Objective : To examine the effect of asbestos exposure on oxidative stress, so as to provide insights into the elucidation of pathogenesis and management of asbestos-related diseases. Methods : Totally 245 subjects were recruited from an asbestos manufacturing area in Zhejiang Province, and their gender, age and history of asbestos exposure were collected through a questionnaire survey. The serum levels of 8-hydroxy-2'deoxyguanosine ( 8-OHdG ), glutathione ( GSH ), malondialdehyde ( MDA ), superoxide dismutase ( SOD ) and total antioxidative capacity ( TAOC ) were measured using an enzyme-linked immunosorbent assay ( ELISA ), and the levels of catalase ( CAT ), peroxiredoxin 2 ( PRX2 ), SOD1, SOD2 and thioredoxin-1 ( TRX1 ) were detected in peripheral white blood cells ( WBCs ) using a liquid-chip assay. Multivariable linear regression analysis was performed to identify the association between asbestos exposure and oxidative stress parameters. Results : There were 50 subjects without a history of asbestos exposure (unexposed group), 102 subjects with asbestos exposure for less than 10 years ( AE<10-year group ) and 93 subjects with asbestos exposure for 10 years and more ( AE≥10-year group ). No significant differences were found among the three groups in terms of age, gender, proportion of smokers or proportion of alcohol consumers ( P>0.05 ). Significantly higher 8-OHdG and MDA in serum, and higher PRX2 in peripheral WBCs were detected in the AE≥10-year group than in the unexposed group ( P<0.05 ); lower GSH and TAOC in serum, and lower CAT in peripheral WBCs were detected in the AE≥10-year group than in the unexposed group ( P<0.05 ); higher 8-OHdG and MDA in serum, and higher PRX2 in peripheral WBCs were detected in the AE≥10-year group than in the AE<10-year group ( P<0.05 ). Multivariable linear regression analysis showed that asbestos exposure significantly correlated with 8-OHdG, MDA and TAOC in serum, and CAT and PRX2 in peripheral WBCs ( P<0.05 ). Conclusion Asbestos exposure may induce the oxidative stress damage, suggesting that oxidative stress may be involved in asbestos-related diseases.

An analytical procedure for the determination of water-and methanol-extractable pentachlorophenol ( PCP) in soils using vortex assisted liquid-liquid extraction ( VALLE ) and gas chromatography ( GC ) was developed. Significant extraction parameters, such as liquid-liquid ratio and vortex speed were optimized. The recovery of PCP was the highest ( 97. 4%) at the vortex rotation speed of 2000 r/min, with good reproducbility and a small relative sdandard deviation ( RSD, 0. 5%), with regard to the volume ratio of derivatization solution to n-hexane, the recovery of PCP was 103% with an RSD of 0. 7% at 10:4. The linearity of the calibration ranged from 1. 25 μg/L to 4000 μg/L, with a correlation coefficient ( R2 ) of 0. 9999. The detection limit of the PCP in water was below 0. 2 μg/L. Compared with traditional liquid-liquid extraction ( LLE) and solid phase extraction ( SPE) , the VALLE method was more simple and more economic in terms of chemical consumption, also with a recovery of 96. 8% and a RSD of 3. 7%. Four different soils were used to check the reliability of this method. The soils were respectively extracted with water once and methanol three times. The total recoveries were 89. 5%-98. 9% and 88. 7%-98. 4% at the PCP spiking level of 1 and 10 mg/kg, respectively. It is concluded that the VALLE method satisfied the extraction and determination of water and methanol extractable PCP in soils with varying PCP polluting rates.

Objective To discuss nursing ways to prevent frontal pressure sores for children after cochlear implantation.Methods A total of 267 cases with sensorineural deafness were selected and divided into the control group (admitted from December 2012 to July 2013,115 cases) and the observation group (admitted from August 2013 to March 2014,152 cases) randomly.Skin preparation was done routinely,general nutritional condition and frontal skin condition were evaluated before operation.The control group used conventional elastic bandage after surgery.The observation group used Allevyn Thin foam dressing to cover the forehead skin before pressure dressing by elastic bandage.The incidence of frontal pressure sores was compared between two groups.Results Two cases occurred frontal pressure sores in the observation group and 31 cases in the control group,x2=39.736,P ＜ 0.01.Patients in the control group with pressure sore focused within 1-2 years old,totally 18 cases.Conclusions Nurses should attach great importance to younger children with cochlear implantation to prevent pressure sores.Nursing staff should keep update knowledge of prevention and treatment of pres-sure ulcers,accurately assess skin conditions and risk factors in patients and adopt precautions predictably.

Objective To investigate the influence of a systematic management model in outpatient department on accompanies and their children with amblyopia in regard to compliance to treatments as well as their satisfaction towards the management. Methods One hundred accompanies, whose children met the inclusion and exclusion criteria for amblyopia, were enrolled into the study, equally assigned into two groups,the control group and the experiment group. The systematic management model was implemented in the experimental group. Data were collected before the intervention and one month later using a questionnaire with validity and reliability. Results In terms of socio-demographics and outpatients' diseased status, two groups were well compared. Before the intervention, in addition to the scores of outpatient department environment,there were significant differences between two groups' scores of staff' s attitudes, their knowledge and behaviors related to amblyopia. One month later, the difference was still significant in respect of relevant knowledge and behaviors. Conclusions The proposed systematic management model in outpatient department was found effective on improving accompanies' and their children' s attitudes and their compliance to treatments. It deserves further investigation for its extensive application in outpatient department.

Objective To investigate the expression of γ-aminobutyric acid A receptor α1 subunit (GABAAα1) in the ventrolateral periaqueductal gray (vLPAG) in rats with formalin-induced acute pain. Methods The rats were randomly divided into two groups:control group(group C) and formalin-induced pain group(group F),12 rats in each group:0.9% sodium chloride solution or 2% formaldehyde 50 μL was injected into the ventral surface of right hind paw respectively. The pain scores were recorded for every 5 minutes and the mechanical pain threshold were recorded for every 10 minutes until 1 h. The expression levels of GABAAα1in vLPAG were determined by Western blot analysis in each group.Results The rats in formalin group showed significant nociceptive behaviors immedi-ately, such as paw withdrawal and/or paw licking. Results demonstrated that the rats exhibited a biphasic response to pain. The pain behavior scores in group F were significantly higher than that in group C (P<0.05),and the mechanical pain threshold in group F was decreased after injection compared with group C(P<0.05). The expression of GABAAα1 protein in group F was significantly higher than that in group C (P<0.05).Conclusions The up-regulation of GABAAα1 expression in ventrolateral periaqueductal gray is associated with the decrease of pain threshold in rats with acute pain.

Objective To describe the clinical features of venous diethylene glycol poisoning and to identify factors correlating with such kind of poisoning.Methods Retrospective chart review was performed to analyze the epidemiology,clinical presentation,hepatorenal functions,bemodynam- ics and pathological characteristics of 64 patients with severe liver diseases who received intravenous diethylene glycol.Comparative analyses of correlating factors and causes of poisoning were based on the presence or absence of poisoning.Results Fifteen cases of poisoning were reported.After a 5 day incubation period,all poisoned patients displayed acute renal failure and 11 cases with digestive tract symptoms and(or) symptom exacerbations were noted.Neurological system impairment was observed in 10 cases after 2 weeks.Metabolic acidosis developed in 13 cases.Poisoned patients exhibited signif- icantly lower red blood celI(RBC)[(2.32?0.76)?10~(12)/L],hemoglobin(Hb) [(79.5?23.6)g/L] value and higher white blood cell(WBC)[(9.78?3.75)?10~9/L] count.Renal biopsy of poisoned patients revealed acute tubular necrosis and interstitial nephritis.Twelve poisoned patients died.Sig nifieant differences were found between groups regarding preexisting severe hepatitis,ascites,renal disease and diuretic therapy.Prior to diethylene glycol injections,mean values of neutrophil,blood urea nitrogen(BUN),creatinine(Cr) and calcium and phosphorousions differed significantly between groups.Conclusions Features of venous diethylene glycol poisoning include oliguric acute renal fail- ure,metabolic acidosis,digestive symptoms,nervous system impairment and a high probability of anemia and WBC proliferation.Mortality is high.Correlative factors include preexisting severe liver disease,renal disease and infection.

Objective To investigate the ostengenie potential of adipose-derived stem cells(AD- SCs)when exposed to adenovirns containing hBMP-2 cDNA(Adv-hBMP-2)and offer a choice of cell source for gene therapy and tissue engineering.Methods Human adipose tissues were obtained from patients who received orthopaedic surgery or liposuction.ADSCs were obtained by digesting the adipose tissues.Firstly,flowcytometric analysis was performed for the confirmation of mesenchymal stem cell ori- gin and the surface markers including CD34,CD44,CD68,CD71,CD90,and CD105.The ADSCs were transfected by Adv-hBMP-2 and the effects were tested in vitro,lmmunoprecipitation and Western blotting and ELISA were performed for confirming BMP gone transduction and its stable expression.The transform of ADSCs was assessed by extracellular ALP staining,intracellular ALP spectrophotometry,von Kossa staining and RT-PCR.In the in vivo experiment ADSC-Adv-hBMP-2 cells were injected into the hind limb of nude mice and analyzed radiographically and histologically.Results ADSCs were successfully isolated from human adipose tissues.The isolated ADSCs expressed CD44,CD71,CD90 and CD105 and CD34 and CD68 were absent.The result confirmed the mesenchymal stem cell origin of the cells.West- ern blotting and ELISA confirmed successful and persistent hBMP-2 production by ADSC-Adv-hBMP-2 cells.Extracellular ALP staining,intracellular ALP spectrophotometry,yon Kossa staining and RT-PCR revealed that ADSCs treated with Adv-hBMP-2 had a tendency of transfering into osteoblast.X-ray and H&E sections from hind limb of nude mice injected with ADSC-Adv-hBMP-2 cells confirmed bone forma- tion at 2 weeks.Conclusions Liposuction aspirates contain abundant ADSCs that can be transduced with hBMP-2 gene,and the tranduced ADSCs differentiate into the osteoblast.ADSCs may be an ideal source of mesenchyme-lineage stem cells for gone therapy and tissue engineering.

OBJECTIVEThe aim of this retrospective investigation was to explore the influence of femoral osteoporosis on short-term curative effects of cementless hip arthroplasty and to evaluate the femoral metaphyseal bone mineral density (BMD) for femoral osteoporosis in order to guide prosthesis choice and rehabilitation.

METHODSWe performed 127 total arthroplasty operations between June 1999 to February 2003 and investigated 49 cementless hip replacements with the Metalcancellous cementless Lubeck II system being used in all hips. There were twenty men and twenty-nine women whose mean age at the time of the operation was 60 years (range, 52~81 years). The patients were divided into osteoporosis or normal groups according to the femoral metaphyseal BMD measured preoperatively. The average duration of follow-up was 30 months (range, 8~52 months). We evaluated all of the patients from a clinical standpoint with use of a standard-terminology questionnaire with respect to the short-term curative effects and patients' satisfaction. Hip pain status and functional ability were important indicators of treatment efficacy.

RESULTSHarris hip score and patients' satisfaction in femoral osteoporosis patients who underwent noncemented hip arthroplasty were lower (P=0.004, P=0.03) while the incidence of thigh pain was higher (P=0.03) than the patients with non-osteoporosis.

CONCLUSIONThe higher incidence of pain, as well as the decrease in function experienced by the patients in osteoporosis group, supports the case that cementless arthroplasty is not a better choice for those patients and that we had better select prosthesis based on the femoral metaphyseal BMD.
