2.Acceleration of the Recovery of Chemotherapy-Induced Immune Suppression by the Intrasplenic Transplantation of GM-CSF Gene-Transfected Fetal Liver Cells

After murine fetal liver cells (FLC) were transfected with granulocyte-macrophage colony-stimulating factor (GM-CSF) gene by recombinant adenovirus and intrasplenically transplanted into allogeneic mice, the effects of GM-CSF gene-transfected FLC on the recovery of immune response inhibited by chemotherapy were observed. The number of CD4 + cells and the ratio of CD4 + /CDS + cells from peripheral blood lymphocytes increased significantly. The cytotoxicity of the NK cells and the proliferation response of splenocytes to ConA, LPS elevated markedly, but the same results were not from bone marrow. These data demonstrated that intrasplenic transplantation of GM-CSF gene-transfected FLC could effectively accelerate the recovery of immune response after high-dose chemotherapy.

Objective To observe the distribution of aerobic bacteria in conjunctival sac of patients with proliferative diabetic retinopathy and the sensitive antimicrobial agents in order to master the distribution of bacteria in the conjunctival sac and select sensitive antibiotics to decrease the infection rate and improve the therapeutic effect. Methods From March 2013 to August 2016, 248 patients with proliferative diabetic retinopathy were treated with aerobic bacterial culture and antibiotic susceptibility test. The distribution characteristics of aerobic bacteria in conjunctival sac and the sensitivity of antibiotics were observed. Results There were 112 strains in this study, the positive rate was 45.16% (including 8 cases of mixed infection), including 104 strains of gram-positive bacteria, 92.86% of gram-negative bacteria and 7.14% of gram-negative bacteria, the difference was statistically significant differences (P<0.05). Among them, the positive rate of staphylococcus epidermidis higher than other types of strains, the difference were all significant differences (P all<0.05). The sensitivity of vancomycin to gram-positive bacteria 100% was higher than that of, ofloxacin 13.46%,ciprofloxacin 21.15%,gentamicin 69.23% and rifampicin 88.46%, (P all<0.05). The sensitivity of gentamicin to gram-negative bacteria 87.50% was higher than that of rifampicin 0 and vancomycin 12.50%, (P all<0.05). Positive rate of culture results and staphylococcal multidrug resistance rate were 72.94%,31.76%, the incidence of >65 years of age were higher than ≤ 65 years old 30.67%,12.27%, male 51.61%,23.12% than female 25.81%,6.45%, there are basic diseases 69.15%,31.91% were higher than those without underlying diseases30.54%,11.04%, (P<0.05). Staphylococcus number composition ratio, the difference was not statistically significant. Conclusion To strengthen the observation of aerobic bacteria and the sensitivity of commonly used antimicrobial agents in patients with proliferative diabetic retinopathy, especially the high risk population, it is helpful to master the distribution characteristics of bacterial conjunctival sac and sensitive antimicrobial agents, so as to achieve the preoperative conjunctival sac Bacteria and reduce the rate of infection and other purposes.

Objective To explore the changes of serum leptin level in patients with diabetic microangiopathy.Methods A total of 90 subjects were assigned to three groups which included 30 healthy controls,30 diabetic patients without microangiopathy and 30 patients with diabetic microangiopathy.The studing groups were matched for sex and BMI.The level of serum leptin ,ⅣC,insulin and glucose of these patients were measured,and the correlation between serum leptin level and other indexes was analyzed.Results The level of serum leptin in controls,diabetic patients without microangiopathy and patients with diabetic microangiopathy were (7.20?2.11) ?g/L, (7.95?3.78)?g/L and (10.26?4.37)?g/L respectively .The patients with diabetic microangiopathy had higher serum leptin levels than in the diabetic patients without microangiopathy( t =2.18, P

Psoriasis is a non-infectious chronic inflammatory skin disease. It′s acknowledged that interleukin (IL)-17 signaling pathway dominantly drives the development of psoriasis. Recently, the role of neuro-immune axis in psoriasis has attracted widespread attention. Lidocaine, a local anesthetic, has ability to block the conduction of nerve impulses, while its therapeutic efficacy on psoriasis remains to be confirmed. Here, we evaluated the therapeutic efficacy of topical application of compound lidocaine cream (LIDO) on imiquimod (IMQ)-induced mouse psoriasis model. Animal welfare and experimental procedures follow the regulations of the Ethics Committee of China Pharmaceutical University. OBJECTIVETo compare the difference of Euphorbia Pekinensis Radix before and after being processed with vinegar in the toxicity on rat small intestinal crypt epithelial cells IEC-6, and make a preliminary study on the mechanism of detoxication of Euphorbia Pekinensis Radix processed with vinegar.
METHODWith rat small intestinal crypt epithelial cells IEC-6 as the study object, the MTT method was adopted to detect the effect of Euphorbia Pekinensis Radix before and after being processed with vinegar on IEC-6 cell activity. The morphology of cells were observed by the inverted microscope. The down-regulated mitochondrial apoptosis pathway of enterocytes caused by the vinegar processing was analyzed by using the high content screening.
RESULTCompared with the negative control group, the proliferation inhibition experiment showed that Euphorbia Pekinensis Radix showed a relatively high intestinal cell toxicity (P < 0.01). The results of HCS analysis showed that Euphorbia Pekinensis Radix could significantly reduce the cell nucleus Hoechst fluorescence intensity and mitochondria membrane (P < 0.05, P < 0.01), and increase Annexin V-FITC and PI fluorescence intensity and membrane permeability (P < 0.01, P < 0.01, P < 0.01). After being processed with vinegar, compared with Euphorbia Pekinensis Radix groups with different doses, Euphorbia Pekinensis Radix processed with vinegar could significantly decrease the cell proliferation inhibition effect on enterocytes, increase the cell nuclear Hoechst fluorescence intensity and mitochondria membrane (P < 0.05, P < 0.05), and decrease Annexin V-FITC and PI fluorescence intensity and membrane permeability (P < 0.01, P < 0.01, P < 0.05), and showed a certain dose-effect relationship.
CONCLUSIONThe vinegar processing can further reduce the toxicity of Euphorbia Pekinensis Radix on enterocytes. Its possible mechanism can decrease the effect of Euphorbia Pekinensis Radix on the permeability of IEC-6 cell membrane, so as to provide a basis for further explanation of the detoxication mechanism of Euphorbia Pekinensis Radix processed with vinegar.

8.The influence of sealing tube with urokinase on the malfunction of central venous catheter with Dacron ring

Objective To explore the effect of sealing tube with regular urokinase for the malfunction of central venous catheter(CVC) in patients with hemodialysis (HD).Methods This was a prospective randomized controlled study,132 HD patients with cuff CVC were studied.In the early stage,52 H D patients without 80 patients with regular catheter-locking solution of urokinase.To analyze effect of cuff CVC malfunction,blood flow(BF),dialysis adequacy,anemia and serum albumin (ALB) with regular catheter-locking solution of urokinase.The incidence of catheter malfunction was calculated based on the catheter dysfunction in the first 3 months.Clinical index was measured,recorded BF and calculated urea clearance rate (Kt/V).Results HD patients catheter-locking solution with regular urokinase could reduce the incidence of dialysis catheter malfunction ( 26.3% vs 32.7%,x2 =32.727,P <0.01 ),increase the BF,Kt/V,levels of hemoglobin(HGB) and ALB(P<0.05 or P <0.01).Conclusion Regular catheter-locking solution with urokinase is effective in reducing HD patients incidence of dialysis CVC malfunction,increasing patients BF,dialysis adequacy and the level of ALB,improving anemia.
9.The in vitro and in vivo experimental study of long-circulating superparamagnetic iron oxide liposome nanoparticles as novel MR contrast agent

Objective To evaluate pharmacodynamics of prepared long-circulating superparamagnetic iron oxide (SPIO) liposomes. Methods Control and experimental groups were established after adding SPIO or long-circulating SPIO liposomes as agents. (1)Macrophages experiment in vitro: the RAW 264.7 macrophage cell strains were recovered, cultured and seeded in the culture plate at a density of 2.5×105 cells/well until they reached 80%-90% confluence. The intracellular Fe uptake of control and experimental group cells were quantified by Ferrozine assay after incubation with different concentrations of drugs. Factorial design analysis of variance was used as statistics method. Prussian blue staining method was used to detect staining of experimental cells.(2)Drug biodistribution in mice: C57BL/6J(n=6) were classified into blank control group (n=2), control group(n=2) and experimental group(n=2).Saline, SPIO and long circulating SPIO were injected via the tail vein in the blank control group, control group and experimental group respectively. Then distribution of drugs in the body was observed by pathological examination.(3) MR imaging of tumor-bearing nude mice: 20 BALB/c nude mice bearing lung cancer models were established and classified into control group and experimental group. After administration of drugs, all animals underwent MR scanning. Signal intensities of livers and tumors were measured, SNR-time dynamic curves were drew. Covariance analysis was used to compare post-enhanced SNR at the 12th hour. (4)Cytotoxicity studies (MTT): cytotoxicity of both drugs on human liver cell line HL-7702 was studied, and statistically analyzed using factorial design analysis of variance. Results (1) Macrophages experiment in vitro: The nanoparticle uptake by macrophage cells evaluated by ferrozine assay showed the uptake of blank SPIO was higher than long-circulating SPIO liposomes. Compared with the blank control group, there was strong blue staining in the macrophages with Prussian staining in the control group and little blue staining in the experimental group. (2) Drug biodistribution in mice: for blue stained cells composed of iron particles, the amounts in the liver, spleen, lung, kidney of the control group were more than those in the experimental group. (3) MR imaging of tumor-bearing nude mice: the non-enhanced SNR of livers and tumors in the control group and experimental group were 31.47 ± 0.56, 30.89 ± 1.41, 58.41 ± 0.61, 58.44 ± 1.08, respectively. After injecting of contrast agents, SNR of livers and tumors in the control group and experimental group were 17.00 ± 0.96, 22.29 ± 0.73, 58.50 ± 0.63, 52.47 ± 1.18, respectively. The covariance analysis showed that SNR of the livers in the control group after 12 hours was significantly lower than the experimental group (F=167.022, P=0.000); while the SNR of the tumors in the experimental group was significantly lower than the control group (F=266.106, P=0.000).(4) Cytotoxicity of nanoparticles by MTT method: the viability of HL-7702 cells tend to decrease with the increase of Fe concentration. Cytotoxicity in the long-circulating SPIO liposomes was lower than the SPIO(F=2256.204,P=0.000). Conclusions Long-circulating SPIO liposomes we prepared reveal suitable sizes, even distribution, and good anti-macrophage ability in vitro and in vivo. They have long circulation characteristic and T2 negative enhancement effect in the transplanted lung cancers, while they still maintain low cytotoxicity.

ObjectiveTo explore a new simple epicanthoplasty method fit for various types of epicanthus with inconspicuous scar after operation.Methods As the latest studies showed that the anatomical structure of epicanthus is mainly due to the eyelid orbicularis muscle's dislocation at the starting point of the medial canthal tendon,which affects the skin appearance,rather than the traditional view that was merely the excess skin,we released the adhesions between the skin and the inner canthus orbicularis muscle,and corrected the dislocation of orbicularis muscle that was key to the operation,incised along with the line A′→B→A″→ C in the field of interior canthus,separated the subcutaneous fascia tissue,relaxed the subcutaneous fibrous cord,disconnect the orbicularis muscle's attachment point in the inner canthal fold,and then sutured and folded the inner canthal ligament.After sewing up point A′ and point A″,the released skin retracted naturally and formed a new inner canthal contour.ResultsFrom June 2008 to June 2009,57 cases of epicanthus patients accepted this surgery.After operation,the appearance of canthus was nice.Eye fissure got widen,and the scar was not obvious.Also there was a high satisfactory rate.ConclusionsThis method has a broad range of applications.It is suitable for various types of epicanthus correction.
