Objective To examine the effect of tetraethylammonium (TEA, K~+ channel blocker) on pancreatic ?-cell apoptosis and explore the mechanism. Methods Mouse ? cells (NIT cells) were exposed to streptozotocin(STZ) to induce apoptosis, and TEA of different concentrations were applied along with STZ to prevent efflux of intracellular K~+. Cells were stained with annexin V, PI and rhodamine 123. Flow cytometer (FCM) was used to determine the percent of apoptotic or viable cells and the change of mitochondrial membrane potential. Culture media was collected to quantify the content of NO and ROS produced by NIT-cells. Cells were collected for detecting the activity of super oxide dismutase (SOD) in cells lysates. Tamoxifen(TAM)is an important therapeutic strategy for the treatment of estrogen receptor(ER)-positive breast cancer.However,tamoxifen resistance is a major cause of endocrine therapy failure.The potential mechanism of TAM resistance is multifactorial and most of them are still unknown.This review presents recent advances in the mechanism of TAM resistance in breast cancer,providing valuable information and ideas for elucidating the mechanism of tamoxifen resistance and overcoming drug resistance.

Objective To observe the changes of the content of ATP,lactate and the activity of phosphofructokinase(PFK) in perihemotoma region in rabbit brain after intracerebral hemorrhage. Methods(The rabbits) were divided into 3 groups: normal group(N),hemorrhage group (H) and the sham group(S). The H and S groups were subdivided into 1 h, 6 h, 12 h, 24 h, 48 h and 72 h groups. A model of cerebral hemorrhage in rabbit with autologous blood infusion was set. The contents of ATP, lactate and water in perihemotoma region were measured and the metabolite concentration in edematous brain regions were corrected for dilution. The activity of phosphofructokinase was also determined.Results There was obvious edema in perihemotoma region at 1 hour postinfusion and getting more serious at 72 hours. ATP was moderately decreased at 1 hour, and remained in a similar level until 12 hours. There appeared another evident decrease in 24 hours [(13.29?2.92) ?g/g, about 58% of N group], and no obvious changes from 24 to 72 hours. The content of lactate increased in 1 hour and reached the peak at 12 hours [(21.01?0.18) ?mol/g]. Until 72 hours , the level of lactate [(12.89?0.25) ?mol/g] in hemorrhage group reach higher than in the normal and sham group. The activity of phosphofructokinase was decreased at 1 hour [(3.98?0.02) U/mg] and remained in a decreasing tendency to 72 hours. Conclusion The failure of energy metabolism occurs in 24 hours postinfusion,coming late somewhat to the brain edema. The result might be related to the changes of some enzymes, maybe a key role in energy metabolism.

Objective To determine the role of double stapling technique(DST) in anus-saving operations for lower rectal cancer. Methods The clinical data of 96 patients with lower rectal carcinoma treated by DST were analyzed retrospectively. Results There were 63 males and 33 females,aged from 22 to 84 (an average of 57.7 yesars). The distance from the lower end to dens line was 4～7cm.Dukes classfication: 24 patients belonged to A class, 30 B class, 30 C class,and 12 D class. Histological type: 47patients had high differentiation adenocarcinoma, 27 median differentiation adenocarcinoma, 17 low differentiation adenocarcinoma,and 5 mucous adenocarcinoma. No one died after the operation but 12 had anastomotic leakage(12.5%)postoperatively and all healed after treatment; 5 patients had anastomotic constriction(5.2%)postoperatively and all healed with expanding anus. Conclusions DST is a safe alternation for anus-saving operation for patients with rectal cancer.Conservative therapy is effective in the treatment of anastomotic constriction and anastomotic leakage.

To study the clinical significance of combined measurement of neuron specific enolase(NSE), carcinoembryonic antigen(CEA) and Cytokeratin19 fragment 21 1 (CYFRA 21 1 ) in the diagnosis of lung cancer. The serum levels of CEA, NSE, and CYFRA 21 1 in 75 lung cancer patients as well as in 22 benign pulmonary diseases patients were measured by radioimmunoassay. Patients with pulmonary adenocarcinoma had a highest CEA level. Small cell lung carcinoma (SCLC) patients had a highest NSE level, while patients with squamous cell carcinoma of lung had a highest CYFRA 21 1 level. The positive rate of the combination of CEA, NSE, and CYFRA 21 1 for lung cancers was 82 67%. The results suggested that the combined measurement of CEA, NSE, and CYFRA 21 1 was helpful in the diagnosis of lung cancer.

Objective:To assess the preventive and therapeutic effects and primary mechanisms of Danshen injections in the treat-ment of hepatic encephalopathy ( HE) . Methods:The acute ammonia poisoning model induced by ammonium chloride was established in mice, and HE model with liver injury induced by carbon tetrachlorie and high blood ammonia induced by thioacetamide was made in rats. The time of death was detected in the first model, and the histamine levels in blood and brain, liver function and liver pathologi-cal histology changes were detected respectively in the other model. Results:In the acute ammonia poisoning experiment, the time of death in Danshen injections group was longer than that in the control group. Furthermore, Danshen injections could notably decrease the histamine levels in blood and brain, reduce the levels of ALT, AST, ALP and Tbil in serum and improve the function of liver to show the preventive and therapeutic effects on HE. Conclusion:Danshen injections exhibit promising preventive and therapeutic effects on HE, which may be related to decreasing histamine levels in blood and brain.

OBJECTIVE To report malignant hematologic patients suffered in fungal infections treated with voriconazole(VOR),compare them with other similar patients treated with aluphotericin B liposome(L-AMB) as control,and then review the references.METHODS Fifty patients were treated with VOR and 57 patients were treated with L-AMB.According to the clinical performance and the changes in the CT,we could find the difference of between them.RESULTS Forty cases were effective in VOR group and most of them were in good tolerance.In the control group,37 cases were effective,most of them were in poor tolerance,and some of them stopped the treatment.CONCLUSIONS Instead of L-AMB,VOR can be a useful drug in the treatment of hematologic malignancies patients suffered in fungal infections.

AIM: To establish a HPLC method for the determination of tanshinone Ⅱ A in Compound Danguifukang Capsule. METHODS: Analytic column: Reliasil C18(5μm, 250mm×4.6mm), protective column: (5μm, 12.50mm×4.6mm); mobile phase: methnaol-ultrapure water (80: 20); detective wavelength was at 270a good linearity within the range of 1×10-2-0.16μg, r=0.9995. The average recovery was 99.73% and RSD was 2.91%. CONCLUSION: The method is feasible, and simple to operate and suitable for the quality control of Compound Dangguifukang Capsule.
