1.ANALYSIS OF COMPREHENSIVE BENEFIT ON INSTALLATION OBSTRUCTED SNAILS IN LARGE IRRIGATION CANALS FOR PREVENTING SNAILS SPREADING

Objective To evaluate the comprehensive benefit about the installation obstructed snails in large irrigation canals for preventing snails spreading in lake-swamp areas of schistosomiasis.Methods The installation obstructed snails was set up in upper reaches of main canal Shilong, Tianmen City Data of snails and schistosomiasis prevalance were collected and analysed. The comprehesive benefit was analysed with the cost-benefit methods of economic hygienics. Results After the installation obstructed snails was set up the snails areas and human infection rate decreased markedly. New patients with acute and advanced schistosomiasis were not found in the irrigation areas. The ratio of comprehesive cost-benefit was 1∶9.84,the ratio of net benefit was 8.84∶1. Conclusion Setting up the installation obstracted snails to cut off the way of snails spreading was the effective measure of control schistosomiasis in lake-swamp areas.

A s an im portant part of epigenetic m arker, D N A m ethylation involves in the gene regulation and attracts a w ide spread attention in biological auxology, geratology and oncology fields. In forensic science, because of the relative stable, heritable, abundant, and age-related characteristics, D N A m ethyla-tion is considered to be a useful com plem ent to the classic genetic m arkers for age-prediction, tissue-identification, and m onozygotic tw ins' discrim ination. V arious m ethods for D N A m ethylation detection have been validated based on m ethylation sensitive restriction endonuclease, bisulfite m odification and m ethylation-C pG binding protein. In recent years, it is reported that the third generation sequencing m ethod can be used to detect D N A m ethylation. T his paper aim s to m ake a review on the detection m ethod of D N A m ethylation and its applications in forensic science.
3.Codon optimization and eukaryotic expression analysis of the analgesic peptide gene BmK AngM1 from Buthus martensii Karsch.

Codon bias is an important factor which influences heterologous gene expression. Optimizing codon sequence could improve expression level of heterologous gene. In order to improve the expression level of BmK AngM1 gene encoding the analgesic peptide from Buthus martensii Karsch in Pichia pastoris, the codon-optimized BmK AngM1 gene according to its cDNA sequence and the preference codon usage of P. pastoris were cloned into expression vector pPIC9K and then transformed into P. pastoris. The expersion of recombinant BmK AngM1 (rBmK AngM1) was inducced by methanol in the medium, and the expression level of the optimized BmK AngM1 gene was 3.7 times of the native one. These results suggested that the expression of BmK AngM1 in P. pastoris could be successfully improved by codon optimization.
4.Drug-coated balloon vs standard angioplasty balloon in the treatment of postoperative in-stent restenosis in patients with arteriosclerosis obliterans of the lower extremity

Objective:To compare drug-coated balloon (DCB) and standard angioplasty balloon (SAB) in the treatment of postoperative in-stent restenosis (ISR) in patients with arteriosclerosis obliterans (ASO) of the lower extremity.Methods:From Jan 2017 to Dec 2018, 43 ISR patients after percutaneous transluminal angioplasty for ASO of the lower extremity at our hospital were enrolled.Patients were divided into 2 groups with 18 patients treated by DCB and 25 by SAB. The patients were followed up for 6~12 months.Results:There was no significant difference in the incidence of complications between DCB group and SAB group ( P>0.05).Compared with that in SAB group, the plasma level of ET-1 in DCB group was lower while NO was higher at 6, 24 h and 2 weeks after surgery ( P<0.05), there was no significant difference in P-selectin ( P>0.05). The ABI values in both groups increased, and that in DCB group were higher than SAB group at 6 and 12 months after surgery ( P<0.05). The lumen loss in DCB group at 6 and 12 months after surgery was significantly lower ( P<0.05). At 6 and 12 months, the primary patency of target lesions in the DCB group was 100.00% and 88.89%, which was higher than the 72.00% and 52.00% in the SAB group ( P<0.05); the CD-TLR rate in the DCB group was 11.11%, which was lower than 48.00% in the SAB group ( P<0.05). Conclusion:DCB comes with lower postoperative ISR in ASO patients of the lower extremity.
5.Study on the preparation method of capillary electrophoresis sieving medium based on domestic genetic analyzer

Objective To prepare a capillary electrophoresis sieving medium and apply it in GA118-16A genetic analyzer. Methods The white solid polyacrylamide (LPA) was prepared by polymerization and lyophilized. Through the swelling of the sol buffer, the sieving medium was obtained. The sieving medium was evaluated by 1) characterizing the parameters, including molecular weight, structure and viscosity, 2) applying in the GA118-16A genetic analyzer, including the spatial calibration, the spectral calibration and the STR analysis.. Results The prepared sieving medium Mw 1.8 x 105Da, Mn 1.2 x 105 Da, is of correct structure and high purity. The polydispersity was 1.5The spatial calibration and spectral calibration files can be established successfully in GA118-16A genetic analyzer, and the sieving medium can effectively separate the DNA fragments with 1bp difference. The STR profile is of sharp peaks, no impurity peaks, no tail, and no peak loss. Conclusion The sieving medium prepared by the method can be applied to domestic genetic analyzer such as GA118-16A.
6.Clinical and electrophysiological analysis of limb-girdle muscular dystrophy:A geneological report

Objective To analyze the clinical and electrophysiological features of one geneology with limbgirdle muscular dystrophy(LGMD). Methods Twenty-seven members of one family with limb-girdle muscular dystrophy(LGMD)were investigated.Fourteen of them were examined with electromyography(EMG)and their motor conduction velocities(MCV)and sensory conduction velocities(SCV)were measured.Among them,10 had no clinical manifestations,while 4 demonstrated symptoms and signs of LGMD. Results Three of the 4 patients had suffered from LGMD when young.They demonstrated the typical clinical features,including the progressive muscle weakness in the upper and lower extremities,positive Gower signs,duck gait,muscle atrophy distributed tO the proximal extremity,and no gastrocnemius hypertrophy.One subject presented atypical characteristics.The MCVs and SCVs of the 4 patients were normal,but neuropathic manifestations were found in the EMGS of 3 of them.and mixed neuropathic and myopathic manifestations were found in the EMG of the other.Conclusion LGMD patients in the same family can vary in their clinical characteristics.The longer the duration,the more severe the clinical features.Electrophysiological examination can reveal normal MCV and SCV but abnormal elctromyography.
7.Double scanning method for computer aided design and computer aided manufacture all-ceramic restorations.

OBJECTIVETo introduce the double scanning for producing computer aided design and computer aided manufacture (CAD-CAM) zirconia crowns, and to compare Procera with Everest CAD-CAM system by observing clinical effects of the crowns.
METHODSForty CAD-CAM zirconia crowns (10 crowns by Procera, and 30 crowns by Everest) were evaluated, and the examination criteria included: marginal fit, aesthetics, gingival health, retention, and fracture. All of the zirconia copings were produced using double scanning. Procera system and Everest system use contact scanner and optical scanner respectively.
RESULTSThe observation time for the crowns ranged from 6 to 24 months. The results of clinic evaluation were acceptable in all restorations.
CONCLUSIONSDouble scanning could be used to produce complicated zirconia copings by CAD-CAM in order to get proper symmetrical porcelain length. It was easier to make complicated coping with contact scanner than with optical scanner.

9.Relationship between leptin gene of adipose tissues and nonalcoholic fatty liver disease.

OBJECTIVETo investigate leptin mRNA expressions in subcutaneous (SC) and omental (OM) adipose tissues of patients with nonalcoholic fatty liver disease (NAFLD), and their relationships with insulin resistance (IR), blood leptin, blood triglyceride, total blood cholesterol, blood glucose, body weight index and waist-hip ratio.
METHODSSC and OM adipose tissues were obtained from 10 obese and 11 nonobese NAFLD patients and from 11 obese and 13 nonobese patients without NAFLD, who served as controls. Leptin mRNA expression levels in the subcutaneous and omental adipose tissues were measured using SYBR Green I quantitative real-time PCR. IR was estimated using homeostasis assessment (HOMA). The levels of plasma leptin and insulin were measured using ELISA.
RESULTSThe relative mRNA expression of leptin, HOMA-IR and blood leptin levels in NAFLD differed significantly from those of the controls (P < 0.05). The leptin/GAPDH ratio of the obese and nonobese NAFLD and control cases were 1.32 +/- 0.12, 0.99 +/- 0.05, 1.10 +/- 0.09, 0.87 +/- 0.13 respectively. The expression levels of SC and OM adipose leptin mRNA in NAFLD patients were positively correlated with HOMA-IR (r=0.72, P < 0.05), blood leptin (r=0.69, P < 0.05), blood triglyceride (r=0.32, P < 0.05), body weight index (r=0.57, P < 0.05) and waist-hip ratio (r=0.50, P <0.05).
CONCLUSIONThe primary reason for high levels of blood leptin is high leptin mRNA expression in adipose tissues; in both obese and nonobese patients with NAFLD; high levels of blood leptin and the leptin mRNA expression in adipose tissues and IR exist. These findings suggest that leptin resistance exists in patients with NAFLD and leptin resistance is positively correlated with NAFLD, the same as in insulin resistance.

10.Analysis of qualifications of medical and health institutions and certified doctors for providing occupational disease diagnosis in China.

OBJECTIVETo investigate the qualifications and current situations of the medical and health institutions and certified doctors for providing occupational disease diagnosis in China and to provide a reference for developing relevant policies.
METHODSWork reports and questionnaires survey were used to investigate the qualifications of all medical and health institutions and certified doctors for providing occupational disease diagnosis in China and their acceptance and diagnosis of occupational disease cases from 2006 to 2010. The rate for the work reports was 100%, and the response rate for the questionnaires was 71.0%.
RESULTSBy the end of 2010, in the 31 provincial-level regions (excluding Hong Kong, Macao, and Taiwan) in China, there had been 503 medical and health institutions which were qualified for providing occupational disease diagnosis, including 207 centers for disease control and prevention, accounting for 41.2%, 145 general hospitals, accounting for 28.8%, 69 enterprise-owned hospitals, accounting for 13.7%, and 64 institutes or centers for occupational disease prevention and control, accounting for 12.7%; 4986 certified doctors got the qualification for providing occupational disease diagnosis, with 9.4 certified doctors on average in each institution, and there was 0.65 certified doctor per 100 000 employees. In addition, 16.5% of the institutions got all the qualifications for diagnosing 9 occupational diseases, and 17.1% of the institutions got the qualification for diagnosing one occupational disease. Each certified doctor accepted diagnosis of 16.8 cases of occupational diseases on average every year.
CONCLUSIONA national occupational disease diagnosis network has been established in China, but the imbalance in regional distribution and specialty programs still exists among the qualified medical and health institutions and certified doctors. It is essential to further strengthen the development of regional qualified medical and health institutions and training of qualified doctors.
