Objective To explore the relationship between the expression of matrix metalloproteinase (MMP) and tissue inhibitor of metalloproteinase (TIMP) in gestational trophoblastie diseases and its invasiveness.Methods The expression and distribution of MMP-7,MMP-9,TIMP-1 and TIMP-2 were determined by immunohistochemistry PV-6000 in 49 cases of hydatidiform mole (eight of them with malignant transformation),six cases of invasive hydatidiform mole,two cases of choriocarcinoma and 18 cases with normal villi in early pregnancy.Results The expression of MMP-7,MMP-9,TIMP-2 and MMP- 9/TIMP-1 was significantly higher in choriocareinoma,invasive mole and malignant transformed mole than that in normal villi in early pregnancy and non-malignant transformed mole (F=10.950,7.760,7.304, 3.442;P
2.A clinical controlled study of mesh and non-mesh technique in laparoscopic repair of large hiatal hernias

Objective To investigate the safety and effectiveness of mesh and non-mesh techniques in laparoscopic repair of large hiatal hernias (LRLHH).Methods A retrospective clinical controlled study of mesh and non-mesh techniques in LRLHH form January 2006 to August 2014 was performed.Results A total of 83 and 36 patients were recruited to mesh and non-mesh group respectively.There were no significant differences in operation time,operation bleeding volume,hospitalization time or complications between the two groups.Main symptoms were significantly improved during the postoperative long-term follow-up in both groups.The improvement of dysphagia in mesh group showed no significant difference [22.9% (19/83) VS 12.0% (10/83),P=0.066],however,non-mesh group showed significant difference [30.6% (11/36) VS 5.6% (2/36),P=0.006].Rate of dysphagia alleviation in non-mesh group was significantly higher than that in mesh group [25.0% (9/36) VS 10.8% (9/83),P =0.048].Mesh-related complications of esophageal erosions occurred in 5 patients (6.0%) in mesh group,including esophageal stenosis in 3 patients,esophageal-cardiac stricture in 1 patient.Recurrent hernia occurred in 1 patient (1.2%) in mesh group and 3 patients (8.3%) in non-mesh group (P =0.082).Conclusion LRLHH with mesh should be individualized.The use of mesh in LRLHH reduces the recurrence rate,but may lead to some complications.
3.Expressions of Let-7 in Normal Breast Tissue and Breast Cancer Tissue

Objective To study the expression characteristics of Let-7 genes in breast cancer.Methods Twenty-eight patients with breast cancer were randomly selected,and their cancer tissue and adjacent normal tissue were collected.TRIzol was used to extract the total RNA and real-time quantitative PCR was used to detect the relative gene expression levels.Results The expression of precursor Let-7 in cancer tissue was (9.65 ± 2.31),which was lower than that in normal tissue (10.05 ± 2.81),P =0.048.Precursor Let-7 had dependence relationship with the long menstruation (b =0.816,P =0.029).The menarche age showed a positive correlation with precursor Let-7 in normal tissue (r =0.502,P =0.048) and a negative correlation in cancer tissue (r =-0.484,P =0.049) Conclusions The expression of precursor Let-7 in cancer tissue is lower than that in normal tissue.The period of menstruation is a protective factor to breast cancer.

5.Ultrasonographic characteristics of ovarian carcinosarcoma.

OBJECTIVETo summarize the ultrasonographic characteristics of ovarian carcinosarcoma (OCS).
METHODSTotally 12 patients with OCS have been detected in our department. All patients underwent surgical resection and the OCSs were confirmed pathologically. Ultrasound examination was performed before operation. Imaging features were analyzed retrospectively for bilaterality, size, and tumor solidity (cystic or solid). Presence of ascites and other evidence of peritoneal seeding, adjacent organ invasion, and surgical staging were also evaluated.
RESULTSUnilateral OCSs were found in 11 patients and bilateral OCSs in one patient one side was missed by ultrasound). Nine OCSs were solid, and 3 were mixed (solid and cystic). The diameter of the largest dimension was less than 10 cm in 4 cases, 10-20 cm in 7 cases, and larger than 20 cm in one case. Other ultrasonographic findings including peritoneal seeding and direct invasion into the adjacent organs were seen in 8 patients. Surgical stages were International Federation of Gynecology and Obstetrics (FIGO) classification c and b in one patient each, c in 9 patients, and in one patient.
CONCLUSIONOCSs usually present as large, hypoechoic, solid, and aggressive tumors, combined with frequent peritoneal seeding and adjacent organ invasion.

6.Effect of asymmetric dimethylarginine on platelet-aggregation and losartan intervention in spontaneous hypertensive rat models.

OBJECTIVE:
To evaluate the effect of asymmetric dimethylarginine (ADMA) and the results of losartan intervention on platelet-aggregation in spontaneous hypertensive rats.
METHODS:
Spontaneous hypertensive rats (SHR) were randomly assigned into 3 groups: SHR control group, L-arginine treatment group (L-arg) and losartan (los) treatment group, each group consisting of 16 rats. Another 16 Wistar Kyoto rats (WKY) served as normal control group. The L-arginine and losartan treatment groups received 1.0 g/kg L-arginine or 30 mg/kg losartan in 10 mL/kg distilled water daily through gastric tube for 2 weeks respectively, while the SHR and WKY groups received distilled water alone. All the rats took tap water and standard feed freely during the experimental period. Systolic blood pressure (SBP) was monitored by the tail-cuff method. At the end of the 2-week intervention, all the rats were sacrificed and the blood samples were collected from the carotid artery. The platelet-aggregation-rate, NO levels, eNOS activity, and ADMA levels both in the plasma and the platelets were measured. We got other platelet samples from the SD rats and incubated the platelets with blood vascular endothelium from the above 4 groups of experimental rats and the platelet-aggregation-rate was monitored as well.
RESULTS:
(1) Systolic blood pressure of the SHR was significantly higher, compared with that of the WKY (P<0.01), which were significantly reduced both in the L-arginine and losartan groups (P<0.01). (2) Platelet-aggregation-rate of the SHR was significantly higher, compared with that of WKY (P<0.01), which was significantly reduced both in the L-arginine and losartan groups (P<0.01). (3) NO levels both in the plasma and the platelets of the SHR were lower, compared with those of the WKY (P<0.05); and were elevated significantly both in the L-arginine and losartan groups,compared with those of the SHR (P<0.05); (4) Both the plasma and the platelet eNOS activities of SHR followed the same pattern of the NO levels in these groups (P<0.01). (5) In contrast, the plasma and platelet ADMA levels showed a reverse pattern (P<0.05). (6) Platelets from the SD rats incubated with vascular endothelium of WKY exhibited lower platelet-aggregation-rate,compared with the platelets incubated with SHR vascular endothelium (P<0.05); Platelet-aggregation-rate of the SHR group increased, compared with that of the WKY group (P<0.05); Platelet-aggregation-rate both of L-arginine and losartan groups reduced, compared with that of the SHR group (P<0.05).
CONCLUSION
High levels of ADMA both in the plasma and in the platelets of SHR are associated with the decline of eNOS activity and NO levels, which might be an important reason for the increased platelet-aggregation-rate. Intervention with Losartan can reduce the platelet-aggregation-rate simultaneously with its known anti-hypertensive effect. The possible mechanism might be that losartan can enhance the eNOS activity and elevate NO levels through the suppression of ADMA.

9.Effects of shangke jiegu tablet on the gene expressions of osteoprotegerin and osteoprotegerin ligand in the repairing process of mandibular defect rabbits.

OBJECTIVETo explore the mechanism of Shangke Jiegu Tablet (SJT)in repairing the mandibular defect.
METHODSTotally 72 healthy male New Zealand rabbits were randomly divided into the normal control group (n = 24), the model group (n = 24), and the SJT group (n = 24). Then the mandibular defect model was established. Animals in the normal control group and the model group were fed with normal forage, while those in the SJT group were fed with SJT forage. On the day 7, 14, 28, and 56 after model establishment, 6 rabbits were killed in each group. The bone was collected from the mandibular defect. The gene expressions of osteoprotegerin (OPG) and osteoprotegerin ligand (OPGL) were detected by means of RT-PCR. The positive dyeing strength and area of the bone tissue were detected by means of immunohistochemical technique.
RESULTSCompared with the normal control group, the degree of OPGmRNA expression was remarkably up-regulated on day 7 after model establishment (P < 0.05) and the degree of OPGLmRNA expression was remarkably up-regulated on day 14 after model establishment (P < 0.05) in the model group. Compared with the model group, the degree of OPGmRNA expression was remarkably up-regulated (P < 0.05), and the positive dyeing strength and area of bone tissue were stronger and broader on day 14, 28, and 56 after model establishment in the SJT group. The degree of OPGLmRNA expression was remarkably down-regulated (P < 0.05), and the positive dyeing strength and area of bone tissue were weaker and smaller on day 14 after model establishment in the SJT group. The ratio of OPGmRNA/OPGLmRNA was remarkably up-regulated on day 14, 28, and 56 after model establishment (P < 0.05).
CONCLUSIONThe effect mechanism of promoting mandibular defect repairing by SJT may be correlated to regulating the expressions of OPGmRNA and OPGLmRNA.

10.Investigation on the migration and biologic effects of nano FeOx powders under the exposure of extremely low frequency altering electric magnetic field in human heptoma-bearing nude mice in vivo.

To investigate the mechanism and biologic effects of 37 nm magnetic nano FeOx powders (MNPs) on human hepatoma-bearing nude mice. 37 nm MNPs were prepared by coprecipitation methods and then injected into human hepatoma (Bel-7402) bearing-nude mice through the tail vein. After injection of MNPs, the mice were first exposed under static magnetic field and then treated under extremely low frequency altering-electric magnetic field directing to the tumor area. The migration and trafficking of MNPs were determined by MMR. Tumor growth was monitored with calipers every 5 days and tumor volume was calculated on the basis of three-dimensioned measurements. The apoptosis of tumor cells was analyzed by flow cytometry analysis. The expressions of apoptosis-associated proteins Bcl-2, Bax and HSP27 were determined using western-blot analysis. Static magnetic field could direct the migration and trafficking of MNPs to the tumor site with a higher ratio of 98.9%. Extremely Low Frequency Electric-Magnetic Field (EMF) treatment could inhibit the proliferation of tumor cells and prolong the survive time of tumor-bearing mice injected with MNPs. In addition, the survival time of tumor-bearing mice and percentages of prohibition on tumor cell growth were 27.4+/-0.7 days and 37.5+/-0.8% (F = 0.005, P is less than to 0.05), respectively. The results of flow cytometry analyses showed that about 18.1+/-0.6% (F = 0.030, P is less than to 0.05) of tumor cells were induced into early apoptosis. Furthermore, expressions of apoptosis-associated proteins Bcl-2 and Bax were significantly induced by MNPs under EMF treatment. The ratio of Bcl/Bax in both MNPs and EMF treatment group was 0.07+/-0.01, which was much lower than that of control group (0.23+/-0.02) (F = 0.016, P is less than to 0.05). Heat shock protein-27 (Hsp-27) was not significantly induced in different treatment groups. Injection of MNPs with EMF exposure on human hepatoma-bearing nude mice could significantly prolong the survival time, inhibit the tumor proliferation and growth, and induce tumor cells into apoptosis.
