2.Application of micro transesophageal echocardiography (micro-TEE) in neonat and infant cardiac surgery

Objective To evaluate the value of phased array probe of transesophageal echocardiography miniature (micro-TEE) in the neonatal and infant cardiac surgery intraoperation.Methods Micro-TEE probe was used in children with weight ≤5 kg undergoing cardiac surgery to detect the atrial and ventricular cavity size,the left and right ventricular outflow tract diameter were detected by transthoracic echocardiography(TTE) 3-7 d after operation,and compared with the measured value immediately after the TEE.Results Before and after intubation,airway index not statistically significant (P > 0.05).Preoperative TEE and TTE inspection results were consistent with intraoperative diagnosis.Postoperative TEE measuring intracardiac structural data (each room,chamber size and left and right ventricular outflow tract diameter) and postoperative TTE measurement value representing the difference was not statistically significant (P > 0.05).Conclusions Micro TEE intraoperative guardianship of low birth weight infants is safe and feasible,and can supplement and improve the preoperative diagnosis,and can accurately and timely evaluate postoperative curative effect.
3.Expression of telomerase in skin lesions of condyloma acuminatum,bowenoid papulosis,Bowen's disease and vulvar squamous carcinoma

Objective To study the expression of telomerase and its possible clinical significance in patients with condyloma acuminatum,bowenoid papulosis,Bowen's disease and vulvar squamous cell car- cinoma.Methods The human telomerase reverse transcriptase (hTERT) was detected by immunohisto- chemistry technique in all patients.Histopathological analysis was done in patients with bowenoid papulosis or Bowen's disease.Results There was a significant difference in the expression of hTERT among normal tissue,condyloma acuminatum and squamous carcinoma,and the expression rate increased successively from normal control to squamous carcinoma.In bowenoid papulosis,the expression of bTERT was significantly higher as compared to that in condyloma acuminatum,significantly lower as compared to that in squamous carcinoma,and was as the same as that in Bowen's disease.Histopathologically,the degree of cell atypia was significantly higher in Bowen's disease than that in bowenoid papulosis.Conclusions The expression of telomerase increases successively from normal skin,benign tumors to malignant carcinomas,which sug- gests that the expression of telomerase may play an important role in both cell proliferation and immortaliza- tion.
4.Effect of Caprini risk score-based care on the prevention of venous thromboembolism of patients with major orthopedic surgery

Objective:To investigate the effect of Caprini risk score-based care on the prevention of venous thromboembolism of patients with major orthopedic surgery.Methods:Totally 124 patients with major orthopedic surgery were randomly divided into experimental group and control group, each group contained 62 cases. The control group received routine thrombosis prevention nursing, patients in the experimental group carried out Caprini risk score-based intervention. The rate of VTE was compared between two groups, the levels of D-Dimer as well as thigh circumference was measured after 24 hours of admission, 3 days after surgery, 5 days after surgery and at discharge day. At discharge day, the inpatients satisfaction with nursing service scale was used to assessment the satisfaction with nursing service of inpatients.Results:The rate of VTE in the experimental group was 1.6% (1/62) and 6.5% (4/62) in the control group, the difference was not statistically significant ( χ2 value was 1.876, P=0.171). Three days after surgery, 5 days after surgery and at discharge day, the D-Dimer were (1.26±0.65) μg/ml, (1.27±0.35) μg/ml, (0.96±0.27) μg/ml in the experimental group, 5 days after surgery and at discharge day, the thigh circumference were (29.81±4.15) cm, (28.86±3.40) cm in the experimental group, significantly lower than those in the control group [(1.64±0.58) μg/ml, (1.51±0.55) μg/ml, (1.25±0.44) μg/ml, (31.84±5.22) cm, (30.83±3.91) cm], the difference was statistically significant ( t value was 2.405-4.485, P<0.05). The health education and guidance, personality traits, service quality, discharge guidance scores and total satisfaction scores were 26.31±3.06, 35.73±3.64, 27.89±3.11, 8.47±1.18 and 111.08±6.33, significantly higher than those in the control group (24.15±2.31, 33.92±2.66, 26.27±3.41, 7.42±1.82 and 106.52±5.85), the difference was statistically significant ( t value was 2.750-4.433, P<0.05). Conclusions:Caprini risk score-based nursing care significantly alleviates the hypercoagulable states and relieves the degree of swelling of the affected of patients with major orthopedic surgery, as well as improves patients' satisfaction with nursing work.
5.Therapeutic value of Shuxuetong injection in patients with acute coronary syndrome

Objective To investigate the therapeutic value of Shuxuetong injection in patients with acute coronary syndrome(ACS).Methods 89 ACS patients were divided randomly into two groups: control group(n =44) and experimental group(n=45).The conventional therapy was used in the control group while Shuxuetong was added in the experimental group.The course of treatment was two weeks.Thromboxane B2(TXB2) and 6-keto-prosta-glandin F1a (6-K-PGF1a) were detected by radio-immunity before and after therapy.Homocysteine were tested by corpuscle enzyme linked immunosorbent assay.Results TXB2 was obviously lower in the experimental group [(554.2±303.2) ng/L] and the control group [(737.2±299.3 ) ng/L ] after treatment than that before treatment (P<0.05).Homocysteine was obviously lower in the experimental group [(10.42±2.99 )μmol/L ] and the control group [(16.39±2.29) μmol/L ] after treatment than that before treatment (P < 0.05 ).6-K-PGF1a was obviously higher in the experimental group[(328.4±99.6) ng/L] and the control group[ (234.5±96.2) ng/L] after treat-ment than that before treatment (P<0.05).TXB2 and homoeysteine in the experimental group were obviously lower than those in the control group after treatment(P<0.05). 6-K-PGF1 a in the experimental group was obviously high-er than those in the control group after treatment (P<0.05).Conclusion Shuxuetong injection could degrade the level of TXB2 and HCY, increase the level of PGF1 a and improve the balance of TXB2/PGF1a, so Shuxuetong is ef-fective in treating acute coronary syndrome.
6.The relationship between EphB receptor and the development of tumor

Currently ,EphB receptor has been demonstrated as an important factor on the biological be -havior of normal cells .Some studies found that it plays an important role in the regulation of normal cell morpholo -gy,cell adhesion ,migration and angiogenesis .There is a certain relationship between abnormal expression of EphB receptor with a variety of tumor development .Visiblely,EphB receptor is important research direction in cancer studies,understanding the relationship between EphB receptor and tumor can better realize the progress of the tumor,and further to provide more scientific basis for cancer treatment and prevention .
7.Synthesization, expression, purification and activity assay of hIGF-1

AIM: To express the synthesized human insul in like growth factor I (hIGF-1) gene in E.coli with high expression level a nd explore the way to increase the efficiency of factor Xa cleavage. METHODS: The gene of hIGF-1 was designed and synthesized accordi n g to the preference of E.coli. A fusion protein with a recognized site of fa ctor Xa between CBD (cellulose binding domain) and hIGF-1 was expressed and puri fied by cellulose affinity chromatography. MTT method was used to assay the bioa ctivity of CBD-IGF fusion protein. hIGF-1 was released by factor Xa. In order to improve the sensitivity of fusion protein to factor Xa, the short flexible pept ide (Gly-Thr-Gly- Gly-Gly-Ser-Gly) was added before the recognized site of fac tor Xa. RESULTS: SDS-PAGE results indicated that the CBD-IGF fusion prot ein was expressed and purified . Biological assay results indicated CBD-IGF fusi on protein could promote the growth of NIH3T3 cell. The short flexible peptide (Gly-Thr-Gly-Gly-Gly-Ser-Gly), which was added before the recognized site of f actor Xa, improved the sensitivity of fusion protein to factor Xa. CONCLUSION: CBD-IGF fusion protein with bioactivite are expresse d and purified. The amio acid sequences changes between the site recognize of fa ctor Xa can help to improve the cleavage efficiency of Factor Xa.
8.Study on chemical constituents from stem of Photinia parvifolia

Object To study the chemical constituents from the stem of Photinia parvifolia (Pritz.) Schneid. Methods The compounds were isolated by silica gel column chromatography and their structures were elucidated by means of spectral analysis. Results Seven compounds were identified as lupeol (Ⅰ), ?-sitosterol (Ⅱ), betulinic acid (Ⅲ), pyracrenic acid (Ⅳ), epicatechin (Ⅴ), daucosterin (Ⅵ), pruasin (Ⅶ). Conclusion Compounds Ⅰ, Ⅲ-Ⅵ are isolated from the plants of Photinia Lindl. and compounds Ⅱ and Ⅶ are isolated from P. parvifolia for the first time. Compound Ⅳ shows anticancer effect.
10.Experimental study of the cytokeratin expression pattern of regenerated epithelia of dermal tissue flaps transferred for intraoral mucosal defect reconstruction

Objective To observe the changes in cytokeratin expression pattern of the regenerated epithelia of dermal tissue flaps transferred for intraoral mucosal reconstruction. Methods Mucosal defects were made on the right buccal mucosa of 12 healthy adult dogs, and then were reconstructed immediately by pedicled platysma myodermal flaps (PMDF). Changes in the expression patterns of CK10 and CK19 of the epithelia on the flaps were observed by immunohistochemical staining. Results Low level of CK19 expression was found in the basal layer and the layers superior to the basal layer of the regenerated epithelia at 4 week after transferring the dermal tissue flaps. The expression pattern of keratin in PMDF was identical to that of the normal skin 12 weeks later. Conclusion The regenerated epithelia remain the original structure without mucosal changes.
