2. Expression of suppressor of cytokine signaling 2 in breast carcinoma by tissue microarray

Objective: To investigate the expression of suppressor of cytokine signaling 2 (SOCS2) in breast carcinoma tissues and to discuss its relationship with clinical pathological data of breast carcinoma patients. Methods: The tissue microarray for 171 cases of breast carcinoma specimens, 18 adjacent tissues and 20 breast benign lesions were established. Then the expression of SOCS2, ER, PR, cerbB2, p53 and Ki-67 was detected by tissue microarray technique and S-P immunohistochemistry. Results: Positive rates of the SOCS2 protein in the breast carcinoma specimens, adjacent tissues and breast benign lesion were 57.89% (99/ 171), 94.44% (17/18), and 75% (15/20), respectively. The expression of SOCS2 was significantly different in breast carcinoma tissues of different TNM classification, different histological grades, and with or without Ki-67 expression and lymphatic metastasis (P<0.05). Conclusion: The lower expression of SOCS2 might contribute to oncogenesis of breast carcinoma and poor prognosis.

4.Effects of Tiam 1 antisense oligodeoxynucleotides transfection on the invasive and migratory potentials of gastric cancer cells in vitro and vivo

Background and purpose:As a regulatory factor of cytoskeletal structure,a close correlation existed between Tiam 1(T lymphoma invasion and metastasis inducing factor 1) and the invasion and metastasis of gastroenteric tumor.We aimed to observe the effects of Tiam 1 antisense oligodeoxynucleotides(Tiam 1 ASODN) transfection on the invasive and migratory potentials of gastric cancer cells in vitro and vivo.Methods:The higher invasive and migratory subgroup(MH) were separated from human gastric cancer cell line MKN-45(M0) by laminin adhesion method in vitro.Tiam 1 ASODN was transfected into MH cells with liposome,and the expression of Tiam 1 mRNA and protein was determined by RT-PCR and ELISA separately.Changes in the invasive and migratory potentials of MH cells in vitro and vivo after transfected with ASODN were observed by Boyden chamber test and inoculation into nude mice respectively.Results:The expression of Tiam 1 mRNA and protein in MH cells after transfected with 0.43 ?mol/L ASODN(0.162?0.018,0.982?0.119) was significantly lower than that of either Liposome-transfected cells(0.789?0.054,1.237?0.108),sense oligodeoxynucleotides(SODN)-Liposome-transfected cells(0.754?0.039,1.234?0.103) or No-transfected cells(0.801?0.065,1.290?0.182)(P
5.Relationship between the expression of Tiam1, Rac1 and the pathobiological behavior of gastric cancer

Objective To investigate the relationship between the expression of T lymphoma invasion and (metastasis) inducing factor 1 (Tiam1), ras-related C3 botulinum toxin substrate 1 (Rac1) with the invasion and metastasis of gastric cancer. Methods The expressions of Tiam1 and Rac1 proteins in 60 cases of (gastric) cancer and paracarcioma gastric mucosa tissues were detected using Strept Avidin-Biotin Complex(SABC) immunohistochemical method,and the correlation between the expression of Tiam1, Rac1 and (clinicopathological) parameters of gastric cancer were analysed. Results (1)There was negative staining of Tiam1 protein in paracarcioma gastric mucosa tissues, while positive staining was detected in gastric cancer (tissues)(78.33%)(P 0.05). 6.Effects of PI3-K/AKT signal transduction pathway in rat neural stem cells

Objective To incubate rat neural stem cells (NSCs) with selection-specific inhibitor ( LY294002) of PI3-K, and explore the effects of PI3-K/AKT signal transduction pathway in NSCs. Methods NSCs derived from E15-16 rats were isolated and cultured. After incubation of NSCs with different concentrations of LY294002 (0 to 40 μmol/L), cell survival ( WTS-8 assay), proliferation ( BrdU immunohistochemical identification), differentiation (β Tubulin-III immunohistochemical identification) and AKP phosphorylated protein (Western blotting) were detected. Results The effects of LY294002 on the survival, proliferation and differentiation of NSCs was concentration-dependent. The survival of the NSCs decreased significantly with a higher concentrations of LY294002 (25, 30, 35, 40 μmol/L) (P < 0. 05). The numbers of BrdU positive cells with LY294002 of 30, 35 and 40 μmol/L were significantly smaller than that of the control (LY294002 0 μmol/L) (P < 0.05). The numbers of β Tubulin-III positive cells with LY294002 of 35 and 40 μmol/L were significantly smaller than that of the control (P < 0. 05). The expression of AKT was blocked by LY294002 in a concentration-dependent manner. Conclusion PI3-K/AKT signal transduction pathway plays a vital role in the survival, proliferation and differentiation of NSCs.
7.Efficacy of different doses of ulinastatin for prevention of insulin resistance in patients undergoing partialhepatectomy

Objective To evaluate the efficacy of different doses of ulinastatin for prevention of insulin resistance in the patients undergoing partial hepatectomy.Methods Sixty patients of both sexes,aged 33-71 yr,weighing 44-70 kg,of ASA physical status Ⅰ or Ⅱ,scheduled for electivepartial hepatectomy,were randomly assigned into 3 groups (n =20 each) using a random number table:control group (group C),low dose of ulinastatin group (group UL) and high dose of ulinastatin group (group UH).Before induction of anesthesia and at the beginning of operation,ulinastatin 2 500 U/kg and 5 000 U/kg (in 20 ml of normal saline) were injected intravenously in UL and UH groups,respectively.The equal volume of normalsaline was given instead of ulinastatin in group C.Before skin incision,before removal of the liver,after removal of the liver,and after closure of the abdominal cavity,blood samples were taken from the right internal jugular vein for determination of plasma glucose,insulin and glucagon concentrations.Insulin sensitivity index was calculated.Results Compared with group C,insulin sensitivity index was significantly increased,and the plasma glucose and insulin concentrations were decreased in UL and UH groups.Conclusion Ulinastatin 2 500 U/kg injected intravenously before induction of anesthesia and at beginning of operation can effectively prevent insulin resistance in the patients undergoing partial hepatectomy.
9.The role and mechanisms ofω-3 polyunsaturated fatty acids in inducing cell apoptosis and anti-prolif-eration in multiple myeloma

Objective To explore the role and mechanisms of ω-3 polyunsaturated fatty acids (ω-3PUFA) alone or in combination with dexamethasone ( DEX) in inducing cell apoptosis and anti -proliferation in multiple myeloma(MM).Methods DEX resistance MM cell line MM1R were treated with different concentra-tions of Eicosapentaenoic acid(EPA)or Docosahexaenoic acid(DHA)alone or in combination with DEX for 24hrs or 48hrs.Cell proliferation was detected by MTT .Cell cycle and apoptosis were measured by flow cytometry .The levels of apoptosis related proteins were analyzed by Western blot .Results The proliferation of MM1R was in-hibited by different concentrations (10,20,50,100μM)of EPA or DHA alone or in combination with 10μM DEX in a dose-and time-dependent manner .Inhibition effect was significantly higher in combinative groups than in single agent groups(P<0.05).The percentage of G0/G1 phase and cell apoptosis rate in MM1R treated with dif-ferent concentrations of EPA or DHA alone was increased in a dose -dependent manner ,and being significantly higher in combinative groups than in single agent groups (P<0.05).The expressive levels of cleaved caspase -3 and Bax were up-regulated ,while pro-caspase-3 and BCL-2 were down-regulated in a dose-dependent manner.Conclusion ω-3PUFA can inhibit DEX resistant MM cell proliferation ,arrest cell cycle and induce cell apoptosis ,and has a synergistic anti -resistant effect in combination with DEX ,may serve as a new ,effective MM drugs.
10.CT analysis of classification of external nasal fracture and the influence of fractured position to nasal septum.

OBJECTIVE:
To investigate the classification and distribution of external nasal fracture, and its influence to the nasal septum.
METHOD:
We randomly chose 60 patients who received nasal CT scan because of nasal trauma and diagnosed as external nasal fracture. We reviewed their CT data in PACS system with computer.
RESULT:
Of the 60 cases of nasal trauma, 90 sides got external nasal fracture, among which 58 sides (64.4%) had only nasal bone fracture, 16 sides (17.8%) had only maxillary frontal process fracture, and 16 sides (17.8%) had both. Half of these 60 patients got unilateral external nasal fracture, among whom 14 patients (46.7%) had traumatic nasal septum deviation meanwhile. The other 30 patients suffered from bilateral external nasal fracture, among whom 26 patients (86.7%) had traumatic nasal septum deviation. The difference between these two groups was statistically significant (P < 0.01). Moreover, 24 patients got fracture of either nasal bone or maxillary frontal process, 11 of whom (45.8%) got traumatic nasal septum deviation at the same time. The other 36 patients suffered fracture of both these two bones, 29 of whom (80.6%) had traumatic nasal septum deviation. The difference between these two groups was statistically significant (P < 0.01). We classified the nasal bone fracture as below: 16 patients with only unilateral external nasal fracture belonged to Type I, of whom 15 patients (25.0%) were type Ia with nasal bone or maxillary frontal process fracture and 1 patient (1.7%) fell into type Ib with fracture occurred on both of these two bones at the same side. Four patients suffered bilateral external nasal fracture belonged to type II, among whom 2 patients (3.3%) belonged to type IIa with nasal bone or maxillary frontal process fracture and 2 patients fell into type IIb with fracture of both of these two bones at different sides. The other 40 patients (66.7%) belonged to Tpye III, who suffered external nasal fracture accompanied with traumatic nasal septum deviation.
CONCLUSION
Traumatic nasal septum deviation was extremely concomitant in patients with external nasal fracture. It will be better to perform external nasal reconstructive surgery and plasty of nasal septum deviation by using the endoscope within 1 month for the patients whose symptom were serious or who cared a lot about the appearance.
