Chinese Journal of Behavioral Medicine and Brain Science 2013;22(8):677-679
Objective To investigate the effect of streptozotocin (STZ) on learning, memory and expression of p-MKK4,p-JNK and p-Jun in APP transgenic mice. Methods Three-month-old APP/PS1 mice were intraperitoneally injected with STZ.Learning and memory function was performed by Morris water maze. The changes of neuronal morphology in hippocampum were observed by Nissl staining. The expression of MKK4,JNK and c-Jun were detected in hippocampal tissues by Western blotting. Results In the hidden platform experiment of Morris maze, the escape latencies and the pathway length of STZ-induced diabetic mice were longer than those in controls from the 2nd to 5th day(P < 0.05 or P < 0.01).The Nissl staining showed that some neurons in hippocampum of STZ-treated mice were displayed changes of injury.Compared with the vehicle control, the expression levels of pJNK and p-c-Jun were increased by 217.78 ± 23.49 and 189.27 ± 17.80,respectively (P < 0.01).Conclusion STZ can decrease the learning and memory function and injury the neurons, which maybe relate to the JNK signal pathway.

3.Effects of Blood-cooling and Blood-stasis-removing Decoction on Autoantibody and Serum Levels of Interleukins 6, 17, 21 in MRL/lpr Experimental Rats
Journal of Guangzhou University of Traditional Chinese Medicine 2015;32(6):1055-1058
Objective To observe the effect of blood-cooling and blood-stasis-removing Decoction on anti-double-strain DNA ( anti-dsDNA) antibody, anti-nucleosome antibody ( AnuA) and serum levels of interleukin ( IL) -6, 17, 21 in spontaneous systemic lupus erythematosus ( SLE) MRL/lpr experimental rats. Methods The experimental rats were divided into blank control group, model group, and high- and low- dose Chinese medicine groups ( 25.2, 12.6 g/kg respectively) , the treatment lasting 8 weeks. At the end of the experiment, the blood taken from the orbital veins was separated for obtaining serum, and then the serum anti-dsDNA antibody, AnuA, IL-6,17,21 levels were tested by enzyme-linked immunosorbent assay ( ELISA). Results The serum autoantibody and IL-6, 17, 21 levels of rats in the model group were increased significantly as compared with the blank control group ( P<0.01). The serum antibody and cytokine levels of Chinese medicine groups were reduced as compared with the model group, the difference between high-dose Chinese medicine group and model group being significant ( P<0.01). Conclusion Blood-cooling and blood-stasis-removing Decoction has certain effect on reducing the serum levels of anti-dsDNA antibody, AnuA, and IL-6,17,21, which may coniribute to one of its therapeutic mechanisms for SLE.

Journal of International Oncology 2013;(3):163-165
Indoleamine 2,3-dioxygenase (IDO) is known as an endogenous immunosuppressive enzyme which plays a significant role in the process of tumor.IDO is not only found in tumor cells but also detected in dendritic cell (DC) in tumor microenvironment, which participates in the formation of tumor immune tolerance through expressing IDO enzyme.Signal transducer and activator of transcription (STAT) is the main signal protein family which participates in the IDO transcriptional regulation of DC.It is necessary to detail the signaling pathway in regulating IDO expression, which will help us develop high specific and more active IDO inhibitors and provide new options for anti-cancer targeted therapy.

Chinese Journal of Thoracic and Cardiovascular Surgery 2003;0(06):-
Objective: To assess the peculiarity in fluorine-18 fluorodeoxyglucose (FDG) uptake in patients with bronchial alveolar carcinoma. Methods: From December, 1998 to April, 2002, 22 patients with bronchial alveolar carcinoma (BAC) were imaged with FDG-PET (positron emission tomography) before surgery. Their maximum and mean standard uptake value (SUVmax and SUVmean ) of tumor and SUV of normal lung (SUVlung) were measured. Results: All tumors were detected by FDG-PET. FDG uptake of tumor was higher than that of normal lung (P

Journal of International Oncology 2017;44(6):468-471
Radiotherapy is the principle method of non-surgical treatment for esophageal cancer. The 5-year survival rate ranges from 20% to 40%.In recent years, the studies have found that anti-angiogenesis therapy can not only directly inhibit tumor nutritional intake, but also turn around tumor hypoxia by normalizing tumor vessels so as to enhance the radio-sensitivity of tumor ceils, which is expected to become an effective way to improve the curative effect of esophageal cancer radiotherapy.

Objective To understand the relationship between embryonic chromosomal abnormalities and missed abortion by high-throughput gene sequencing, so as to provide a basis for guiding the next pregnancy.Meth-ods By chromosome detection on villi chromosome of 52 cases of missed abortion by high-throughput gene sequen-cing technique, the types of chromosome abnormalities and the proportion were investigated. Results Among villi samples from the 52 cases of missed abortion, chromosomal abnormalities were founded in 28 cases, including 24 ca-ses of number abnormality (17 cases of trisomy, 6 cases of triploid and a case of monomer) and 4 cases of structural abnormalities. Conclusion Embryonic chromosomal abnormality is the main cause of missed abortion in early preg-nancy.High-throughput gene sequencing technology used to detect the villi chromosome is comprehensive, fast and accurate, helping to clarify the cause of missed abortion and guide the next pregnancy reasonably.

Objective To construct a eukaryotic expressing vector harboring human DC-SIGN, and establish a BHK21 cell line stably and highly expressing DC-SIGN. Methods The DC-SIGN gene fragment which contained Not I and BamH I sites was amplified by PCR from pUNO-hDCSIGN1Aa plasmid, digested with Not I and BamH I, and then cloned into an eukaryotic expression vector pIRES-neo to construct eukaryotic expression vector pIRES-neo-DC-SIGN. The recombined plasmid was identified with Not I and BamH I enzyme digestion and sequencing, the latter was then transfected to BHK21 cells by LipofectamineTM 2000. After screening culture by G418, BHK21 cell line stably expressing DC-SIGN was established. The expression of DC-SIGN was identified by flow cytometry, Western blotting and immunofluorescence method. Results The gene sequence of DC-SIGN was consistent with that of design. PCR and double enzyme digestion analysis showed that the recombinant plasmid pIRES-neo-DC-SIGN was constructed successfully. After transfection, positive clones were selected with G418. After limiting dilution assay, BKH21 cell lines stably expressing DC-SIGN were established. The detection result of flow cytometry showed that the expression ratio of DC-SIGN positive clones was close to 90%. The result of immunofluorescence displayed that the expression of DC-SIGN was mostly located on the surface of cell membrane. Western blotting displayed the specific band of DC-SIGN protein. It showed that the BHK21 cells stably expressing DC-SIGN were successfully established. Conclusion DC-SIGN eukaryotic expression vector has been successfully constructed. The successful establishment of BHK21 cell lines which can stably express DC-SIGN provides a substantial foundation for further study on the DC targeting vaccines.
