1.Study on Angiotensin Converting Enzyme Gene Polymorphism in Children with Congenital Heart Disease

Objective To investigate the relationship between the insertion/deletion(I/D) polymorphism of the angiotensin convcer-(ting) enzyme(ACE) gene and congenital heart disease(CHD) and to quest the predisposing gene of congenital heart disease by analyzing the polymorphism of the ACE gene in patients with CHD.Methods Sixty-eight patients with CHD were served as observing group,sixty-four patients without CHD as control group.We drew DNA of leukocyte from blood by the method of saturation phenol and chloroform and we amplified the ACE gene using the DNA polymerase chain reaction technique.The amplification products were employed electrohoresis in 2% sepharose.the polymorphism of the angiotensin converting enzyme gene was analyzed under ultraviolet lump.Results In the obser-(ving) group,the genotype distribution frequency of the ACE was II in 12(17.65%),DD in 32(47.06%) and ID in 24(35.29%),and in the control group,it was II in 20(31.25%),DD in 10(15.63%) and ID in 34(53.12%).Conclusions The study result clarifies that CHD has close correlation to the polymorphism of the angiotensin converting enzyme gene.The relative risk of DD genotype is higher than that of the others,and DD genotype possibly is the predisposing genotype of CHD D allele perhaps is the susceptibility gene of CHD.

Objective To investigate the changes of dopaminergic(DA) neurons in substantia nigra compact of the aged rats.Methods Using RT-PCR,Western blot,immunohistochemistry and electron microscope to detect the levels of TH mRNA and TH protein,the number and the ultrastructure of TH-positive neurons in the substantia nigra compact of the aged rats(≥24 months),and comparing those with adult rats(4~5 months).Results The level of TH mRNA(0.66?0.12) in substantia nigra compact of the aged rats was much lower than that in the adult rats(1.09?0.08)(P0.05).However,the number of TH-positive neurons in the caudal of substantia nigra compact of the aged rats was much lower than that of the adult rats(P

4.~1H-MRS in bilaterlis frontal lobe and hippocampus of patients with first episode major depression after SSRIs antidepressant treatment

Objective To measure neuro-biochemical changes in brain of first episode major depression (MD) patients. Methods Single-voxel proton magnetic resonance spectroscopic (1H-MRS) examination of bilateralis frontal lobe and hippocampus was conducted in 21 first episode major depression patients and 21 age-, sex-and education-matched healthy controls. After this, major depression patients took selectivity serotonin reuptake inhibitors (SSRIs) for three months. Then, we examined the changes in NAA, Cho, Cr, Glx and mI in bilaterlis frontal lobe and hippocampus of patients. Finally, we compared the metabolism of the subjects with that of the controls. Results ① Bilateralis frontal lobe NAA/Cr, right frontal lobe Glx/Cr and left hippocampus NAA/Cr and Glx/Cr were significantly lower in MD patients than in the controls, but right frontal lobe and right hippocampus mI/Cr significantly were higher than those in controls. ② After treatment left frontal lobe and left hippocampus NAA/Cr significantly increased compared with pretherapy. Right frontal lobe mI/Cr significantly decreased. Conclusion Nerve cell activity disorder, abnormal second messenger and glutamicacid and glutamine may be involved in the pathogenesis of MD. Antidepressant can regulate abnormal metabolism and improve nerve cell activity.
5.Survey and Practice on the Construction of Medical Ethics Emotion Education Model for Clinical Trainees

Objective:To analyze the sympathy,responsibility and rational emotion of clinical trainees and to construct the medical ethics emotion education system.Methods:A questionnaire survey was conducted among undergraduate clinical medical students.Of the 307 selected students,302 returned valid questionnaire.After eight months in the field trip,the educational outcome was assessed using questionnaire survey again,and the data was analyzed with descriptive statistics analysis.Result:According to the results of questionnaire survey,we came up with effective countermeasures to improve the medical ethics emotion of clinical medical students,and then strengthened students' sympathy,responsibility and rational emotion quality education.Conclusions:It suggests that cultivating the students' learning interests and motivating the subjective initiative of trainees on ethics learning with various teaching methods promoted the transformation from medical ethics emotion to behaviors and customs,and thus constructed a medical ethics emotion education model and improved quality medical ethics education.
6.Clinical and pathological characteristics of multiple system atrophy

Objective To study the clinical,pathological characteristics and diagnosis of multiple system atrophy(MSA).Methods Among 4 cases of MSA,2 of them were from clinical data,the other 2were frum pathological data.Two cases verified by autopsy were investigated using Hematoxylin-eosi,Kltiver.Barrera and Gallvas-Braak silver staining and confirmed by immunohistochemistry using ubiquitin,α-synuclein staining.Results All 4 patients were male.aged 51-76.Case No.1 wag diagnosed as Parkinson's svndrome.Case No.2 was diagnosed as spinal ataxia cerebella,and the other two eases were diagnosed as MSA.The following changes were found by pathological studies.Macroscopic atrophies were presented in pens and cerebella.as well as putamen,globus pallidus and substantia nigra.Cerebral ventricles were dilated.Neuronal lOSS and gliosis could be seen at cerebral cortex.nigrostriatal.globus pallidus,pontine nuclei,subslantia innominata,inferior olives,doral motor nucleus of vagus,cerebellum antl intermediolateral column of the spinal cord.In the white matter of these regions cytoplasmic inclusions bodies were extensively present in oligodendrocytes.Conclusions Olivopontocerebellar atrophy mainly shows the,clinical symptoms of pens,cerebellum,and autonomic nerves damage;Shy-Drager syndrome presents mainly with the erecting hypotenstion symptom,while striatonigral degeneration mainly involves extrapyramidal system.As these three diseases share the common basic pathological changes,they are preferred to be classified as the subtype of MSA.

10.Development of medical consortiums at Zhejiang University and its affiliated hospitals:practice and ;insights
