By using intrathecal administration of antisense oligodeoxynucleotide (AS-ODN) against preprodynorphin mRNA in rats, we observed that this treatment could block both the formalin-induced behavioral nociceptive responses and the increased expression of dynorphin A (1-8) in the dorsal horn, with the increased expression of c-Fos protein being unchanged. For we have reported that intrathecal administration of AS-ODN against c-fos mRNA blocks the nociceptive responses and both the increased Fos protein and dynorphin A (1-8), the results of the present study suggest that: (1) Nociceptively induced spinal expression of dyorphin and Fos protein is involved in the transmission of nociceptive information at the spinal level and the expression of Fos protein is the up-stream event. (2) dynorphin may act as a pronociceptive, not an antinociceptive, factor in the modulation of the spinal hyperalgesic state.
2.Influence of Rapid Reduction of Physical Activity on Bone Mass and Biochemical Markers of Bone Metabolism in Growing Rats

Objective To observe the effect of rapid reduction of physical activity on bone mass (BMC), bone mineral density (BMD)and bone metabolism in growing rats. Methods 48 male SD rats were divided into the high-intensity exercise group (T1, n=12), high tolow-intensity exercise group (T2, n=12), low-intensity exercise group (T3, n=12) and control group (Con, n=12). During the first 6 weeks,the rats in T1 and T2 group ran on treadmill with high intensity, while rats in T3 group took low-intensity treadmill exercise. 6 rats were sacrificedfrom each group at the end of 6th week. BMC, BMD of femur were measured with DEXA. Alkaline phosphatase (ALP), osteocalcin(BGP), tartrate-resistant acid phosphatase (TRACP) were detected with ELISA. The exercise intensity of T2 group was reduced rapidly to alow level as in T3 group, but remained in the other 2 groups. At the end of the second 6 weeks, the other rats were investigated again. ResultsCompared with the first measurements, ALP and BGP of the T2 group decreased significantly (P<0.05), and TRACP increased (P<0.05). There was no significant difference in BMC and BMD of femur (P>0.05). Conclusion Rapid reduction of physical activity can suppressbone formation and stimulate bone resorption of long bone among growing rats.
3.Clinical Observation of E_2 Transdermal Patch in the Treatment of Menopaus-al Syndrome

Objective:To evaluate the security, efficacy and side effects of E_2 transdermal patch (Fem-7) in the treatment of menopausal syndrome. Methods:A total of 60 cases with menopausal syndrome (blood serum FSH >30 U/L,E_2 <109.8 pmol/L) were randomly divided into treatment group ( Fem-7 group) and control group (estrodiol valerate (E_2V) group) for 12 weeks treatment. Fem-7 group changed patch every 7 days and took the 2 mg medroxyprogesterone (MPA) every day. While the E_2V group took the 2 mg MPA and 1.5 mg E_2 V every day. Serum E_2, FSH, lipoprotein and coagulation function were evaluated at baseline and at the 12th week of the treatment. In addition, menopausal symptoms and some other index of security and side effects were also observed. Results:Kupperman indix in both groups was decreased and the mean value of E_2 was increased obviously. Hepatic function, renal function, coagulation function and blood glucose were no obvious change. Lipoproteina was descended markedly in Fem-7 group. Other index of security in both groups was no obvious change. Conclusions:Fem-7 patch is secure and valid for relieving symptom of menopause, especially for the women who is not sutable for oral medication.

5.An exploration of contributing factors to the oocyte utility rate of high responders in an in vitro fertilization cycle

Objective:To figure out the clinical factors contributing to the oocytes utility rate (OUR) of high responders in in vitro fertilization (IVF)-embryo transfer treatment.Methods:OUR was defined by the number of usable embryos for transfer and (or) cryopreservation divided by the number of oocytes retrieved in a freeze-all cycle. The cycles with ≥15 eggs were included from January 2013 to December 2019. Those with OUR at the top 10% (Group A) and the bottom 10% (Group B) were picked and compared for patients′ characteristics, parameters relating to ovary stimulation and pregnant outcomes. Multifactorial logistic regression was applied to reveal the risk factors affecting OUR in them.Results:A total of 43 patients were included in Group A (OUR: 77.4%, 601/776) and 47 for Group B (OUR: 11.9%, 104/874). Previous IVF/intracytoplasmic sperm injection (ICSI; OR=0.10, 95% CI: 0.01-0.81) and endometriosis ( OR=0.16, 95% CI: 0.03-0.84) were negative factors for OUR ( P<0.05); dual suppression protocol ( OR=3.74, 95% CI: 1.06-26.86) and longer days of stimulation ( OR=3.24, 95% CI: 1.25-8.42) were protective factors in terms of ovarian stimulation ( P<0.05), on contrary to that, any decline of estradiol during the stimulation led to poorer OUR ( OR=0.16, 95% CI: 0.04-0.64). Although two groups had similar quantities of eggs and metaphase of meiosis Ⅱ (MⅡ) oocytes, distinguished cumulative clinical pregnancy rate and cumulative live birth rate were seen in Group A and group B respectively [95.3% (41/43) vs 40.4% (19/47) and 90.7% (39/43) vs 31.9% (15/47), all P<0.01]. Conclusions:High responders with attempted IVF/ICSI and endometriosis should be considered as risk factors for OUR. Over ovarian stimulation, dual suppression and a slightly longer stimulating duration could be tried; besides, estradiol decline should be prevented for a better OUR.

Phosphodiesterase(PDE) is hydrolase of 3',5'-cyclic adenosine monophosphate(cAMP) and 3',5'-cyclic guanosine monophosphate(cGMP).PDE3 are composed of two genes and distributed widely in vivo.Its inhibitors have been applied in antiplatelet aggregation and vasodilation.It has been reported that PDE3 inhibitors have neuroprotective effects on cerebral ischemia,which may provide new methods to prevention and therapy of cerebral ischemia.

8.Progress on the roles of TGF-βSmad signaling pathway in HIV pathogenesis

Abstract: TGF-β/Smad signaling pathway has a wide range of biological activities and plays an important roles in regulating cell growth, adhesion, differentiation, cell dynamic balance, and immune responses. The higher activity of TGF-β/Smad signaling pathway may promote scar formation, organ fibrosis, immunosuppression, and late-stage cancer progression, while low activity may lead to inflammation, dysplasia, poor healing and oncogenesis. The function of the TGF-β/Smad signaling pathway is extremely complex and can exhibit inhibitory or enhancing effects on immunity and inflammation under different circumstances, but immunosuppressive and anti-inflammatory effects are dominant. During HIV infection, the TGF-β/Smad signaling pathway interacts with HIV in a complex manner as HIV proteins tat and gp120 can induce TGF-β expression. Meanwhile, this signaling pathway may also play a role in HIV infection and replication, latent virus reservoir, host immune deficiency and HIV-related inflammation. It is worth noting that even though TGF-β, which mainly exhibits anti-inflammatory effects, is induced by HIV, high levels of TGF-β do not seem to inhibit HIV-related inflammation. So far, the relationship between TGF-β/Smad signaling pathway and HIV infection has not been elucidated, and its role and mechanism in HIV infection and related illnesses need further exploration and validation. This review summarizes the relevant research progress on the TGF-β/Smad signaling pathway and HIV infection, and provides a reference for further understanding of HIV pathogenesis and exploring strategies of AIDS treatment.
9.Effect of panax notoginseng saponins on growth and metastasis of malignant melanoma

Objective To investigate the effect of PNS on malignant melanoma and the expression of connexin32 in melanoma. Methods The spontaneous and experimental lung metastasis models of B16 melanoma were used to investigate the inhibitory effect of PNS on tumor growth and metastasis. The expression of connexin in melanoma were detected by immunohistoehemistry. Results (1) PNS can obviously inhibit the growth of B16 melanoma. The inhibition rate of the 480mg/kg PNS group was 50. 85%. (2) PNS can obviously inhibit the lung metastasis of B16 melanoma. The number of tumor colonies in lung of the 240 mg/kg PNS group and 480 mg/kg PNS group were lower than that in the negative control group. ( 3 ) The expression of connexin32 in melanoma was detected by immunohistochemistry:PNS could up-regulate the expression of connexin32 in membrane of melanoma. Conclusion PNS can inhibit malignant melanoma growth and metastasis and could also up-regulate the expression of connexin32 in membrane of melanoma.

Picture archiving and communication system ( PACS ) based on digital teaching platform of medical imaging consists of web teaching system and teaching resource library of medical imaging.The web teaching system includes four function modules of classroom management,work management,teaching management and test management.The teaching resource library includes script,pictures,and audio,video and animation materials of theoretical books,typical cases,examination techniques and medical imaging equipments.Utilized in theoretical and practical classes,the teaching characteristic of medical imaging is made prominent,and the subjective motility of the students is brought into play.
