1.Preparation nucleic acid vaccine of brucella melitensis and its immune effect

Abstract Objective:To construct bmcella melitensis Br.melitensis genomic DNA expression library and study its immune effect.Meth-ods: Hind Ⅲ digested Br.melitensis genomic DNA fragments were cloned into pSV-?-Gal plasmds.The recombinant plasmids were transformedinto JM109 host bacteria. After large-scale isolation, the recombinant plasmids were inoculated into the quadriceps muscle of mice. In the 9thday after third boost immunization .agglutination antibodies against Br. melitensis and lymphocyte mitogenic ability of mice were determined. Re-sults : Expression library nucleic acid vaccine can induce the production of agglutination antibodies against Br. melitensis and stimulate the lym-phocyte porliferation primed by ConA. Conclusion:The genomic DNA expression library may provide a new vaccine against Br. melitensis infec-tion.
2.Evaluation of the regeneration of chemically extracted acellular facial nerve allograft for the whole facial nerve defect in rat with multiple retrograde tracing

[Objective]To evaluate the axonal transport function of the regenerate nerve of buccal branch,marginal mandibular branch and cervical branch after chemically extracted acellular facial nerve allograft to the whole facial nerve defect in rat using fluoro-gold(FG),quantum dot(QD) and fast blue(FB).[Method]The extracranial section of facial nerve and branches were dissected under light microscope,and 10-mm-long segments of facial nerve gap were made on the left side.The defect was repaired by acellular facial nerve allografts with epineural suture method.Two months later,fluoro-gold(FG),quantum dot(QD) and fast blue(FB) were injected into distal bridged side of buccal branch,marginal mandibular branch and cervical branch.After three days,the slices of brain stem were collected and the facial motoneuron was observed under fluorescences microscope.[Result]There were three kinds of different fluorescences in the frozen sections of the facial nerve originated from the brainstem nuclei under the fluorescence microscope.Fluoro-gold(FG),quantum dot(QD) and fast blue(FB) labeled neurons were shown yellow,red and blue colors,respectively.[Conclusion]According to estimation of axonal branching by triple retrograde labeling,it was found that the regenerate facial nerve repaired the gap,and nerve continuity obtained restoration.The evaluation method of this study is simple,reliable and easy.It is an ideal evaluation method.
3.Effects of intrinsic ganglionated plexi on the susceptibility to atrial fibrillation in canines induced by rapid atrial pacing

Objective To study the effects of intrinsic ganglionated plexi on the susceptibility to atrial fibrillation(AF)in canines induced by rapid atrial pacing.Methods Atrial efficient refractory period(AERP)and the dispersion of AERP were obtained in 13 healthy mongrel dogs before paced at 600 bpm.The fat pads containing the ganglionated plexi located on the junction of right superior pulmonary vein and atrium in 7 dogs were excided(denervation group),while the fat pad in the other 6 dogs were kept(control group).The time for the emerge of sustained AF was recorded.Atrial efficient refractory period(AERP)and the dispersion of AERP were also determined.Results There was statistical difference in the time for the emergence of sustained AF.In denervation group,it took more time than the control group(120?67 h vs 80?52 h,P
4.Application of ureteral grasping forceps in ureteroscopic operations

Objective To study the clinical application of ureteral grasping forceps in ureteroscopic operations. Methods A total of 74 cases treated under ureteroscope by using ureteral grasping forceps from April 2001 to July 2003 in this hospital was retrospectively analyzed. There were 25 cases of polyp resection and pneumatic lithotrity, 21 cases of rigid guidance through ureteral retortion, 19 cases of ureteral stricture dilation and ureteroscopic lithotrity, 3 cases of whole ureteral membranous obstruction, 3 cases of ascending shift of double “J” tube, 2 cases of intraoperative biopsy,4 cases of removal of double “J” tube with urethral stricture,and 2 cases of pneumatic lithotrity. Results The procedures were successfully accomplished in 69 cases.A conversion to open operation was required because of malfunction of ureteral grasping forceps in 1 case, and ureteral perforation occurred in 4 cases. Conclusions Ureteral grasping forceps is indispensable in ureteroscopic operations, which should be skillfully operated.
5.Paracentesis using jugular vein indwelling catheter for simple renal cysts

Objective To investigate the efficacy of paracentesis using jugular vein indwelling catheter (Angiocath) in the treatment of simple renal cysts. Methods We retrospectively analyzed clinical data of 61 patients with simple renal cyst treated by Angiocath paracentesis combined with sclerosing agent injection under the guidance of ultrasonography. Results Fifty-two patients were cured on one session, while a recurrence was seen in 9 patients, among which 6 patients were cured by a re-paracentesis and 3 frequently recurrent patients were cured by the unroofing operation. No complications occurred except for 3 cases of mild transient hematuria. Conclusions Paracentesis using Angiocath in the treatment of simple renal cysts is minimally invasive, safe, effective and economical.
6.Effects of some drugs for resolving phlegm on blood rheological property in rats

In order to study on the theory of " phlegm and blood stasis having same source" and effects of drugs for resolving phlegm on blood rheology, effects of Ban Xia, Gua Lou, Zhe Bei Mu and Shi Chang Pu on whole blood viscosity, erythrocyte deformation index and plasma fibrinogen content in normal rats. Results indicated that the drugs.for resolving phlegm had functions of decreasing whole blood viscosity, inhibiting hemoagglutination (except Gua Lou) and increasing capability of erythrocyte deformation; at high shearing rate, Shi Chang Pu and Zhe Bei Mu could slightly increase whole blood viscosity: Shi Chang Pu increased slightly plasma fibrinogen content. It is suggested that resolving phlegm also can removing blood stasis, and the mechanism of the drug for resolving phlegm in decreasing blood viscosity is related with decrease of blood lipids and anti - peroxidation of lipids.
7.The protective effect of tumor necrosis factor-?antibody on lung injury after cardiopulmonary bypass

Objective: To study the protective effect of anti-tumor necrosis factor-?antibody (TNF-? Ab) on lung injury after cardiopulmonary bypass(CPB). Methods: 20 patients with valve diseases underwent valve replacement under CPB. The patients were randomly divided into two groups: control group and TNF-? Ab group. In the TNF-? Ab group, human TNF-? Ab was dropped into the intracheal tube before operation and just after releasing the aortic clamp, respectively. Lung dynamic compliance, blood neutrophils count and TNF-? from the right and left atrium were determined perioperatively. Results: The dynamic compliance of the lung in TNF-? Ab group was higher than that in control group. TNF-? Ab can reduce releasing of TNF-? and restrain leukocyte accumulation in the lung. Conclusion: TNF-? Ab has markedly protective effect on lung injury after CPB.
8.Evaluation of Target Controlled Infusion of Midazolam for Sedation During Combined Spinal and Epidural Anesthesia

Objective To evaluate the performance of the target controlled infusion(TCI) system with midazolam during combined spinal and epidural anesthesia. Methods Twenty female patients scheduled for selective lower abdominal or pelvic surgery under combined spinal and epidural anesthesia were enrolled in this study. They use combined spinal and epidural anesthesia with target controlled infusion of midazolam sedation. Midazolam plasma concentration was set at 100ng/ml. Blood pressure,heart rate, pulse oxygen saturation were monitored during anesthesia and operation. We use BIS as a pharmacodynamic value of midazolam sedation. Blood samples were taken from radial arterial for analysis of plasma midazolam concentration during infusion. Midazolam plasma concentration were tested by high performance liquid chromatography. Results MDPE of target controlled infusion with midazolam with Burher parameters was 38.7%, MDAPE was 38.7%,and wobble is 24.9% in Chinese female patients. Conclusions Burher parameters of midazolam target controlled infusion system should be correct, then can be used for our country female patients conscious sedation accurately.
9.Current study on diagnostic values of various inflammatory cytokines in postoperative infection related to orthopedic implants

In recent years,along with social economic development and population aging,more and more fracture patients have received internal fixation and artificial joint replacement.Postoperative infection related to orthopedic implants is a catastrophic complication which imposes serious psychological,physiological and economic burdens on the patients.Therefore,it is necessary to be able to make an early diagnosis of the infection.However,diagnosis of implant-related infection is always a clinical problem for orthopedists.Currently,it is thought to be a valuable way to judge orthopedic implant infection by monitoring inflammatory cytokines.This article will offer an overview on the progress in research of diagnosis values of different inflammatory cytokines in postoperative infection related to orthopedic implants.
