1.Single-agent vinorelbine in the treatment of elderly patients with advanced non-small-cell lung cancer

Purpose:This report evaluate whether the single-agent therapy with vinorelbine (V group) may obtain a high response rate, with acceptable toxicity and improvement in survival and quality of life, comparing with the therapy with Kanglaite (a traditional Chinese medicine) (K group) and other supportive care among elderly patients.Methods:Forty patients with advanced NSCLC were included,20 of whom were allocated to receive continual infusional NVB 7.5mg/m 2 /24hr delivered via a central venous line on days 1-5, and NVB 7.5mg /m 2 was given as a 20 min intravenous infusion on days 1 every 3 weeks; 20 of whom were allocated to receive infusional Kanglaite 200 ml on days 1-20 every month. Patients received a minimum of two courses unless progressive disease was detected. Results:V group: CR 0,PR 7,CR+PR 35%(7/20), median survival time was 7.2 months and projected 1-year survival was 25%; K group:CR 0,PR2,CR+PR 10%(2/20),median survival time was 4.8 months and projected 1-year survival was 5%. Conclusions:In elderly patients with NSCLC, single-agent vinorelbine treatment is associated with better effective,better survival and improved quality of life than Kanglaite .Its toxicity is mild and acceptable.
2.Effect of hydrogen sulfide postconditioning on systolic function of left ventricle during myocardial ischemia-reperfusion in rats

Objective To investigate the effect of hydrogen sulfide postconditioning on the systolic function of left ventricle during myocardial ischemia-reperfusion (IR) in rats. Methods Part Ⅰ Adult male SD rats weighing 200-250 g were anesthetized with pentobarbital 40 mg/kg and heparin 250 U/kg. Their hearts were excised and perfused in a Langendorff apparatus with K-H solution saturated with 95% O2-5% CO2 at 37 ℃. Forty isolated rat hearts were randomly divided into 5 groups ( n = 8 each) after 20 min of equilibration: control group (group C); IR group; sodium hydrosulfide 1,10, 100 μmol/L postconditioning group (group SP1, SP10, SP100 ).In group Cthe hearts were perfused continuously for another 100 min. In group IR, the hearts were reperfused for 60 min after 40 min ischemia induced by 10 ml/kg ST. Thomas solution. In group SP1 , SP10 and SP100 the hearts were perfused with K-H solution containing sodium hydrosulfide 1, 10, 100 μmol/L for 2 min before reperfusion.LVDP and ± dp/dtmax were recorded at the end of equilibration and reperfusion. Part Ⅱ Cardiomyocytes were isolated from the male SD rats (weighing 200-250 g) and then cultured in CO2 incubator for 4 h. Sixty-four dishes of cultured myocytes were randomly divided into 4 groups( n = 16 each): control group (group C), hypoxia/reoxygenation group (group HR), hydrogen sulfide postconditioning group (group SP) and hypoxia postconditioning group (group HP). Group C were cultured continuously for 2 h. Group HR, SP and HP were exposed to 1 h hypoxia (95%N2-5%CO2 ) followed by 1 h reoxygenation. In group SP 10 μmol/L sodium hydrosulfide was added and the myocytes were then incubated for 2 min before reoxygenation. In group HP the cultured myocytes were expased to 3 min reoxygenation followed by 3 min hypoxia for 3 times before the 1 h reoxygenation. Mitochondrial membrane potential and F-actin expression were determined. Results Part Ⅰ Compared with group C, LVDP and ± dp/dtmax were significantly decreased at the end of reperfusion in group IR (P < 0.05), while no significant difference was found in group SP1 , SP10 and SP100(P >0.05). Compared with group IR, LVDP and ± dp/dtmax were significantly increased in group SP ( P < 0.05). There was no significant difference in LVDP and ± dp/dtmax among group SP1, SP10 and SP100(P >0.05). Part H Compared with group C, the mitochondrial membrane potential was significantly decreased in group HR and HP, and the expression of F-actin was significantly up-regulated in group HR, SP and HP ( P < 0.05). Compared with group HR, the mitochondrial membrane potential was significantly increased and the expression of F-actin up-regulated in group SP and HP ( P < 0.05 ). There were no significant difference in the mitochondrial membrane potential and expression of F-actin between group SP and HP ( P >0.05).Conclusion Hydrogen sulfide postconditioning can improve left ventricular systolic function during IR in rats by stabilizing mitochondrial membrane potential and promoting aggregation of F-actin.

6.The Effects of Suanzaoren decoction on hippocampus, cortex BDNF and TrKB geneexpression in depression rats

Objective To investigate the effect of Suanzaoren Decoction on hippocampus, cortex BDNF and TrKB gene expression in depression model rats. Methods Depression rat models were established by social-isolated raise and chronic stress stimulation. Suanzaoren decoction was administrated to the models. RT-PCR was adopted to detect the expression of mRNA BDNF and TrKB genes. Results The mRNA expression of BDNF and TrKB in cortex of Suanzaoren decoction high dose group、medium dose group and clomipramine group(0.213±0.094, 0.639±0.023, 1.032±0.015, 1.089±0.014, 1.580±0.012, 1.860±0.019)were all higher than the model group(0.032±0.008, 0.001±0.000), showing a significant difference among four groups (P<0.01), and the mRNA expression of BDNF and TrKB in hippocampus of Suanzaoren decoction high dose group, medium dose group and clomipramine group(0.213±0.094, 0.639±0.023, 1.032±0.015, 1.089±0.014, 1.580± 0.012, 1.860±0.019)were higher than the model group (0.021±0.015, 0.125±0.013), there was a significant difference between four groups(P<0.01). Compared with the model group, the mRNA expression of low-dose group of Suanzaoren decoction in both cortex and hippocampus of BDNF and TrKB was not significantly different to the model group(P>0.05). Conclusion Suanzaoren decoction can increase the expression of BDNF and TrKB gene, promote neuronal proliferation, and resist depression.
7.EFFECTS OF NICOTINAMIDE SUPPLEMENT ON BEHAVIORS IN OPEN-FIELD TEST OF RESTRAINED RATS

Objective: To study the effects of different niacin nutritures on behaviors of stressed rats. Methods: 100 Wistar male rats were assigned into 10 groups:control, pair-fed, nicotinamide (NAM) deficiency (ND), low-dose NAM supplement (LNS 40 mg/kg diet) and high-dose NAM supplement (HNS 500 mg/kg diet), and the corresponding restrained groups. 24 h urinary excretion of N1-methylnicotinamide (NMN), body weight changes, behaviors in the open-field test, plasma cortisol, nitric oxide(NO) and liver nitric oxide synthase(NOS) were determined. Results: Compared with the control group, restrained rats had relatively lower urinary NMN. Both ND and restrainted stress retarded the body weight gains . Rats in the control and LNS groups had relatively shorter latency time in the open-field test. Restrained rats had higher plasma cortisol and NO than those in the corresponding control group. However, rats with HNS had the lowest plasma cortisol concentrations. Dietary supplement of NAM increased liver NOS . Conclusions: Restraint stress may increase niacin depletion and appropriate NAM supplement has positive effects on stress adaptation in rats.
8.An analysis of correlation between post-stroke depression and relevant factors

Objective To investigate correlation between post-stroke depression (PSD) and multiple factors during onset of acute cerebral infarction. Methods Depression was measured with Hamilton Depression Rating Scales (HAMD) in 58 patients with acute cerebral infarction, and their neurological function were evaluated by neurological function defect (NFD) score. Their immunoglobulin G (IgG) index was calculated and level of nitric oxide (NO) in cerebrospinal fluid (CSF) was measured. Lesion and nature of cerebral infarction in 58 patients with acute stroke were located by CT. All the data were statistically analyzed with student-t test and ? 2 test, as well as linear regression model. Results Seventeen of 58 patients of stroke appeared PSD with an occurrence rate of 29.3%. Objective To explore the relationship between the protective effect of hydrogen sulfide (H2 S ) postconditioning against myocardial hypoxia-reoxygenation and the dynamics of actin. Methods Adult rat cardiomyocytes were isolated and the same hatch cells were divided into 4 groups (n =3):normal group (group N),hypoxia-reoxygenation group (group HR),ischemia postconditioning group (group IPTC)and H2 S postconditioning group (group S).The four groups were divided into two sub-groups with or without cytochalasin D (CyD).At the end of reoxygenation,F-actin/G-actin,intracellular calcium ion concentration (Ca2+ )and pH value were detected with laser scanning confocal microscopy, meanwhile the level of p-p38MAPK was detected with Western blot.Results The fluorescence intensity of F-actin/G-actin of group HR,group IPTC and group S were significantly higher than that of group N (P <0.05).The fluorescence intensity of Ca2+ of group HR was higher than that of group N,group IPTC and group S (P <0.05);The intensity of Ca2+ of all group with CyD treatment was higher than those without (P <0.05);The fluorescence intensity of pHi of group N and group HR with CyD treatment was higher than those without;The fluorescence intensity of pHi of group IPTC and group S was lower than those without;The flu-orescence intensity of pHi of group HR was lower than that of group N,group IPTC and group S (P <0.05), respectively;the pHi of group N and group HR sub-group with CyD treatment was higher than those without correspondingly (P <0.05),however,the pHi of IPTC and S sub-groups were lower than their corresponding groups (P <0.05).The level of p-p38MAPK in group HR was significantly higher than those of other groups (P <0.05);there was no difference among groups N,IPTC and S.Conclusion Hydrogen sulfide postcondi-tioning could promote F-actin to remodel and stabilize the cellular environment.Hypoxia-reoxygenation pro-motes the phosphorylation level of p38MAPK,which could be surpressed by H2 S postconditioning.
