1.All-trans retinoic acid diminishes collagen production in a hepatic stellate cell line via suppression of active protein-1 and c-Jun N-terminal kinase signal.

Following acute and chronic liver injury, hepatic stellate cells (HSCs) become activated to undergo a phenotypic transformation into myofibroblast-like cells and lose their retinol content, but the mechanisms of retinoid loss and its potential roles in HSCs activation and liver fibrosis are not understood. The influence of retinoids on HSCs and hepatic fibrosis remains controversial. The purpose of this study was to evaluate the effects of all-trans retinoid acid (ATRA) on cell proliferation, mRNA expression of collagen genes [procollagen α1 (I), procollagen α1 (III)], profibrogenic genes (TGF-β(1), CTGF, MMP-2, TIMP-1, TIMP-2, PAI-1), fibrolytic genes (MMP-3, MMP-13) and the upstream element (JNK and AP-1) in the rat hepatic stellate cell line (CFSC-2G). Cell proliferation was evaluated by measuring BrdU incorporation. The mRNA expression levels of collagen genes [procollagen α1 (I), procollagen α1 (III)], profibrogenic genes (TGF-β(1), CTGF, MMP-2, TIMP-1, TIMP-2, PAI-1), and fibrolytic genes (MMP-3, MMP-13) were quantitatively detected by using real-time PCR. The mRNA expression of JNK and AP-1 was quantified by RT-PCR. The results showed that ATRA inhibited HSCs proliferation and diminished the mRNA expression of collagen genes [procollagen α1 (I), procollagen α1 (III)] and profibrogenic genes (TGF-β(1), CTGF, MMP-2, TIMP-1, TIMP-2, PAI-1), and significantly stimulated the mRNA expression of MMP-3 and MMP-13 in HSCs by suppressing the mRNA expression of JNK and AP-1. These findings suggested that ATRA could inhibit proliferation and collagen production of HSCs via the suppression of active protein-1 and c-Jun N-terminal kinase signal, then decrease the mRNAs expression of profibrogenic genes (TGF-β(1), CTGF, MMP-2, TIMP-1, TIMP-2, PAI-1), and significantly induce the mRNA expression of MMP-3 and MMP-13.
2.Relationship between Onset of Malnutrition and Growth Hormone-Insulin-Like Growth Factor Axis in Children with Congenital Heart Disease

Objective To investigate the relationship between onset of malnutrition and growth hormone-insulin-like growth factor(GH-IGF) axis in children with congenital heart disease(CHD).Methods Fifty cases with CHD were divided into two groups,13 cases of cyanotic group and 37 cases of acyanotic group.And the acyanotic group was then subdivided into two groups,25 cases of heart failure group and 12 cases of without heart failure group,and 20 normal children were selected as control group.The nutritional status of these children were evaluated by detecting the serum total protein,serum protein,body height and weight.IGF-Ⅰ and insulin-like growth factor binding protein-3(IGFBP-3) and growth hormone binding protein(GHBP) were detected by immunoradiometric assay.Results The acute malnutrition ratio and chronic malnutrition ratio in the children with CHD were 62% and 28%,respectively.The acute malnutrition ratio was similar between cyanotic group and acyanotic group(P=0.105);the chronic malnutrition ratio significantly increased in cyanotic group than that in cyanotic group(P=0.006).The levels of serum IGF-Ⅰ and IGFBP-3 decreased in the patients with CHD,which significantly decreased in cyanotic group and heart failure group.The level of serum GHBP decreased in cyanotic group and heart failure group.Conclusions The levels of serum IGF-Ⅰ and IGFBP-3 in patients with CHD decrease,which are related to growth hormone receptor down regulated,that is one of the molecular mechanisms of growth hormone resistant or insensitivity,growth hormone counteract or insensitivity are one of the reasons of acute and chronic malnutrition in children with CHD.
3.Effect of BDNF gene modified umbilical cord mesenchymal stem cells transplantation on neurological function improvement in rats subjected to brain trauma

Objective To explore the effect of brain derived neurtrophic factor(BDNF) gene modified umbilical cord mesenchymal stem cells ( UCMSC ) transplantation on neurological functional improvement in rats after brain trauma.Methods Cerebral contusion model in motor-sensory cortex in rats was established by a weight hammer falling method.UCMSC were cultured and transferred with BDNF gene.After BDNF expression and activity were determined,the BDNF gene modified UCMSC were implanted into traumatic brain.The neurological function was evaluated for 2 weeks after brain injury.And the BDNF expression was determined by using immunohistochemistry.Results Severe neurological dysfunction was seen in animals that had been subjected to contusion brain injury( 10.50 ±0.53 ).A significant improvement on neurological function was found in the UCMSC transplantaion animal( 7.75 ± 0.71 ), compared with only brain injury group (P < 0.01 ).Moreover, rats in BDNF gene modified UCMSC showed the most behavior improvement ( 5.50 ± 0.76 ) (P < 0.01 ).Conclusion BDNF gene modified UCMSC transplantation can survive and migrate, and improve neurological function in brain traumatic rats.
4.Transplantation of NGF modified umbilical cord mesenchymal stem cells improves neurological deficits in rats subjected to brain trauma

Objective To investigate the effect of NGF gene modified umbilical cord mesenchymal stem cells (UCMSC) transplantation on neurological functional improvement in traumatic brain rats. Methods Cerebral contusion model in motor-sensory cortex in rats was established by a weight hammer falling method. UCMSC were culutred and transferred with NGF gene. After NGF expression and activity was identified,the NGF gene modified UCMSC were engrafted into injured brain. The neurological function was evaluated 2 weeks after brain injury. And the NGF immunostaining was also performed to explore the level of NGF expression. Results Severe neurological dysfunction( 10.50 ± 0.53 )occurred in rats after traumatic brain injury, while the UCMSC transplantaion led to a significant functional improvement( 7.75 ± 0. 71 )(P < 0. 01 ). Moreover, the best functional improvement was found in rats receiveing UCMSC grafts modified with NGF gene (5.38 ± 0. 52 ) (P < 0.01 ). Conclusion NGF gene modified UCMSC transplantation can improve neural behavior in rats with brain trauma.
5.Advance in studies on regulatory mechanism of phenotype polarization of macrophages and intervention with traditional Chinese medicines.

Macrophages are heterogeneous and diversified, and can be polarized into different phenotypes in various microenvironments and physiological or pathological conditions. Major macrophage subpopulations including classically activated(M1) and alternatively activated(M2) macrophages, which represent different surface receptors, secret different cytokines and chemokines, are regulated by different signal paths of transcriptions and epigenetic levels, and play distinctive roles in tumor progress. TCMs may improve the microenvironment by regulating phenotype polarization of macrophages. So far, specific biomarkers and polarized molecules mechanisms generated through the macrophage polarization approach are still unclear. In this article, we merely summarize the advance in domestic and foreign studies on phenotype polarization of macrophages and regulatory mechanisms and look into the future of intervention with TCMs.

6.The current situation of nursing research based on Omaha system in China:a content analysis

Objective To investigate the current situation of nursing research based on Omaha system in China. Methods All the nursing research based on Omaha System were retrieved and collected by using China National Knowledge Infrastructure, Wanfang database and VIP database, NoteExpress software for the management of documents was used. Based on the classification of nursing research literature, a content analysis of system analysis model analyzed the documents had been retrieved. Results Sixty-four pieces of literature were included, and thirteen categories system were formed. The transverse reliability coefficient of all kinds of system were 0.940-1.000, the longitudinal reliability coefficient were 0.658-1.000. Conclusions The study of nursing research based on Omaha system in China is booming, which is also facing challenges, and has a broad space for exploration.
7.Adhesion of Lactobacillus protects the enterocyte-like cell HT-29 against the invasion of enteropathogenic E. coli

Objective To investigate the beneficial effect on enterocyte-like cells HT-29 through adhesion of Lactobacillus. Methods Using viable bacteria counting method to evaluate inhibition of Escherichia coli adhesion and invasion to enterocyte-like cells by incubation with Lactobacillus together. Cellular membrane permeability and cell viability were examined in vitro by LDH level and MTT method respectively. Results Adhesion and invasion of enteropathogenic Escherichia coli to enterocyte-like cells was inhibited by high adhesiveness strain Lactobacillus reuteri JCM1081. In vitro, no significant changes of the morphology, structure and function of the HT-29 cells after being treated with Lactobacillus alone for 3 hours, especially cellular LDH level, viability and membrane permeability. Lactobacillus reuteri JCM1081 did not alter cell integrity and prevented the increase in permeability induced by enteropathogenic Escherichia coli infection. Conclusion Lactobacillus reuteri JCM1081 interact with intestinal epithelial cell receptor to competitively inhibit the adhesion and invasion of enteropathogenic Escherichia coli to HT-29 cells by incubation and enhance the integrity of the cells.
8.Operative treatment of fractures of the calcaneus for earthquake patients

[Objective]To investigate the characteristics and clinical results of open reduction and internal fixation for fractures of the calcaneus for earthquake patients.[Method]The fractures were classified according to Sanders classification.Fifteen cases(24 fractures) were treated by open reduction and internal fixation.Thirteen cases(21 fractures) were followed up from four,four and half months(average of 4.2 months).[Result]All fractures had been healed.Functional results were evaluated using Marryland Foot Score and excellent results occured in 9(42.9%),good in 10(47.6%),fair in 2(9.5%) and the satisfactory rate was 90.5%.Two cases of incision infection were cured by skin flap transplantation.[Conclusion]Open reduction and internal fixation is an effective method for treatment of displaced intra-articular fractures of the calcaneus.Earthquake patients are not fit for early operation.They should be treated with operation after normal functional index and stable psychological situation.
9.Determination of Emodin in Changtong Oral Liquid by HPLC

OBJECTIVE:To determine the content of emodin in Changtong oral liquid.METHODS:HPLC was used.The stationary phase was Nova-Pak C 18 column and the mobile phase was methanol-0.1%phosphoric acid solution(85∶15).The UV detection wavelength was254nm.RESULTS:There was a good linear relationship within the concentration range of0.48~2.4?g/ml of emodin in Changtong oral liquid(r=0.9998).The average recovery was99.73%(n=5),RSD=1.42%. CONCLUSION:This method is simple,fast and accurate.
