2.Changes of Ultrastructure of Muscle in Rats with Spastic Palsy after Injected with Botulinum Toxin

Objective To observe the ultrastructural changes of end-plate and gastrocnemius muscle of rats with spastic palsy after injected with botulinum toxin type A in order to provide scientific base of histomorphology for clinical use of botulinum toxin type A.Methods Wistar rats′ pyramidal tracts in experiment group were injured by electric current 2.5 mA for 30 s,twice,but 36 normal control rats were only drilled hole in skull(control group).Seventy-two rats were successful in building spastic palsy model.Botulinum toxin type A (6 U/kg) were injected into right gastrocnemius muscles of 36 spastic palsy rats (group B),and physiological saline were injected into the same muscles in 36 spastic palsy rats as control(group A).Control group were not injected.The rats of group A,group B were sacrificed on 3,7,15,30,60 and 90 days after botulinum toxin therapy,and right gastrocnemius muscles was separated for histological analysis.Results Under the electron microscope,the structure of muscle fiber in group B had changed(Z line rupture,disorder and abolition,myofilament lysis,triad tract diso-rder,vacuolar degeneration) and these changes in group B occurred earlier and were severer than those in group A.There was compensatory hypertrophy of muscle fiber at later stages in group B.During the first 15 days after botulinum toxin type A injection,there were many synaptic vesicle without pre-synaptic membranes near terminal portion of nonmedullated nerve fibers.Following this,between 15 to 30 days,there were many folds similar to postsynaptic membranes in the cellular membranes and there were many grains in it.But there was no synaptic vesicle and pre-synaptic membranes near them.Compensatory hypertrophy and atrophy of muscle fiber coexist at 90 days after injection in group B.Conclusions Injection of botulinum toxin type A maybe induce the sprout of nerve and degeneration of the partial postsynaptic where there are many grains.It also accelerates muscle atrophy,but it induces compensatory hypertrophy of muscle fiber at later stage.It implies that injection of botulinum toxin type A can improve spasm symptom of the gastrocnemius muscles and this is good for muscle fiber repairing.

4.Effect of Omegaven on blood clotting system and inflammatory reaction in ICU patients with parenteral nutrition

Objective: To observe the changes of blood coagulation factors and inflammatory factors and the regulatin effect of Omegaven.Methods: Thirty ICU patients were randomly allocated into two groups: Omegaven group and control group.Serum concentrations of NF-κB、PGF1α、Eselectin、sICAM-1、TAT、D-dimer、TMD、PF1+2 were measured before and after parenteral nutrition.Results: Patients in control group had significantly higher NF-κB、Eselectin、sICAM-1、TAT、D-dimer、TMD、PF1+2 concentration,lower PGF1α concentration.Patients treated with Omegaven had significantly lower NF-κB、Eselectin、sICAM-1、TAT、D-dimer、TMD、PF1+2 concentration,higher PGF1α concentration.Conclusion: Omegaven can suppress inflammatory reaction and improve coagulopathy.
5.Cost-effectiveness analysis of two therapeutic methods for prolactinoma

Objective To evaluate the therapeutic responses to transsphenoidal surgery and medical therapy in terms of normalization of prolactin(PRL),mortality,morbidity and the cost-effectiveness of PRL normalization in order to establish an individualized therapeutic protocol for the patients with prolactinoma.Methods A retrospective study was undertaken of a consecutive series of patients with prolactinoma who were followed for at least 1 year after transsphenoidal surgery or medical treatment.The clinical characteristics and the long-term outcomes(normalization of PRL,morbidity or mortality)were assessed.Utilizing the principle of medical economics and data from the two types of treatment,we worked out a Markov chain and calculated the lowest cost of two kinds of therapeutic protocols.Results(1)The success rate of normalizing serum PRL through surgical treatment in microadenoma was 85%(22/26),and that of medical treatment was 95%(19/20).There was no statistical difference between the two therapies(P>0.05).The success rate of normalizing serum PRL through surgical treatment in macroadenoma was45%(19/42),and that of medical treatment was 5/5.There was a statistical difierence between the two therapies(P<0.05).(2)According to the Markov model,it would cost a microprolactinoma patient 25 129.25 yuan to normalize serum PRL by surgical treatment.This is comparable to the cost of medical treatment which would be 24 943.99 yuan.Whereas for a macroprolactinoma patient surgery would cost 35 208.20 yuan and medical treatment would cost 25 344.38 yuan.Conclusions Medical therapy is superior to surgical treatment in regard to complication rate and cost-effectiveness for macro-and extra big prolactinomas.Transsphenoidal surgery remains an option for patients with microadenomas.Markov model is an effective way to predict the treatment cost for patients with hyperprolactinoma at different ages and with different canses
6.Mouse-bovine Somatic Cell Interspecies Nuclear Transfer

Simplified interspecies nuclear transfer in mouse and bovine and the possibilities of the embryos reprogramming were developed. The zona and nucleus of the bovine oocyte were removed by pronase and bisection method respectively. When the skin fibroblast of mouse was fused with the nucleus-free oocyte of the bovine using 40?s and 1. 5 kv/cm direct current pulse, the rates of fusion and cleavage were 67. 44% and 30.23% respectively. Reconstructed heterogenous embryos finally developed to 8-cell stage when they were previously activated by ionomycin and 6-DMAP and subsequently cultured in press-made micro-well in the drop of the medium. Zona-free bisection method could be used for mouse heterogenous embryos cloning, and the micro well culture method could be used to avoid zona-free embryos aggregation.

Objective To study the chemical constituents from the roots of Euphorbia sonngarica. Methods Compounds were isolated by Sephadex LH-20,MPLC,and silica gel column chromatographies. Their structures were identified by spectral methods together with physicochemical analysis.Results Eleven compounds were isolated from the roots of E.sonngarica.They were identified as cryptomeridiol (Ⅰ),betulin(Ⅱ),betulinic acid(Ⅲ),3?-hydroxy-olean-12-en-28-oic acid(Ⅳ),7-oxo-?sitosterol (Ⅴ),erythrinasinate(Ⅵ),octaeosanoie acid(Ⅶ),1-octacosene(Ⅷ),24-methene-cycloartenol(Ⅸ),eu- phol(Ⅹ),?-sitosterol(Ⅺ).Conclusion CompoundsⅠ-Ⅷare isolated from this plant for the first time.

AIM: To explore the influence of CD47 molecules on the maturation and function of cultured dendritic cells (DCs). METHODS: Monocyte cell-derived DCs were propagated in granulocyte-macrophage colony-stimulating factor (GM-CSF) and lipopolysaccharide (LPS) plus interleukin (IL)-4, in the presence or absence of anti CD47 monoclonal antibodies (anti-CD47 mAbs). 10.Study on gallnuts development and its tannin amount.

OBJECTIVETo find out the suitable time for collecting gallnuts. When Kaburgaia rhusicola parasailed on leaflets of Rhus potainii, the parts of the leaflets developed into gallnuts.
METHODThe regulation of development and tannin amount during whole growing stage were studied.
RESULTK. rhusicola lived in gallnuts for about 80 days. The gallnuts developed quickly during earlier stage and slowly during later stage. Gallnuts maturated from 12th, July to 30th, July, which had no relations with the size and the time for forming, but had relations with development at earlier stage. It developed quickly with earlier maturation and developed slowly with later maturation. Its tannin amount kept stable during the whole growing stage.
CONCLUSIONThe best harvesting days are those shortly after 7th, July.
