1.The verification of industry standard for growth hormone quantitative labeling immunoassay kit

Objective:To establish the industry standard for growth hormone quantitative labelling immunoassay kit, and to validate it by chemiluminescence labeling and time-resolved fluorescence labeling method which are suitable for the standard.Methods: Different assay method kits, including magnetic particle chemiluminescent assay, electrochemiluminescence assay, chemiluminescence assay and time-resolved fluorescent assay, were used to verified the blank limitation, linearity, accuracy, precision, specificity and stability in accordance with protocol industry standard.Results: Other verification results could meet requirements of the protocol industry standard besides accuracy in part of kits couldn't achieve to anticipative remand (relative deviation couldn't be more than ±10%).Conclusion: According to the verification results, the accuracy requirements was adjusted to ±15%. The other items of industry standard were maintained. The industry standard for growth hormone quantitative labeling immunoassay kit is ultimately established. The standard would contribute to unity quality standard of growth hormone quantitative labeling immunoassay kit, and provide the basis for the supervision and administration of its production, examination, circulation, clinical application and other areas.
2.Effects of intermittent change the angle of operating table on the skin in pressure areas of patients in prone position

Objective:To evaluate the effects of intermittent change the angle of operating table on the skin in pressure areas of patients in prone position.Methods:A total of 150 hospitalized patients for elective surgical treatment in Yantai Yuhuangding Hospital from January 2019 to June 2019 were selected as subjects. The patients were divided into control group, 15° test group and 30° test group by random number table method, with 50 patients in each group. The control group received routine intraoperative pressure ulcer prevention measures, the 15° test group on the control group, the itinerant nurses adjusted the angle of the operating table according to the left 15°-supine-right 15° every half hour, and the 30° test group on the control group, the itinerant nurses adjusted the angle of the operating table according to the left 30°-supine-right 30° every half hour. The incidence of surgically related pressure ulcers and doctors' satisfaction with the surgical position of the three groups of patients were evaluated.Results:Pressure ulcers occurred in 8 patients of the control group, the incidence of pressure ulcers was 16%. Pressure ulcer occurred in 0 patient of the 15° test group, the incidence of pressure ulcers was 0. Pressure ulcers occurred in 2 cases in the 30° test group, the incidence of pressure ulcers was 4%. The difference was statistically significant ( χ2 value was 10.120, P<0.05). Further comparison showed that the incidence of pressure ulcers was statistically significant between the control group and the 15° test group ( P<0.016 7), while the incidence of pressure ulcers was not statistically significant between the control group and the 30° test group, between the 15° test group and the 30° test group ( P>0.016 7). The results of the postoperative doctors' satisfaction with the surgical body position were (27.880 ± 1.637), (27.520 ± 1.693), (26.920 ± 1.947) points in the control group, the 15° test group and the 30° test group, there was a statistically significant difference among the three groups ( F value was 3.779, P<0.05). Further comparison showed that there was no statistically significant difference in the score of surgical position satisfaction between the control group and the 15° test group, between the 15° test group and the 30° test group ( P>0.05). The satisfaction score of surgeons in the control group was higher than that in the 30° test group, and the difference was statistically significant( P<0.05). Conclusions:Intermittent adjustment of surgical position can reduce the incidence of pressure ulcers and improve the quality of nursing in the operating room without affecting the convenience of the surgeon. The optimal effect was achieved by adjusting the angle of the operating bed according to the left-leaning 15°-supine-right-leaning 15°at an interval of 30 minutes.
3.Effect of Acupuncture Combined with Vibroacoustic Therapy on Upper Limb Spasticity Post Stroke

Objective To observe the clinical effect of acupuncture combined with vibroacoustic therapy on upper limb spasticity post stroke. Methods From November, 2014 to November, 2015, 90 patients with upper limb spasticity post stroke were randomized into treat-ment group (acupuncture combined with vibroacoustic therapy), control 1 group (simple vibroacoustic therapy) and control 2 group (simple acupuncture) with 30 cases in each group. All the patients accepted routine fundamental treatment and rehabilitation training. The modified Ashworth Scale (MAS), Fugl-Meyer Assessment (FMA) and modified Barthel Index (MBI) were used to assess the upper limb muscle ten-sion, motor function and activities of daily living before and four weeks after treatment. Results After treatment, MAS score decreased in all groups (Z>3.067, P<0.001), and FMA score and MBI score increased (t>3.326, P<0.01). MAS score decreased more (Z>2.919, P<0.01), and FMA score and MBI score increased more (P<0.05) in the treatment group than in the control groups after treatment. However, there was no significant difference between the control groups (P>0.05). The total effective rate was 80%in the treatment group, 50%in the control group 1 and 53%in the control group 2 (χ2=8.573, P=0.014), respectively. Conclusion Acupuncture combined with vibroacoustic therapy can alleviate the upper limb spasticity, and promote the recovery of motor function and activities of daily living.
4.Application of EWGSOP Consensus in Evaluating Elderly Female Patients with Sarcopenia Ranging in age from 70 to 79 from Communities of Shanghai

Objective To explore the feasibility of using the consensus of European Working Group on Sarcopenia in Older People(EWGSOP) to diagnose sarcopenia among community-dwelling women aged from seventy to seventy-nine in Shanghai.Methods Twenty-two healthy community-dwelling women were recruited as the subject group(group S),and another ten young girls were randomly selected as the control group(group C).The anthropometry,bioelectrical impedance analysis(BIA)and dual energy X-ray absorptionmetry(DEXA)were used to evaluate the circumferences of upper and lower limbs,and the dimension of muscle mass,according to relative appendicular skeletal mass(RASM)and skeletal muscle index(SMI).The grip dynamometer and isokinetic dynamometer were used to measure the muscle strength of upper and lower limbs.The ten meters gait speed,functional reach test,timed up and go test,Berg balance scale and short physical performance battery were all applied to assess the physical performance.The criteria of EWGSOP consensus were applied to diagnose the situation of sarcopenia.Results Firstly,the average biceps circumference and fat mass of upper limbs in group S were significantly higher than group C (P<0.01),while no significant differences were observed in the muscle mass(P>0.05).Secondly,RASM values gained from BIA and DEXA in S group were significantly higher than C group(P<0.05),while the SMI values of the former were significantly lower than the latter(P< 0.01).Thirdly,absolute grip strength,relative grip strength and other bio-mechanical indexes of lower limbs in the s group were significantly lower than the C group(P<0.01);the relative grip strength showed significant correlation with most bio-mechanical indexes of lower limbs.Fourthly,the physical performance in group S declined significantly compared with that of the c group(P<O.05),and no significant correlation was found between muscle strength and physical performance of upper limbs except the absolute grip strength and gait speed.The prevalence was 0% according to the RASM index,while the prevalence was 63.6% and 45.4% according to the BIA and DEXA respectively.Conclusion The assessing pattern from EWGSOP consensus could not be simply copied to applying on Chinese old people due to serious difference between RASM and SMI.The combination of SMI+relative grip strength+ TUG/SPPB/Berg may be better than the classical mode of RASM+absolute grip strength+gait speed in diagnosing sarcopenia.
5.Estimating the copy numbers of exogenous gene in transgenic cashmere goats by real-time fluorescence quantitative PCR

The copy numbers of exogenous gene in transgenic animals is always regarded as an important information of transgenic animals.Thus,simple and sensitive methods are required for the detection of the copy numbers of exogenous gene.Three kinds of transgenic Shanbei white cashmere goats,containing Tβ4-GFP,FGF5s-GFP and VEGF164-GFP,has been obtained by using PiggyBac(PB) transposon system.Fluorescence quantitative PCR was carried out to detect the copy numbers of copGFP.Using Gluc as reference gene,the double standard curves of exogenous gene and reference gene were mapped and the genomic DNA of transgenic goats were analysized by real-time fluorescence quantitative PCR.Moreover,the copGFP/Gluc ratio in the samples was calculated as the copy numbers of copGFP.In addition,Tβ4-GFP transgenic cashmere goats were selected to detect the integration sites by using the genomic walking kit.The results showed that the standard curve equation of copGFP was y=-3.230 6x+39.216 (R2 =0.998 8) and the standard curve equation of Gluc was y=-3.564 8x+38.440 (R2 =0.996 0).The copy numbers of exogenous gene in the transgenic cashmere goats were obtained and the numbers of integration sites in the selected Tβ4-GFP transgenic goats were consistent with the copy numbers of copGFP.As a conclusion,the high throughput,fast and sensitive real-time fluorescence quantitative PCR is an efficient and convenient method for the copy number of exogenous gene in transgenic cashmere goats.
6.Effects of Dong's Extraordinary Acupoints Acupuncture and Rehabilitation on Neural Function in Patients with Upper Limb Spastic Hemiplegia after Stroke

Objective To observe the effects of Dong's Extraordinary Acupoints acupuncture and rehabilitation on upper limb spastic hemiplegia after stroke. Methods 105 patients with upper limb spastic hemiplegia after stroke were randomized into 3 groups as groups A, B, C equally, and receiving Baclofen and rehabilitation training, acupuncture at Dong's Extraordinary Acupoints, and both acupuncture at Dong's Extraordinary Acupoints and rehabilitation training for 8 weeks, respectively. They were assessed with China Stroke Scale (CSS) and modified Ashworth Scale (MAS) before and after treatment Results The CSS and MAS scores obviously improved after treatment in each group (P<0.01), and improved more in the group C than in the group A and B for CSS (P<0.05). Conclusion Both acupuncture at Dong's Extraordinary Acupoints and rehabilitation can improve the neural function and upper limb muscle tension in patients with upper limb spastic hemiplegia after stroke, with the synergistic effects.
7.The Quality Analysis of National Supervising Sampling for Rubella Virus IgM Diagnostic Kits in 2014.

OBJECTIVETo evaluate the quality status of rubella virus IgM diagnostic kits by national supervising sampling.
METHODSUsing legal inspection combining with exploratory study, the positive and negative coincidence rate, detection limit and repeatability of kits were verified.
RESULTSThe results showed that 15 of 16 batches of kits were qualified using legal inspection, and the passing rate was 93.8%. The unqualified item was negative coincidence rate. In exploratory study, only 11 batches (68.8%) complied with industry standard. The unqualified items were negative coincidence rate, detection limit and repeatability.
CONCLUSIONAt present, rubella virus IgM diagnostic Kits have some quality problems in the market. It is recommended that we adopt industry standard and national reference panel in the registration inspection for the future, which will prompt enterprises to improve quality.

8.Protective effects of melatonin on learning and memory in rats by noise stress

Objective To explore the effect of melatonin(MT) on the behavior of rats treated with noise stress and the related bio-mechanism. Methods Fifty rats were randomly divided into a blank group,two experimental groups and two control groups. The blank group was untreated. The experimental and control groups were exposed to 120dB noise stress for 1 day or 3 days, 8 hours per day, and treated with 15 mg/kg melatonin by intraperitoneal injection,or the same volume of physiological saline 30 minutes before noise stress. After noise stress,the rats' behavior was measured by open field test, learning and memory ability of rats was investigated with the method of Morris water maze and then nitric oxide (NO), superoxide dismutase (SOD) and malondialdehyde (MDA) contents in cerebral cortex and hippocampus of the rats were measured by TBA and Griess method respectively. Results No matter noise stress time was 1 day or 3 days, the excitability and explorative behavior of the 2 experimental groups(total movement distance (TMD) (1322.50 ± 504.32) cm, (1819.55 ± 458.37) cm, faster movement time (FMT) (68.49 ± 23.90) s, (87.34 ± 16.01) s, distance to center (DTC) (63.56 ± 2. 75) cm, (60. 13 ±1.87)cm, inner toriod time(ITT) (7.87 ±2.06)s,(9.60 ±2.89)s) in the open field test decreased significantly compared with those of the control group (TMD (2042.03 ± 449. 19) cm, (2325.73 ± 384.90) cm,FMT (109.32 ±21.84)s,(124.65 ± 16.74)s, DTC (58.00± 1.53)cm,(55.05 ±5.13)cm, ITT (12.84 ±3.62) s, (14.92 ± 2.75) s, P < 0. 05, P < 0.01);the escape latency of the experimental groups (( 10. 69 ±3.37) s, (18.87 ± 4.74) s) in Morris water maze was significantly shorter than that of the control group (( 23.86± 7.66)s, (33.55 ± 7.20)s, P< 0.05, P<0.01). The contents of NO or MDA in cerebral cortex and hippocampus of the experimental groups (NO in cerebral cortex (3.35 ± 0.40) μmol/gprot, (4.50 ± 0.41) μmol/gprot, NO in hippocampus (2.24 ±0.18) μmol/gprot,(3.15 ±0.21) μmol/gprot, MDA in cerebral cortex(1.34 ±0.44)nmol/mgprot, (2.39 ± 0. 18) nmol/mgprot, MDA in hippocampus (0. 13 ± 0. 07) nmol/mgprot, (0.53 ± 0. 10)nmol/mgprot) were lower than those of the control group (NO in cerebral cortex (3.35 ± 0. 40) μmol/mgprot,(5.03 ± 0.44)μmol/mgprot, NO in hippocampus (2.93 ± 0. 31) μmol/gprot, (3.38 ± 0.24) μmol/gprot, MDA in cerebral cortex (2.24 ± 0.26) nmol/mgprot, (4.21 ± 0.21) nmol/mgprot, MDA in hippocampus (0.47 ± 0.29)nmol/mgprot, (1.33 ± 0. 187) nmol/mgprot, P < 0.05, P < 0. 01) respectively and the contents of SOD in cerebral cortex and hippocampus of the experimental groups (in cerebral cortex (763.95 ± 214.36) U/mgprot, (491.33 ±35.85) U/mgprot, in hippocampus (817.02 ± 232.39) U/mgprot, (644.85 ± 28.02) U/mgprot) were higher than those of the control group(in cerebral cortex (556.50 ± 101.51) U/mgprot, (327.35 ± 30.54) U/mgprot, in hippocampus (279.74 ± 117.02) U/mgprot, (108.75 ± 15.52) U/mgprot, P < 0.05, P< 0.01) respectively. Conclusion Melatonin is effective in improving the ability of learning and memory in the rats of noise stress,possibly by inhibiting the increase of NO and MDA and increasing the SOD activity in cerebral cortex and hippocampus of the rats.
9.Cannulated screws combined with transposition of cuboid periosteal flap pedicled with fascia and lateral tarsal artery in the treatment of talus neck fractures

Objective To investigate the surgical techniques and clinical efficacy of cannulated screws combined with transposition of cuboid periosteal flap pedicled with fascia and lateral tarsal artery in the treatment of talus neck fractures.Methods From March 2008 to June 2011,12 cases with talus neck fractures were treated with cannulated screws combined with transposition of cuboid periosteal flap pedicled with fascia and lateral tarsal artery.External fixation in functional position for 12-14 weeks,and the load time was determined by X-ray fracture healing.Functional results were assessed according to AOFAS (American Orthopaedic Foot and Ankle Society) score at last follow-up.Results Twelve cases were followed up for an average of 24 months (12 to 48 months).One case of skin flap necrosis healed by dressing.All the fractures healed,and the mean healing time was 20 weeks (16 to 24 weeks).AOFAS ankle and hindfoot mean postoperative score was 82.5 (ranging from 55 to 96) points.Among them 4 cases were excellent,good in 5 cases,fair in 3 cases.Two cases presented with mild posttraumatic subtalar arthritis and pain relief after oral anti-inflammatory analgesic.One case with avascular necrosis of talus body,and X-ray displayed sclerosis of talus body,but no collapse,and the patient was told to reduce weight and regularly followed up.Conclusion The application of cannulated screws combined with transposition of cuboid periosteal flap pedicled with fascia and lateral tarsal artery for the treatment of talus neck fractures is an effective treatment,which can improves the blood supply of the talus body and reduces the occurrence of avascular necrosis of the talus.
10.Polymorphisms in HIF1A gene are associated with prognosis of hepatocelluar carcinoma

Objective To assess the association between polymorphisms in HIF1α gene and prognosis of advanced hepatocelluar carcinoma.Methods We collected prognosis data from a cohort of 448 advanced HCC patients treated by transarterial chemoembo-lisation,and used 5ml peripheral blood from patients for extraction DNA.Three SNPs (rs2301 1 13、rs2057482 and rs1 957757 )in HIF1αgene were selected and genotyped.Multivariate Cox proportional hazards model,Kaplan-Meier curve and log-rank test were used for prognosis analyses.Results The variant-containing genotypes (WV+VV)of SNP rs2301 1 13 exhibited a significant associ-ation with a better overall survival in HCC patients who had tumor size smaller than 5 cm (hazard ratio [HR],0.58,95% confidence interval [CI],0.35-0.96,P =0.036).In the patients taken single tumor subgroup,the variant-containing genotypes (WV+VV) of SNP rs2301 1 13 exhibited a significant association with a better overall survival (log-rank P =0.048),comparing to those carrying wild-type genotype.Conclusion Our results suggest that polymorphisms in HIF1αgene may serve as an independent prognosis bio-marker for advanced HCC patient.
