1.Therapeutic research progress in rat models of vascular dementia

Vascular dementia is one of the common causes of cognitive impairment.Occlusion of bilateral common carotid arteries, middle cerebral artery occlusion, as well as induced hypertension are major methods to make rat model for vascular dementia.The present review will summarize research progress in medical and non-medical treatment of vascular dementia rat models in order to provide theoretical direction for clinical practice.

5.Effect of JAK2/STAT1 on acute kidney injury induced by liver cold ischemia reperfusion in rats

Objective To investigate the effect of Janus kinase2/signal transducer and activator of transcription 1 (JAK2/STAT1) signaling pathway on acute kidney injury induced by liver cold ischemia reperfusion (IR) in rats.Methods Thirty healthy male Sprague-Dawley rats,weighing 220-250 g,were assigned randomly to 3 groups (n =10/group):sham operation group (Sham group);liver cold ischemia reperfusion model group (I/R group);JAK2 kinase inhibitor AG490 group (AG490 group) (AG490 at dose of 10 mg/kg was intraperitoneally injected 30 min before establishment of the model).Other groups were given the equal volume of normal saline at the same time points.Then the rats were sacrificed at 6 h after reperfusion (at 6 h after the end of operation in Sham group).The renal function and oxidative stress level were observed.The pathological changes of the renal tissues and nephritic cell apoptosis were analyzed,and the expression of p-JAK2,pSTAT1,Bcl-2 and Bax was detected by Western blotting.Results As compared with Sham operation group,renal histological lesion and renal dysfunction were aggravated,level of oxidative stress and apoptosis rate were increased in I/R group,the Bcl-2/Bax ratio was decreased and the expression of pJAK2 and p-STAT1 was up-regulated.As compared with I/R group,AG490 dramatically attenuated histological lesions and oxidative stress,restored the renal function,and reduced the number of apoptotic tubular epithelial cells.AG490 significantly increased the Bcl-2/Bax ratio,and inhibited the expression of p-JAK2 and p-STAT1.Conclusion Blockage of JAK2/STAT1 signaling pathway can alleviate acute kidney injury after liver cold ischemia reperfusion probable through inhibiting the oxidative stress and apoptosis.
6.CT,MR and MRS Diagnosis of Basal Ganglia Infarction

Objective To discuss imaging differentiating diagnosis of basal ganglia infarction by summarizing the features of 17 lesions on CT and MRI.Methods Among 17 cases, 6 were children occurred after their injury, and ranged in age from 5 to 11 years old with average 8 years old. Others were adults ranged in age from 53 to 78 years old with average 63 years old. All patients were examined with CT plain scans. 11 cases were underwent MRI and 5 were underwent 1 H MRS.Results 6 lesions in children were all single lateral cases. Among 11 adult lesions, 10 were single lateral and 1 was bilateral case. Basal ganglia infarctions demonstrated hyposignal on T_1WI, hypersignal on T_2WI and hypodense on CT. The lateral ventricles' anterior horns of the same side were pressed and narrow. On 1 H MRS, Lac was obviously observed in all lesions. NAA decreased markedly in 4 lesions and slightly in 1 lesion. 2 basal ganglia infarctions were together with hemorrhage.Conclusion Basal ganglia infarctions were mainly observed in the elder with arteriosclerosis and injured children. Its features on CT and MRI are characteristic, but it is in need of differentiating from basal ganglia lesions caused by other reasons. Marked Lac on 1 H MRS is its character.
7.The Study of ~1 HMRS in Diagnosis of Astrocytoma

Objective To assess the value of ~1 HMRS in the classification and differential diagnosis of astrocytoma. Methods 58 cases of astrocytoma including 18 cases of diffuse astrocytoma, 24 cases of anaplastic astrocytoma and 16 cases of glioblastoma were all examined by routine MR scan and ~1 HMRS, statistical analysis was done on Cho/Cr, Cho/NAA and NAA/Cr ratio. Results Diffuse astrocytoma showed slightly elevated Cho, a bit decreased NAA and Cr. Lac wave did not appear. Anaplastic astrocytoma and glioblastoma both manifested distinctly elevated Cho, obviously decreased NAA and Cr. Lac wave existed in 6 cases. In diffuse astrocytoma the ratio of Cho/NAA was 2.72?1.16, Cho/Cr was 2.66?1.21, NAA/Cr was 1.02?0.34. In anaplastic astrocytoma the ratio of Cho/NAA was 5.69?1.32, Cho/Cr was 4.92?1.46, NAA/Cr was 0.92?0.16. In glioblastoma the value of Cho/NAA was 5.71?1.08, Cho/Cr was 5.12?1.76, NAA/Cr was 0.87?0.13.Conclusion It is very significant of ~1 HMRS in the classification and differential diagnosis of astrocytoma.
8.Biological Characteristics of Marine Bacterium Strain E18 and the Stability of its Indigo Pigment

One strain of Pseudoalteromonas sp. E18 was isolated from the sea mud of Ningbo, Zhejiang. It can produce indigo pigment. The morphological, cultural and biochemical characteristics of the bacterium were studied. The indigo pigment was also extracted. The results showed that the maximum absorption peak of the pigment was at 579nm. The pigment was stable to UV, Na_2SO_3,It was also stable at pH 3~9. The pigment was unstable to the sunlight and high concentration H_2O_2. Temperature of 60℃~80℃ could increase the hue while temperature higher than 90℃ could reduce the hue.
9. Discovery of quality markers and quality evaluation of Poria cocos based on “in vitro-in vivo” multidimensional chemical group associated network

Objective: The quality markers of Poria cocos were identified based on the “in vitro-in vivo” multidimensional chemical group associated network and in vivo pharmacokinetics, and the mass spectrometry method for quantitative detection of quality markers was established to evaluate the quality of P. cocos. Methods: A quantitative analysis method of triterpenoids in P. cocos was established by ultra-high performance liquid chromatography-triple quadruple tandem mass spectrometry (UHPLC-MS/MS) while dioscin was chosen as internal reference substance. The pharmacokinetic curves of active ingredients in vivo were drawn. Seven quality markers of P. cocos including dehydrotumulosic acid, tumulosic acid, and poricoic acid B were identified based on the results of pharmacokinetics. The content of high content components and quality markers in 10 batches of P. cocos were detected and used as variables for cluster analysis of 10 batches of P. cocos. Results: There were differences in the results of clustering analysis with different indexes as variables in evaluating the quality of P. cocos. Conclusion: To a certain extent, the method of quality control of P. cocos with high content components is one-sided. In order to comprehensively and accurately control the quality of P. cocos, it is necessary to take into full account the in vivo and in vitro changes and the in vivo dynamic process of P. cocos composition, and select the effective quality markers related to its pharmacodynamics for its quality control.
10.Criteria for performing peripheral blood smear review and its clinical application

Objective To find a proper way for accurate results on complete blood counts.Methods Based on the results from automatic hematology analyzer, set up criteria for blood cell microscopic examination. 9 992 blood specimens were detected and the results were analyzed according to the criteria, statistics on the data were made to find out the reliability of the policy.Results The criteria could help to find more abnormal cells, it was very useful for clinical diagnosis and proper treatment.There were 29.3% of CBC specimens needed for microscopic examination, 32.2% of which had increased bands or increased atypical lymphocytes or other abnormalities.Conclusions It was very important to set up the criteria for performing peripheral blood smear. It can provide more accurate and reliable information to clinical doctors.
