1.Evaluation of enzyme-linked immunosorbent assay for diagnosing bullous dermatosis

Objective To evaluate the clinical application and significance of enzyme-linked immunosorbent assay(ELISA)for serologically diagnosing bullous dermatosis.Methods Serum Dsg1,Dsg3 and BP180 expressions were detected in 34 cases of pem-phigus,32 cases of bullous pemphigoid and 33 cases of non-bullous dermatosis by ELISA.Results The sensitivity and specificity of ELISA for detecting the bullous dermatosis antibody were 87.88% and 93.94% respectively.Conclusion The ELISA method has the diagnostic value on bullous dermatosis and the important significance in the disease classification.
2.Application of endoscopy combined with 125I seed implantation in the pancreatic cancer surgery in elderly patients

Objective To observe the application of endoscopy combined with 125I seed implantation in the pancreatic cancer palliative surgery in elderly patients.Methods A total of 78 elderly patients with pancreatic cancer who can not be treated with radical operation were divided into observation group and control group (n=39 each group).All patients were treated with stent intervention.Patients in the ation group were treated with 125I seed implantation guided by CT or ultrasonangraphy additionally.The clinical symptoms were compared between groups.The survival time and complications of the patients were recorded during the follow-up.Results The clinical symptoms of patients in both two groups were greatly improved (x2 =0.06,P>0.05).Compared with the control group,the pain remission was better and the incidence of pain caused by pancreatic cancer was lower in the observation group (46.2% vs.76.9%,x2 =7.80,P<0.05).There was a significant difference in the average of survival time between the observation group and control group [(13.7±3.4) months vs.(8.4±3.8) months,t=3.73,P<0.01],but no statistical difference was found in the long-term survival rate between groups.There was no significant difference in the incidence of complications between the two groups (10.3% vs.7.7%,x2=0.15,P>0.05).Conclusions Endoscopy combined with 125Ⅰ seed implantation is an effective treatment for elderly patients with advanced pancreatic cancer,which can effectively relieve the clinical symptoms,proiong the survival time,reduce the pain and complications.
3.The clinical utility of virtual non-contrast in the diagnosis of solitary pulmonary nodule

Objective To investigate the clinical utility of dual-energy virtual non-contrast CT(VNCT)of dual source CT in the di-agnosis and differential diagnosis of solitary pulmonary nodule(SPN).Methods Thirty-six patients with suspected SPN underwent chest plain single energy CT and dual-phase contrast enhanced CT (DECT)(25 and 90 s).The images of dual energy were sent to a commercial workstation for VNC image generation.CT values of SPN on VNC and true non-contrast(TNC),as well as signal-to-noise ratio(SNR)and contrast-to-noise ratio(CNR)were compared.The accuracy of calcification detection was compared according to the diameter and density of the calcification.The TNC images were used as the reference.Results CT values,SNR and CNR of VNC had no statistical difference among TNC and VNC at 25 s and 90 s(P >0.05).DECT VNC(5mm slice)could accurate detected calci-fication(diameter>2 mm,CT value>1 50 HU)in SPN.The 2 mm slice performed significantly better than 5 mm slice VNC images. Conclusion The VNC could provide consistent diagnostic information with TNC.The thin slice DECT VNC images can be used in clinical practice to replace TNC without losing small calcification in SPN,which has potential to reduce the patient radiation dose.
4.Isolation and structural elucidation of flavones from flower of Paulownia tomentosa

Object To isolate and identify the flavones from the flower of Paulownia tomentosa (Thunb.) Steud., which has been used as traditonal herb medicine in the treatment of some diseases. Methods The flavones were isolated by column chromatography and their structures were elucidated through spectroscopic analysis (NMR). Results Five flavones were isolated and identified as 5, 4′-dihydroxy-7, 3′-dimenthoxyflavanone (Ⅰ), 5-hydroxy-7, 3′, 4′-trimenthoxyflavanone (Ⅱ), diplacone (Ⅲ), mimulone (Ⅳ) and apigenin (Ⅴ). Conclusion All these compounds are obtained from the flower for the first time and the flavones should be as bioactive constituents.

Objective: To study the pharmacological action and clinical curative effect of Linglongqingre microenema. Methods: The actions of relieving fever and calming hypnotism. anti convulsions of Linglongqingre Microenema on rabbit, mouse and rat were observed. Also children's fever illess was treated to judge preliminary curative effect. Results: Linglongqingre Microenema has obvious action of relieving fever, more effective than Chaihu injection, as same as Antipyrine (80mg/kg) on subcutaneous injection. It was used to treat children's feverillness on 38 cases, markedly effective rate 60.53%, total effective rate 86.84%. Among them body temperature of one third case fell over 1.5?C in two hours. Conclusion: Linglongqingre microenema is a new quick effective pharmaceutical preparation which is suitable for emergency treatment of traditional Chinese medicine

Objective To study the influence of carboxymethyl-chitosan(CM-chitosan)on chondro- cyte apoptosis induced by recombinant human interleukin-1?(rhIL-1?)and explore its mechanism.Methods Rabbit chondrocytes were isolated and cultured.Chondrocytes were pretreated with different concentrations of CM-chitosan for 1 h,then 10 ng/ml IL-1?were added into the culture medium.After 24 h,the apoptotic rates of chondrocytes were measured by flow cytometry with AnnexinⅤ-FITC and PI staining.The morphology of nuclei was observed by fluorescent microscopy with Hoechst 33342 staining.The mitochondrial membrane po- tentials were tested by confocal laser scanning microsocopy with Rhodamine-123 and ATP contents were mea- sured by luciferase reaction.Results CM-chitosan could inhibit chondrocyte apoptosis and restore the func- tion of mitochondria induced by IL-113 in a dose-dependent manner.Conclusion CM-ehitosan can inhibit chondrocyte apoptosis by protecting mitochondrial function.
7.Effects of ABO blood group factors on erythrocyte suspension transfusion reactions

Objective To investigate the effects of ABO blood group factors on erythrocyte suspension ( RCS) transfusion reactions in patients. Methods TestⅠA total of 12 600 patients in whom RCS was transfused during operation at the department of anesthesiology of 11 hospitals of Inner Mongolia from January 2006 to January 2014 were selected. The occurrence of transfusion reactions ( fever [ an increase in body temperature>1 ℃ than that before transfusion] , allergy, hemolysis) was recorded in the patients. Test Ⅱ A total of 120 RCS?transfused patients of both sexes, aged 18-55 yr, weighing 45-75 kg, of American Society of Anesthesiologists physical statusⅠ or Ⅱ, who underwent surgical operation, were divided into 4 groups ( n=30 each) according to the blood group: blood group A group ( group A) , blood group B group ( group B ) , blood group O group ( group O ) and blood group AB group ( group AB) . The standard for RCS transfusion was defined as hemoglobin ( Hb) <7 g∕L, and Hb was maintained>10 g∕L. Before induction of anesthesia ( T1 ) , before blood transfusion ( T2 ) , and at 5 min, and 1, 6 and 24 h after blood transfusion ( T3?6 ) , blood samples were collected from the central vein for determination of the plasma tumor necrosis factor?alpha ( TNF?α) , interleukin?4 ( IL?4 ) and IL?10 concentrations by enzyme?linked immunosorbent assay. Results Among the 12 600 RCS?transfused patients, 216 cases developed transfusion reactions, and the incidence of transfusion reactions was 1.714%. For the patients of different blood groups, the incidence of transfusion reactions from the high to the low was blood group B, blood group AB, blood group A, and blood group O in turn ( P<0. 05 or 0.01) . Compared with group B, the plasma TNF?α and IL?10 concentrations were significantly decreased, and the plasma IL?4 concentrations were increased at T3?T6 in the other three groups ( P<0.05) . Compared with group AB, the plasma TNF?α and IL?10 concentrations were significantly decreased, and the plasma IL?4 concentrations were increased at T3?T6 in A and O groups (P<0.05). Compared with group O, the plasma TNF?α and IL?10 concentrations were significantly decreased, and the plasma IL?4 concentrations were increased at T3?T6 in group A ( P<0. 05 ) . Conclusion ABO blood group factors affect RCS transfusion reactions in the patients, and the incidence of transfusion reactions from the high to the low is blood group B, blood group AB, blood group A, and blood group O in turn.

Whennearinfraredspectroscopyisappliedtoon-linemonitoringandcontroloftobaccoflavors,the variation in temperature can severely deteriorate the predictive performance of near infrared spectroscopic calibration models and results in a significant increases of the root mean square error value for the main constituents in syrup samples from 2. 4% to 29. 0%. In this paper, near infrared spectroscopy has been incorporated with an advanced calibration transfer method-loading space standardization to effectively eliminate the deteriorate effects of temperature variation on quantitative results and finally realize the fast and accurate on-line quantitative monitoring and control of tobacco flavors. The root mean square error value for the main constituents in syrup samples is successfully retained at a satisfying low level of 3 . 8%. The results of this paper will provide technical support for the preparation, preservation and use of tobacco flavors, and realize on-line process quality control of cigarettes.

Objective To evaluate the role of autophagy in hydrogen-induced inhibition of apoptosis in hippocampal neurons in a rat model of orthotopic liver transplantation (OLT).Methods Fifty-six pathogen-free healthy adult male Sprague-Dawley rats,aged 8-10 weeks,weighing 220-250 g,were used in the study.Thirty-two rats were selected and assigned into 4 groups (n =8 each) using a random number table:sham operation group (group S),OLT group,hydrogen-rich saline group (group HS) and chloroquine group (group CQ).The other 24 rats severed as the donors.In group S,laparotomy was performed,and the related blood vessels were isolated.The model of OLT was established in OLT,HS and CQ groups.In group OLT,normal saline 6 ml/kg was slowly injected via the inferior vena cava at 5 min before anhepatic phase.In group HS,hydrogen-rich saline 6 ml/kg was slowly injected via the inferior vena cava at 5 min before anhepatic phase.In group CQ,autophagy inhibitor chloroquine 60 mg/kg was injected intraperitoneally at 1 h before establishment of the model,and the other treatments were similar to those previously described in group HS.At 6 h of reperfusion,the rats were sacrificed and hippocampi were isolated for determination of malondialdehyde (MDA) content and superoxide dismutase (SOD) activity,for pathological examination (with light microscope),and for detection of cell apoptosis (by TUNEL staining) and expression of autophagy-and apoptosis-related proteins caspase-3,cytochrome c (Cyt c),microtubule-associated protein 1 light chain 3 Ⅱ (LC3 Ⅱ),Beclin-1 and p53 in hippocampal tissues (by Western blot analysis).Apoptosis index (AI) was calculated.Results Compared with group S,the MDA content and AI were significantly increased,the SOD activity was decreased,and the expression of caspase-3,Cyt c,LC3 Ⅱ,Beclin-1 and p53 was up-regulated in OLT,HS and CQ groups (P<0.05).Compared with group OLT,the MDA content and AI were significantly decreased,the SOD activity was increased,the expression of caspase-3 and Cyt c was down-regulated,and the expression of LC3 Ⅱ,Beclin-1 and p53 was up-regulated in group HS (P<0.05).Compared with group HS,the MDA content and AI were significantly increased,the SOD activity was decreased,and the expression of caspase-3 and Cyt c was up-regulated,and the expression of LC3 Ⅱ,Beclin-1 and p53 was down-regulated in group CQ (P<0.05).Conclusion The mechanism by which hydrogen inhibits apoptosis in hippocampal neurons is related to promotion of autophagy in a rat model of OLT.
