1.Effect of Different Needle-retaining Lengths on the Therapeutic Efficacy of Acupuncture in Treating Post-stroke Shoulder Pain

ObjectiveTo observe the effect of different needle-retaining lengths on the clinical efficacy of acupuncture in treating post-stroke shoulder pain.MethodSixty patients were randomized into a 30 min group and a 60 min group, 30 cases in each group. The same acupuncture treatment protocol was adopted in the two groups, and the acupuncture treatment was given once a day, 5 sessions a week, for 20 times in total. The Visual Analogue Scale (VAS), shoulder pain frequency, and Fugl-Meyer Assessment (FMA) were observed before treatment and after 4-week treatment.ResultThe scores of each index were significantly improved in both groups after treatment (P<0.01), and there were significant differences in comparing each index score between the two groups after treatment (P<0.05,P<0.01).ConclusionIn the treatment of post-stroke shoulder pain with acupuncture, 30 min needle-retaining is better than 60 min in comparing the analgesic effect.
2.Systematic Review of Acupuncture-moxibustion for Deglutition Disorders after Cerebral Stroke

Objective To evaluate the clinical efficacy of acupuncture-moxibustion in treating deglutition disorders after cerebral stroke.Method By computer and manual literature retrieval, eligible randomized controlled trials (RCTs) were collected for systematic review by using the Cochrane method, and the meta analysis was performed by using Revman 5.2.Result Nine studies were recruited, covering 577 subjects of deglutition disorders due to cerebral stroke. The total effective rate of the treatment group was significantly higher than that of the control group [RR=0.36, 95%CI (0.25,0.50), Z=5.80 (P<0.00001)]. The treatment group was superior to the control group in improving the water drinking test (χ2=108.73,P<0.00001).Conclusion Acupuncture can produce a content therapeutic efficacy in treating deglutition disorders due to cerebral stroke, which still requires high-quality large-sample-size clinical RCTs for further verification.

4.Clinical Observation of Synergism of TCM Medicinal Oxygen Therapy in Treating Acute Cerebral Infarction

Objective To observe the clinical efficacy of synergism of TCM medicinal oxygen therapy in treating acute cerebral infarction.Methods Eighty cases were randomly divided into treatment group and control group, 40 cases in each group, and 1 case was lost in treatment group. Based on routine treatment, the control group was treated with low flow oxygen inhalation only, while the treatment group was treated with TCM medicinal oxygen therapy. The course of treatment was 2 weeks. The two groups were scored with National Institutes of Health Stroke Scale (NIHSS) and Stroke Symptom Scale of TCM (SSTCM) before and after the course to evaluate the therapeutic effect. Results The scores of NIHSS and SSTCM in both groups after the treatment were with statistical significance (P<0.05). There was no statistical difference in the scores of NIHSS between two groups after the treatment (P=0.10). There were statistical difference in the scores of SSTCM between two groups after the treatment (P<0.05). The clinically curative effect rate in treatment group was76.9% (30/39), which was superior to that of 60.0% (24/40) in the control group, with statistical significance (P=0.27).Conclusion TCM medicinal oxygen therapy can improve the neurological deficit and enhance the efficacy.
5.Effect of Different Needle-retaining time on Post-stroke Hypermyotonia in Acupuncture Treatment

Objective To compare the effect of different needle-retaining time on post-stroke hypermyotonia in acupuncture treatment.Method Ninety patients with post-stoke hypermyotonia were randomized into group A, group B, and group C, 30 in each group. The three groups were all treated by the twelve hand-foot needling method from the thirteen therapies invented by acupuncture master WANG Le-ting, once a day, 5 times a week, 20 times in total. For group A, needles were removed right after needling qi arrived; for group B, needles were retained for 30 min after needling qi arrived; for group C, needles were retained for 60 min after needling qi arrived. The modified Ashworth Scale (MAS), Clinical Spasticity Index (CSI), and Fugl-Meyer assessment scale (FAS) were adopted for observation before and after intervention.Result After treatment, the MAS was significantly changed in all three groups (P<0.05). The CSI score and FAS score of the affected limb were significantly changed in all three groups after intervention (P<0.05). Both group A and B were significantly different from group C in comparing the CSI and FAS scores (P<0.05).Conclusion Compared to retaining needles for 60 min, acupuncture without retaining needles or retaining needles for 30 min can produce better effect in improving post-stroke hypermyotonia, spasticity, and motions of limbs.
6.Construction and Identification of Lentiviral-Mediated RNA Interference Vector of Rat Suppressors of Cytokine Signaling 3 Gene

Objective To study the construction of the lentiviral-mediated RNA interference(RNAi) vector targering rat suppressors of cytokine signaling 3(SOCS3) gene.Methods Three target sequences were selected by on-line designer software on Ambion according to rat SOCS3 mRNA sequence(NM053565),the complementary DNA contained both sense and antisense oligonucleotides were designed and synthesized.After annealing,these double strands DNA were cloned to pRNA-Lenti-green fluorescent protein(GFP),which contained U6 promoter and GFP.The resulting Lentiviral vector containing SOCS3 shRNA was named pRNA-Lenti-SOCS3-GFP.After the rat glioma cells(C6)were transduced with the constructed 1entiviral vectors,real-time polymerase chain reaction was used to evaluate the level of SOCS3 expression(including siRNA1 group,siRNA2 group,siRNA3 group,vacuity group and siRNA-Negative group).The pRNA-Lenti-SOCS3-GFP and Lentivector Pakaging plasmid mix were cotransfected into 293T to package Lentivirus particles.Culture supematant was harvested,then the virus titer was determined by serial dilution assay.Results The SOCS3 mRNA sequence was successfully cloned to pRNA-Lenti-GFP,which was proved by PCR and DNA sequence.Compared with control group,the SOCS3 mRNA expressions were obviously suppressed in all 3 experimental groups,especially the expression rate in siRNA1 group was reduced by 80%.The Lentiviral particle titer was determined by serial dilution assay with 1.0?1010 TU?L-1.Conclusion The lentiviral-mediated RNAi vector of rat SOCS3 gene has been constructed successfully,this may provide a potential tool for studying and treating SOCS3-related diseases.

8.Production of VEGF induced by GMCSF via ERK-NF-KB singling 'pathway in human fibroblasts during wound healing

ObjectiveTo observe production of vascular endothelial growth factor (VEGF) induced by granulocyte/macrophage colony-stimulating factor (GMCSF)via ERK nerve growth factor (NF)-κB singling pathway in human fibroblasts during wound healing and explore relating mechanism.MethodsHuman fibroblasts from the injured skin were used for this study and treated with GMCSF.RT-PCR was used for analyzing the protein and mRNA levels of VEGF and Western blotting was employed to determine the phosphorylation of ERK. The fibroblasts were pre-treated with ERK specific inhibitor PD98059 and further treated with GMCSF, then the fibroblasts and the supernatant were collected for detection of protein level of VEGF by means of Western blot. ERK signal pathway was inhibited to detect the activation of NF-κB by means of immunofluorescence staining. Furthermore, the nuclear and cytoplasmic extraction kit was used to separate the cytoplasm and nucleus and Western blot employed for observation of the NF-κB activation. ResultsThe mRNA level and protein level of VEGF were increased significantly with treatment with higher concentration of GMCSF in a dose-dependent manner. VEGF mRNA level was increased two hours after administration with GMCSF and reached peak at 4-6 hours. GMCSF could remarkably activate the ERK phosphorylation. Compared with GMCSF, the ERK specific inhibitor PD98059inhibited significantly the effect of GMCSF in inducing VEGF expression (P < 0.05). Western blot and immunofluorescence staining analyses showed that the activation of NF-ΚB was inhibited with reduced production of VEGF after GMCSF treatment.Conclusion GMCSF up-regulates production of VEGF through activating NF-κB via ERK signal pathway in the human fibroblasts.
9.The effects of low frequency ultrasound on the proliferation and apoptosis of smooth muscle cells in carotid atherosclerosis

Objective To investigate the effects of low frequency ultrasound (LFU) on the proliferation and apoptosis of smooth muscle cells (SMCs) in rabbits with carotid atherosclerosis(CA).Methods CA models were established in 30 New Zealand rabbits using a high fat diet and air-drying.They were randomly divided into a control group and four LFU groups:group A received 0.5 W/cm~2 LFU for 5 min/d,group B 0.5 W/cm~2 for 10 min/d, group C 1 W/cm~2,5 min/d,and group D 1 W/cm~2,10 min/d.The rabbits' carotid arteries were autopsied after 20 d of the LFU treatment.The expression of PCNA and TUNEL staining were used to explore the proliferation and apopto- sis of SMCs,and the proliferation rates (PRs) and apoptosis rates (ARs) were calculated.Results Compared with the control group,the PRs in groups B,C and D were significantly lower,while the ARs in groups B,C and D were significantly higher.There was no significant difference in ARs or PRs among groups B,C and D.Con- clusion LFU can induce SMC apoptosis and inhibit SMC proliferation in rabbits with CA.
10.Effect of high glucose on expression of MIP-2 and MCP-1 in macrophages cultured in vitro

0.05).The protein and mRNA expression of MIP-2 in high glucose group significantly increased after culture for 4 h,and guadually decreased then.The protein and mRNA expression of MCP-1 began to increase significantly after culture for 8 h,reached peak at 12 h,and slightly decreased after culture for 24 and 48 h. Conclusion High glucose promotes the protein and mRNA expression of MIP-2 and MCP-1 from mouse peritoneal macrophages cultured in vitro,which indicates that high glucose may delay the wound healing by increasing the expression of chemokines in diabetic mice.
