1.Inhibition of clotrimazole on Paneth cell defensins

Aim To investigate the Clotrimazoles (CLT) inhibitive effect on the secretion of ?-defensins of the Paneth cells.Methods The ?-defensin mRNA expression is determined by semi-quantitative RT-PCR. The secretion of defensins is determined by dot-blot and ELISA. Results CLT has no influence on the defensin mRNA expression. All of 125 nmol?L -1, 250 nmol?L -1 and 500 nmol?L -1 CLT can inhibit the secretion of ?-defensins. Conclusion CLT could inhibit the secretion of defensins, which suggests that CLT maybe influence the function of small intestine
2.Effects of Candesartan on the Expression of Cerebral NAD(P) H Oxidase mRNA in Salt-loaded Stroke-Prone Spontaneously Hypertensive Rats

Background Oxidative stress has been implicated in the pathogenesis of angiotensin Ⅱ(Ang Ⅱ)related hypertension.Superoxide anion,produced by NAD(P)H oxidase,plays important roles in hypertension and its complications.Ang Ⅱ infusion increases cerebral NAD(P)H oxidase activity,which is inhibited by angiotensin Ⅱ receptor blocker(ARB)candesartan and NAD(P)H oxidase inhibitor.However,the underlying mechanism of ARB in preventing hypertensive stroke is not elucidated clearly.Objective To investigate the effects of candesartan on the expression of cerebral NAD(P)H oxidase mRNA in salt-loaded stroke-prone spontaneously hypertensive rats(SHRsp).Methods Twelve-week-old salt-loaded SHRsp were treated with candesartan [1.0 mg/(kg?d)],trichlormethiazide [TCM,1.6 mg/(kg?d)] or vehicle(n=12 in each)for 2 weeks.Age-matched salt-loaded WKY rats were served as control(n=12).Blood pressure was measured every week.After two weeks,cerebrums were harvested and 24 h urine was collected.Urinary albumin was examined by ELISA.Cerebral cortex NAD(P)H oxidase subunits(p22phox,p47phox and gp91phox)mRNA expression were assayed by real-time PCR.Results The systolic blood pressure was increased significantly in salt-load SHRsp.Candesartan and TCM reduced SBP to the similar level.Urinary albumin excretion and cerebral cortex NAD(P)H oxidase subunits were markedly higher in salt-loaded SHRsp than those in salt-loaded WKY rats.Incidence of stroke was significantly reduced in candesartan treated group as compared with TCM treated SHRsp(P
3.Effects of radix salvia miitiorrhizae on the apoptosis of hepatocytes during cold preservation and reperfusion injury in rat donor liver

Objective To investigate the effects of radix salvia miltiorrhizae on the apoptosis of hepatoeytes during cold preservation and reperfusion injury in rat donor liver. Methods Forty male SD rats were divided into experimental group, control group and sham operation group. The rat model of liver transplantation was established according to the Kamada method. The grafts were preserved in lactated Ringer's solution with radix salvia miltiorrhizae in experimental group and in lactated Ringer's solution in control group for 5 hours, then they were transplanted orthotopically. Six hours after transplantation, the recipients were sacrificed, and the serum ALT and AST were detected by automatic biochemistry analyzer, the hepatocyte apoptosis by TUNEL, the expression of Bcl-2 and FasL protein by flow cytometry. The histopathological changes of the liver grafts were observed under light microscope. Results The levels of ALT and AST in the experimental group were significantly lower than those in the control group after reperfusion. Compared with that in the control group, the apoptosis index of the hepatoeyte was signifieandy decreased in the experimental group ( F = 133.802, P <0.05 ), while the level of Bel-2 protein expression was increased ( F = 91.063, P < 0.01 ). No statistical difference upon FasL protein expression was detected between the 2 groups( F = 1.329, P >0.05). The histopathological injury of the liver grafts in the experimental group was significantly slighter than that in the control group. Conclusions Radix salvia mihiorrhizae inhibits the apoptosis of hepatocytes by increasing the Bcl-2 protein expression during cold preservation and reperfusion injury, so it has a protective effect on the liver graft against ischemia reperfusion injury.
4.Mesenchymal stem cells: their phenotype, immunological features, and potential for homing

Mesenehymal stem cells CMSCS are nonhematopoietie stromal cells that are capable of differentiating into, and contribute to the regeneration of, mesenchymal tissues such as bone, cartilage, muscle, ligament, tendon and adipose. MSCs have been isolated from human first- and second-trimester fetal blood, liver, spleen, bone marrow, adipose tissue, amniotie fluid, periosteum and embryo. Not only can they express many phynotypes involving adhesion molecules and chemokine receptors, but also have character of immune tolerance, so they have potential of migration and transportation to target issue and feature of immune suppressing. There are wide application prospects for a number of areas in the cardiovascular, neurological, blood, surgical injury and other diseases. The current review describes the phynotypes, the immune tolerance and the potential for homing of MSCs.
5.Progress on the origin of hepatocyte stem cell

Generally speaking,the hepatocyte stem cell is not a specific cell type.but an overall name of all kinds of cells that possess stem cell characters about embryonic development and regeneration of liver.HSC is a pluripotential stem cells which have self-renewal capacity and could differentiate into mature hepatocytes and bile duct cells.According to the different origin of hepatocyte stem cell,it can be usually divided into two kinds:non liver-derived hepatocyte stem cell and liver-derived hepatocyte stem cell.
6.Effects of music intervention on growth and development of premature infants

Objective To explore the effects of music intervention on growth and development of premature infants. Methods 112 premature infants were randomly divided into the observation group and the control group with 56 cases in each. The control group received conventional nursing measures, and the observation group was simultaneously given music intervention besides the conventional routine nursing measures. Weight increase, hospitalization time, milk-intake volume and feeding tolerance were compared between the two groups. Results The weight increase in the observation group was higher than that in the control group. Hospitalization time and rate of feeding intolerance for the observation group was lower compared with that of the control group. The premature infants in the observation group took in more milk than those in the control group. Conclusions Music intervention can elevate feeding tolerance, facilitate nutrition and increase weight of premature infant, so it is beneficial to the growth and development of premature infant.

Retinopathy of prematurity (ROP) is a potentially blinding retinal vascular disease that occurs in premature infants and low birth weight infants.Along with the development of perinatal medicine,the incidence of ROP has increased.If infants with ROP could get treatment in time,the disability rate due to ROP would significantly reduce.Now,the present situation and advances in treatment of ROP were reviewed.

Objective To explore the expression of interleukin 13(IL-13) mRNA in peripheral blood mononuclear cells (PBMCs) and its association with serum C-reaction protein(CRP) in patients with ulcerative colitis(UC). Methods IL-13 mRNA expression in PBMCs was detected by RT-PCR in patients with UC(21 cases of active UC and 10 cases of quiescent UC)and 20 healthy subjects. At the same time, the serum level of CRP was detected by ELISA. Results The expression level of IL-13 mRNA in PBMCs of patients with active UC was significantly lower than that in PBMCs of patients with quiescent UC and healthy subjects (6.45?1.23 vs 14.72?2.12 or 15.17?2.38,P0.05). Objective To assess the role of matrix metalloproteinase-9(MMP-9) and interleukin-6 (IL-6) of amniotic fluid (AF) in predicting infection of amniotic cavity. Methods Eighty-four cases of sigle foetus women with premature delivery, premature rupture of foetal membranes,or suspicious infection of amnitotic cavity were enrolled in this study. AF was obtained by transabdominal amniocentesis before starting any treatment.Infection of amniotic cavity was defined when AF bacterial culture was positive. The contents of glucose, leukocytes, MMP-9,and IF-6 in AF were determined. Results The levels of MMP-9 and IL-6 in AF were significantly higher in women with infection of amniotic cavity than those in women without. OBJECTIVETo investigate whether the genistein can increase the radiosensitizing effect on laryngeal squamous carcinoma Hep-2 cells.METHODS Hep-2 cells were treated with genistein, radiation, and genistein plus radiation respectively. DMSO was used as the control group. EdU assay was performed to assess the short-term effect of genistein and (or) radiation on the proliferation of Hep-2 cells. Clonegenic assay was used to detect the survival rate of Hep-2 cells after treatment with radiation doses of 0, 2, 4, 6, 8 Gy and radiation combined with genistein. The data was fitted into the classic single-hit multi-target mathematical model to analyze the long-term effect on cell proliferation death of Hep-2 cells.RESULTSIt was observed that radiation combined with genistein could significantly inhibit the proliferation of Hep-2 cells. And the SER of 10μmol/L genistein was 1.412.CONCLUTIONGenistein can inhibit the proliferation of Hep-2 cells by DNA synthesis inhibition, and can be an adjunct agent of radiotherapy.
