1.Screening and preliminary research of an actinomyces strain with antitumor effect

The apoptosis was observed under a fluoroscope when the tested tumor cells were treated with the supernatant derived from the liquid culture of an actinomyces strain AK 11, which had showed cell toxicity by means of MTT assay, and was further confirmed by the correspondent band of ”DNA ladder” in the agarose gel electrophoresis. In a bioassay employed nude mouse, the pure compound produced by the strain AK 11 showed tumor inhibition in rates of 52.01% and 33.64% corresponding to the dosages of 0.016 ?g?kg -1 and 0.008 ?g?kg -1 ip, respectively. According to the indexes IC 50 of human hepatocarcinoma SMMC7721 and stomach cancer MGC 803 cells, it was estimated that the inhibition effect of the pure compound was slightly stronger than that of Adriamycin. The results showed high antitumor effect of the tested compound from the strain AK 11. When the cells were treated with AK 11, it was found by flow cytometry analysis that cell division was ceased at S phase due to the DNA synthesis block .
2.Intralesional curettage and wide excision for treatment of giant cell tumors (GCTs) of the distal radius: A Meta-analysis.

OBJECTIVETo search all studies that had been published in the world with regarding to the effectiveness of the extent of intralesional curettage and wide excision for recurrence rate and complications and comparative functional outcomes in patients with giant cell tumours (GCT) of the distal radius and analyze them which were in high quality by means of Meta analysis, in order to give some evidences for the choice of method dealing with giant cell tumors GCT in surgery.
METHODSCochrane central register of controlled trials(Issue 8 2014), PubMed(1970-01-01/2013-01-01), Ovid (1970-01-01/2013- 01-01), Elsevier (1970-01-01/2013-01-01), CNKI (1970-01-01/2013-01-01) were searched. Including intralesional curettage and wide excision were performed to treat giant cell tumors (GCTs) of the distal radius in the literatures, selecting on meet eligibility in the standard literatures underwent strict quality assessment. The Meta-analysis was performed with software RevMan5.0 from the Cochrane collaboration. Additionally, the analysis checked the heterogeneity of data. The effectiveness of the extent of intralesional curettage and wide excision for recurrence rate and complication in patients with giant cell tumours of the distal radius were evaluated and Odds Ratio was calculated.
RESULTSSeven relevant articles were identified involving total 163 cases. Among them, 92 cases were intralesional curettage (PMMA, n = 54; bone graft, n = 33; no PMMA or bone grafts, n = 5) and 71 cases were wide excision. The patients in the intralesional curettage group had a higher recurrence rate [OR = 3.87, 95% CI (1.42, 10.53)],especially for Campanacci grade 3 GCTs [OR = 10.12, 95% CI (1.57, 65.27)], yet fewer major complications [OR = 0.13, 95% CI (0.04, 0.40)] than the wide excision group. The use of PMMA versus bone graft did not affect the recur- rence rate [OR = 0.96, 95% CI (0.26, 3.56)]. By selecting the system evaluation of MSTS, the VAS and dynamometer, the result showed that the intralesional curettage group was equivalent or preferable to wide excision in terms of function rehabilitation.
CONCLUSIONBased on data obtained from the limited number of studies available, intralesional curettage appears to be moreappropriate for the treatment of local lesions (Grade 1 and 2) than Grade 3 GCTs of the distal radius. Moreover, PMMA was not additionally effective as an adjuvant, the intralesional curettage group was found to be equivalent or preferable to wide excision in terms of function rehabilitation.

3.Effects of Fengshi Bitong Prescription on Cytokines in Serum and Synovial Fas/FasL mRNA Expression in Rats with Collagen-Induced Arthritis

Objective To observe the effects of Fengshi Bitong Prescription (FSBT) on cytokines in serum and Fas/FasL mRNA expression system in synovium; To discuss relevant mechanism of action. Methods Rats were given foot intradermal injection of bovine type Ⅱ collagen emulsion to establish collagen-induced arthritis (CIA) model. After successful modeling, they were randomly divided into 6 groups: normal control group, CIA model group, tripterygium glycosides group, FSBT high-, medium-, and low-dose groups. Each medication group was given relevant medicine for gavage for 14 d. The weight and arthritis scores of CIA rats were observed. Serum levels of IL-1β, IL-15, and TGF-β1 were detected by ELISA. Expressions of Fas and FasL mRNA in synovium of CIA rats were detected by real-time PCR. Results Compared with model group, the foot arthritis score and serum levels of IL-15 in FSBT high- and medium-dose groups significantly decreased (P<0.01), and the serum level of TGF-β1 significantly increased (P<0.05). Expression of Fas significantly decreased and FasL significantly increased in FSBT high-dose group (P<0.05). Conclusion FSBT may have certain immune regulation effects on rheumatoid arthritis. Its mechanism may be in regulating Fas/FasL apoptosis system, thereby inducing apoptosis of synovial cells and inhibiting synovial hyperplasia.
4.Development of hip joint simulators.

Currently, hip replacement is an effective treatment for some hip joint diseases. The friction and wear of a prothesis in the human body are the main causes for the failure of joint arthroplasty. It is, therefore, very important to simulate the working conditions of a hip prosthesis in order to get an optimal design and successful clinical applications. This article summarizes wear testing methods of hip prostheses and the developing status of their simulators. Three key aspects of the simulators, i.e., the structures, motions and lubrications, are analyzed in detail. At the end, the developing trend of the simulators are discussed.

We present a rare case of colonic metastasis of renal cell carcinoma (RCC) and review the literature.A 54-year-old male was referred to our hospital with a history of bloody stools and fever.A dght kidney tumor measuring about 10 cm in diameter was found by abdominal computed tomography.Right radical nephrectomy and a right hemicolectomy with ileotransversostomy were performed.Pathological diagnosis was chromophobe RCC with sarcomotoid change involving the colon.Chromophobe RCC with sarcomotoid change is very rare.
7.Melatonin attenuates the intensity of β-endorphin immunoreactivity in the arcuate nucleus of rat hypothalamus

The present study was undertaken to observe the effect of exogenously administered melatonin on the intensity of β-endorphin (β-Ep) immunoreactivity of the neuron in the arcuate nucleus of rat hypothalamus with an aim to explore the possible mechanisms of the analgesic effect of melatonin. The experimental rats were divided into two groups, one injected intraperitoneally with melatonin (110 mg/kg) and the other with only a vehicle. One hour after injection, the brain was processed for coronal sections, which were stained with immunohistochemical ABC technique. The integral optical density (IOD) and mean optical density (OD) of the stained sections were measured with a computer-assisted image-processing and analytical system. β-Ep immunoreactivity was much decreased in the sections treated with melatonin and the IOD and OD were also decreased significantly (P<0.01; P<0.05). The above results suggest that melatonin may result in a decrease of β-Ep content in the arcuate nucleus, as a result of increased β-Ep release induced by administration of melatonin. It is likely that the analgesic effect of melatonin may be in part mediated by the release of β-endorphin from the arcuate nucleus.
8.Optimization of matrix formulation of effective parts cataplasm of Pogostemon cablin by uniform design.

OBJECTIVETo optimize the matrix formulation of the effective part Cataplasm of Pogostemon Cablin.
METHODThe optimal preparation prescription was selected by U17 (17(11)) uniform design,and the tacking strength, cohesive strength and transdermal speed constant were used as test indexes. The equations of three test indexes were established by SPSS. With analysis of the contribution of factors by SPSS regression, the optimal matrix formulation was acquired.
RESULTThe optimal matrix formulation is carbopol U10-NoveriteTM7s-glycerine-sorbitol-kaolin-citric acid-aluminum trichloride (1.0:5.0:20:2.0:2.0:0.25:0.2).
CONCLUSIONThe matrix has good adhesive property, proper drug release rate, desirable hemocompatibility with the extractions of Pogostemon cablin.

9.Literature survey on botanical origin and clinical application of traditional Tibetan medicine "Shengdeng".

10.Determination of puerarin, daidzin and daidzein in root of Pueraria lobata of different origin by HPLC.

OBJECTIVETo develop an HPLC method to determine Puerarin, Daidzin and Daidzein in Pueraria lobata.
METHODThe separation was performed in a SYMMETRY C18 column with a mobile phase of methanol-1% HAC solution (gradient elution), the The detection wavelength being 250 nm.
RESULTThe average recovery was respectively 101.7%, 100.7% and 101.7% (n = 3), RSD 0.43%, 0.82% and 1.50% (n = 3) for Puerarin, Daidzin and Daidzein.
CONCLUSIONThis method is suitable for the determination of Puerarin, Daidzin and Daidzein in Pueraria lobata and its preparation.
