1.Determination of Methionine,Vitamin B1 and Vitamin B2 in Lingzhi Erwei Methionine Capsules by HPLC

Objective:To establish an HPLC method for the simultaneous determination of methionine, vitamin B1 and vitamin B2 in Lingzhi Erwei methionine capsules. Methods:The determination was performed on a Ultimate? AQ-C18 column(250 mm × 4. 6 mm, 5 μm,Welch Inc. ) with the detection wavelength at 220 nm. The mobile phase was composed of methanol and 0. 07 mol·L-1 sodium heptanesulfonate solution (14 ml triethylamine diluted with water to1 000ml, adjusting pH to 3. 5 with diluted orthophosphoric acid) with gradient elution, and the flow rate was 1. 0 ml·min-1. Results: Methionine, vitamin B1 and vitamin B2 was in good linearity within the range of 0.527 0-1.510 0 mg·ml-1 - (r =0.999 9), 0.045 52-0.136 56 mg·ml-1 (r =0.999 9) and 0.010 10-0. 060 58 mg·ml-1(r=0. 999 9), respectively. The mean recovery was 100. 5%, 97. 7% and 101. 5% with RSD of 0. 5%, 1. 0%and 1. 4%(n=9) accordingly. Conclusion:The method is simple, accurate, reliable and appropriate in the simultaneous determina-tion of methionine,vitamin B1 and vitamin B2 in Lingzhi Erwei methionine capsules.
2.Studies on a new experimental system of erythrocyte regulating IL-8 in patients with primary hepatocarcinoma

Objective To approach the capacity of erythrocyte regulating IL-8 in patients with primary hepatocarcinoma (PHC) by a new experimental system of hemaimmune. Methods 0.2ml suspension of cancer cells (S180: 5?10~6/ml) or NS were added into 0.2ml anticoagulant suspension of whole blood cells or leukocytes and 0.3ml plasma, then incubated for 1 h at 37℃. The content of IL-8 was determined by ELISA. Results In the patients with PHC, the IL-8 levels (pg/ml) in experimental and control groups of whole blood cells, and in experimental and control groups of leukocytes were 376.35?243.96, 353.64?271.92, 461.27?277.11 and 424.97?278.93, respectively; while in the normal human, they were 11.36?6.93, 4.98?4.35, 29.41?30.66 and 20.77?24.20, respectively. In the patients with PHC, the activation rates of cancer cells in the experimental groups of both whole blood cell and leukocyte were 0.22?0.24 and 0.25?0.53, respectively; while in normal human, they were 2.49?2.33 and 0.75?0.21, respectively. In the patients with PHC, the IL-8 adsorption rate of erythrocyte in both experimental and control groups of whole blood cell were 0.22?0.18 and 0.17?0.33, respectively; while in normal human, they were 1.18?2.29 and 0.86?0.49, respectively. 3.The Relationship between the Sign of the Lumbar Disc Protrusion on MRI and the Clinical Types of the Lumbar Disc Protrusion

Objective To research the relationship among the disc protrusion on MRI, sex, age, course and the clinical types. Methods MRI was performed for 102 patients. The disc protrusion was divided into three types. According to clinical types, the patients were divided into four types. Statistically, age, course, clinical type and protrusion on MRI were analyzed by SPSS13.0. Results The age was not related with the clinical types and disc protrusion on MRI. There were obvious relations between clinical types and course. There were obvious relations between clinical types and disc protrusion. Conclusion The clinical types are very important on diagnosis of disease stages and choice of therapy in treating lumbar disc protrusion.
5.Modified Brisson technique for concealed penis in children

Objective To investigate the corrective effect of modified Brisson technique to treat concealed penis in children.Methods From July 2014 to July 2016,selected 12 patients (3-10 years old,mean 7.2 years old) with concealed penis were surgically treated with the modified Brisson technique in the Second Affiliated Hospital of Nanjing Medical University.Their clinical manifestations included the penis buried below the surface of prepubic skin which can be palpated and visualized through pushing the skin around the penis to the pubis and the short foreskin which was not attached to the penile corpus cavernosum.Patients with other conditions such as obesity and penial agenesis that caused buried penis were excluded from the study.The modified Brisson technique consisted of 4 steps.Ventral incision was used to gradually slit the narrow ring of the prepuce and to expose the glans followed by circumfe-rential inner foreskin incision made below the coronary sulcus of penis then the foreskin stripping released the corpus cavernosum.Finally the skin coverage was accomplished by posthioplasty.The operation time,blood loss,complications,postoperative treatments and followed-ups were retrospectively analyzed.Results For all the 12 patients,the operations were successful,lasting 40 to 60 minutes.The bleeding was about 5 to 10 mL and there was no operation-related complications like bleeding and infection of incision.Satisfactory cosmetic outcomes were obtained without retraction and the scars were similar to those of circumcision.All patients had penile lymphedema postoperatively which disappeared within 6 months after operation.Follow-up ranged from 6 months to 1 year,and there were no complications like skin necrosis and contracture or voiding difficulty.Conclusions The modified Brisson technique for concealed penis repair is technically easy and safe.In addition,it provides a good cosmetic appearance,functional outcomes and excellent postoperative satisfaction grades.
6.Transplantation of marrow mesenchymal stem cells through renal artery in repair of acute tubular necrosis in nude mice

Objective:To investigate whether transplantation of mesenchymal stem cells(MSCs)through renal arteries can protect kidney from acute tubular necrosis(ATN),so as to lay a foundation for MSC transplantation in treatment of ATN.Methods:Five-week-old nude mice were randomly divided into three groups:normal control group(n=10),acute tubular necrosis(ATN)model group without(n=10)and with MSCs treatment group(n=11).ATN nude mice were induced with 50% glycerin.MSCs labeled with enhanced green fluorescent proteins(EGFP)were injected into kidney through renal arteries.Serum creatinine was determined in all groups and pathological changes of renal tissues were detected using H-E staining.The amount and distribution of the EGFP-marked MSCs in renal tissues were determined with fluorescence microscope.Results:Degeneration and exfoliation of renal tubular epithelial cells,and even renal tubular tamponade with cast-off cells were observed in the ATN group;these pathological changes were mainly located at renal cortex and juncture of renal cortex and medulla.The damages were greatly alleviated in the ATN+MSCs transplantation group,with no swelling of epithelial cells,nuclear condensation or edema.Fourteen days after MSCs transplantation,EGFP positive cells were increased in renal tubules of recipient mice.Conclusion:The MSCs transplantation via renal artery can locate in renal tubular epithelium,and promote the repair of injured renal tubular epithelial cells.

9.Brain mechanisms of hypoxic preconditioning.

A Review: A concept of tissue adaptation to hypoxia (i. e. hypoxic preconditioning) was developed and its corresponding animal models were reproduced in 1966s. The methods of model reproduction in rat, rabbit, and mouse in particular and the main results are briefly introduced in this review. The tolerance to hypoxia of preconditioned animals is significantly increased. Regular changes in animals' behavior, neurophysiology, respiratory and circulatory physiology, neuron morphology in vivo and function of brain and spinal cord in vitro are briefly demonstrated. The protective effects in vivo and in vitro of homogenate extract taken from the brain of preconditioned animals, neurochemicals and molecular neurobiological alterations are briefly presented. The essence and significance of tissue adaptation to hypoxia/hypoxic preconditioning are discussed in the review in terms of evolution and practical implication.
