Objective: To explore the association of survivin gene with hepatocellular carcinoma(HCC) in Han nationality in east China. Methods: A case-control study was designed, which included 176 HCC cases and 196 healthy controls, all from Haimen city of Jiangsu province. The rs1042489 and rs9904341 loci of survivin gene were genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique, and the association of the genotypes and haplotypes of the two loci with HCC was analyzed under different genetic models. Results: Univariate analysis showed no significant difference in the genotypes or allele distribution between HCC case group and the control group under different genetic models in either rs9904341 or rs1042489 loci (P>0.05). Linkage disequilibrium (LD) analysis showed that the two loci were in LD (χ2 = 4.777, P = 0.03), with the D' value being 0.188 and 0.183 in the case and control group, respectively. The recessive genetic model was found to be the optimal model by multivariate Logistic regression, according to the lowest value of Akaike's information criteria (AIC). With no rs9904341C-rs1042489T (C-T) haplotype as reference, it was found that the haplotype of C-T from the two loci was associated with a lower risk for HCC under the recessive genetic model, with the factors such as alcohol drinking and HBV infection controlled (OR = 0.48, P = 0.04), and no other haplotypes were found associated with HCC. Multivariate analysis showed that HBsAg+ and hepatitis B were the risk factors for HCC, but without haplotype-environment interaction. Conclusion: No association has been found for rs9904341 or rs1042489 in survivin gene with HCC in Han nationality in east China, and rs9904341C-rs1042489T might be a protective haplotype for HCC.

Objective: To transfect a recombinant short hairpin RNA(shRNA)expression vector targeting Livin gene isoform(BIRC71,BIRC72)into the cervical cancer cell line(Hela cell),in an attempt to observe RNAi-mediated silen- cing on Livin gene and the induction of Hela apoptosis.Methods: Hela cells were transfected with the recombinant plas- mid pGenesil-1-BIRC71,pGenesil-1-BIRC72 and pGenesil-1-HK via Lipofectamine~(TM)2000.The expression levels of Livin was determined in Hela cells before and after transfection by fluorescence quantitative real-time PCR and Western blotting. Objective To evaluate the therapy and turnover of Kawasaki disease(KD)with glucose-6-phosphate dehydrogenase(G-6-PD)deficiency.Methods Six hundred and twenty-four patients with KD were selected including 32 patients who had G-6-PD defected.The same dose intravenous immunoglobulin(IVIG)was used in all 624 patients(2 g/kg),but used Dipyridamole in 32 patients had G-6-PD deficiency which replaced the role of acetylsalicylic acid(ASA)after acute period.The coronary artery of these patients were checked and followed-up through echocardiograph.The turnover of 32 patients with G-6-PD deficiency and 356 case selected randomly from all the KD patients were compared.SPSS 10.0 software was used to analyze the data.Results In 32 cases of KD with G-6-PD deficiency,4 children had coronary aneurysm(12.5%).After 6-12 months follow-up,the coronary lesions were recovered in 62.5% children,improved in 21.9% children and not improved in 15.6% childern,which were not significantly different from all the KD patients(Z=-1.604 P=1.09).Conclusions IVIG and Dipyridamole are feasible in treating KD with G-6-PD deficiency.J Appl Clin Pediatr,2009,24(1):26-27

Objective To investigate the correlation of interleukin-1 family genotypes,including interleukin-1 (IL-1α,IL-1β) and interleukin-1 receptor antagonist (IL-1Ra),with coronary heart disease (CHD) and serum lipoprotein level in the elderly.Methods Interleukin-1 family genotypes were detected in 318 elderly controls and 329 elderly CHD patients by polymerase chain reaction and restriction fragment length polymorphisms method.Serum levels of lipoproteins were inspected simultaneously.Results The TT and Ⅰ / Ⅰ or Ⅰ/Ⅳ genotype frequency of IL-1Ra was 90.3％ in elderly CHD patients,but 82.4％ in controls.Carriers with TT,Ⅰ / Ⅰ or Ⅰ/Ⅳ genotype of IL-1Ra were at an increased risk with an odds ratio of 1.98 in elderly CHD patients as compared with controls (x2=8.55,95％ CI:1.25-3.16).The TT and Ⅰ/Ⅰ or Ⅰ/Ⅳ genotype frequency of IL-1Ra was 96.2％ in elderly CHD patients with acute coronary syndrome,but 84.8％ in elderly CHD patients with stable angina.Carriers with TT,Ⅰ / Ⅰ or Ⅰ/Ⅳ genotype of IL-1Ra were at an increased risk with an odds ratio of 4.54 in acute coronary syndrome group as compared with stable angina group (x2=12.17,95％CI:1.81-11.36).The CT or TT genotype frequency of IL-1α-889 was 22.8％ in acute coronary syndrome group,but 7.6 ％ in stable angina group.Carriers with CT or TT genotype of IL-1α-889 were at an increased risk with an odds ratio of 3.59 as compared with stable angina group (x2 =14.93,95％CI:1.82-7.03).There were no significant differences in levels of serum lipoproteins among the different genotypes (P＞0.05).Conclusions In elderly patients with coronary heart disease,IL-1α(-889) CT or TT genotype carriers are at high risk for acute coronary syndrome,but IL-1Ra CC,TC,Ⅰ / Ⅱ or Ⅱ / Ⅱ genotype carriers are at a low risk for CHD or severe CHD.

Objective To investigate the effects of three procedures for hysterectomy.Methods The clinical characteristics of 370 cases who respectively received hysterectomy by total transabdominal hysterectomy(TAH,150 cases),transvaginal hysterectomy(TVH,120 cases),or laparoscopically assisted vaginal hysterectomy(LAVH,100 cases)were analyzed and their effects and outcomes and the expenses for medical treatment were compared from 2000 January to 2005 December.Results The average operation time of TAH,TVH and LAVH was(78?28) minutes,(102?35)minutes,(119?28)minutes respectively.The average operation time of TAH was significantly shorter than that of TVH and LAVH groups(P＜0.01).The volume of hemorrhage during operation in TAH group was less than that of other two groups significantly[TAH(53?29)ml,TVH(110?35)ml,LAVH(93?50 )ml](P＜0.01).The average time of bowel function recovery in TAH group[(42?7)hours]was significantly longer than that of TVH[(23?5)hours]or LAVH[(25?4) hours]groups(P＜0.01).The average hospital stay after opera- tion in TAH[(8.9?1.1)days]group was significantly longer than that of TVH[(5.7?2.4)days]or LAVH [(6.3?1.9)days] groups(P＜0.01).The average expenses for medical treatment in LAVH[(7273?938)yuans] group was significantly more than that of TAH[(5594?308)yuans]or TVH[(5450?167)yuans]groups(P＜0.01).But the lesion and the complication after operation of three groups had no difference.Conclusion Different procedures for total hysterectomy have their own advantage and disadvantage.The operation procedure must take ac- count of the economy,echnique of medical treatment and the condition of the hospital.

ObjectiveTo study the effects and mechanism of survivin gene on invasion of prostate cancer cell.MethodsAfter PC-3 prostate cancer cell lines were transfected by survivin small interfering RNA (siRNA), the mRNA and protein of survivin and matrix metalloproteinase-2 and-9 were determined by real time RT-PCR and western blot assay, respectively. The anchorage-independent growth was examined by clon formation in soft agar, and invasion ability was evaluated by boyden chamber model.The invasion ability of cancer cells in vivo was determined by nude mice model. ResultsThe results from clon formation in soft agar showed that the colonies numbers of group of 3.125,6.250 and 12.500 nmol/L of siRNA were 17.8±1.6,13.6±1.5 and 8.8±1.4, and the control group was 22.6±1.8(P<0.05). The results from boyden chamber model exhibited that the cells numbers crossing filter membrane in group of 3.125,6.25 and 12.5 nmol/L of siRNA were 33.6±2.1,19.5±1.9,8.1±1.83, and the control group was 49.4±2.3(all P<0.05). The results in vivo showed that cancer cells of control groups invaded into striped muscle and blood vessel, and there were no these phenomenons in transferred group with survivin siRNA. Survivin siRNA could reduce expression level of MMP-2 and MMP-9 in prostate cancer cells(P<0. 01).Conclusions Survivin-directed RNA interference can inhibit invasion of human prostate cancer cell through down-regualting MMP-2,-9 genes.

Objective To study the relationship between serum leptin and thyroid function by investigating the change of serum leptin concentration in children with abnormal thyroid function (hyperthyroidism and hypothyroidism).Methods The levels of serum leptin was determined by radio-immunoassay(RIA) in 20 cases of hyperthyroidism, 17 cases of hypothyroidism, 25 cases of normal controls, respectively .Meanwhile, the serum levels of free triiodothyronine(FT 3), free thyroxine(FT 4) and thyroid-stimulating hormone(TSH) were measured by micro-particle-chemiluminescence immunoassay.Results The levels of serum leptin in hypothyroidism group before treatment was significantly lower than that of normal control groups (P0.05).Conclusions The levels of serum leptin in hypothyroidism markedly descend and no change in the hyperthyroidism. Thyroid hormone can promote the secretion of serum leptin eligibly.

Background Meibomain gland is a specially differentiated sebaceous gland lying in the tarsus of upper and lower eyelid.The morphological changes of the gland is associated with a variety of ocular surface diseases.Studying the relationship of morphological and functional change of meibomain gland with ocular surface is of great significance.Objective This study was to observe the change of morphology,structure and function of meibomain gland over aging and investigate the assocation of meibomain gland abnormality with ocular surface.Methods A prospectively cases-observational study was performed.Ninety-three eyes of 93 patients with age-related cataract aged 45 and older were enrolled in Shanxi Eye Hospital from March to September 2016 under the informed consent.The patients were divided into 45 to 59-year group and ≥60-year group according to age or meibomian gland loss ≥ 1/3 group and meibomian gland loss ＜ 1/3 group.The ocular anterior segment,lid margin,meibomian gland orifices and lipids traits were examined by slit-lamp microscope.The ocular surface symptoms were assessed and scored by Ocular Surface Disease Index (OSDI) scale.The break-up time of tear film (BUT),tear meniscus height,meibomian gland dropout degree,conjunctival hyperemia and corneal fluorescence staining scores were measured using ocular surface analyzer.Results No dry eye symptom was complained in all the subjects,and their OSDI scores were ＜12.No abnormal changes at the lid margin and the muco-cutaneous junction were observed.No abnormality of the meibomian gland orifices,the lipids traits and drainage was observed under the slit-lamp microscope.BUT was shortened in 42 eyes (45.16％);tear meniscus height was lowed in 52 eyes (55.91％);meibomian gland loss range was ≥1/3 in 58 eyes (62.27％).The meibomian gland loss scores were 1.65±0.79 in the 45 to 59-year group and 1.86±0.72 in the ≥60-year group,showing an insignificant difference between them (t =1.301,P =0.197).But when coming to the correlation analysis,a positive correlation was found between meibomian gland loss scores and age (rs =0.323,P=0.002),and no correlations were seen between age and BUT or tear meniscus height (rs =0.154,P =0.141;rs=-0.024,P =0.821).In addition,meibomian gland loss scores showed a negative correlation with mean BUT (rs =-0.251,P =0.015).The eye number of BUT abnormality in the meibomian gland loss ≥ 1/3 group was more than that in meibomian gland loss ＜1/3 group (P =0.018).Conclusions Meibomian gland loss is more serious over aging in middle aged and elderly population,and serious meibomian gland loss increases the risk of tear film instability.The early meibomain gland dysfunction-like signs occur prior to symptoms,which should raise concern in clinical work.
