2.Changes of antimicrobial susceptibility of Pseudomonas aeruginosa induced by disinfectants

Objective To investigate the minimal inhibitory concentration (MIC) of Pseudomonas aeruginosa to antibacterial agents which were induced by disinfectants.Methods The susceptibility test of 68 strains from clinic was determined by VITEK-2 system.The MIC of povidone iodine,glutaraldehyde and chlorhexidine was determined by agar dilution method.68 strains were induced by different concentrations of three kinds of disinfectants for eight times.The concentration of three kinds of disinfectants was lower than their MIC.The MIC before and after inducement was determined by micro dilution.In the mean-while,strains of their MIC raised to antibiotics were inoculated on plate agar containing the PAβN which was the efflux pump inhibitor,in order to determine the phenotype of efflux pump.Results Six strains,MIC in 68 strains significantly changed after induction of disinfectants.The MIC of these six strains was higher than pre-induced to some antibacterial agents such as FQNs,cephems and carbapenem.The induction of povidone iodine and gilutaraldehyde were more obvious,and five strains,phenotype of efflux pump were positive after being induced by these two disinfectants.Conclusions The antimicrobial susceptibility of some Pseu-domonas aeruginosa will change after being induced by some disinfectants and the efflux pump will play an important role in this process.So the reasonable application of disinfectants were indispensable,in order to decrease the resistance and the opportunity of nosocomial infection.
3.Micro-implant anchorage for orthodontic intervention of buccal impacted maxillary canine

Micro-implant anchorage was used for orthodontic intervention of 3 patients with buccal impacted maxillary canine,good clinical outcome was obtained.The micro-implant anchorage may provided a new approach for the treatment of this kind of teeth.
4.Intraoperative opioid-sparing effect of different duration transcutaneous electrical acupoint stimulation in video-assisted thoracoscopic lobectomy

Objective To evaluate the intraoperative opioid?sparing effect of different duration transcutaneous electrical acupoint stimulation ( TEAS ) in video?assisted thoracoscopic lobectomy. Methods Seventy?five patients, aged 18-64 yr, weighing 40-96 kg, of ASA physical status Ⅰ or Ⅱ, scheduled for elective video?assisted thoracoscopic lobectomy under general anesthesia, were randomly divided into 3 groups (n=25 each) using a random number table: control group (group C), 30 min of stimulation before induction of anesthesia group ( group B) , and stimulation throughout surgery ( group T) . In group B, the patients received TEAS ( frequency 2∕100 Hz ) on acupoints Xinshu ( BL15 ) , Feishu (BL13), Neiguan (PC6), Hegu (LI4) on the operated side starting from 30 min before induction of anesthesia until the beginning of induction, and the intensity was the maximum current that could be tolerated. The intensity for Neiguan ( PC6) and Hegu ( LI4) was 6-12 mA, and for Xinshu ( BL15) and Feishu ( BL13 ) was 9-18 mA. In group T, the patients received TEAS on the four acupoints mentioned above starting from 30 min before induction of anesthesia until the end of surgery. The patients had the electrodes applied, but received no stimulation in group C. After anesthesia was induced with propofol?sufentanil?cisatracurium, double lumen endotracheal tube was inserted. Propofol was given by target?controlled infusion to maintain BIS value within the range of 40-60. Cisatracurium was infused continuously to facilitate muscle relaxation. The infusion rate of remifentanil was adjusted to maintain analgesia nociception index value within the range of 50-70. The intraoperative consumption of remifentanil ( the intraoperative consumption of sufentanil was converted to the consumption of remifentanil producing the equivalent effect by 1∶ 10) was recorded. Results Compared with group C, the intraoperative consumption of remifentanil was significantly decreased in B and T groups. The intraoperative consumption of remifentanil was significantly lower in group T than in group B. Conclusion TEAS on Xinshu ( BL15 ) , Feishu (BL13), Neiguan ( PC6) and Hegu acupoints throughout surgery and for 30 min before induction of anesthesia significantly reduces intraoperative opioid consumption in the patients undergoing video?assisted thoracoscopic lobectomy, while TEAS throughout surgery provides better effect.
5.Clinical value of analgesia/nociception index in evaluating analgesic effect during lobectomy performed via video-assisted thoracoscope

Objective To primarily investigate the clinical value of analgesia/nociception index (ANI) in evaluating the analgesic effect during lobectomy performed via video-assisted thoracoscope.Methods Forty ASA physical status Ⅰ or Ⅱ patients,aged 25-64 yr,weighing 45-80 kg,undergoing elective lobectomy performed via video-assisted thoracoscope,were enrolled in this study.After induction of anesthesia with propofol,sufentanil and cisatracurium,patients received double lumen endotracheal intubation.Anesthesia was maintained with targetcontrolled infusion of propofol,and iv infusion of remifentanil and cisatracurium.The concentration of propofol was adjusted to maintain the bispectral index (BIS) value in the range of 40-60.ANI,HR,systolic blood pressure (SBP),diastolic blood pressure (DBP) and BIS value were recorded within 5 min before and after the predefined time points including posture change between lateral and supine position,ventilatory pattern change between onelung and double-lung ventilation,skin incision and trocars insertion,lymph node dissection and pleural lavage.At skin incision and during trocars insertion,lymph node dissection and pleural lavage,the development of hemodynamic responses (increase in HR and SBP > 20% of baseline value) were recorded.Results The incidence of hemodynamic responses was 100% at skin incision and trocars insertion,and 84 % during No.4,7,10 groups of lymph node dissection and after pleural lavage and difference was found in ANI during these stimuli.ANI was significantly decreased within 5 min after skin incision,trocars insertion,No.4,7,10 groups of lymph node dissection and pleural lavage than that before the procedures (P < 0.05).The BIS value was maintained at 40-60,and no significant changes were found between before and after the procedures (P > 0.05).No significant changes were found in ANI,HR,SBP,and DBP between before and after the changes of posture and respiratory pattern (P > 0.05).Conclusion ANI can be used to evaluate the analgesic effect during lobectomy performed via video-assisted thoracoscope in patients and is unaffected by the changes of posture and ventilatory pattern.
6.Effects of montmorillonite and mannitol on early pulmonary fibrosis in rats with acute paraquat poisoning

Objective To observe the intervene effects of montmorillonite powder and mannitol on rat with acute paraquat poi-soning by intragastric administration .Methods 40 male SD rats were divided into four groups randomly ,control group(C) ,pa-raquat group(P) ,PT1 groups were treated with montmorillonite powder and mannitol gavage after paraquat exposure one hour ,PT2 group was treated after paraquat exposure four hours .P ,PT1 ,and PT2 groups were exposed to paraquat 50 mg/kg .one and four hour later ,PT1 and PT2 group rats were given 7 g/kg montmorillonite-mannitol solution respectively ,C group and P group with sa-line instead .then to observe general state in rats .Give rats run training before and after intragastric administration ,comparison the fall times from the treadmill among groups before and 5 ,10 ,15 days after administration ,rats were killed in 15 th days exposed pa-raquat ,collect their lung tissue ,hydroxyproline content were measured in lung tissue of rat by alkaline hydrolysis ,observe the path-ological changes by HE staining in rat lungs .Results the general condition were poor in P ,PT1 and PT2 group rats .And those mean food intakes were significantly lower than the control group (P<0 .01) .there are no significant of the weight gain and the fall times from the treadmill before and 5 th ,10 th after administration among four groups(P>0 .05) ,but P group′s fall times increased significantly than the others group′s at 15 th day after exposed(P< 0 .05) .the all group rat lung tissue hydroxyproline overall difference significant(P<0 .01) ,from the two-two compared results ,three paraquat exposed groups higher than control group ,and P group also higher than PT1 and PT2 group(P<0 .05) .From the lung tissue slices ,P ,PT1 and PT2 group showed significant pul-monary fibrosis than C group ,and the P group′s was the most severe .Conclusion Pulmonary interstitial fibrosis may be alleviated by early use of montmorillonite powder and mannitol on paraquat poisoning rats .
7.Effects of Bingcha Embolus on the proliferation,apoptosis and cell cycle of astrocytes

AIM: To study the effects of Bingcha Embolus on the proliferation, apoptosis and cell cycle of astrocytes in vitro in terms of serum pharmacology. METHODS: The cerebral cortex astrocytes of neonatal Sprague-Dawley rats were cultivated in vitro, and the purified astrocytes were randomly devided into Bingcha Embolus groups (high dose, middle dose, low dose) and control group after being identified by means of immunocytochemical method. After the incubation of 48 h with sera, MTT was employed as a proper method to detect the proliferation of astrocytes. The ratio of apoptosis and different cell cycle phases were measured by Flow Cytometry. Objective To study the evaluation between the X-ray findings and ultrasound(US) manifestations of upper digestiveobstruction in infancy.Methods X-ray and US manifestations of 20 cases of infant with upper digestive obstruction proved by operationwere analysed respectively.Results In 20 cases,12 patients had pyloric stenosis,3 duodenal stresia,3 midgut malformation,2 hiatalhernia.Pyloric stenosis was diagnosed by X-ray in 11 cases,by US in 9,misdiagnosed by US in 1 case.Duodenal stresia was observed by X-ray and US in 2 cases separately,not observed by X-ray and US in 1 case separately.Midgut malformation was demonstrated by X-ray and US in 1 case separately,not demonstrated in 2 cases separately.Hiatal hernia was found by X-ray in 2 cases,misdiagnosed by US in 2 cases.Conclusion The two methods can't substitute with each other but complement.X-ray is better than US in the diagnosis of pyloric stenosis and hiatal hernia.US is better than X-ray in the diagnosis of midgut malformation.
9.Study on structure-activity relationship of flavonoids' multidrug resistance-associated protein inhibitory activity.

To study the quantitative structure-activity relationship (QSAR) between the stuctures of 29 flavonoids and the inhibitory activity of their multidrug resistance-associated protein (MRP) 1 and 2 by using the comparative molecular similarity index analysis (CoMSIA). By studying the impact of the combination of different molecular force fields, researchers obtained the molecular force fields that played an important role in inhibiting the activity of MRP1 and MRP2, built the optimized QSAR model, and discussed the structural modification method for flavonoids' multidrug resistance-associated protein inhibitor. The results of the study could not only provide the guidance for new drug R&D, but also help partially discuss the synergy mechanism between MRP1 and MRP2 receptors and traditional Chinese medicines containing flavonoids.

10.Discovery of potential nicotinic acid receptor agonists from Chinese herbal medicines based on molecular simulation.

Nicotinic acid could increase high density lipoprotein and reduce serum total cholesterol, low density lipoprotein cholesterol and triglycerides in human bodies, thus is frequently applied in treating low high-density lipoprotein cholesterol and hypertriglyceridemia in clinic. However, according to the findings, nicotinic acid could also cause adverse effects, such as skin flush, beside its curative effects. In this study, bioisosterism, fragment-based search and Lipinski's Rule of Five were used to preliminarily screen out potential TCM ingredients that may have similar pharmacological effects with nicotinic acid from Traditional Chinese medicine database (TCMD). Afterwards, homology modeling and flexible docking were used to further screen out potential nicotinic acid receptor agonists. As a result, eleven candidate compounds were derived from eight commonly used traditional Chinese medicines. Specifically, all of the candidate compounds' interaction with nicotinic acid receptor was similar to nicotinic acid, and their docking scores were all higher than that of nicotinic acid, but their druggability remained to be further studied. Some of the eight source traditional Chinese medicines were used to lower lipid according to literature studies, implying that they may show effect through above means. In summary, this study provides basis and reference for extracting new nicotinic acid receptor agonists from traditional Chinese medicines and improving the medication status of hyperlipidemia.
