Objective To investigate the clinical results of resection of periacetabular tumors and reconstruction with fibular autograft. Methods Eight patients with tumors around acetabular were treated surgically in Tongji Hospital between January 1999 and April 2003. The series comprised 5 males and 3 females, with an average age of 39.6 years old (range, 21-65 years old). One patient was diagnosed with osteosarcoma, 2 with chondrosarcoma, 3 with metastatic bone tumor, 1 with giant cell tumor(GCT), and 1 with aneurysmoid bone cyst. There were ⅠB in one patient, ⅡB in 3 patients according to Enneking surgical staging in four primary malignant tumors patients. The tumors involved the zones Ⅱ of Enneking classification in 5 patients, the zonesⅠand Ⅱin 3 patients. All the 8 patients with periacetabular tumors underwent resection and reconstruction with fibular autograft(including 3 cases of a free vascularized fibular graft). Functional results were evaluated according to the system of the Musculoskeletal Tumor Society. Results Bony union occurred 15.2 weeks (range 14-16 weeks) in 5 patients reconstructed with non-vascularized fibular graft and 13.6 weeks (range 12-15 weeks) in 3 patients reconstructed with vascularized fibular graft after the initial surgery. Five of them were followed up ranged from 14 to 42 months (mean 28 months). There were continuously disease-free without complications during follow-up. At the final followup, the average functional score was 22 (range 18-27) according to the system of the Musculoskeletal Tumor Society. The good and excellent rate of functional results was 100%. Conclusion Tumor resection and reconstruction by fibular autograft is an effective treatment for patients with periacetabular tumors. Satisfactory postoperative function was obtained, and relief of pain and walking independently could be achieved with a high level of patient satisfaction.

Objective: To express glycosyl-phosphatidylinositol (GPI) modified Met- RANTES fusion protein on Chinese hamster ovary (CHO) cells and to develop a novel immunosuppressant GPI anchored form of Met-RANTES. Methods: The eukaryotic expression vector PEF/GPI-Met-RANTES were constructed and transfected into CHO cells by electroporation. The transfectants were selected with methotrexate (MTX). Expression of the recombinant protein was assessed by flow cytometric analysis, cell immunofluorescence staining and immunogold electron microscopy. Results: The chimeric molecules of GPI anchored form of Met-RANTES including the whole reading frame were constructed, and the sequence was identical to the designed sequence. GPI anchored form of Met-RANTES was stably expressed on CHO- DHFR- cells. Conclusion: A large amount of GPI modified Met-RANTES fusion protein was expressed on CHO cells. GPI anchored form of Met-RANTES may be used as novel immunosuppressant for suppressing reaction in graft rejection.

Objective To investigate the effects of the serum from chronic renal failure (CRF) rats on endoplasmic reticulum stress (ERS) and activation of activating transcription factor 4 (ATF4) of rat arterial smooth muscle cells (ASMCs) cultured in vitro and explore the possible mechanism. Methods The rat model of CRF was established by 5/6 nephrectomy. ASMCs were incubated in the media with 10% or 20% serum of CRF rats cultured in vitro. ATF4 nuclear translocation were analyzed by immunofluorescence. GRP78 (glucose regulated protein 78), p-PERK (pancreatic ER kinase) and ATF4 proteins in response to the CRF serum in ASMCs were determined by Western blot. The ATF4-DNA binding activity was analyzed by electrophoretic mobility shift assay (EMSA). Results The CRF serum could significantly promote the expression of GRP78 of ASMCs in dose-dependent manner. Under the stimulation of 20% CRF serum, nuclear translocation of ATF4 in ASMCs was found. Compared with control serum group, the expression of ATF4 and p-PERK of CRF serum group was increased significantly (P<0.01). The CRF serum could increase the ATF4-DNA binding activity. Conclusion The CRF rat serum can effectively induce ERS of ASMCs and activate the pathway of PERK-ATF4.

Abstract: Objective To collect the cases of laboratory-acquired infections (LAI) reported in literatures in China, summarize the infection routes and causes of LAI in China, in order to improve laboratory staff's understanding of its occupational health and safety risks. Methods The cases of laboratory-acquired infection reported in domestic literatures were collected from PubMed, CNKI, Wanfang Database, CBM China Biomedical Literature Database up to April 11, 2022, retrospectively analyze the number and causes of LAI reports, the main risk factors of LAI and its harm to society, the consequences of LAI or the leakage of pathogenic microorganisms, and put forward the relevant countermeasures of biological safety. Results A total of 22 LAI reports were collected, reviewed and integrated into 21 reports. There were 7 kinds of pathogenic microorganisms. The main pathogenic microorganisms were hantavirus (42.86%, n=9) and Brucella (33.33%, n=7). There were 122 cases and 3 deaths in the laboratory. Most of the reports came from research laboratories (66.67%, n=14). The main route of infection was inhalation of aerosol (42.86%, n=9), followed by transdermal route (38.09%, n=8). Conclusions Failure to report LAI events will increase the risk of pathogenic microorganisms spreading to people outside the laboratory and the environment through infected laboratory staff. Local health institutions and laboratories should be encouraged to report LAI cases as a powerful tool for monitoring accidental leakage of pathogenic microorganisms and further improving laboratory biosafety. The laboratory needs strong biosafety measures to protect staff's health and prevent environmental pollution caused by accidental leakage of pathogenic microorganisms.
