Dendritic cells (DC) are antigen presenting cells (APC) that play critical roles in the initiation of T cell response and development of T cell-dependent antibodies in vivo. CD34_+ hematopoietic progenitor cells of bone marrow and peripheral blood can differentiate into DC when cultured with GM-CSF and TNF-? in vitro. In the present study, we cultured monocytes isolated from human peripheral blood with 100ng/ml hGM-CSF and 500U/ml hIL-4 for one week, and then found that a large number of DC with high purity were gengerated. DC expressed MHC I , MHC II and costimuladng moleculers highly on cell surface and cound stimulate proliferation of allogeneic T lymphocytes. Self serum or fetal calf serum are best for generation of DC. When hGM-CSF was used alone, the monocytes differentiated into macrophages but not to DC. TNF-? could induce further maturation of DC when added in late period of the culture. Generation of DC from human peripheral blood may facilitate further studies on DC and their clinical applications.

The blocking effect of selenium-enriched malt (Se-malt) on AFB1-in-duced UDS was reported. When the rats were fed with food containing lppm Se in form of Se-malt for two weeks, the UDS values of leukocytes treated with 6?10-7M AFB1 in vitro was decreased from 54.8 ?14.0 opm/106 WBC to 35.1?10.0 cpm/106 WBC (p

Objective To investigate the teaching effect of the PCMC-type (problem-originated clinical medical curriculum) pathology experiment course and clinical pathological conference (CPC) by using microscope digital system. Methods 384 undergraduates of second grade in clinical medicine major and imaging diagnosis major were chosen for the teaching reform project, all of whom were divided into experimental group (192) and control group (192),randomly,and they were taught by PCMC pathological experiment teaching and the traditional pathological experiment teaching separately. After the curriculum, statistic analysis of test score was used to analyze the teaching effect.SPSS 13.0 software diagram method was used to perform Levene variance analysis and t test to the achievement of two groups of students and the students' ability of self evaluation was investigated through questionnaire. Results The average score of the students in the experimental group (86.16 ± 3.28) in the theory exam was significantly higher than that of control group (75.63 ± 2.24) (P=0.000).And the average score of the students in the experimental group in experiment (27.10 ± 0.61) was significantly higher than that in control group (19.87 ± 0.25) (P=0.000).The questionnaire showed that compared with control group, the comprehensive ability of the students in the experimental group was obviously enhanced. Conclusion The PCMC pathology teaching is beneficial to improving the students' ability of linking theory with practice and enhancing their ability to analyze and solve problems, and obviously stimulate their interest in learning. It is worth promoting.

Objective:To investigate the protective effects of nuclear factor ?B inhibition on liver graft reperfusion injury during transplantation. Methods:Orthotopic liver transplantation using modified cuff technique was established in rats, animals were divided according to the grafts cold storaged in 4℃ Ringer's lactated solution with(PDTC group)or without 0.1 mol/L PDTC(control group)for 6 h.During the early stage of reperfusion,DNA binding activities of NF ?B in liver grafts were analyzed by EMSA(electrophoretic mobility shift assay),mRNA level of TNF ? and ICAM 1 by RT PCR,and activities of ALT and LDH were also detected. Results:NF ?B in liver grafts was activated at early stage of reperfusion during transplantation; PDTC treated liver displayed lower activation of NF ?B 1 h after reperfusion, whereas no difference was shown between 2 groups 6 h after reperfusion.Up regulation of TNF ? and ICAM 1 transcription, high level of ALT and LDH activities were observed in both groups during reperfusion, whereas the transcriptional up regulation and the activities of ALT and LDH in PDTC group were reduced compared with those of control group( P

ObjectiveTo explore a new simple epicanthoplasty method fit for various types of epicanthus with inconspicuous scar after operation. Methods As the latest studies showed that the anatomical structure of epicanthus is mainly due to the eyelid orbicularis muscle's dislocation at the starting point of the medial canthal tendon, which affects the skin appearance, rather than the traditional view that was merely the excess skin,we released the adhesions between the skin and the inner canthus orbicularis muscle, and corrected the dislocation of orbicularis muscle that was key to the operation, incised along with the line A′→B→A″→ C in the field of interior canthus, separated the subcutaneous fascia tissue, relaxed the subcutaneous fibrous cord, disconnect the orbicularis muscle's attachment point in the inner canthal fold, and then sutured and folded the inner canthal ligament. After sewing up point A′ and point A″,the released skin retracted naturally and formed a new inner canthal contour.ResultsFrom June 2008 to June 2009,57 cases of epicanthus patients accepted this surgery. After operation, the appearance of canthus was nice.Eye fissure got widen, and the scar was not obvious. Also there was a high satisfactory rate.ConclusionsThis method has a broad range of applications. It is suitable for various types of epicanthus correction.

Objective To establish a porcine model of penetrating-suture pancreaticojejunostomy. Methods Ten domestic pigs were selected, and after general anesthesia they underwent laparotomy and the pancreas was visualized. Then the pancreas was dissected at the level of superior mesenteric vascular, and the proximal pancreatic stump was sewed up.The anastomosis between the distal pancreatic stump and the intestinal wall adopted penetrating-suture pancreaticojejunostomy; the digestive tract was reconstructed by Roux-en-Y.Results The anastomosis with penetrating-suture pancreaticojejunostomy was successful in ten domestic pigs. The mean pancreatic stump diameter was 2.5 cm,and the mean pancreatic duct diameter was 1.5 mm,the mean time for operation was 1.0 ~ 2.5 h,and the average time of pancreaticojejunostomy was 8 minutes. The mean blood loss was 25 ml.After operation, diarrhea occurred in 2 pigs and wound infection occurred in 1 pig,and all were cured with appropriate management. No pig died intra-operatively,and no pancreatic fistula or death occurred after operation. Conclusions A porcine model of penetrating-suture pancreaticojejunostomy is successfully established.
