BACKGROUND: During vascular dementia, ischemia, hypoxia, energy expenditure, abnormal metabolism of neurons, decrease of generation of Adenosine Triphosphate and disorder of ionic environment in and out of cells are observed in brain tissue, which can cause abnormal release of monoamine transmitter.OBJECTIVE: To probe into effect of shenma yizhi capsule on content of monoamine transmitter in brain tissue of rat models with vascular dementia induced by multiple cerebral infarction.DESIGN: Randomized controlled study.SETTING: Pathological and Physiological Department o Liaoning Basic Medical Institute.MATERIALS: Totally 96 Wistar rats of either gender, aged 8-12 months, weighing 270-500 g, were selected. METHODS: The experiment was completed in the Pathological and Physiological Department of Vocational-technical College, Liaoning College of Traditional Chinese Medicine from April to July 2001. All rats of either gender were divided into 6 groups with 16 in each group. Wistar rats in 5 groups were injected with cruor embolus in internal carotid artery to make animal model of vascular dementia induced by multiple cerebral infarction. After modeling, rats were randomly divided into 3.2 g/kg, 1.6 g/kg and 0.8 g/kg shenma yizhi capsule groups (shenma yizhi capsule was extracted from ginseng and tall gastrodia tuber with 2.7 g raw materials and provide by Xiyuan Hospital of Chinese Academy of Traditional Chinese Medicine),positive control group and dementia control group. Animals without modeling were regarded as normal control group. Rats in each dosage group were perfused with the corresponding dosage of shenma yizhi capsule; rats in positive control group were perfused with 1 mg/kg hydergine dihydroergotoxine; rats in dementia control group and normal control group were perfused with the same volume of saline solution. One week after modeling, rats were medicated once a day for 6 weeks. Content of monoamine transmitter was measured with high performance liquid chromatograpy.MAIN OUTCOME MEASURES: Content of monoamine transmitter such as levarterenol, adnephrin, dopamine, indoleacetic acid, homovanillic acid and 5-serotonin.RESULTS: Nine rats died because of self-body quality and environmental change, and totally 87 animals entered the final analysis. Content of levarterenol of rats in dementia control group was lower than that in normal control group [(0.40±0.23), (0.70±0.14) ng/g, t=2.712, P < 0.01]; content of levarterenol of rats of 3.2 and 1.6 g/kg dosage groups was higher than that of dementia control group [(0.57±0.09), (0.58±0.19), (0.40±0.23) ng/g,t=2.211, P < 0.05], but was lower than that of normal control group. Content of levarterenol of rats of 0.8 g/kg dosage group was lower than that of normal control group [(0.48±0.23) ng/g, t=2.213, P < 0.05], but was higher than that of dementia control group and positive control group [(0.41 ±0.19) ng/g]. Differences of other neurotransmitters were not significant.CONCLUSION: Content of levarterenol in brain tissue of rats of dementia control group is decreased obviously, but shenma yizhi capsule can increase content of levarterenol in brain tissue of rats. The mechanism of shenma yizhi capsule on treating vascular dementia is possibly related with increasing content of levarterenol in brain tissue.

OBJECTIVETo investigate the effect of four traditional Chinese medicines distributed along lung meridian, namely Ephedrae Hebra, Zingiberis Rhizoma, Scutellariae Radix and Mori Cortex, on TLR2 and NF-κB expressions in mice with lung heat syndrome, in order to study the intervention effect of the four traditional Chinese medicines (TCMs) on the lung heat syndrome.
METHODOne hundred KM mice were randomly divided into the normal control group, the model group, the Ephedrae Hebra group, the Zingiberis Rhizoma group, the Scutellariae Radix group and the Mori Cortex group (20, 10 g x kg(-1)), nasally dripped with streptococcus pneumoniae to establish the mouse lung heat syndrome model, and then administered with different TCMs. The expressions of TLR2, NF-κB p65 proteins in lung tissues were analyzed by the immunohistochemical method. The expressions of TLR2, NF-κB p65 mRNA were measured by real time PCR.
RESULTCompared with the normal control group, the expressions of TLR2 and NF-κB p65 proteins in lung tissues in the model group were higher (P < 0.01), and the expressions of TLR2 and NF-κB p65 mRNA in lung tissues were up-regulated (P < 0.01 or P < 0.05). Compared with the model group, Ephedrae Hebra high and low dose groups, the Zingiberis Rhizoma low dose group and the Scutellariae Radix high dose group showed decreased expression of TLR2 protein (P < 0.05 or P < 0.01); Ephedrae Hebra high and low dose groups, the Zingiberis Rhizoma low dose group, Scutellariae Radix high and low dose groups and Mori Cortex high and low dose groups showed reduced expression of NF-κB p65 protein (P < 0.05 or P < 0.01). Ephedrae Hebra high and low dose groups, Zingiberis Rhizoma high and low dose groups, Scutellariae Radix high dose group and Mori Cortex high dose group showed down-regulated expression of TLR2 mRNA (P < 0.01 or P < 0.05).
CONCLUSIONEphedrae Hebra, Zingiberis Rhizoma, Scutellariae Radix and Mori Cortex can induce the TLR2/NF-κB inflammatory signal pathways by down-regulating the expressions of TLR2 and NF-κB p65 in protein and mRNA, so as to alleviate the lung tissue injury in mice with lung heat syndrome.

OBJECTIVETo record the electrical activities of Antirior cingulate cortex (ACC) neurons by in vivo multi-channel recording methods using the model of complete freund's adjuvant (CFA) induced conditioned place avoidance (C-CPA), which has been set up in our previous studies.
METHODSThe electrode was self-made and the CPA responses were recorded by in vivo multi-channel recording method.
RESULTS(1) The electrical activities of ACC neurons could be successfully recorded by the self-made electrode. (2) Before or after the injection of CFA, rats were respectively conditioned to the different place. The firing rates of ACC neurons in the CFA-paired place vs that in the non-CFA-paired place was (0.853 ± 1.377) imp/s vs (0.221 ± 0.971) imp/s (P < 0.05, n = 26). (3) The CPA responses in the CFA-paired place vs that in the non-CFA-paired place were (303.55 ± 61.77)s vs (140.32 ± 33.52)s(P < 0.05, n = 6).
CONCLUSIONThe firing rates of rACC (rostral Anterior Cingulate Cortex) neurons were involved in the occurrence of the affective pain.

ObjectiveTo study the relationship between C-erbB-2 and estrogen (ER) and progesterone (PR) receptors, and the relationship between C-erbB-2, ER, PR with histologic grade. MethodsTo detect ER, PR and C-erbB-2 states by using immunohistochemical analysis and fluorescence in situ hybridization for C-erbB-2 in 163 unselected invasive breast carcinomas. ResultsC-erbB-2, ER ,PR were expressed in 21.5% ,64.4% ,44.2% of 163 cases respectivly . 5 pure mucinous carcinomas , 3 tubular carcinomas and 1 micropapillary carcinoma were ER + ( 100.0% ) 、C-erbB-2 - ( 100.0% ) and PR + (40.0% ,66.7%, 100.0% ). C-erbB-2 was positive in 22.3% of grade Ⅱ and 27.0% of grade Ⅲ invasive ductal carcinomas and negative in all grade Ⅰ invasive ductal carcinomas.ER and PR expression were decreased significantly in C-erbB-2 + tumors compared with C-erbB-2 - tumors( ER,25. 7% vs 75.0% ; PR,25.7% vs 49.2% ). Although ER or PR expression is decreased in C-erbB-2 + tumors, a substantial proportion of them still express ER or PR. ConclusionC-erbB-2 overexpression or amplifcation was limited to a minority of invasive breast carcinomas. Tumour grade was an independent predictor for ER expression. ER was expressed in small number of high-grade and in large number of grade Ⅰ invasive ductal carcinomas. C-erbB-2 overexpression or amplification essentially was limited to grades Ⅱ and Ⅲ ductal carcinomas and correlated inversely with ER or PR expression.
