1.Role of angiotensin Ⅱ receptors in collagen synthesis of fibroblast in human hypertrophic scars

Objective To observe the expression of both type 1 (AT1) and type 2 (AT2) receptors of angiotensin Ⅱ (AngⅡ) in human hypertrophic scars, and to explore theirs role in collagen synthesis of fibroblasts in human hypertrophic scars. Methods The expression of both AT1 and AT2 receptors in fibroblasts of hypertrophic scars was detected with immunohistochemical staining and radioligand receptor binding assay. Collagen synthesis was examined in cultured in vitro fibroblasts of hypertrophic scars by measuring [~3H]proline incorporation into collagenous proteins. Results Positive staining signals of both AT1 and AT2 receptors were found in fibroblasts of hypertrophic scars. Similar results were also got in cultured in vitro fibroblasts of hypertrophic scars, expression level of AT1 and AT2 receptors were 10.69?2.15fmol/10~6cells and 4.9?1.05fmol/10~6cells respectively. 2.Mechanism of dermal fibroblasts transdifferentiation into myofibroblasts induced by estrogen

Objective To explore the possible pathway and regulatory mechanism of dermal fibroblasts' transdifferentiation into myofibroblasts induced by estrogen. Methods Human dermal fibroblasts were divided into six groups (A: control; B: estrogen; C: estrogen + ICI-182780; D: estrogen + SB203580; E: estrogen + PD98059; F: estrogen + SP600125). The cells were collected for RNA extraction and the expression of α-SMA was detected by real time quantitative RT-PCR. Some cells were analyzed by single cell RT-PCR to detect positive expression percentage of α-SMA.Results The expression and positive rate of α-SMA in estrogen group were significantly increased (Group B vs. Group A, 7. 385±0. 246 vs 1. 367±0. 034, P<0.01) and those in ICI-182780 group and SP600125 group were significantly inhibited (Groups C and F vs. Group B, 4. 619 ±0. 164,2. 409±0. 091 vs 7. 385±0. 246, P<0. 05). Conclusions In the process of fibroblast transdifferentiation into myofibroblasts induced by estrogen, estrogen β receptor and JNK-MAPK signal transduction pathway may play an important role.
3.Effect of δ-opioid receptor on bioactivity of human epidermal stem cells in vitro

Objective To observe the effect of δ-opioid receptor on proliferation and migration of human epidermal stem cells (hESCs) in vitro so as to offer treatment theory for skin injury.Methods hESCs from fresh foreskin tissues of normal young volunteers were isolated and cultured by enzyme digestion and differential adherence technique.Immunofluorescent staining was used to determine expression of integrin β1 and cytokeratin 19 (CK19) and flow cytometry was used for cell count.Second generation of cells were cultured for 5 consecutive days with keratinocyte serum-free medium (K-SFM) supplemented with 1 nmol/L (D-Ala2,K-Leu5)-enkephalin in Group A,with K-SFM supplemented with 1 nmol/L naltrindole and 1 nmol/L (D-Ala2,K-Leu5)-enkephalin in Group B,and with isolated K-SFM in Group C.Cellular division and proliferation were detected by MTT method.An in vitro 100 μm scratch-wound model was created on the confluent monolayer cells at 24 hours of incubation.Cells migrating from the wound margin were determined by inverted phase contrast microscope at 24,48,72,and 96 hours after wound formation,while wound closure rate was calculated at 72 hours.Results Primary cultured hESCs presented cobblestone-like shape after adherence growth,Immunofluorescence staining showed positive results for integrin β1 and CK19 and cell purity reached 95.6%.Moreover,MTT findings revealed proliferation of hESCs enhanced significantly in Group A,but lowered in Group B as compared to Group C (P < 0.05).hESCs migrated from the wound margin in all groups at 24 hours.However,more migrated cells were seen in Group A than in Group C and less in Group B than in Group C.Rate of wound closure was (89.5 ±0.7)% in Group A,(76.1 ±0.3)% in Group B,and (81.1 ±0.6)% in Group C at 72 hours,indicating significant differences among groups (P < 0.05).Conclusion Activation of δ-opioid receptor promotes the proliferation and migration of hESCs in vitro and may be implicated in wound healing.
4.Effect of p-nitrophenol Shock on Sludge Activity and Microbial Populations and in UASB Reactor

Effect of p-nitrophenol shock on microbial populations and sludge activity in UASB reactor was studied by DGGE-PCR of 16S rDNA fragments and detection of COD removing and biogas yield.The results showed that p-nitrophenol seriously inhibited the sludge activity,resulting in the drop of biogas and COD removing rate.The 40mg/L p-nitrophenol had more inhibition than 20mg/L p-nitrophenol.It would take 27 and 16 days respectively for reactor to recover after 40mg/L and 20mg/L p-nitrophenol shock.The diversity of eubacteria and methanogens were also effected by the p-nitrophenol shock.The variation of eubacteria was more than that of methanogens after p-nitrophenol shock.The drop of biogas was mainly related to the variation of Methanosaeta sp.and Methanomicrobia sp.after p-nitrophenol shock.Among the eubacteria the population of Chloroflexi sp.、Bacteroide sp.and Anaerovibrio sp.decreased greatly after p-nitrophenol shock.And more,the Rheinheimera sp disappeared after 40mg/L p-NP treatment.But the Flavobacteria sp.appeared after p-nitrophenol shock,which was probably related to the degradation of p-NP.
5.Observation on therapeutic effect of acupuncture at abdomen acupoints plus tuina for lumbar intervertebral disc herniation

Objective:To observe the clinical efficacy of acupuncture at abdomen acupoints plus tuina for lumbar intervertebral disc herniation (LIDH).Methods:A total of 70 patients with LIDH were randomized into an observation group and a control group,with 35 cases in each group.The observation group was treated with acupuncture at abdomen acupoints plus tuina,while the control group was treated only with tuina treatment.The clinical efficacy was observed after one course of treatment.Results:The cure rate and the total effective rate of the observation group were 83.3% and 96.7%,respectively.The cure rate and the total effective rate of the control group were 39.4% and 78.8%,respectively.There were significant differences in the cured rate and the total effective rate between the two groups (both P<0.05).There was no significant difference in Japanese Orthopedic Association (JOA) score between the two groups before treatment (P>0.05).After treatment,the JOA scores of both groups increased significantly,and the intra-group differences were statistically significant (both P<0.05);the JOA score of the observation group was significantly higher than that of the control group (P<0.05).Conclusion:Acupuncture at abdomen acupoints plus tuina has a better therapeutic effect than tuina alone in the treatment of LIDH.
6.Therapeutic Observation of Warm Needling at Yintang (GV 29) plus Ginger-partitioned Moxibustion for Allergic Rhinitis

Objective To observe the clinical efficacy of warm needling at Yintang (GV 29) plus ginger-partitioned moxibustion in treating allergic rhinitis.Method Seventy patients with allergic rhinitis were randomized into a treatment group and a control group,35 cases each.The treatment group was intervened by warm needling at Yintang (GV 29) plus ginger-partitioned moxibustion;the control group was treated with conventional medication.After 4-week treatment,the clinical efficacies of the two groups were compared.Result The markedly effective rate and total effective rate were respectively 83.9% and 96.8% in the treatment group,versus 56.3% and 75.0% in the control group,and the between-group differences were statistically significant (P<0.05).Conclusion Warm needling at Yintang (GV 29) plus ginger-partitioned moxibustion is an effective approach in treating allergic rhinitis.
7.Study on chemical constituents in rhizome of Smilax perfoliate.

OBJECTIVETo investigate the components of Smilax perfoliate.
METHODThe chemical components were isolated by solvent extraction and column chromatography. The chemical structures were identified on the base of physical and spectral data.
RESULTNine compounds were isolated and identified as palmitic acid(I), beta-sitosterol(II), delta 7-5 alpha-cholesten-3-beta-ol(III), naringenin(IV), succinic acid(V), apigenin(VI), resveratrol(VII), daucosterol(VIII) and juncusyl ester B(IX).
CONCLUSIONCompound(I), (III), (IV), (VI) and (IX) were obtained from the genus Smilax for the first time.

9.Psychological well-being among adolescents of parents living with AIDS or HIV.

OBJECTIVETo investigate the psychological well-being among adolescents of HIV-positive parents.
METHODSThis study was involving 298 adolescents from 10 to 18 years old, including 54 adolescents of both parents living with AIDS or HIV, 114 adolescents of one parent living with AIDS or HIV and another 130 adolescents from normal families. Only one adolescent was studied per family. All participants completed an anonymous questionnaire. Psychological health, satisfaction on life, self-esteem and coping style were evaluated by SCL-90, Multidimensional Students' Life Satisfaction Scale, Self-esteem Scale and Simplified Coping Style Questionnaire.
RESULTSThe total score and 10 factors of the SCL-90 in the adolescents of parents living with HIV or AIDS was higher than the adolescents from normal families and those scores in the adolescents of both parents living with HIV or AIDS were the highest. There were 27.8% of the adolescents with both HIV-positive parents were having the total score of SCL-90 more than 160 or at lest one of the 10 factors' score more than 3. The detected rate of adolescents with one HIV-positive parent was 16.7%, higher than those adolescents from normal families (8.5%) and with significant difference (chi2 = 11.457, P = 0.003). 50.0% of the adolescents with both HIV-positive parents, were having one or more factor' score between 2.0 and 3.0, compared to the rate (37.0%) of adolescents with one HIV-positive parent. The rate was higher than the adolescents from normal families (29.8%) and with significant difference (chi2 = 7.250, P = 0.027). Among the students from normal families, the total score and the 5 dimensionalities' score of the MSLSS were lower than those adolescents of parents living with AIDS or HIV. In the friend dimensionality, the score of the adolescents from normal families was significantly higher than the adolescents of parents living with AIDS or HIV. The scores of self-esteem, positive coping style and negative coping style were not significantly different in three different subjects.
CONCLUSIONThe psychological status of the adolescents of parents living with AIDS or HIV indicated unoptimistic situation.

10.Therapeutic Observation of Moxibustion plus Tuina for Early-stage Diabetic Foot

Objective To observe the clinical efficacy of moxibustion plus tuina in treating early-stage diabetic foot. Method Seventy patients with early-stage diabetic foot were randomized into a treatment group and a control group, 35 cases each. The control group was intervened by conventional medication, while the treatment group was additionally intervened by moxibustion plus tuina. After 2 treatment courses, the changes of ankle bronchial index (ABI) in the two groups were observed, and the clinical efficacies were compared. Result Patients in the treatment group showed significant improvement in symptoms, and the total effective rate was 96.9%. The total effective rate was 75.8% in the control group. The difference between the two groups was statistically significant (P<0.05). Conclusion Moxibustion plus tuina is safe and effective in treating diabetic foot. With significant Chinese medicine feature and acceptable to patients, this method is a good choice in treating early-stage diabetic foot.
