Objective To assess the value of color Doppler flow imaging (CDFI) in indicating the recurrence and prognosis of liver cancer.Methods Two hundred and eighty-seven patients with primary liver cancer were enrolled,thirty-seven cases received operations ( 12.9% ).The size,profile,margin,echogen,blood supply of the tumors were observed preoperatively by CDFI. Twenty-eight patients were followed for 0.5 to 5 years.Results Recurrent nodules were seen in 24 cases (85.7%) during five years.Lesions larger than 5 cm recurred much earlier than those less than 5 cm in the diameter of the patients( P

4.Metabolomics study of doxorubicin induced hepatotoxicity.

To reveal the underlying mechanism of doxorubicin induced hepatotoxicity, an NMR-based metabolomic approach combined with multivariate statistical analysis was used to observe its metabolic alternations of rat liver. Sixteen differential metabolites between model rats and normal rats were characterized as potential pathological biomarkers related to doxorubicin induced hepatotoxicity. Six pathways, including phenylalanine, tyrosine and tryptophan biosynthesis, valine, leucine and isoleucine biosynthesis, phenylalanine metabolism, glycine, serine and threonine metabolism, alanine, aspartate and glutamate metabolism, and tyrosine metabolism were regarded as the targeted metabolic pathways according to Metabolic Pathway Analysis (MetPA). The results suggested that the metabolic perturbations in rats with doxorubicin induced hepatotoxicity were mainly involved in amino acid metabolism, lipid pathways, purine metabolism, energy metabolism, dysfunction of biotransformation and oxidative stress. The investigation revealed the effects of doxorubicin on liver in a holistic metabolic way, which laid a foundation for further studies on its toxicity mechanism.

5.Application study on multicolor combinational probe coding real-time PCR in detection of foodborne pathogens.

OBJECTIVETo investigate the detection limit of multicolor combinational probe coding real-time PCR (MCPC-PCR) in detection of Salmonella and Staphylococcus aureus suspended in the food samples, and to apply MCPC-PCR to detect the samples of food poisoning.
METHODSSeries concentration of bacterium suspension (10(1) - 10(9) CFU/ml) was prepared by using 22 simulated samples including fresh meat and cakes and then MCPC-PCR was applied to detect Salmonella and Staphylococcus aureus in 22 samples. Enrichment broth of 101 frozen samples and 5 early patients' anal swabs in food poisoning cases were detected after the DNA samples were extracted.
RESULTSThe limits of MCPC-PCR assay in detecting Salmonella and Staphylococcus aureus were about 10(2) copies/test; 101 frozen enrichment broth of samples in food poisoning cases were detected by MCPC-PCR assay, of 23 positive samples, 18 were confirmed by bacteriology techniques; 96 samples detected by MCPC-PCR and bacteriology techniques had the same results, and the coincidence rate was 95.05%. Anal swabs, collected from 5 of early patients in a food poisoning case gave a clue to be Vibrio parahaemolyticus by MCPC-PCR assay and then were perfectly consistent with bacteriology assay.
CONCLUSIONAs a method of high sensitivity and good specificity, MCPC-PCR assay can quickly and conveniently detect multiple pathogens existing in food samples, therefore we recommend it to be used in rapidly screening or simultaneous detection of food-borne diseases.

6.Cleavage of HCV by HCV specific deoxyribozyme in vitro.

OBJECTIVETo study the cleavage activity of specific deoxyribozyme to hepatitis C virus in vitro.
METHODSThree deoxyribozymes were designed to cleave at sites 157, 168, 173 in HCV 5'-noncoding region with the active region of 5'-GGCTAGCTACAACGA-3' respectively. Plasmid pCMV/T7-NCRC -Delta Luc was completely linearized with restriction endonuclease Xba I. HCV RNA5'-NCRC was transcribed in vitro from the linearized products and radiolabelled with [alpha-32P] UTP. Under the conditions of 37 degrees C, pH7.5, Mg2+ 10 mmol/L, the three deoxyribozymes were mixed with substrate RNA individually for 120 minutes and then the reactions were terminated. The cleavaged products were separated with 8% denaturated polyacrylamide gel electrophoresis and displayed by autoradiography. DRz3 was mixed with the substrate RNA at different Mg2+ concentrations. The cleavage efficiency was analyzed with a gel document action analyzing systems.
RESULTSUnder the adopted conditions the three deoxyribozymes efficiently cleaved to the target RNA in vitro and the cleavage activity of DRz3 was increased with the increase of Mg2+ concentration.
CONCLUSIONThe designed deoxyribozymes can cleave 5'-NCR mRNA of HCV efficiently in vitro and it is dose-respondent to Mg2+ concentration.

7.Correlation between immunochemical level and patient with caries.

OBJECTIVETo explore the relationship between immunochemical level of salivary and caries in children aged 4-6 years old.
METHODSTwo groups were assorted as patients with caries and without caries. Every group included 45 people. Measurements of salivary secretory immunoglobulin A (SIgA)were performed by using radio-immunoassay and single agar diffusion assay. The levels of lysozyme (LZ), alkaline phosphatase (ALP) and lactic dehydrogenase (LDH) were studied with colorimetry and turbidimentry.
RESULTSThe levels of LDH, SigA, ALP, LZ had significant difference between the two groups (P < 0.05). The level of LDH between patients and peoples without caries had little difference (P > 0.05).
CONCLUSIONThe incidence of caries is associated with age, and it may have association with immunochemical levels of salivary.

8.Experimental study on effect of pre-acupuncture and moxibustion on molecular markers in pre-thrombosis state of rats.

OBJECTIVETo explore the best method for prevention and treatment of thrombosis and its mechanism.
METHODSForty SD rats were randomly divided into a blank group, a model group, an electroacupuncture group and a crude herb moxibustion group. In the electroacupuncture group and the crude herb moxibustion group. "Zusanli" (ST 36), "Sanyinjiao" (SP 6), "Pishu" (BL 20) and "Geshu" (BL 17) were punctured or moxibusted for 2 weeks, then pre-thrombosis model was induced by Adrenalin Hydrochloride and ice water method in the model group, the electroacupuncture group and the crude herb moxibustion group, respectively. Molecular markers in venous blood after the model made in 18 hours were detected.
RESULTSAct: vaty of tissue-type plasminogen activator (t-PA) and content of alpha-granule membrane protein (GMP-140) decreased and content of nitrogen monoxidum (NO) increased after electroacupuncture or crude herb moxibustion. The levels of t-PA and GMP-140 in the model group were higher than those in the electroacupuncture group, the crude herb moxibustion group and the blank group (all P < 0.05), and the content of NO in the model group was lower than those in the electroacupuncture group, the crude herb moxibustion group and the blank group (all P < 0.05), while there was no significant difference in t- PA, GMP-140 and NO among the crude herb moxibustion group, electroacupuncture group and blank group (all P > 0.05).
CONCLUSIONElectroacupuncture and crude herb moxibustion can significantly change the contents of t-PA, GMP-140 and NO and there was no significant difference between the two therapies.

9.Inhibition of HCV replication by HCV specific U1 small nuclear RNA chimeric ribozyme in vivo.

OBJECTIVETo determine the advantage of U1 small nuclear RNA as a ribozyme vector (U1-Rz) to inhibit HCV replication in vivo.
METHODSThe 3rd stem-loop was substituted by HCV core specific ribozyme to construct an U1-Rz eucaryotic expression plasmid. Then it was co-transfected with pCMV/T7-NCRC Delta-luc into Huh7 cell line mediated by lipofectin. The cell lysis supernatant was subjected to Western blot and lumenometer to determine the luciferase levels.
RESULTSA U1 snRNA chimeric ribozyme was constructed successfully. Both Rz and U1-Rz inhibited luciferase expression in Huh7 by 48.64% and 87.46%, respectively.
CONCLUSIONRz has more efficacy in cells when using U1 snRNA delivery system. U1 can be an efficient vector for HCV specific ribozyme.

10.Simultaneous occlusal orthodontics during mandibular distraction osteogenesis.

OBJECTIVETo study the significance and principle of simultaneous orthodontics during mandibular distraction osteogenesis.
METHODSTotally 11 patients simultaneously underwent occlusal orthodontic treatment for 3-4 months during mandibular distraction osteogenesis. Square-wire and elastic loops were adapted to perform the orthodontics by ways of more frequent adjustment of orthodontic device than routine method.
RESULTSAll 11 patients with mandibular micronathia obtained the improved occlusion with their mandibular expected elongation, for instance, their open-bite and teeth displacement were partially corrected.
CONCLUSIONSimultanous orthodontics with mandibular distraction osteogenesis may improve the malocclusion, decrease the orthodontic time, and lead the mandibular distraction direction.
