Background and purpose: MicroRNA(miRNA) is a class of small non-coding RNA playing an important regulatory role in many diseases. In this study, we explore the levels of miR-192 and zinc ifnger E-box binding homeobox 2 (ZEB2) in CRC, the clinical signiifcance of miR-192 and the correlation between the expression of miR-192 and ZEB2 protein. Methods: The expression levels of miR-192 and ZEB2 mRNA in 30 colorectal carcinoma samples, 30 corresponding cancer-adjacent tissue samples (from the edge of tumor≥5 cm), 25 colorectal adenoma samples, and 15 normal tissue samples were quantiifed using quantitative real-time polymerase chain reaction (qRT-PCR). ZEB2 protein expression was determined using Western blot. The relationship between the miR-192 and clinicopathological factors, miR-192 and ZEB2 protein expression was analyzed. Results:Signiifcant upregulation of miR-192 expression and reduction of ZEB2 mRNA and protein expression were identiifed in CRC tissues, compared to cancer-adjacent tissues, colorectal adenoma samples, and normal tissues (P<0.05). Low miR-192 levels were signiifcantly associated with lymph node (P=0.021) and distant metastasis (P=0.023). An inverse relationship between miR-192 and ZEB2 protein expression was identified in CRC group (r=-0.365, P<0.05). Conclusion: MiR-192 downexpression was correlated with CRC metastasis. MiR-192 may play a role in the development and progression of CRC through ZEB2.

Objective To investigate the role of platelet rich plasma (PRP) on free fat graft's survival in a rat model.Methods Twenty-four rats were used in the experiment.For each rat,three groups of free fat grafts were injected into the subcutaneous layer of dorsum.Group A was the control group with 0.5 ml fat and 0.12 ml normal saline.Group B received 0.5 ml fat and 0.1 ml PRP with 0.02 ml normal saline.Group C was given combination of 0.5 ml fat and 0.1 ml PRP which was activated by 0.02 ml calcium chloride.Eight rats were euthanised using pentobarbital sodium each time,1,2 and 3 months after fat transplantation.Specimens were acquired and stained with haematoxylineeosin,and then the histological changes examined using light microscopy.All the data were statistically analysed using paired t test to determine if there were differences between the study groups.Results Compared the volume of the fat grafts at 1 month,there was no deference between the groups(P＞0.05).However,the diameter of group C was significantly higher than group A (P＜0.05).On histological examination,larger vascular density was observed in group C compared with group A at 1 month (P＜0.05).And reduced inflammatory reaction was also observed in group C at 3 months(P＜0.05).While other histological parameters were not statistically significant (P＞0.05).Conclusions PRP does not enhance the survival of fat graft in the Wistar rat model.

During the period of peri-operation on gallbladder,T2MD patients with calculous cholecystitis were randomly divided into groups of continuous subcutaneous insulin infusion(CSII,n=32),multiple subcutaneous insulin injection(MSII,n=48) and OHA(n=64).It is found that CSII has better glucose control than MSII and OHA.

Objective:To observe the efficacy of contact lenses on a wide range of corneal epithelial shedding. Methods:The information has collected a wide range of corneal epithelium of thirty-six patients admitted to our hospital in the January 2010-December 2010, tracking observed in patients with epithelial wear contact lenses after the healing. Results:The patients wear contact lens immediate relief of symptoms with eye pain, foreign body sensation. Patients with continuous wear contact lenses after five to seven days, including twenty-eight patients with corneal epithelial fully restored, the remaining eight cases without contact lens continue to use eye drops of one to two days corneal epithelial fully restored in the ointment;thirty-six patients were followed up in three to six months, not fall off again. Conclusion:Therefore, use contact lenses to treat corneal epithelial shedding, the patients suffered less pain, treatment is easy to operate, low cost, the treatment effect is significant.

Objective To establish a rapid, specific and sensitive diagnostic technique for the human T. gondii infection with hematopoietic stem cell transplantation and discuss its clinical significance. Me- thods Fifty-six patients subject to hematopoietic stem cell transplantation were detected with ELISA and PCR. Results Among 56 recipients of hematopoietic stem cell transplantation, 7 were positive for T. gondii antigen and 10 were positive for SAG3 gene fragment respectively with the positive rate being 14.3 % and 17.8 % in the ELISA and PCR screening respectively. Twenty healthy people were negative for anti-Toxo antibody.Conclusion PCR is an accurate, relatively rapid, sensitive and specific method for detecting SAG3 gene of T. gondii, and can be considered a valuable additional tool for identification of T. gondii infections.

OBJECTIVE:To investigate the effect of arsenic trioxide(As2O3)on human hepatoma cells SMMC-7721 viability and the expression of urokinase plasminogen activator receptor(uPAR).METHODS:SMMC-7721 cells were cultured and treated with As2O3 at different concentrations for different time with untreated SMMC-7721 cells(without As2O3 treatment)served as control.The proliferation rate of the SMMC-7721 cells was measured by MTT assay;the change of cell cycle was detected by flow cytometry analysis;the effect of As2O3 on uPAR mRNA levels was analyzed by real-time fluorescent quantitative polymerase chain reaction.RESULTS:As2O3 had significant inhibitory effect on SMMC-7721 proliferation in time-dependent and concentration-dependent manner.As compared with control group,As2O3 significantly decreased the proportion of cells in G0/G1 phase but increased the cell proportion in G2/M phase,and it markedly down-regulated the level of uPAR mRNA expression(P

Objective To study the effects of genistein on the biological characteristics of and collagen synthesis by human skin fibroblasts in vitro.Methods Human skin fibroblasts(HSFBs)were obtained from the prepuce of healthy adolescents,and suhjected to primary culture.Atier 5 to 15 passages of culture,HSFBs were treated with various concentmtions(0.03125,0.0625,0.125,0.25,0.5,1mg/L)of genistein for different durations.The potential of cell proliferation was detected by MTT assay and growth curves were drawn for HSFBs.Flow cytometry(FCM)and RT-PCR were used to estimate cell cycle phases and mRNA expression of type Ⅰ procollagen.respectively.Results The proliferation rate of HSFBs was 97.7%,113.8%,132.5%,116.4%,94.5%and 83.3%after treatment with genistein of 0.03125,0.0625,0.125,0.25,0.5 and 1 mg/L,respectively.The genistein of more than 0.5 mg/L displayed an inhibitive effect on the proliferation of HSFBs.while that between 0.0625 and 0.25 mg/L showed a promotive effect.Atier treatment with genistein at 0.0625,0.1 25 and 0.25 mg/L,the percentage of HSFBs in S phase and G2 phase significantly increased compared with untreated HSFBs(S phase:41.15%±2.88%,61.89%±3.16%,48.18%±1.68%vs30.12%±0.60%,P＜0.05;G2 phase:9.76%±3.99%,10.40%±0.54%,7.46%±2.47%vs 0.61%±0.16%,P＜0.05).Compared with the untreated HSFBs.the relative mRNA expression level of type Ⅰ procollagen was increased with genistein of 0.0625,0.125 and 0.25 mg/L(0.4814±0.0138,0.5767±0.0291,0.5675±0.0272 vs 0.4101±0.0236,P＜0.01),but decreased with genistein of Ⅰ and 0.5 mg/L(0.1662±0.0165 and 0.2017±0.0203 vs 0.4101±0.0236,P＜0.01).ConclusionCertain concentrations of genistein could enhance the proliferation and growth of as well as mRNA expression of type Ⅰ procollagen in HSFBs.
