1.Effects of curcumin combined with cisplatin on growth and lymphatic metastasis of cervical cancer xeno-grafts in nude mice

Objective To analyze the effects of curcumin combined with cisplatin on the growth and lymphatic metastasis of cervical cancer xenografts in nude mice.Methods The human cervical carcinoma Caski cells xenotransplanted tumor models were established.Inoculated mice were randomly divided into 4 groups according to random number table:control (normal saline 10 ml/kg),Cur (curcumin 100 mg/kg),Cis (cisplatin 3 mg/kg) and Cur + Cis group (100 mg/kg curcumin +3 mg/kg cisplatin).The tumor volume and anti-tumor rate were calculated.The mRNA levels of macrophage migration inhibitory factor (MIF) and vascular endothelial growth factor-C (VEGF-C) were analyzed by real-time fluorescent quantitative polymerase chain reaction (PCR).Results The inhibitory rates of Cur group,Cis group and Cur + Cis group were 40.8%,53.3% and 60.0%.After 15 days treatment,the tumor volumes of the control group,Cur group,Cis group and Cur + Cis group were (123.44 ± 35.62),(71.72 ± 28.36),(65.47 ± 18.32),(53.44 ± 10.79) mm3.The growth rates of tumor volume in Cur group,Cis group and Cur + Cis group were slower,Cur + Cis group could more effectively inhibit tumor volume growth.The difference among the four groups had statistically significance (F =16.890,P =0.000).Real-time fluorescent quantitative PCR detection showed that compared with the control group (1.000),the MIF mRNA expressions in Cur group,Cis group and Cur + Cis group were 0.322 ±0.094,0.154 ± 0.006 and 0.136 ± 0.007,VEGF-C mRNA expressions in the three groups were 0.312 ± 0.068,0.263 ± 0.072 and 0.221 ± 0.041.The differences among groups had statistically significance (F =220.279,P =0.000;F =143.250,P =0.000).MIF mRNA was positively related with VEGF-C mRNA (r =0.815,P =0.001).Conclusion Curcumin combined with cisplatin can inhibit the growth of Carski cell xenografts in nude mice.Through the down-regulating expression of MIF mRNA and VEGF-C mRNA,cisplatin and curcumin can inhibit the lymphatic metastasis of cervical cancer,and it may be one of the important mechanisms of its anti-tumor effects.
2.Establishment of subcutaneous and orthotopic transplatation tumor model of human esophageal cancer in nude mice

Objective To establish a subcutaneous and orthotopic transplatation tumor model of human esophageal cancer in nude mice .Methods Fresh tissues of human esophageal cancer were collected and transplanted into subcutaneous places of nude mice .The successfully transplanted primary generation tumor was passed down three times .Then tumor were implanted into the subcutaneous and esophageal submucous tissues of nude mice .The formation rate ,morphology ,invasion and metastasis of tumor were observed .Results The tumor formation rate of subcutaneous transplatation model was 60% ,it was low grade squamous cell carcinoma without lymph node and distant organ metastasis .The tumor formation rate of orthotopic transplatation model was 80% , the morphological characteristics were consistent with human original tumor .Lymph node and organ metastasis could be found ex-tensive .Conclusion We preliminary established effective subcutaneous and orthotopic transplatation tumor model of human esopha-geal cancer in nude mice .It might be an available model for further research of esophageal cancer .
3.Research in the treatment of hilar cholangiocarcinoma with hemihepatectomy in 50 cases

Objective To compare the clinical and follow-up results of different ways of operation for the treatment of patients with hilar cholangiocarcinoma ,and to explore its e key technology and related prognosis .Methods 31 out of 50 patients with biliary and partial liver resection(control group) ,19 patients with hemihepatectomy (observation group) .Radical resection rate and surviv-al rate were compared in the two groups .Results radical resection rate control group and observation group was 35 .48% (11/31) and 68 .42% (13/19) ,the difference was statistically significant (P=0 .014);the difference of 1 ,3 ,5 year survival rates between the two groups were statistically significant (P= 0 .03) ,observation group :85 .4% ,58 .6% ,32 .2% ,control group:86 .1% ,65 .2% , 20 .0% .Conclusion Radical resection is still the most important measures the patients for cure and long-term survival ,radical re-section rate ,survival rate ,and prognosis could be improved greatly by improving operation skills and experience accumulation ,com-bined with hepatectomy .
4.Expression and significance of estrogen receptor alpha in renal tissue of IgA nephropathy

Objective To investigate the expression and significance of estrogen receptor alpha in renal tissue of immunoglobulin A (IgA) nephropathy.Methods Fifty renal tissue samples including forty five cases of IgA nephropathy and five cases of normal expression were collected.The expression of estrogen receptor αt (ERα) was detected by immunohistochemistry method,and its relationship with clinical parameters and the degree of glomerular damage were analyzed.Results ERα was located in the glomerular and renal tubules.With the severity of the lesion,the expression of ERα in renal tissue of IgA nephropathy decreased gradually (P =0.001) and its expression was associated with estimated glomerular filtration rate (eGFR),serum creatinine (Scr),and pathologic grade of IgA nephropathy (r =0.876,-0.818,and -0.736,P < 0.05),The expression of ERα was significantly decreased in hypertensive patients with IgA nephropathy,and the difference was statistically significant compared with that in non-hypertensive group (P =0.011).Conclusions The expression of ERα in renal tissue of IgA nephropathy was significantly decreased and there was a correlation between the degree of renal tissue damage,suggesting that ERα might play a role in the development of renal disease.
5.Change of bacteria and enzymes in the drainage fluid in patients with intestinal fistulas

ObjectiveTo investigate the amount of bacteria and the expression of amylase and lipases in the drainage fluid in patients with intestinal fistulas with time courses.MethodsThe samples were collected from 16 patients with high intestinal fistulas from July 1998 to January 2008.The amounts of bacteria from the drainage fluid were measured 0,2 and 4 hours after taking out from the patients.At the respective time points,the intestinal juices were also collected to measure the amylase and lipase expressions.After reinfusion of succus entericus,thelevels of albumin,prealbumin,transferring,and fibronectin were measured at 0,7,and 14 days,ResultsThere was no significant increase of bacteria in the drainage fluid within 4 hours ( F(0,2) =18 812.50,P > 0.05 ; F(0,4) =387 625.00,P > 0.05).and there was no change in the expressions of amylase ( F(0,2) =190.60,P > 0.05 ;F(0,4) =631.75,P>0.05) and lipase within 4 hours (F(0,2) =204.10,P>0.05; F(0,4) =1080.05,P>0.05).After succus entericus reinfusion,the fibronectin (F(0,14) =74.24,P < 0.01 ; F(7,14) =59.78,P <0.01),transferring (F(0,14) =0.46,P < 0.01 ; F(7,14) =0.39,P < 0.05 ),and prealbumin ( F(0,14) =54.37,P < 0.05) were increased significantly.ConclusionsBacteria and enzymes do not increase in the drainage fluid within 4 hours in patients with intestinal fistulas.Therefore,it is safe and effective to reinfuse succus entericus.

8.Surgical treatment for pulmonary carcinoid tumors: a single-center analysis of 62 patients

ObjectiveTo evaluate prognostic factors and the surgical results of pulmonary carcinoid tumors.Methods We retrospectively reviewed the medical records of 62 patients who were diagnosed as pulmonary carcinoid tumors between January 2000 and October 2010 at Department of Thoracic Surgery,Shanghai Chest Hospital.The following information was available for each of the 62 patients:age,sex,pathological type,and TNM stage.ResultsThere were no operative death.The 3-year and 5-year survival rates after surgery were 92.1% and 77.8%,respectively.Of the 62 patients,42 were diagnosed as typical carcinoid tumor,and among them,4 patients (8.3%) had lymph node metastases.Their 3-year and 5-year survival rates were 97.8% and 94.7%,respectively.The remaining 20 patients were diagnosed as atypical carcinoid tumor,and among them,6 patients (37.5%) had lymph node metastases.Their 3-year and 5-year survival rates were 84.4% and 58.8%,which were statistically significant compared with typical carcinoid tumor( P =0.0047 ).There was significant difference in survival rate between the patients with lymph node metastases and the patients without lymph node metastases (P =0.0048).CondusionThe main risk factors affecting survival rate of those patients who were diagnosed as pulmonary carcinoid tumors were pathological types and lymph node metastases.
9.Effect of P38MAPK pathway on autophagy in rat hippocampus following cerebral ischemic postcoditioning

Objective To explore the mechanism of ischemic postconditioning in relieving cerebral ischemia reperfusion (IR) by regulating autophagy through P38MAPK pathway.Methods Cerebral ischemia reperfusion model was established by using modified Pulsinelli four-vessel occlusion (4-VO).Totally 128 male SD rats were divided into 4 groups randomly:control group (sham),cerebral ischemia reperfusion model group (CIR),cerebral ischemic postconditioning group (CIP),and cerebral ischemic postconditioning + P38MAPK inhibitor group (SB203580 group).Each group was subdivided into four time points:6 h,24 h,48 h,and 72 h.The morphological changes of the hippocampus CA1 area neurons at each time point and the number of surviving nerve cells were detected with HE staining.The expression of the hippocampus CA1 area phosphorylated P38MAPK and the autophagy-related genes of Beclin-1 and LC3-Ⅱ were detected with immunohistochemistry.The protein content of the hippocampus phosphorylated P38MAPK and autophagy-related genes of Beclin-1 and LC3-Ⅱ were detected with Western blotting.Results Compared with those in sham group,the damage of rats' hippocampal neuron structure and the survival rate of neurons at each time point decreased in CIR group,the expressions of p-P38MAPK,LC3-Ⅱ and Beclin-1 increased.Compared with those in CIR group,in CIP and SB203580 groups the structure of rats hippocampal neurons was improved,the survival rate of neurons increased,the expression of p-P38MAPK decreased and the expressions of LC3-Ⅱ and Beclin-1 increased at each time point.Compared with CIP group,SB203580 grouphad improved structure of rats' hippocampal neurons,increased survival rate of neurons,decreased expression of p-P38MAPK,and increased expressions of LC3-Ⅱ and Beclin-1 at each time point.Conclusion Cerebral ischemic postconditioning through inhibiting P38MAPK pathway can regulate autophagy and exert its nerve-protective effect.

Objective To observe the efficacy of combinated inhaled glucocorticosteroid and long- ?2 - agonist for asthmatic children.
