatory cytokine IL-37 eukaryotic expression vector pEGFP-N1/IL-37b, which lays a foun-dation for further study on IL-37 functions and its association with related diseases.
8.THE EFFECT OF hCG STIMULATION GIVEN AT DIFFERENT INTERVIALS ON TESTOSTERONE BIOSYNTHESIS IN CULTURED HUMAN TESTICULAR TISSUE

Cultured human testicular tissues were grouped as follows:control group,group tracted with single dosc of human chorionic gonadotropin(hCG), group treated with double doses of hCG given at three-day-interval and group treated with multiple doses of hCG. The dose of hCG was 20kIU/L,and it was added into the medium and incubated for 24 hours. Testosterone was measured by RIA. The result indicated that all three hCG-treated groups had a testosterone secretion peak at 48-72 hours after the first dose of hCG. After the second hCG treatment given in three days interval the testosterone increased again. but its maximal rise reduced. Multiple doses of hCG given successively inhibited the response of Leydig cells to hCG stimulation.

Mesenchymal stem cells(MSCs)have a unique role in immune regulation and focus to T-cells.In the mixed lymphocyte reactions,MSCs inhibit T-cells proliferation by cycle arrest,but they do not increase T-cell apoptosis and the suppress T-cell activation.In addition,MSCs can reduce CD8~+T cells and Thl cells,and simultaneously increase Th2 cells in the reaction system to suppress the inflammatory response,which may play a therapeutic effect on the T-cells mediated autoimmune diseases.
10.The effect of hyperxia liquid on posttraumatic vasospasm in patients with traumatic brain injury using transcranial Doppler

Objective To study the role of transcranial Doppler (TCD) in early diagnosis of posttraumatic vasospasm in traumatic brain injury patients and in treatment effect monitoring of hyperxia liquid for this condition. Methods Seventy-four patients with posttraumatic vasospasm were divided into two groups. The control group (n=42) received the general treatment, while the treatment group (n=32) received the treatment of hyperxia liquid in addition to the general treatment. Their cerebral blood flow velocities of bilateral MCA and extra-cranial portion of ICA were monitored regularly by TCD, starting from the first day after head injury until 14th day. The changes of physiological and neurofunctional parameters in both groups were compared, including cerebral vasospasm(CVS),arterial blood oxygen pressure (PaO 2),arterial blood oxygen saturation (SaO 2), the Glasgow coma scale (GCS)and the ultimate effects of treatment as indicated by Glasgow outcome scale(GOS). Results Cerebral vasospasm occurred in 1 to 3 days and peaked to the 3 to 7 days after injury, then markedly relieved at 14 days after injury. After infusion of hyperxia liquid, the PaO 2 and SaO 2 in the treatment group were significantly higher than those of the control group. The degree of vasospasm was significantly higher in the control group than that in the treatment group. GCS and GOS of the treatment group were significantly higher than those of the control group. Poor outcome was common in patients with severe cerebral vasospasm. Conclusion Early posttraumatic vasospasm can be detected by TCD. High-oxygen liquid is effective for treating posttraumatic vasospasm.
