1. Establishment of cell suspension culture system and its alkaloid accumulation of Lycoris radiate

Objective: To investigate the culture conditions for the establishment of cell suspension culture system for Lycoris radiata callus and its alkaloid accumulation. Methods: Using the bulbs of L. radiata as explants for callus induction, calli with good quality were selected for suspension cultivation. The factors influencing the establishment of L. radiata cell suspension culture system, such as hormone, inoculum amount, inositol, and conditional culture, were investigated. The alkaloid content in cell suspension culture system of L. radiata was determined by HPLC. Results: The optimal medium for the cell suspension culture was MS+0.5 mg/L 2, 4-D+0.1 mg/L KT+300 mg/L inositol and the inoculum size was 15%. The lag phase of the cell suspension culture system could be evidently shortened by the medium of logarithmic growth. The galanthamine contents in the cell suspension culture system and the medium were 4.18 and 0.69 times of those in the bulbs of L. radiata, respectively. Conclusion: The cell suspension culture system of L. radiata is established and it is a feasible way to gain galanthamine.
2.Investigation of the cognition and self management about osteoporosis and quality of life in elderly osteoporosis patients

Objective To investigate the cognition, self-management and quality of life of osteoporosis in elderly patients, and to provide the basic theory for health education in elderly patients with osteoporosis.Methods 565 elderly patients with osteoporosis in our hospital from May 2012 to Mar.2015 were enrolled.The cognition, health beliefs, self-efficacy and quality of life were investigated by questionnaires and analyzed by Microsoft Office Excel 2007.Results 565 osteoporosis patients, the symptom score was (15.6±2.0), and the score was lower in sporting knowledge (4.3 ± 1.8) than in calcium knowledge (5.7 ± 1.5) (t=2.784, P=0.011).Health beliefs score was (139.0±9.6), containing exercise benefits score of (21.5±3.1), exercise barriers score of (13.8±3.9), susceptible score of (19.4±4.2), serious score of (18.8±4.4), health motivation score of (25.0±3.0), calcium intake benefits score of (20.7±3.5) and calcium intake disorder score of (19.8 ± 4.0).Self efficacy score was (63.7 ± 19.4), containing exercise efficacy score of (58.6 ±18.7) and calcium efficacy score of (69.2± 18.5).Quality of life score included the general health (16.5± 2.9 score), social function (5.8 ± 1.3 score), vitality (14.8 ± 3.2 score), mental health (20.7± 4.2 score), physiological function (20.5 ± 4.1 score), physiological role limit (6.3 ± 1.6 score), emotional role limit (5.1 ± 1.3 score) and body pains (4.3 ± 1.8 score).Conclusions The score in cognitive function is low in elderly osteoporosis patients.It's useful to improve elderly osteoporosis awareness and quality of life by carrying out relative education.
3.Quality Control on Whole Process in Sterilization Supply Department

OBJECTIVE To investigate the method of quality control on the whole process in sterilization supply department. METHODS According to issued by Ministry of Health all kinds of criteria and inspections,the whole process in sterilization supply department was analyzed,and then the quality was controlled. RESULTS The whole process of sterilization was standardized,systematized and programmed. CONCLUSIONS The quality control on the whole process is an effective measure to ensure the rate of supply eligibility and support.
4.Identification of proteins related to proliferation and regulated by Epstein Barr virus encoded latent membrane protein 1 in nasopharyngeal epithelail cells

AIM: To investigate the molecular mechanism of Epstein-Barr virus encoded latent membrane protein 1 regulated cellular proliferation in nasopharyngeal epithelial cells. METHODS: The nasopharyngeal epithelial cells NP69 were infected with RV-pLNSX (the empty vector) and RV-LMP1 retroviruses, respectively. Therefore, the NP69-pLNSX and NP69-LMP1 cell lines were established. Sequentially, cellular proliferation of NP69-pLNSX and NP69-LMP1 cells was compared to draw the cellular growth curve. The experiments of plate clone formation and forming of soft agar colony were conducted. Meanwhile, the differential expression of proteins were identified between NP69-pLNSX and NP69-LMP1 cell lines by proteomic methods, and the expression levels of partial identified proteins were verified. RESULTS: (1) LMP1 was able to accelerate cellular proliferation of nasopharyngeal epithelial cell NP69 (n=3, P<0.05). (2) Twenty two proteins (9 up-and 13 down-regulated) of LMP1 mediated regulation were identified from infected NP69 cell lines, and the differential expression of partial identified proteins was confirmed by Western blotting and fluorescent real-time quantitative RT-PCR. CONCLUSION: LMP1 probably mediates the regulation of vimentin protein and keratin 19 protein expression to promote cellular proliferation in NP69 cells.
5.The comparison of ticagrelor and clopidogrel on patients undergoing percutaneous coronary intervention with acute ST elevated myocardial infarction

Objective To compare the effects of ticagrelor and clopidogrel on patients undergoing percutane-ous coronary intervention(PCI)with acute ST elevated myocardial infarction (STEMI).Methods 120 patients with STEMI received PCI within 12h of symptom onset in our hospital were randomly divided into clopidogrel treated group (n=60)and ticagrelor treated group (n=60).Serum was collected before surgery and 36 hours after PCI for ALT, Cr,CK-MB,and MA.Cardiac ultrasound was examined,too.All patients were followed 6 months post-PCI for main adverse cardiovascular and cerebrovascular events (MACCE)and medicine side effect.Results No significantly difference was noted in baseline between the two groups.The level of CK-MB and MA in the ticagrelor treated group [CK-MB(56.5 ±8.3)U/L,MA (45.9 ±6.4)mm[and clopidogrel treated group[CK-MB(74.3 ±9.6)U/L,MA (35.6 ±7.3)mm]were significant difference (CK-MB,P=0.043;MA,P=0.038).The MACCE of patients in ticagrelor treated group were significantly lower than patients in clopidogrel treated group during post-PCI 6 months follow-up(The ratio of angina in ticagrelor group was 1.7%,while in clopidogrel group was 6.7%,P=0.042). Conclusion Ticagrelor is more effective in suppress the function of platelet,decrease MACCE in patients with STE-MI undergoing PCI.
6.Effects of 5-Aza-CdR on cell apoptosis in lung cancer A549/DDP cell line and expression of tumor suppressor gene hMLH1

Objective To investigate the effects of 5-Aza-2'-deoxycytidine (5-Aza-CdR) on the apoptosis of A549/DDP cells and the expression of hMLH1 gene.Methods A549/DDP cells were treated with 5-Aza-CdR at 0.5,5,50 μmol/L.The growth curve of A549/DDP cells was investigated by MTT assay.The methylation status of hMLH1 gene was detected by methylation specific PCR (MSP).The expression of hMLH1 mRNA was evaluated by FQ-PCR.The apoptosis rate of A549/DDP cells was analyzed by flow cytometry.Results A549/DDP cells treated with 5-Aza-CdR showed a slow growth in comparison with the control cells,and the growth rates were decreased with the increasing of 5-Aza-CdR concentration.The apoptosis rate after treatment was higher than that before treatment in A549/DDP cells (P < 0.05),and had a positive correlation with 5-Aza-CdR dose (P < 0.001).hMLH1 mRNA expression level was increased in a 5-Aza-CdR concentration dependent manner (P < 0.05).hMLH1 promoter in A549/DDP cells was methylated and hMLH1 mRNA was negatively expressed before treatment,but the mRNA was positively expressed after treatment with 5-Aza-CdR.Conclusions 5-Aza-2'-CdR can induce apoptosis of A549/DDP cells by inducing demethylation of hMLH1 promoter and thereby enhancing hMLH1 gene expression and its tumor suppressor function.
7.Role of EGFR mutation status in patients with advanced lung adenocarcinoma treated with pemetrexed

Objective:To assess the role of EGFR mutations on pemetrexed response in patients with advanced lung adenocarcino-ma. Method: Forty pulmonary adenocarcinoma patients with evaluable lesions were retrospectively screened .They had been treated with pemetrexed-included chemotherapy and had EGFR gene test results. The evaluation endpoints were overall response rate,disease control rate and progression free survival. Result:No significant statistical difference was seen in overall response rate(ORR) (44.4%VS 31.8%, respectively) and disease control rate(DCR) (88.9%VS 81.8%, respectively ) between EGFR wild group and EGFR muta-tion group, but patients in EGFR wild group had longer progression free survival(PFS) ( 8.9 months VS 5.3 months;P=0.046). Conclu-sion:EGFR mutation status can influence the efficacy of pemetrexed.

Objective To investigate the reason of persisting positive rapid plasma reagin (RPR) in serofast syphilis patients, and to provide reference for clinical treatment and prognosis.Methods A total of 33 serofast patients and 23 healthy controls were enrolled in this study.The percentages and absolute counts of CD3+, CD4+, CD8+ T lymphocytes and natural killer (NK) cells were detected by flow cytometry.The comparison of two groups was analyzed by independent sample t test, and the correlation between change of lymphocyte subgroups and RPR titer in serofast syphilis patients was analyzed by bivariate linear correlation method.Results Compared with healthy controls, the percentages of CD3+, CD8+ T lymphocytes in serofast syphilis group were both increased significantly (75.75±5.76)% vs (68.37±5.80)%, (t=4.69, P<0.01);(27.34±7.02)% vs (24.33±1.95)%, (t=2.34, P=0.025), while both the percentage and absolute count of NK cells were significantly decreased (7.32±4.48)% vs (14.87±6.26)%, (t=5.269, P<0.01);(136.2±83.4)/μL vs (298.8±166.9)/μL, (t=4.311, P<0.01).RPR titer of the patients was negatively correlated with both percentage and absolute count of CD4+ T lymphocytes (r=-0.476 and-0.515, respectively, both P<0.01), and it was positively correlated of CD8+ lymphocytes (r=0.588 and 0.305, P<0.01 and P=0.804).Conclusion The imbalance of immune response of lymphocyte subsets observed in serofast syphilis may explain the RPR titers change.
10.Self-Control Study on 51 Children with Epilepsy Treated by Levetiracetam as Add-on Therapy

Objective To evaluate the long-term clinical efficacy and security of levetiracetam (Lev) as add-on therapy in patients with different types of epilepsies from an observational study.Methods Fifty-one patients were evaluated (14 female,37male,age range from 7months to 16 years,mean age 8.7 years) with different types of epilepsies ( 20 complex partial seizure,10 tonic-clonic seizure,1 tonic seizure,6 myoclonic epilepsy,2 Lennox-Gastaut syndrome,4 infantile spasms and 2 unspecified epileptic syndromes).The basis for comparison was defined as the seizure frequency in the 3 months prior to the commencement of treatment.Patients received Lev as add-on therapy.The initial dosage was 20 mg/(kg?d),and it was increased 10 mg/(kg?d) every 2 weeks.The maintenance dosage was 30-40 mg/(kg?d).Seizure frequency changes and adverse events were observed.Follow-up was conducted for a period of 6.8 months after treatment.SPSS 14.0 software was used to compare the difference between the seizure frequency before the Lev treatment and that after the Lev treatment.Results Thirteen (25.5%) out of the 51 patients reduced seizure frequency,16 (31.4%) patients had no reoccurrence;While another 9 (17.6%) patients seizure frequencied were reduced,8 patients' remained the same,and 5 patients' condition was got wor-sened.Six cases ceased treatment because of the worsening of the disease and the intolerance of Lev.The difference and after seizure frequency before in Lev treatment is statistically significant(P
